Expression of eIF(iso)4E having mutations in cap binding pocket give broad TuMV resistance

eIF4E family is well known for recessive resistance gene of potyvirus in many crops. And Turnip mosaic virus (TuMV) is one of the major viruses in Brassicaceae crops which belong to the genus Potyvirus. To elucidate the key amino acids in the interaction between TuMV VPg and Brassica eIF(iso)4E, amino acids of eIF(iso)4E were mutated. Seven amino acids in cap binding pocket were chose for the candidate amino acid that may play a role in the interaction of TuMV VPg. We demonstrated that a single amino acid mutation in cap binding pocket of Brassica eIF(iso)4E can abolish the interaction with TuMV VPg. eIF(iso)4E which has a mutation at each W49, W95 and K150 positions impaired in its interaction with VPg prominently according to the yeast two hybrid analysis. Complementation of an eIF4E knockout yeast strain by mutated eIF(iso)4E proteins showed that all eIF(iso)4E mutants were able to complement eIF4E of yeast. To find out if these mutations affect the susceptibility of Chinese cabbage, transformant analysis was performed. eIF(iso)4E W95L, W95L/K150E and susceptible wild type were over-expressed in susceptible Chinese cabbage. According to the TuMV screening result of T1 and T2 transformants, over-expression of the eIF(iso)4E mutants showed resistance to four TuMV strains (CHN2, 3, 4 and 5). Our results support that the mutations in eIF(iso)4E may control the broad spectrum TuMV resistance.

목적: The forkhead box protein 1 (FOXP1) is considered as both a tumor suppressor candidate and a potential oncogene. Here, we investigated FOXP1 expression in cervical cancer, and the clinical significance of FOXP1 and it`s mechanism of action in cervical cancer. 방법: FOXP1`s functional role was investigated by employing lentiviral-mediated overexpression and knockdown in cervical cancer cell lines. Immunohistochemical staining for FOXP1 was performed on a cervical cancer tissue microarray consisting of 158 primary cervical cancers, 280 cervical intraepithelial neoplasias (CINs), and 378 matched normal tissues. 결과: FOXP1 overexpression promoted cell proliferation and tumorigenesis, whereas FOXP1 knockdown inhibited these properties in HeLa and CaSki cell lines. By immunohistochemical staining, FOXP1 expression increased during the normal to tumor transition of cervical carcinoma (p< 0.001), and this increased expression was significantly associated with tumor stage (p=0.009) and tumor grade (p< 0.001). In multivariate analysis, FOXP1+ (p=0.031) and tumor stage (p=0.032) were independent prognostic factors for overall survival. 결론: Taken together, our data indicate that FOXP1 has a crucial role in cervical cancer progression, and its overexpression is associated with poor prognosis, supporting that FOXP1 may be used as a promising novel target for therapeutic interventions.

Teratoma originate from one or more germ cell layer and commonly arise from sacrococcygeal area in neonate. Teratoma arising from the oropharyngeal cavity is called epignathus tumor and is extremely rare in neonate. Clinical manifestation of epignathus tumor vary from asymptomatic to severe respiratory distress symptom. It is reported that most of the tumor are benign in nature. Large teratoma can be diagnosed by prenatal ultrasonography, but most cases were diagnosed with computed tomography or magnetic resonance image after birth. Prognosis is determined by the need for neonatal resuscitation for respiratory distress at the time of birth and the extent of tumor, involving large vessles, skull base or communication with the brain. We experienced a case of epignathus tumor in a neonate with severe respiratory distress immediately after birth, so that reported with review of the literature.
