      • Molecular Cloning, Characterization and Expression Analysis of an ILF2 Homologue from Tetraodon nigroviridis

        Interleukin-2 enhancer binding factor 2 (ILF2) was reported to regulate transcription of interleukin-2 (IL-2), a central cytokine in the regulation of T-cell responses. This property of ILF2 was well characterized in human and mammals, but little is known in bony fish. In this paper, an ILF2 homologue was cloned and well characterized from Tetraodon nigrovirid is for the further investigation of the function of ILF2 in bony fish. The full-length Tetraodon ILF2 cDNA was 1380 bp in size and contained an open reading frame (ORF) of 1164 bp that translates into a 387 amino-acid peptide with a molecular weight of 42.9 kDa, a 5' untranslated region (UTR) of 57 bp, and a 3' UTR of 159 bp containing a poly A tail. The deduced peptide of Tetraodon ILF2 shared an overall identity of 58%~93% with other known ILF2 sequences, and contained two N-glycosylation sites, two N-myristoylation sites, one RGD cell attachment sequence, six protein kinase C phosphorylation sites, one amino-terminal RGG-rich single-stranded RNA-binding domain, and a DZF zinc-finger nucleic acid binding domain, most of which were highly conserved through species compared. Constitutive expression of Tetraodon ILF2 was observed in all tissues examined, including gill, gut, head kidney, spleen, liver, brain and heart. The highest expression was detected in heart, followed by liver, head kidney and brain. Stimulation with LPS did not significantly alter the expression of Tetraodon ILF2. Gene organization analysis showed that the Tetraodon ILF2 gene have fifteen exons, one more than other known ILF2 genes in human and mouse. Genes up- and down-stream from the Tetraodon ILF2 were Rpa12, Peroxin-11b, Smad4, Snapap and Txnip homologue, which were different from that in human and mouse.

        Genetic diversity and population structure of the amylolytic yeast Saccharomycopsis fibuligera associated with Baijiu fermentation in China

        The amylolytic yeast Saccharomycopsis fibuligera is a predominant species in starters and the early fermentation stage of Chinese liquor (Baijiu). However, the genetic diversity of the species remains largely unknown. Here we sequenced the genomes of 97 S. fibuligera strains from different Chinese Baijiu companies. The genetic diversity and population structure of the strains were analyzed based on 1,133 orthologous genes and the whole genome single nucleotide polymorphisms (SNPs). Four main lineages were recognized. One lineage contains 60 Chinese strains which are exclusively homozygous with relatively small genome sizes (18.55–18.72 Mb) and low sequence diversity. The strains clustered in the other three lineages are heterozygous with larger genomes (21.85–23.72 Mb) and higher sequence diversity. The genomes of the homozygous strains showed nearly 100% coverage with the genome of the reference strain KPH12 and the sub-genome A of the hybrid strain KJJ81 at the above 98% sequence identity level. The genomes of the heterozygous strains showed nearly 80% coverage with both the sub-genome A and the whole genome of KJJ81, suggesting that the Chinese heterozygous strains are also hybrids with nearly 20% genomes from an unidentified source. Eighty-three genes were found to show significant copy number variation between different lineages. However, remarkable lineage specific variations in glucoamylase and α-amylase activities and growth profiles in different carbon sources and under different environmental conditions were not observed, though strains exhibiting relatively high glucoamylase activity were mainly found from the homozygous lineage.

        Natural fracture prediction in Keshen 2 ultra-deep tight gas reservoir based on R/S analysis, Kuqa Depression, Tarim Basin

        Within ultra-deep tight sandstone gas reservoirs, natural fractures indicate significant effects on reservoir performance because they can serve as storage spaces for hydrocarbons and improve the permeability. Accurate prediction of natural fractures in ultra-deep reservoirs is critical to the exploration and development of tight gas in Keshen 2 Block of Kuqa Depression. In this study, several conventional logs are selected and the R/S analysis method is introduced to predict the development and distribution of natural fractures in ultra-deep tight sandstone reservoirs. Fracture development coefficient Q is calculated based on the second derivatives of lg(R/S) function and corresponding weight of each log parameter, which matches well with linear fracture density measured from image logs. There is a positive correlation between parameter Q and fracture density, higher Q values usually indicate larger fracture density. Factors causing mismatches between calculated parameter Q and fracture density are also analyzed. Fracture dip angle, fracture filling condition, vertical resolution of logs, multiple interpretations of conventional logs, and lateral detection difference between image and conventional logs may contribute to generate those false appearances. The results in this study are expected to provide new geological references for the exploration and development of ultra-deep tight gas in Keshen 2 Block of Kuqa Depression.

      • Recurrent Epidural Fluid Collection after Cranioplasty

        Cranioplasty following decompressive craniectomy is regarded as a relatively simple and safe procedure to protect the brain and improve neurological function. However, some serious complications, such as infection, hematoma, bone resorption, seizure, and epidural or subdural fluid collection may develop after cranioplasty. Among these complications, epidural fluid collection after cranioplasty is not well described and with limited reports on the same. Here, we report recurrent epidural fluid collection after cranioplasty, eventually necessitating the removal of the bone flap. This study discusses the possible pathological mechanisms of this undesirable complication with a review of the literature.

        Media Optimization for Laccase Production by Trichoderma harzianum ZF-2 Using Response Surface Methodology

        Trichoderma harzianum ZF-2 producing laccase was isolated from decaying samples from Shandong, China, and showed dye decolorization activities. The objective of this study was to optimize its culture conditions using a statistical analysis of its laccase production. The interactions between different fermentation parameters for laccase production were characterized using a Plackett-Burman design and the response surface methodology. The different media components were initially optimized using the conventional one-factor-at-atime method and an orthogonal test design, and a Plackett-Burman experiment was then performed to evaluate the effects on laccase production. Wheat straw powder, soybean meal, and CuSO4 were all found to have a significant influence on laccase production, and the optimal concentrations of these three factors were then sequentially investigated using the response surface methodology with a central composite design. The resulting optimal medium components for laccase production were determined as follows: wheat straw powder 7.63 g/l, soybean meal 23.07 g/l, (NH4)2SO4 1 g/l, CuSO4 0.51 g/l, Tween-20 1 g/l, MgSO4 1 g/l, and KH2PO4 0.6 g/l. Using this optimized fermentation method, the yield of laccase was increased 59.68 times to 67.258 U/ml compared with the laccase production with an unoptimized medium. This is the first report on the statistical optimization of laccase production by Trichoderma harzianum ZF-2.

        Serum Uric Acid Relation for Hearing Threshold Shiftt

        Objectives. The effects of serum uric acid (UA) level on a variety of diseases were found from experimental and observational studies via oxidative stress and anti-oxidants. However, research on the association of UA and hearing thresholds is relatively sparse. We investigated this issue in the U.S. general population to evaluate the relationship of serum UA levels and pure tone threshold of hearing. Methods. Forty four thousand eighty four eligible participants aged 20 to 69 years who have serum UA data and received Audiometry Examination Component were enrolled from the National Health and Nutrition Examination Survey 1999–2004. Hearing thresholds (dB) as a pure tone average at low frequencies (0.5, 1, 2 kHz) and at high frequencies (3, 4, 6, and 8 kHz) were computed. Multivariate linear regression models and tertile-based analysis with an extended-model approach for covariates adjustment were used to assess the correlation between serum UA level and hearing thresholds. Results. In the adjusted mode of tertile-based analysis, the regression coefficients elucidated as the change of log-transformed mean hearing thresholds upon comparing participants in the highest tertile of serum UA to those in the lowest tertile were –0.067 (P=0.023) in high frequency and –0.058 (P=0.054) in low frequency. After adjusting for multiple pertinent covariates, inverse association between tertiles of serum UA and hearing thresholds remained essentially unchanged. The negative trends between serum UA and hearing thresholds were statistically significant (P for trends <0.05) in tertile-based multiple linear regressions. Conclusion. Individuals with elevated UA levels independently were found to be inversely associated with hearing thresholds for pure tone audiometry in a nationally representative sample of U.S. adults.

      • Safeguarding Millimeter Wave Communications Against Randomly Located Eavesdroppers

        <P>Mm-wave offers a sensible solution to the capacity crunch faced by 5G wireless communications. This paper comprehensively studies physical layer security in a multi-input single-output mm-wave system, where multiple single-antenna eavesdroppers are randomly located. Concerning the specific propagation characteristics of mm-wave, we investigate two secure transmission schemes, namely maximum ratio transmitting beamforming and artificial noise (AN) beamforming. Specifically, we first derive closed-form expressions of the connection probability for both schemes. We then analyze the secrecy outage probability in both non-colluding eavesdroppers and colluding eavesdroppers scenarios. Also, we maximize the secrecy throughput under a secrecy outage probability constraint, and obtain optimal transmission parameters, especially the power allocation between AN and the information signal for AN beamforming. Numerical results are provided to verify our theoretical analysis. We observe that the density of eavesdroppers, the spatially resolvable paths of the destination and eavesdroppers all contribute to the secrecy performance and the parameter design of mm-wave systems.</P>

        Hyperglycemia-Suppressed SMARCA5 Disrupts Transcriptional Homeostasis to Facilitate Endothelial Dysfunction in Diabetes

        Background: Dysfunction of vascular endothelial cells (ECs) plays a central role in the pathogenesis of cardiovascular complications in diabetes. SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily A member 5 (SMARCA5) is a key regulator of chromatin structure and DNA repair, but its role in ECs remains surprisingly unexplored. The current study was designed to elucidate the regulated expression and function of SMARCA5 in diabetic ECs.Methods: SMARCA5 expression was evaluated in ECs from diabetic mouse and human circulating CD34<sup>+</sup> cells using quantitative reverse transcription polymerase chain reaction and Western blot. Effects of SMARCA5 manipulation on ECs function were evaluated using cell migration, <i>in vitro</i> tube formation and <i>in vivo</i> wound healing assays. Interaction among oxidative stress, SMARCA5 and transcriptional reprogramming was elucidated using luciferase reporter assay, electrophoretic mobility shift assay and chromatin immunoprecipitation.Results: Endothelial SMARCA5 expression was significantly decreased in diabetic rodents and humans. Hyperglycemia-suppressed SMARCA5 impaired EC migration and tube formation <i>in vitro</i>, and blunted vasculogenesis <i>in vivo</i>. Contrarily, overexpression of SMARCA5 <i>in situ</i> by a SMARCA5 adenovirus-incorporated hydrogel effectively promoted the rate of wound healing in a dorsal skin punch injury model of diabetic mice. Mechanistically, hyperglycemia-elicited oxidative stress suppressed <i>SMARCA5</i> transactivation in a signal transducer and activator of transcription 3 (STAT3)-dependent manner. Moreover, SMARCA5 maintained transcriptional homeostasis of several pro-angiogenic factors through both direct and indirect chromatin-remodeling mechanisms. In contrast, depletion of SMARCA5 disrupted transcriptional homeostasis to render ECs unresponsive to established angiogenic factors, which ultimately resulted in endothelial dysfunction in diabetes.Conclusion: Suppression of endothelial SMARCA5 contributes to, at least in part, multiple aspects of endothelial dysfunction, which may thereby exacerbate cardiovascular complications in diabetes.
