      • 백서에서 신 허혈성 손상에 미치는 칼슘 길항제의 효과

        Renal ischemia is one of the most common causes of acute renal failure. Four factors related to the pathogenesis of acute renal failure are vasoconstriction, decreased glomerular filtration rate, tubular back leak of filtrate, and intratubular obstruction. The cellular response to renal ischemic insults include decreased content of adenosine trihosphate, lipid peroxidation induced membrane degradation, alteration in cellular pH, and calcium or phospholipase induced mitochondrial dysfunction. Much attention has been given to the role of increased cellular calcium as a pathogenetic contributor to cell injury during ischemia. Author studied the protective effects of calcium antagonists on cellular injury during renal ischemia in rat. To investigate the protective role of these agents, author measured the amount of malondialdehyde(MDA) and the enzyme activities of free radical scarvengers-superoxide dismutase(SOD), catalase and glutathione peroxidase from non-pretreated group and calcium antagonists pretreated groups in control, ischemia and reflow subgroups. The results were summerized as follows: 1) The amount of MDA in non-pretreated group was higher in the reflow compared with the control(<p<0.01). But, in all pretreated groups, there was no statistically difference in the amount of MDA. 2) The SOD activity in non-pretreated group was lower in both the ischemia and the reflow compared with the control (P<0.05). But, in both verapamil and trifluoperazine-pretreated groups, there was no statistically difference in the SOD activity. 3) Both catalase and glutathione peroxidase activities in non-pretreated group were lower in both the ischemia and the reflow compared with the control (P<0.05). But in all pretreated groups, there was no statically difference in both catalase and glutathione peroxidase activities. These results suggest that free radical mediated ischemic injury by renal artery clamp in rat can be protected by intraperitoneal pretreatment with calcium antagonists. As trifluoperazine has a protective effect in renal ischemia, the calcium activated calmodulin dependent enzyme may play a role in renal ischemic injury.

      • 반건식-백필터에 의한 소각배가스 중의 HCI, SOx 제거 (Ⅰ)

        This study has been carried out to investigate the removal efficiencies of HCl/SOx in a Spray-Dryer/Fabric-Filter system. In HCl/SOx removal, we could identify the key factors such as spray dryer outlet temperature, molar equivalence ratio(MER) and fabric filter velocity, but we couldn't find out the correlation of HCl and SOx. The removals of HCl/SOx were inversely proportional to spray dryer outlet temperature(l30~170℃) and filter velocity(l.0~1.5 m/min), at the MER, HCl removal efficiency was strongly proportional to MER, but SOx, removal efficiency was not particularly increased by MER over 1.5. The removal efficiencies of HCl/SOx were improved over 10% at the fabric filter. In this Spray-Dryer/Fabric-Filter system, HCl/SOx removal efficiencies were about 99%, 96% respectively.

        치주조직 재생을 위한 칼슘 포스페이트 글라스 분말이 백서 두개골 결손부의 골조직 재생에 미치는 영향

        The treatment of the tone defects resulting from trauma, neoplasm, surgery, or infection is one of the major concern in dentistry. The major goal is the functional, esthetical regeneration of supporting structures already destructed by disease. Transplantation technique have been used to provide a scaffold for bone regeneration, to augment bony defects resulting from trauma or surgery, to restore bone loss caused by dental disease, to prevent the collapse the alveolar ridge in recent extraction sites, to replace bone loss by periodontal disease, to augment the alveolar ridge in implant surgery. There are autogeneous, allogenic, xenogenic and alloplastic bone-grafts in transplantation. Among the alloplastic bone-graft materials, calcium phosphates have been received the most attention. In this study, the critical size defects were surgically produced in the calvarial bone of Sprague-Dawley rats using the 8 ㎜ trephine bur. Calcium phosphate glass powders with mean diameter 400 ㎛ transplanted onto the produced detects in the experimental group, while sutured without grafting anything in the control group. Histomorphometric as well as radiodensitometric analyses were performed after sacrifice at 2, 4 and 8 weeks following operation. The prepared calcium phosphate glass powder with average size 400 ㎛ in CaO-CaF_2-P_2O_5-MgO-Al_2O_3 promoted new bone formation in the calvarial defects in the Sprague-Dawley rats. New bone was formed in the upper side of the detects as well as the defect margin and dura mater. Experimental group always exhibited higher values in the length, area and density of the newly formed bone than that of the control group. There were significant differences between the experimental and the control groups, except the density after 4 weeks (p<0.05).

        The growth in interchange of business and other sensitive data has led to increasing interest in access control. Access control for XML is receiving significant attention as XML is gaining popularity for storing and exchanging information. Until now, research on XML security has been focused on the security of data communication using digital signatures or encryption technologies. As XML is also used for a data representation of data storage, XML security comes to involve not only communication security but also managerial security. In this paper, we discuss an XML access control model and propose a technique that supports not only read operations but also update operations.

      • Benzoyl peroxide의 환경에서의 초기 위해성 평가

        Benzoyl peroxide is a High Production Volume Chemical, which is produced about 1,375 tons/year in Korea as of 2001 survey. The substance is mainly used as initiators in polymerization, catalysts in hte plastics industry, bleaching agents for flour nad medication for acne vulgaris. In this study, quantitative Structure-Activity Relationships(QSAR) are used for getting adequate information on the physical-chemical properties of this chemical. And hydrolysis in water, acute toxicity to aquatic and terrestrial organisms for benzoyl peroxide were studied. The physical-chemical properties of benzoyl peroxide were estimated as followed; vapor pressure=0.00929 pa, Log K_(ow)=3.43, Henry's Law constant=3.54×10^(-6) atm-m³/mole at 25℃, the half-life of photodegradation=3days and bioconcentration factor(BCF)=92. Hydrolysis half-life of benzoyl peroxide in water was 5.2hr at pH 7 at 25℃ and according to the structure of this substance hydrolysis product was expected to benzoic acid. Benzoyl peroxide has toxic effects on the aquatic organisms. 72 hr-ErC_(50)(growth rate) for algae was 0.44mg/L, 48hr-EC_(50) for daphnia was 0.07mg/L and the 96hr-LC_(50) of acute toxicity to fish was 0.24mg/L. Acute toxicity to terrestrial organisms(earth worm) of benzoyl peroxide was low(14 day-LC_(50)=>1,000mg/kg). Although benzoyl peroxide is high toxic to aquatic organisms, the substance is not bioaccumulated because of the rapid removal by hydrolysis (half life=5.2hr at pH 7 at 25℃) and biodegradation(8.% by BOD after 21days). The toxicity observed is assumed to be due to benzoyl peroxide rather than benzoic acid, which shows much lower toxicity to aquatic organisms. One can assume that effects occur before hydrolysis takes place. From the acute toxicity value of algae, daphnia and fish, an assessment factor of 100 was used to determine the predicted no effect concentration(PNEC). The PNEC was calculated to be 0.7㎍/L based on the 48hr-EC_(50) daphnia(0.07mg/L). The substance shows high acute toxicity to aquatic organisms and some information indicates wide-dispersive use of this substance. So this substance is a candidate for further work, even if it hydrolysis rapidly and has a low bioaccumulation potential. This could lead to local concern for the aquatic environment and therefore environmental exposure assessment is recommended.

      • 원예치료 및 미술치료가 학교 부적응 청소년의 자아개념 향상에 미치는 영향연구

        본 연구는 원예치료와 미술치료가 학교부적응 청소년의 자아개념, 내적 통제력, 불안감소에 미치는 영향을 조사하고자 실시되었으며, 그 결과는 각 치료의 효과성이 비교·분석되었다. 인천 남구에 위치한 W공업고등학교에서 대조군 15명, 원예치료군 15명, 미술치료군 15명을 주 2회씩 2001년 9월 27일부터 12월 13일까지 원예치료군에는 원예치료 프로그램을 미술치료군에는 미술치료프로그램을 실시하였다. 원예치료 및 미술치료는 각각 전문 치료사에 의해서 행해졌다. 원예치료 및 미술치료 프로그램의 효과를 알아보고자 자아개념(Self-Concept Scale), 내적 통제력 척도(Inner-Control Scale), 특성불안 척도(Trait-Anxiety Scale)을 각 프로그램의 실시전과 실시후로 조사하여 비교하였다. 원예치료군에 있어서는 자아개념척도 중 자아수용, 성격적 자아, 총 자아 긍정 항목에서 통계적으로 유의한 향상을 보였고, 특성불안척도에서는 우울 및 불안 항목에서 매우 유의한 감소를 나타내었다. 한편, 미술치료군에 있어서는 내적통제력과 특성불안척도중 자신감이 유의하게 향상된 것으로 나타났다. 이상의 결과를 종합하여 볼 때, 원예치료군은 프로그램 과정중 자기표현의 기회를 제공함으로써 자아개념이 향상되었으며, 또한, 공동작업을 하는 집단 프로그램을 통하여 내면의 불안과 갈등이 완화되었다고 판단된다. 반면, 미술치료군은 치료자와의 개인적인 상담과정을 통하여 내면의 불안과 갈등이 완화되었고, 지점토 및 칼라믹스 만들기 등 활동적인 프로그램을 통하여 내면의 감정을 통제하는 능력이 향상되었다고 판단된다. This study was conducted to evaluate the effect of HT (horticultural therapy) and AT (art therapy) on the changes of self-concept, inner control, trait anxiety of the school maladjusted adolescents. 45 school maladaptive of teenage students participated in therapy program at the W high school were randomly assigned to 3 groups: 15 for the horticultural therapy, 15 for art therapy, 15 for control group. The program were carried out from Sep. 27 to Dec. 13, 2001 as an activity group. Activity group attended twice a week for 1 hour. For evaluation of the effectiveness of each therapy, Self-Concept Scale, Inner-Control Scale, Trait-anxiety Scale were used 2 times, that is, before/after HT or AT program. According to the results, self satisfiction, personal self, and total positive self items among Self-Concept Scale were significantly improved, and tension and anxiety item among Trait anxiety Scale were significantly decreased in HT group. On the other hand, in AT group, both inner control and self confidence item in Trait-anxiety Scale were significantly improved. In conclusion, in HT group it was suggested that the improvement of self concept could be brought about by being provided the chance of self expression during program and the alleviation of inner anxiety and discord resulted from group therapy program in which joint work have been provided. In case of AT group, it was logically postulated that the alleviation of inner anxiety and discord resulted from personal counselling with AT therapist during program and the improvement of inner control was caused by active program such as painting paper clay, making color mix, and etc.

        Background : The cun measurement System, an essential and convenient method in locating acupoints, has been widely used in the practice of acupuncture. However, traditional cun measurement has been criticized for its lack of reliability. Objectives : The purposes of this study are to determine if one cun measured by the directional methods have a consistency with that of proportional methods and to investigate which factors are related with these differences, especially in forearm. Methods : The distance between the elbow crease and the wrist crease of forearm was compared to a reference value of one cun obtained by the directional method. In this method, one cun is one third of the distance between index finger and Small finger of a subject, measured at proximal interphalangeal joint. In addition, to investigate the factors influencing the differences between these two methods, we measured the height and body weight and calculated body mass index (BMI). Finally we analyzed the factors correlated with these lengths by linear regression test. Results : The results showed that one cun obtained by the directional methods were significantly different from one cun by the proportional methods in forearm. It was demonstrated that the length acquired with the directional method was more correlated with body weight and body mass index, while the length obtained by the proportional method was more correlated with the height.

        Correlation between Capillary Blood-spotted Filter Paper Thyrotropin Results and Serum Thyroid Function Tests in Premature Neonates

        Objective: The interpretation of thyroid function test results in preterm infants is affected by various conditions. Neither a definite thyrotropin cut-off nor intercurrent conditions have been clearly determined for premature neonates. This study aimed to determine the specific considerations required in the interpretation of capillary blood thyrotropin concentration by comparing it to serum thyrotropin when screening for congenital hypothyroidism in preterm neonates. Methods: A retrospective review was performed on 57 premature newborns who were screened for thyroid function test. The comparison of thyrotropin measured by two different methods, dried capillary blood-spotted filter paper using an immunofluorometric assay and serum radioimmunoassay, was performed. Clinical and biochemical factors affecting the difference between the two methods were investigated. Results: Filter paper thyrotropin concentration showed a positive correlation with the serum thyrotropin concentration. In total, 50.9% of premature neonates had a difference in the concentration ≥3.0 mIU/L between the methods. In these subjects, the serum thyrotropin concentration was higher, whereas other factors, such as maternal factors, neonatal anthropometric parameters, Apgar score, and corrected age, were not different when compared to that in subjects with a difference of <3.0 mIU/L. Conclusion: The filter paper thyrotropin concentration differed from the serum level in a substantial portion of premature neonates, and the degree of deviation may be affected by the serum thyrotropin concentration and gestational age at birth. Further investigations and longitudinal studies are warranted to establish clinical consensus guidelines for hypothyroidism screening in preterm neonates.

      • Egr-1 Mediates Transcriptional Activation of IGF-Ⅱ Gene in Response to Hypoxia

        We have previously reported that the exposure of human HepG2 cells to hypoxic conditions results in the overexpression of human insulin-like growth factor Ⅱ (IGF_Ⅱ) mRNA whose size is 6.0 kb. This particular size of IGF-Ⅱ mRNA is transcribed under the control of the IGF-Ⅱ P3 promoter. In the present study, to delineate the molecular mechanism for the activation of the IGF-Ⅱ gene, we examined the induction of P3 promoter activity in HepG2-cells by hypoxia in the transient expression system. In this system, thpoxia induced a linear increase within 24 h in the expression of luciferase that was driven by the IGF-Ⅱ P3 promoter. To pattern of regulators of the P3 promoter, Egr-1, Sp-1, and WT1, were analyzed by reverse transcription-PCR and Northern blot analysis. We contrast, the level of WT1, a repressor of IGF-Ⅱ expression, was markedly decreased during hypoxia. The mRNA stability assay revealed that the induction of transcription is the mechanism of underlying Egr-1 mRNA elevation. We then investigated the effects of hypoxia on the DNA binding activity of Egr-1. Both electrophoretic mobility shift assay and supershift assay demonstrated that the DNA binding activity of the Egr-1 protein was increased by hypoxia. In addition, the level of Egr-1 protein increased under the hypoxia as determined by Western blot analysis. Cotransfection of HepG2 cells with an Egr-1 expression vector and an IGF-Ⅱ P3 promoter-luciferase reporter plasmid showed that the transcription of IGF-Ⅱ was activated by Egr-1 in a dose-dependent manner. Moreover, the elecation of IGF-Ⅱ P3 promoter activity was induced synergistically by the cotreatment of hypoxia whth Egr-1 overexpression. Deletion of sequences in the IGF-Ⅱ P3 promoter containing Egr-1 binding sites did not respond to hypoxic stress. Taken together, these data strongly indicate that hypoxia-induced IGF-Ⅱ expression in HepG2 cells is due to the enhanced activity of Egr-1 on the IGF-Ⅱ P3 promoter and that the Egr-1 binding site in the IGF-Ⅱ P3 promoter is essential for the transcriptional regulation of IGF-Ⅱ under hypoxic conditions.

        Mutant p53-Notch1 Signaling Axis Is Involved in Curcumin-Induced Apoptosis of Breast Cancer Cells

        Notch1 has been reported to be highly expressed in triple-negative and other subtypes of breast cancer. Mutant p53 (R280K) is overexpressed in MDA-MB-231 triple-negative human breast cancer cells. The present study aimed to determine whether the mutant p53 can be a potent transcriptional activator of the Notch1 in MDA-MB-231 cells, and explore the role of this mutant p53-Notch1 axis in curcumin-induced apoptosis. We found that curcumin treatment resulted in an induction of apoptosis in MDA-MB-231 cells, together with downregulation of Notch1 and its downstream target, Hes1. This reduction in Notch1 expression was determined to be due to the decreased activity of endogenous mutant p53. We confirmed the suppressive effect of curcumin on Notch1 transcription by performing a Notch1 promoter-driven reporter assay and identified a putative p53-binding site in the Notch1 promoter by EMSA and chromatin immunoprecipitation analysis. Overexpression of mutant p53 increased Notch1 promoter activity, whereas knockdown of mutant p53 by small interfering RNA suppressed Notch1 expression, leading to the induction of cellular apoptosis. Moreover, curcumin-induced apoptosis was further enhanced by the knockdown of Notch1 or mutant p53, but it was decreased by the overexpression of active Notch1. Taken together, our results demonstrate, for the first time, that Notch1 is a transcriptional target of mutant p53 in breast cancer cells and suggest that the targeting of mutant p53 and/or Notch1 may be combined with a chemotherapeutic strategy to improve the response of breast cancer cells to curcumin.
