CNOT2 promotes proliferation and angiogenesis via VEGF signaling in MDA-MB-231 breast cancer cells

<P><B>Abstract</B></P> <P>Here the underlying role of CNOT2, a subunit of CCR4-NOT complex, was elucidated in cancer progression. CNOT2 was overexpressed in HIT-T15, ASPC-1, BXPC-3, PC-3, LNCaP, MCF-7 and MDA-MB-231 cell lines, which was confirmed by Tissue array in various human tumor tissues. Also, CNOT2 depletion suppressed proliferation and colony formation of MDA-MB-231 cells. Of note, microarray revealed decreased expression of CNOT2, VEGF-A, HIF2 alpha (<0.5 fold) and increased expression of UMOD1, LOC727847, MMP4, hCG and other genes (>2.0 fold) in CNOT2 depleted MDA-MB-231 cells compared to untreated control. Consistently, downregulation of VEGF, CNOT2 and HIF2 alpha was verified in CNOT2 depleted MDA-MB-231 cells by RT-qPCR. Additionally, CNOT2 depletion inhibited VEGF induced tube formation in HUVECs and reduced neovascularization in CAM assay. Furthermore, the growth of CNOT2 depleted MDA-MB-231 cells was significantly reduced in Balb/c nude mice along with decreased expression of VEGF and PCNA by immunohistochemistry compared to untreated control group. Objective-This study aimed to examine the usefulness of each subscale score of the Clinical Dementia Rating (CDR) for predicting Alzheimer’s disease (AD) dementia progression in amnestic mild cognitive impairment (MCI) elderly subjects. Methods-Fifty-nine elderly MCI individuals were recruited from a university dementia and memory disorder clinic. Standardized clinical and neuropsychological tests were performed both at baseline and at the time of 2 years follow-up. Logistic regression analyses were conducted to examine the ability of various clinical measures or their combinations to predict progression to AD dementia in MCI individuals. Results-MCIp individuals showed significantly higher CDR Orientation subscale and CDR sum-of-boxes (SOB) score than MCInp ones, while there were no significant differences in other CDR subscale scores between the two. MCIp individuals also showed marginally higher MMSE scores than MCInp ones. A series of logistic regression analyses demonstrated that the model including CDR Orientation subscale had better AD dementia prediction accuracy than either the model with either MMSE or CDR-SOB. Conclusion-Our findings suggest that CDR Orientation subscale score, a simple and easily available clinical measure, could provide very useful information to predict AD dementia progression in amnestic MCI individuals in real clinical settings.
센서 기반의 인터랙티브 미디어 아트 분석 : 웹캠을 활용한 그래픽 아트를 중심으로

최근 인터랙티브 미디어 아트(interactive media art) 작품들은 센서(sensor)를 주된 매개체로 하여 관객과의 상호작용을 유도한다. 최근 LED, 버튼, 자이로(gyro) 등 다양한 센서들이 관객과의 상호작용을 위해 쓰이고 있지만 그 중에서도 특히 형태, 구조와 같은 시각 정보의 입력을 위해 캠(cam)의 사용이 두드러지고 있다. 본 연구는 센서기반의 미디어 작품들 중에서 특히 웹캠을 활용하는 그래픽 활용 아트작품들의 특성을 분석하고, 그래픽 아트 선행사례들의 분석과 더불어, 웹캠을 다각적으로 활용하는 콘텐츠를 연구하는데 그 목적이 있다. 또한 본 연구는 미디어아트의 변화와 더불어 센서를 활용한 미디어 아트 작품 중 캠을 사용하는 작품 분석을 중심으로 진행한다. 미디어아트의 변화에 따라 웹캠을 사용하기까지의 과정을 알아보고, 웹캠의 종류와 그 제반을 분석한다. 본론 후반부에서는 웹캠의 특성인 인터랙티비티를 연구하고, 그 적용 분류를 모션, 음양, 형태 3가지의 입력방식으로 나누어 작품사례를 분석한다. 또한 표현기법으로서 형상을 직접적으로 표현하는 방식과 형태가 변화되어 표현하는 방식 2가지로 분류하여 사례 분석을 진행한다.

Objective-The Korean Brain Aging Study for the Early Diagnosis and Prediction of Alzheimer’s disease (KBASE) aimed to recruit 650 individuals, aged from 20 to 90 years, to search for new biomarkers of Alzheimer’s disease (AD) and to investigate how multi-faceted lifetime experiences and bodily changes contribute to the brain changes or brain pathologies related to the AD process. Methods-All participants received comprehensive clinical and neuropsychological evaluations, multi-modal brain imaging, including magnetic resonance imaging, magnetic resonance angiography, [11C]Pittsburgh compound B-positron emission tomography (PET), and [18F]fluorodeoxyglucose-PET, blood and genetic marker analyses at baseline, and a subset of participants underwent actigraph monitoring and completed a sleep diary. Participants are to be followed annually with clinical and neuropsychological assessments, and biannually with the full KBASE assessment, including neuroimaging and laboratory tests. Results-As of March 2017, in total, 758 individuals had volunteered for this study. Among them, in total, 591 participants-291 cognitively normal (CN) old-aged individuals, 74 CN young- and middle-aged individuals, 139 individuals with mild cognitive impairment (MCI), and 87 individuals with AD dementia (ADD)-were enrolled at baseline, after excluding 162 individuals. A subset of participants (n=275) underwent actigraph monitoring. Conclusion-The KBASE cohort is a prospective, longitudinal cohort study that recruited participants with a wide age range and a wide distribution of cognitive status (CN, MCI, and ADD) and it has several strengths in its design and methodologies. Details of the recruitment, study methodology, and baseline sample characteristics are described in this paper.
대형 온라인 강좌의 설계와 운영 방안 모색: 재학생, 고등학생, 일반인 대상의 설문조사를 바탕으로

21세기 들어 온라인 테크놀로지의 활성화로 온라인 강좌와 웹기반 커뮤니케이션의 사용이 고등교육 분야에서 증가되어 왔다. 시간과 장소에 구애받지 않는 온라인 교육의 특성은 대학 캠퍼스를 국내외로 확대하고 대학교육의 수혜자도 일반인과 타대학 학생으로 확대할 수 있는 가능성을 열어주었다. 또한 학령인구의 감소, 대학 구조의 재편과 같은 변화도 온라인 교육에 대한 인식을 바꾸는 계기가 되었다. 본 논문에서는 K대학교 재학생, 고등학교 3학년 학생, 일반인을 대상으로 설문조사를 실시하여 온라인 강좌에 대한 만족도, 대형 온라인 강좌를 필요로 하는 영역, 현재 개설된 대형 온라인 강좌의 운영에 대한 학생들의 니즈 등에 대하여 살펴보았다. K대학교 재학생의 온라인 강좌에 대한 만족도는 높게 나타나고 있다. 다만, 각각의 온라인 강좌에 대한 만족도는 높으나 온라인 강좌의 체계 통일성을 높여야 함과 온라인 강좌를 원활하게 수강할 수 있는 환경 개선이 필요함을 알 수 있다. 고등학생의 경우 자연과학에 대한 선호도에서 일반인과 재학생에 비해 상당히 높게 나오고 있는데 수학, 물리와 같은 과목을 선수과목으로 하고 온라인 강좌로 만들어서 전공 수업에 대비하게 해야 할 필요성이 있다고 생각된다. 일반인의 경우 인문학 분야에 대한 선호도가 고등학생과 재학생에 비해 상당히 높게 나타나고 있는데 이는 교양함양이라는 강의 목적을 통해서도 확인할 수 있다. The advancement of online technology in the 21st century has increased online courses and web-based communication in higher education. This type of education is not limited by time or location and has made it possible to expand university campuses globally and broaden the reach of university education to the general public and students from other universities. Changes such as a decrease in the school-age population and a reorganization of the university structure have also created an opportunity to change the perception of online education. In this paper, we conducted surveys on K University students, high school seniors, and the general public to assess their satisfaction with online courses, identify areas that require massive online courses, and determine students’ needs for the operation of massive online courses. The survey showed that K University students are generally satisfied with online courses. However, improvements are needed to ensure a smooth online course-taking environment, increase system uniformity, and enhance the overall online course environment. High school students have a strong preference for natural science and should be offered online courses in subjects such as mathematics and physics as prerequisites to prepare for their major classes. The general public prefers the humanities, which is evident in the purpose of the liberal arts lectures.
