        Phytochemicals and antioxidant capacity of some tropical edible plants

        Objective: To find biological functions such as antibacterial and antioxidant activities in several tropical plants and to investigate the possibility of antibiotic substitute agents to prevent and treat diseases caused by pathogenic bacteria. Methods: Plants such as Poncirus trifoliata fruit (Makrut), Zingiber officinale Rosc (Khing), Areca catechu L. (Mak), Solanum melongena L. I (Makkhuayao), and Solanum melongena L. II (Makhurapro) were extracted by methanol, n-hexane, chloroform, ethyl acetate, butanol and water. The free radical scavenging activities were measured using 2-diphenyl-2-picryl hydrazyl photometric assay. Antibacterial activities with a minimum inhibitory concentration (MIC) were observed by agar diffusion assay against pathogenic strains of Escherichia coli, Burkholderia sp., Haemopilus somnus, Haemopilus parasuis, Clostridium perfringens, and Pantoea agglomerans. Results: Poncirus trifoliata fruit methanol extract showed antibacterial activities against gram-negative and gram-positive pathogens. Additionally, this showed the strongest antibacterial activity against Burkholderia sp. and Haemopilus somnus with MIC $131{\mu}g/mL$, respectively. Areca catechu L. water extract showed antibacterial activities against Burkholderia sp., Haemopilus somnus, and Haemopilus parasuis. The MIC value for Haemopilus parasuis was $105{\mu}g/mL$ in this. Antioxidant activity of Zingiber officinale Rosc n-hexane extract showed 2.23 mg/mL effective concentration 50% ($EC_{50}$) value was the highest activity among tropical plants extracts. Total polyphenol content in Zingiber officinale Rosc methanol extract was $48.4{\mu}g/mL$ and flavonoid content was $22.1{\mu}g/mL$ showed the highest values among tested plants extracts. Conclusion: Taken together, these results suggest that tropical plants used in this study may have a potential benefit as an alternative antibiotics agent through their antibacterial and antioxidant activities.

        Effects of calcium intake, milk and dairy product intake, and blood vitamin D level on osteoporosis risk in Korean adults

        This study was performed to determine the effects of dietary calcium (Ca) intake, milk and dairy product intake, and serum vitamin D level on bone mineral density. The survey data from the 2008-2009 Korea National Health and Nutrition Examination Survey (KNHANES) for adults (3,819 males, 5,625 females) aged > 20 years were examined; osteoporosis was defined according to the standards for Asian populations (T-score < -2.5). The risk for osteoporosis significantly decreased as Ca intake increased; this effect persisted (quartile 4 vs. quartile 1 of Ca intake: odds ratio [OR] 0.66; 95% confidence interval [CI]: 0.50-0.87) even after adjustment for gender, age, and other factors (body mass index, serum vitamin D, menstruation, female hormone intake, menopausal status, and the number of days per week of muscular strength exercise). Additionally, the risk for osteoporosis significantly decreased as the Ca/P ratio increased (quartile 4 vs. quartile 1: OR 0.76; 95% CI: 0.58-0.98). The degree of risk was 0.96 (0.66-1.38) in those who consumed < 1 portion of milk or dairy products daily, and 0.71 (0.53-0.96) in those who consumed > 1 portion per day, compared with those who had zero intake. The risk for osteoporosis significantly decreased as the serum 25(OH) vitamin D level increased. From these results, we advocate an increase in Ca, milk, and dairy product intake, and that serum 25(OH) vitamin D levels be maintained within the normal range, for the maintenance of bone health and the prevention of osteoporosis in adults.

        The difference of castration timing of Korean Hanwoo bulls does not significantly affect the carcass characteristics

        It is already well known that castration improves marbling quality but exact timing of castration is still highly debated in beef cattle production industry. After castration, blood hormonal changes occur in steer and objective of this study was to investigate the effects of growth hormone (GH) levels on adipocyte differentiation in stromal vascular cells (SVCs) and trans-differentiation into adipocytes in C2C12 myoblasts. Total GH concentrations were measured via enzyme-linked immunosorbent assay (ELISA) in 24 male calves and 4 female calves. Cell proliferation, cellular triglyceride (TG) accumulation, and the cell’s lipolytic capability were measured in C2C12 myoblasts and SVCs. Myogenic, adipogenic, and brown adipocyte-specific gene expression was measured via real-time polymerase chain reaction (PCR) using SYBR green. Serum GH levels were the highest in late-castrated calves. Treatment with 5 ng/mL GH resulted in greater TG accumulation as well as increased CCAAT-enhancer-binding protein (C/EBP)α and peroxisome proliferator-activated receptor (PPAR)γ expression compared to that after treatment with 15 ng/mL GH. Treatment with 5 ng/mL GH also resulted in lower myogenin (myo)G and myoD expression compared to that after treatment with 15 ng/mL GH. The expression of bone morphogenetic protein (BMP) 7 after treatment with 5 ng/ mL GH was higher than that after treatment with 15 ng/mL GH. But carcass characteristics data showed no significant difference between early and late castrated steers. Therefore, our results indicate that castration timing does not seem to be inevitable determinate of carcass qualities, particularly carcass weight and marbling score in Hanwoo beef cattle.

        Biochemical and Antioxidant Activity of Yogurt Supplemented with Paprika Juice of Different Colors

        Paprika is known to contain polyphenolic compounds that have good antioxidant properties. This study was conducted to investigate the benefits of adding paprika juice of different colors to yogurt and to determine how paprika affects the quality characteristics of yogurt. Stirred yogurt samples supplemented with different levels of red, orange, or yellow paprika juice were inoculated with mixes of Streptococcus thermophilus and Lactobacillus delbrueckii ssp. bulgaricus. Paprika addition decreased the pH but increased titratable acidity, lactic acid bacteria (LAB) counts, total polyphenol content (TPC), levels of vitamin A and C, and antioxidant activity. Proteolysis and viscosity of paprika-containing yogurts were significantly higher than those of the control yogurt without paprika juice (p<0.05). In particular, the viscosity of red paprika yogurt was significantly higher than that of yogurts containing 5% orange and yellow paprika juices (each p<0.05). The antioxidant activity of the methanol extract of the yogurt containing 2.5% orange paprika juice was the highest. Storage at 4℃ for 15 days only slightly altered LAB counts, antioxidant activity, and TPC of paprika yogurt. These results indicate that paprika could be used as a natural food additive for the development of functional yogurts.

        Antioxidant Properties and Diet-Related α-Glucosidase and Lipase Inhibitory Activities of Yogurt Supplemented with Safflower (Carthamus tinctorius L.) Petal Extract

        Recently, yogurt has been extensively studied to further enhance its functions using edible plant extracts. This study was conducted to investigate whether safflower petal (SP) as a natural food additive can be used to develop functional yogurt with improved health benefits. SPs were extracted with ethanol (SPE) and hot water (SPW), and then safflower yogurt was prepared by adding 0%-1.0% of those extracts to plain yogurt. With an increase in the fermentation duration, the pH of SPE and SPW yogurt samples was decreased, whereas titratable acidity and microbial counts were increased. The concentration of total polyphenols and total flavonoids, the activity of antioxidants, and the inhibitory effect on reactive oxygen species (ROS) were higher in SPW yogurt than SPE yogurt. Furthermore, α-glucosidase and lipase activity inhibitory effects of SPW yogurt were higher than those of SPE yogurt. In particular, free radical-scavenging activities, ROS inhibitory effect, and α-glucosidase activity inhibitory effects were significantly increased in SPW yogurt in a dose-dependent manner. Overall, these results suggest that SP extract possesses antioxidant activities and that it can downregulate α-glucosidase and lipase activities. The SP extract may have potential benefits as a natural food additive for the development of functional yogurt.

        Effects of pumpkin seed oil and saw palmetto oil in Korean men with symptomatic benign prostatic hyperplasia

        This study was to investigate the role of complementary and alternative medicine in the prevention and treatment of benign prostatic hyperplasia. For this purpose, a randomized, double-blind, placebo-controlled trial was performed over 12 months on 47 benign prostatic hyperplasia patients with average age of 53.3 years and international prostate symptom score over 8. Subjects received either sweet potato starch (group A, placebo, 320 ㎎/day), pumpkin seed oil (group B, 320 ㎎/day), saw palmetto oil (group C, 320 ㎎/day) or pumpkin seed oil plus saw palmetto oil (group D, each 320 ㎎/day). International prostate symptom score, quality of life, serum prostate specific antigen, prostate volume and maximal urinary flow rate were measured. In groups B, C and D, the international prostate symptom score were reduced by 3 months. Quality of life score was improved after 6 months in group D, while those of groups B and C were improved after 3 months, compared to the baseline value. Serum prostate specific antigen was reduced only in group D after 3 months, but no difference was observed in prostate volume in all treatment groups. Maximal urinary flow rate were gradually improved in groups B and C, with statistical significance after 6 months in group B and after 12 months in group C. None of the parameters were significantly improved by combined treatment with pumpkin seed oil and saw palmetto oil. From these results, it is suggested that administrations of pumpkin seed oil and saw palmetto oil are clinically safe and may be effective as complementary and alternative medicine treatments for benign prostatic hyperplasia.

        차가버섯 추출물이 당뇨생쥐 (C57BL/KsJ<SUP>-</SUP>m<SUP>+/+</SUP>Lepr<SUP>db</SUP>)의 혈당수준에 미치는 영향

        This study investigated the therapeutic effects of Inonotus obliqua extract on blood glucose, insulin, and other biochemical parameters in genetically diabetic mice (C57BL/KsJ?m<SUP>+/+</SUP> Lepr<SUP>db</SUP>): The mice were divided into four groups - control, Chaga 1 (dose of 0.09 ㎎/㎏ of body weight) , Chaga 5 (5 times of Chaga 1), and Chaga 10 (10 times of Chaga 1) - according to supplemented dose. Inonotus obliqua extract was orally administered to the animals for 6 weeks. The body and organ (liver and kidney) weights were not different among groups. Fasting blood glucose level was significantly lower in the Chaga S group compared with the control (p < 0.05). Hemoglobin Alc content was significantly lower in the Chaga 5 group compared with either the control and Chaga 1 group (p < 0.05). There was no significant difference in serum insulin level among groups. The glucose-6-phosphatase activity in liver was significantly the lowest in Chaga 10 group and was significantly lower in Chaga 5 group as compared with those of control and Chaga 1 groups. Therefore, the results of this study demonstrate that Inonotus obliqua extract alleviates many of the symptoms of diabetes in genetically obese mice and may offer a possibility as a therapeutic supplement for the normalization of blood glucose levels in human with hyperglycemia and have beneficial effects in patients with noninsulin-dependent diabetes mellitus.

        서울 일부지역 여자 고등학생의 식품 및 영양소섭취, Glycemic Index, Glycemic Load와 비만도와의 관련성 연구

        The relationship between food and nutrient intake, glycemic index (GI), glycemic load (GL), and body weight was investigated with high school girls residing in Seoul. As subjects, 159 girls were divided into a normal weight (NW) group (18.5 ㎏/㎠ ≤ BMI < 23 ㎏/㎠, n = 110) and an overweight (OW) group (BMI ≥ 23 ㎏/㎠, n = 49) by body mass index (BMI). The food and nutrient intake data obtained by the 3-day food record were analyzed by Can pro 3.0 software. Anthropometric measurements were collected from each subject. Daily dietary GI (DGI) and dietary GL (DGL) were calculated from the 3-day food record. Body weights and BMI of NW were 52.4 ㎏ and 20.4 ㎏/㎠ and those of OW were 65.2 ㎏ and 25.4 ㎏/㎠, respectively. Total food, animal food, and other food intakes of NW were higher than those of OW, and vegetable food intakes of NW were lower than those of OW. Sugars intake of NW was significantly higher than OW. Nutrient intakes were not different between the two groups. Dietary fiber, calcium, and folate intakes of NW and OW were under 65% of the dietary reference intakes (DRIs). Major food sources of energy intake for both groups were rice, pizza, ice cream, pork, instant noodle, and chicken. Mean adequacy ratio (MAR), an index of overall dietary quality, was higher in NW (0.82) than in OW (0.80). Mean daily DGI of NW and OW was 66.5 and 66.4, respectively. Mean daily DGL of NW and OW was higher in NW (162.0) than in OW (155.9). DGI and DGL adjusted to energy intake were not significantly correlated with anthropometric data.
