        Dual effects of a CpG-DNAzyme targeting mutant EGFR transcripts in lung cancer cells: TLR9 activation and EGFR downregulation

        Non-small-cell lung cancer (NSCLC) is commonly caused by a mutation in the epidermal growth factor receptor (EGFR) and subsequent aberrant EGFR signaling with uncontrolled kinase activity. A deletion mutation in EGFR exon 19 is frequently observed in EGFR gene mutations. We designed a DNAzyme to suppress the expression of mutant EGFR by cleaving the mutant EGFR mRNA. The DNAzyme (named Ex19del Dz) specifically cleaved target RNA and decreased cancer cell viability when transfected into gefitinib-resistant lung cancer cells harboring EGFR exon 19 deletions. The DNAzyme decreased EGFR expression and inhibited its downstream signaling pathway. In addition to EGFR downregulation, Ex19del Dz containing CpG sites activated Toll-like receptor 9 (TLR9) and its downstream signaling pathway via p38 kinase, causing an immunostimulatory effect on EGFR-mutated NSCLC cells. Thus, dual effects of this DNAzyme harboring the CpG site, such as TLR9 activation and EGFR downregulation, leads to apoptosis of EGFR-mutated NSCLC cells. [BMB Reports 2018; 51(1): 27-32]

      • 비정규직의 이직의도에 대한 실증연구: HCCP 패널데이터를 이용하여

        본 연구는 2016년 인적자본기업패널(Human Capital Corporate Panel: HCCP) 데이터를 이용하여 개인특성과 조직특성이 비정규직의 이직의도에 어떤 영향을 미치는지를 분석하였으 며, 이를 통해 비정규직의 이직으로 인한 부정적 영향을 줄이기 위해 개인 및 조직 차원의 해결방안을 모색하고자 하였다. 이 연구의 주요 결과는 다음과 같다. 첫째, 비정규직의 개인요인에서 근속연수가 길어질수록, 인재를 우대하고 있다고 느낄수록, 인간관계에 대한 만족도가 높을수록 이직의도가 낮아지고 커뮤니케이션이 활발할수록 이직의도가 높아지는 것으로 나타났다. 둘째, 비정규직의 기업요인 중 교육훈련이 의무인 경우, 수평적 기업문화를 가진 기업일 경우 이직의도가 낮아지고, 개인성과급을 실시하는 기업일 경우 이직의도가 높아지는 것으로 나타났다. 정규직의 경우, 임금에 대한 만족도가 낮을수록 이직의도가 높게 나타났으나 비정규직의 임금은 이직의도에 전혀 영향을 미치지 못하였다. 이는 안정적 일자리가 수반되지 않은 상황에서 근로자들의 이직의도에 임금은 아무런 영향을 미치지 못하는 것으로 해석된다. 인재를 우대하거나 직장 내 인간관계에 대해 만족할 경우 이직 의도가 낮아지는 것을 통해, 효율적인 인적자본 개발 및 성과달성을 위해 비정규직 근로자의 심리적 자본에 집중할 필요가 있음을 알 수 있다. 교육훈련을 의무로 실시하는 기업의 비정규직 근로자의 이직의도가 그렇지 않은 기업에 비해 낮다는 점을 통해 비정규직 근로자들은 기업에서 제공하는 교육훈련을 긍정적으로 받아들이며 이를 제공하는 기업에 오래 머무르고자 하는 것을 알 수 있다. 전체 근로자를 대상으로 한 분석에서 대부분의 변수가 이직의도에 영향을 미치는 데에 반해, 비정규직만을 대상으로 한 경우 몇 개의 변수만이 영향을 미치는 것으로 나타났다. 이를 통해 비정규직의 이직 의도와 이로 인한 부정적 영향의 개선을 위해서는 정규직과는 다른 접근 방식이 필요함을 알 수 있다.

        배달량 확산과 자율 주행 로봇의 실내외 주행이 가능해지면서 로봇을 활용할 주문 및 배달 서비스가 활발해지고 있다. 따라서 기존의 자율 주행 배달 로봇의 문제점을 개선하여 보다 나은 배달 서비스를 제공하고자 자율 주행 배달 로봇을 연구하였다. 배달 로봇에 AI를 적용하여 장애물 탐지, 최적코스 탐색을 통해 목적지까지 최적 경로로 이동하며, 배달 물품 안전까지 보장한다.

        3D interdigitated electrode array in the microchannel free of reference and counter electrodes

        <P><B>Abstract</B></P> <P>We demonstrate the three-dimensional (3D) interdigitated array (IDA) chip that operates without reference and counter electrodes, which are necessary components to apply enough potential to trigger the intended redox process, but used unwieldy for chip-based electrochemical detection. Using the electrode configuration, we propose a unique electrochemical system that is capable of controlling applied potential to a pair of working electrodes despite absence of reference and counter electrodes by fixing the electron transfer mediator on the electrodes in a microchannel. The electrochemical potential of the 2-electrode (2E) system is defined by the potential of the electron transfer mediator, poly(methylene green) (PMG), immobilized with poly(dopamine) (PDA) on the ITO surface by electropolymerization. The 3D IDA chip in the 2E system successfully acts as an electrochemical immunosensing platform. Creatine Kinase-MB in human serum was measured down to ~ pg / mL level. Therefore, the 3D IDA in the 2E system constitutes a simple and scalable platform that needs only nL level of sample volume for sensitive electrochemical detection in miniaturized multiplex immunoassay field.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A reference and counter electrodes-free system was introduced to 3D interdigitated electrode arrays. </LI> <LI> Poly(methylene green) and poly(dopamine) was immobilized on ITO for immunoassay. </LI> <LI> Creatine Kinase-MB in human sera was detected in the 2-electrode system of the 3D IDA chip. </LI> </UL> </P>

        부모의 사회 경제적 지위가 청소년의 스트레스, 우울, 자살생각에 미치는 영향

        본 연구는 청소년이 인지하는 스트레스, 우울 및 자살생각을 확인하고 일반적 특성에 따른 차이와 부모의 사회경제적 지위에 영향 요인을 파악하기 위한 서술적 조사연구이다. 본 연구 자료는 질병관리본부의 2009년도 국민 건강 영양조사 원시자료 중 건강 설문조사 부분을 응답한 청소년 904명이다. 수집된 자료는 빈도와 백분율, 교차분 석, 다변량 로지스틱 회귀분석을 이용하여 검정하였다. 본 연구의 결과 우울과 자살사고의 경우에서 사회경제적 수준 이 낮은 집단에서 더 높은 분포를 보였고, 스트레스의 경우는 사회경제적 수준이 높은 세대주의 자녀의 경험비율이 그 외의 집단에 비해서 높은 수치를 보였다. 우울과 자살생각을 주요결과로 한 분석모형에서는 부모의 직업 상태에 따라 차이가 있었다. 자살생각에 대한 위험요인으로는 어머니의 교육수준과 기초생활수급가족 여부도 포함되었다. 본 논문의 분석방법을 통해 국가표본조사 데이터를 이용하여 자살을 예방하고 우울, 스트레스를 감소시키기 위한 개별 및 집단 상담, 정규 교육 프로그램의 개발과 적용이 요구되며, 청소년 개인의 건강행태 뿐만 아니라 부모의 정보를 바탕으로 하는 위험 집단 선별과 가정내 환경요인 중재를 강화하는 연구에 기여할 수 있다. This study was to examine the relationship between suicidal ideation, depression, stress and their parent's socioeconomic status. Nation-wide representative data from the Korean National Health and Nutrition Examination Survey 2009 were used in this study. 904 adolescents with parent were analysed. Parent's socioeconomic status, especially mother's low educational level and the beneficiaries for national basic livelihood security were significant risk factor for adolescents' suicidal ideation. These associations remained significant in multiple logistic regression controlling for all covariates. The findings in the current study support the global literature on the importance of socioeconomic status in promoting adolescent's mental health. Future prevention intervention efforts to improve adolescent's suicide risk will need to take into consideration parent's and household's socioeconomic conditions. Future study is needed to explore the possible proximal risk factors and mediators between parent's socioeconomic status and mental health among adolescents.

      • Recommendations of developmental strategies for a marine leisure event using Importance-Performance Analysis

        Purpose: This study was initiated to examine the visitors’ satisfaction factors to a marine leisure event using IPA and provide fundamental information that helps further develop the event. In specific, this study attempted to identify important satisfaction factors for visitors to perceive in consuming a marine leisure event and levels of satisfaction in them. Method: The study employed a survey method with a total of 217 visitors to a marine leisure event held in BMC. The questionnaire was consisted of 5 satisfaction factors including accessibility, facility, operation, publicity and exhibition quality. All the factors and items were derived from previous research (Ryu, 2012) and validated by a panel of experts who checked out factor and item representativeness and appropriateness. With SPSS 20.0, the collected data were analyzed through t-tests and IPA. IPA results provide a strategic direction for each item such as ‘keep up the good work’, ‘concentrate here’, ‘low priority’, and ‘possible overkill’ based on the matrix (Martilla & James, 1997). Result: Findings of current study were as follows. First, the mean score of all 24 importance items were 3.85 at 5 point Likert scale and among others employee kindness, safety and security, amenity, program variety and overall facility quality had high priorities. Second, the mean score of all 24 performance items were 3.53 at 5 point Likert scale and among others safety and security, parking easiness, public transportation, staff kindness and overall facility quality had relatively high levels of satisfaction. Third, IPA results showed that program variety and experience program belonged to II in the quadrant which means ‘concentrate here’. In addition, such items belonged to I in the quadrant as safety and security, parking easiness, staff kindness, overall facility quality, public transportation, amenity, and information signs. Conclusion: Overall performance levels of all 24 factors were lower than overall importance levels of all the items. In conclusion, it was suggested that visitors’ satisfaction factors should be appropriately dealt with based on the IPA matrix. Especially, items with high importance and low performance levels need more marketing and operating efforts from event organizers.

        Multiplex immunoassays using virus-tethered gold microspheres by DC impedance-based flow cytometry

        <P><B>Abstract</B></P> <P>Bead-based multiplex immunoassays for common use require enhanced sensitivity and effective prevention of non-specific adsorption, as well as miniaturization of the detection device. In this work, we have implemented virus-tethered gold microspheres for multiplex immunoassay applications, employing a DC impedance-based flow cytometer as a detection element. The advantages of virus-tethered gold microspheres, including excellent prevention of non-specific adsorption, are extended to signal enhancement arising from the large quantity of antibody loading on each virion, and to flexible movement of filamentous virus. Individual virus-tethered beads generate their own DC impedance and fluorescence signals, which are simultaneously detected by a chip-based microfluidic flow cytometer. This system successfully realized multiplex immunoassays involving four biomarkers: cardiac troponin I (cTnI), prostate specific antigen (PSA), creatine kinase MB (CK-MB), and myoglobin in undiluted human sera, elevating sensitivity by up to 5.7-fold compared to the beads without virus. Constructive integration between filamentous virus-tethered Au-layered microspheres and use of a microfluidic cytometer suggests a promising strategy for competitive multiplex immunoassay development based on suspension arrays.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Filamentous viruses are proved as efficient support for antibody immobilization, increasing antibody loading up to 3.7-fold. </LI> <LI> The sensitivity of virus-tethered bead-based immunoassay was enhanced up to 5.7-times compared to the beads without virus. </LI> <LI> DC impedance-based microfluidic cytometry is successfully integrated with bead-based immunoassay for multiplex analysis. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Ductilization of Nanoporous Ceramics by Crystallinity Control

        <P>Synthesizing ceramic materials with a significant amount of deformability is one of the most important engineering pursuits. In this study, we demonstrate the emergence of metal-like plasticity through the crystallinity control in the monolithic zirconia with the vertically aligned honeycomb-like periodic nanopore structures fabricated using the anodizing technique. The crystalline orders of the nanoporous zirconia films vary between monoclinic, tetragonal, and amorphous phases after the heat treatment and/or proton irradiation, whereas the vertical pore structures are maintained. The micropillar compression tests on those samples reveal a large amount of plasticity, more than 20% of total stains, in the as-anodized and proton-irradiated samples, both of which contain the amorphous phase. In contrast, the fully crystallized zirconia that resulted from annealing at 500 °C shows the brittle failure, the typical characteristic of conventional ceramic foams. These results offer a new opportunity for the nanoporous ceramic materials to be used in various applications, benefited from the tunable structural stability.</P> [FIG OMISSION]</BR>
