        감성돔(Acanthopagrus schlegeli) 유래의 주화세포의 확립과 확립된 세포의 특성

        감성돔 (Acanthopagrus schlegeli)의 비장에서 주화세포인 BSBS를 확립하고 세포의 특성을 검사하였다. BSBS세포는 60회 이상의 계대배양이 가능하였고 형태학적으로는 상피성 세포였다. 세포는 20℃, 10%의 FBS가 든 L-15배지에서 배양하는 조건에서 잘 자랐다. BSBS세포는 해양버나바이러스 (MABY Y-6), 잉어의 봄바이러스병바이러스 (SVCV), 넙치의 랍도바이스(HIRRV)와 연어바이러스 (CSV)를 접종했을 때 세포변성효과가 나타났다. 새로 확립된 주화세포는 앞으로 많은 바이러스병의 연구에 유용하게 사용할 수 있을 것으로 생각된다. A stable cell line, BSBS (black seabream spleen), was established from the cells in spleen of black seabream, Acanthopagrus schlegeli, and characterized. Subculture maintained more than 60 passages and mophologically, BSBS cell was epithelioid cell. The cells grew optimally at 20℃ in Leibovitz's L-15 medium supplemented with 10% fetal bovine serum with incubation temperature of 20℃. BSBS cells supported the growth of marine birnavirus (MABV Y-6), chum salmon reovirus (CSV), spring viremia of carp virus (SVCV) and hirame rhabdovirus (HIRRV). Thus, the new cell line may be useful for studying wide range of fish viruses.

        In this research, we measured Readily Biodegradable COD(Ss) using respiration meter which measured OUR and calculated respiration rate due to calculate organic matter portion of external carbon sources because known organic matter analysis methods i.e. COD, BOD did not measure amount of organic matter which was used in denitrification. Based on the experimental results, leather wastewater had 40.3% Ss in TCOD and food fermentation liquid, alcoholic wastewater and petrochemical wastewater was 38%, 38% and 28.3% respectively. However in case of considering of flowrate and concentration, flood fermentation and alcoholic wastewater was revealed that they were effective external carbon sources

      • 고양이 심장에서 관상동맥 결찰과 재관류로 인한 ATPase 변화에 관한 세포화학적 연구

        To understand the behaviors of Ca^(++)-ATPase and Na^(+)-K^(+) ATPase in ischemic and ischemic-reperfused heart, and to elucidate their roles in those conditions, the authors observed cat myocardium ultracytochemically. Acute ischemia was induced by ligation of the anterior interventricular branch of left coronary artery. Reperfusion was begun after 20minutes of ischemia and discontinued after 1-2 minutes when ventricular fibrillation appeared on EKG. The reaction products of Ca^(++)-ATPase were irregular In shape, and distributed mainly in myocytes in the vicinity of capillaries. In normal cardiac myocytes, they were present at plasmalemma, sarcoplasmic reticulum, transverse tubules, intercalated discs including intermediate junctions, desmosomes and gap junctions. Some were scattered along with inner mitochondrial membranes and myofibrils. After 20 minutes of ischemia. almost all structures had negligible reaction products. except some dilated sarcoplasmic reticulum beneath the plasmalemma, which retained strong reactivities. After reperfusion, the reaction products showed a tendency to be Increased. But there was hardly any reaction products in mitochondria. Patches of myofibrils with much increased enzyme activity than other myofibrils were found scattered. In addition, macrophages showed strong reactivity throughout their nuclei. Endothelial cell nuclei, however, were free of reactions in all conditions. The reaction products of Na^(+)-K^(+) ATPase were fine In nature. In normal cardiac myocytes, they were present at plasmalemma, transverse tubules, sarcoplasmic reticulum, various structures of intercalated discs, myofibrils, mitochondria, inner nuclear membranes and heterochrormatin. In ischemic heart, the reactions decreased generally and increased after reperfusion. Mitochondria in normal states had many fine reaction products scattered throughout them to the degree of masking their details. In ischemic conditions, the reaction products, decreased in number, were located contiguous to inner mitochondrial membrane. After reperfusion each reaction product was increased in size, although the amount was less than normal conditions. Gap junctions were unique structures'in that they lacked reactivity both in ischemic and ischemic-reperfused conditions. Other structures of the intercalated discs, especially desmesomes, showed increased reactions. Some myofibrils showed much strong reactions in ischemic and ischemic-reperfused groups, even though the reactivity at myofibrils as a whole was decreased. Under all conditions, the nuclei of interstitial cails and endothelial cells had their reaction products related to the nuclear membranes and heterochrormatin. The authors, based on the above observations, conclude that Ca^(++)-ATPase may have little role in ischemic and ischemic-reperfused conditions. Na^(+)-K^(+) ATPase during reperfusion, however, even depressed in their activities, may try to normalize the decreased intramitochondrial ATP contents and the increased intracellular Na^(+) concentrations resulted from ischemic conditions.

        Pulmonary blastoma is a very rare primary neoplasm of the lung. We experienced a case of pulmonary blastoma in a 40 years old man. The mediobasal segmentectomy of the right lower lobe was performed. The patient was discharge on the postoperative days 6. There is no evidence of recurrence or metastasis on followup study during 7 months period after operation.

        Usefulness of the Cytomegalovirus Antigenemia Assay in Patients With Ulcerative Colitis

        Background/Aims: Patients with ulcerative colitis (UC) are at high risk for cytomegalovirus (CMV) reactivation. The useful-ness of the CMV antigenemia assay in active UC patients has rarely been studied. We assessed whether the assay detects CMV colitis and predicts clinical outcomes in patients with UC. Methods: We retrospectively reviewed the medical records of pa-tients hospitalized for moderate-to-severe UC from 2003 to 2012. Positive CMV antigenemia was defined as ≥1 pp65-positive cell per 2×105 polymorphonuclear neutrophils. CMV colitis was defined as the presence of inclusion bodies and/or positive im-munohistochemistry in the colonic mucosa. The primary outcome was steroid refractoriness, defined as the absence of clinical improvement after intravenous high-dose steroid administration. Results: A total of 43 patients were enrolled. CMV antigen-emia was detected in 12 (27.9%) patients. Positive CMV antigenemia was significantly associated with CMV colitis (P=0.001). The sensitivity and specificity of positive CMV antigenemia for diagnosing CMV colitis were 66.7% and 87.1%, respectively. Steroid refractoriness was found in 11 of 12 (91.7%) and 12 of 31 (38.7%) patients with positive and negative CMV antigenemia, respectively (P=0.002). The independent predictors for steroid refractoriness were positive CMV antigenemia (adjusted odds ratio [OR], 7.73; 95% confidence interval [CI], 1.22-49.19; P=0.030) and a shorter duration from the diagnosis of UC (adjusted OR, 0.99; 95% CI, 0.98-0.99; P=0.025). Conclusions: The CMV antigenemia assay shows low sensitivity but high specificity for detecting CMV colitis and may predict steroid-refractory UC. Early rescue therapy might be considered in UC patients positive for CMV antigenemia. (Intest Res 2015;13:50-59)

        As a nursing practice involves nurses' actions in a specific context of health care, this study has focused on exploring the espoused theories in nursing practice within the action science perspectives, Espoused theories are the belief, principles, and rationale expressed by the practitioner as guiding her/his actions in a situation of practice. The data were analysed qualitatively and 25 elements of espoused theories of nursing action were identified and clustered into 6 categories. The 25 elements of espoused theories are as follows: The clinical nurse worked in wholistic and individual nursing, focussed on the patient's needed, comfort and supportive nursing (5 theories of nursing goal) ; excellent skills, knowledge based, assessment and data collection, explaining, educating or a scientific basis(6 theories of nursing intervention): advocacy, value oriented, treatment, account- ability and commitment(4 theories of nursing ethics) ; human respect. partnership, trust(3 theories of patient-nurse relationship) : knowledgable, accumulated clinical experiences and personally lived experiences. positive perspectives(4 theories of nurse), role of intervention. rewarding peer relationship(3 theories of situations) The above mentioned espoused theories are similar to that of nursing textbooks which students learned through basic nursing education and almost the same as the Acts ofa Nurse in Korean. However, we are doubtful whether nurses actually do as they think. Therefore. it is recommended to review the theories-in-use in order to and any discrepancies between the espoused theories and the reality of nursing actions

        간호사의 임상 실무 경험

        purpose: This study was conducted to describe qualitatively the entities of nurse's experiences in general hospitals and to suggest basic data guiding research on developing Standards of clinical nursing practice in Korea. Method: Fourteen nurses working at general hospitals with over 300 beds in Seoul were interviewed in-depth until saturation using tape-recorders and transcription. Result: The central theme of clinical nursing practice experienced by subjects was "being with clients" that means accepting client's personal character, solving client's needs and providing client-centered nursing. A also "being with clients" was felt to be the responsibility of nurses which was learned from their nursing schools. The nursing strategies performed in order to be with patients were proving skillful nursing techniques, accepting, educating, emotional support, advocating, and self-reflecting, the subjects experienced somewhat problematic affects such as difficulties in interpersonal relationship, work overload, negative image of nursing, deficit of self-confidence for nursing actions, poor working conditions, and unfair treatment. Nurses at the hospital practiced with pride when they felt that they were accepted by clients. Conclusion: Further research is needed to analysis problems in clinical practice and the comparison of nurses' experiences of clinical practice, with nurses' experiences in various settings.

        Effects of Fetal Mesencephalic Cell Grafts on the Intrastriatal 6-hydroxydoapmine Lesioned Rats

        The effects of fetal mesencephalic cell grafts on the restoration of nigrostriatal dopaminergic function were studied in the intrastriatal 6-hydroxydopamine-lesioned rats. Four weeks after lesioning, transplantation of ventral mesencephalic cells from embryonic day 14 fetuses showed the number of tyrosine hydroxylase (TH) positive cells and fiber outgrowth in the grafted striatum, and significantly ameliorated symptomatic motor behavior of the animals, as determined by apomorphine-induced rotation. Furthermore, in substantia nigra pars compacta (SNc), the numbers of TH + cells and fibers were markedly restored. Dopamine content of ipsilateral SNc was close to that of contralateral SNc $(91.9{\pm}9.8%)$ in the transplanted animals, while the ratio was approximately 32% in sham-grafted animals. These results indicate that grafted cells restored the activity for the dopaminergic neurons located in SNc, although they were transplanted into striatum. In addition, we showed that the implanted fetal cells expressed high level of glial cell line-derived neurotrophic factor (GDNF), suggesting that the transplanted fetal cells might serve as a dopamine producer and a reservoir of neurotrophic factors. These results may be helpful in consideration of the therapeutic transplantation at early stage of PD.

        Ceramide is involved in MPP<SUP>⁢</SUP>-induced Cytotoxicity in Human Neuroblastoma Cells

        <P> To understand the cytotoxic mechanism of MPP<SUP>⁢</SUP>, we examined the involvement of ceramide in MPP<SUP>⁢</SUP>- induced cytotoxicity to human neuroblastoma SH-SY5Y cells. When SH-SY5Y cells were exposed to MPP<SUP>⁢</SUP>, MPP<SUP>⁢</SUP> induced dose-dependent cytotoxicity accompanied by 2-fold elevation of intracellular ceramide levels in SH-SY5Y cells. Three methods were used to test the hypothesis that the elevated intracellular ceramide is related to MPP<SUP>⁢</SUP>-induced cytotoxicity: C<SUB>2</SUB>-ceramide was directly applied to cells, sphingomyelinase (SMase) was exogenously added, and oleoylethanolamine (OE) was used to inhibit degradation of ceramide. Furthermore, inhibition of ceramide-activated protein phosphatase (CAPP), the effector of ceramide, using okadaic acid (OA) attenuated cell death but treatment of fumonisin B<SUB>1</SUB>, the ceramide synthase inhibitor, did not alter the cytotoxic effect of MPP<SUP>⁢</SUP>. Based on these, we suggest that the elevation of intracellular ceramide is one of the important mediators in MPP<SUP>⁢</SUP>- induced cell death.
