      • 생약재 첨가 사료를 투여한 넙치(Paralichthys olivaceus)의 비특이적 면역반응, 혈액성분 및 항병력 효과

        본 연구는 식물성 생약재의 열수추출이 농도별로 첨가된 사료를 넙치(Paralichthys olivaceus)에 경구 투여한 다음 비특이적 면역반응 및 질병 저항성의 향상여부를 알아보고자 하였다. 실험에는 약쑥과 삼지구엽초를 7:3(w/w)의 비율로 열추출한 후, 0%, 0.1%, 0.5% 및 1%의 농도로 첨가하여 만든 2종류 혼합생약재(2-혼합구)와 약쑥, 삼지구엽초, 연교, 구절초, 박하, 지유, 삼백초, 상백피, 팔각의 9종류 생약재가 동일한 농도로 열수추출한 후 제작한 9종류 혼합생약재(9-혼합구)를 사용하였다. 이들 농도별 첨가사료를 넙치(평균 10.3±2.5g)에 12주 동안 경구 투여하면서 혈청내 라이소자임 활성, 혈청의 Escherichia coli에 ??살균능력 및 혈액학적 변화를 조사하였다. 그리고 최종 투여 12주 후에 넙치 에드와드병의 원인균 Edwardsiella tarda의 생균 공격실험을 실시하여 상대생존율(RPS)로써 저항성 향상효과를 살펴보았다. 2-혼합구 0.1%는 비특이적 면역활성을 크게 증가시켰으며, 상대생존율이 67%로 높게 나타나 가장 효과적인 질병 저항성 향상효과를 보였다. 따라서 2-혼합구 0.1%는 넙치 체내 비특이적인 면역기구를 자극함으로써 어체의 자연저향성을 증강시켜 세균성 질병의 감염에 대한 방어력을 상당히 향상시킬 수 있을 것으로 사료된다. 9-혼합구 0.1% 및 0.5%는 비록 비특이적 면역활성은 증가시키지 못하였으나, 상대생존율은 각각 53%를 나타내어 질병 저항성을 일부 향상시키는 효과가 있었다. Effects of medicinal herb extract on nonspecific immune responses, hematology and disease resistance against Edwardsiella tarda in olive flounder, Paralichthys olivaceus were evaluated. Wormwood, Artemisia asiatica NAKAI and barrenwort, Epimedium koreanum NAKAI were mixed at a ratio of 7 : 3 (w/w) for 2-herbs extract and wormwood, barrenwort, Korean forsythia, Forsythia koreana NAKAI, chrysanthemum, Chrysanthemum zawadskii var. latilobum KITAMURA, peppermint, Mentha arvensis L. var, piperascens MALINV., great burnet, Snaguisorba afficinalis L., Lizard tail. Saururus chinensis BAILL., mulberry, Morus alba L., and star anise, Illicium varum HOOK, f, at the same weight for 9-herbs extract. Two-herbs of 9-herbs extract were prepared by heating after adding 10㎖ of distilled water per g of the herb mixtures. Fish (10.3±2.5g) were fed the experimental diets supplemented with the 2-herbs or 9-herbs extract at the different concentrations of 0%, 0.1%, 0.5% and 1% per kg diet for 12 weeks. Lysozyme and bactericidal activities of serum, and hematological characteristics were examined during experimental period. After feeding test period, all experimental groups were challenged with E. tarda. Lysozyme activity from the fish fed the diet supplemented with 0.1% or 0.5% of 2-herbs extract was significantly higher than the control. But there was no difference both in bactericidal activity and hematology among each group. Sixty seven % of relative percent survival values (RPS) in the group fed the diet supplemented with 0.1% of 2-herbs was higher than the other group and the control. These results suggest that supplenmentation of 0.1% of 2-herbs extract to a commercial diet may enhance disease resistance in olive flounder. Although both 0.1% and 0.5% 9-herbs extract did not improve non-specific immune reponses, they could enhance disease resistance of 53% RPS, respectively.

        자갈함량과 질소시비량이 고랭지 배추재배시 침투수에 의한 질소용탈 및 생육에 미치는 영향

        고랭지 배추재배지의 성토재로 사용되고 있는 석비레는 모래와 자갈함량이 높아 토양침식과 양분용탈의 주 요인이 되고 있어 본 연구는 자갈함량에 따른 배추수량과 질소의 용탈양상을 구명하고자 pot 시험을 수행하였다. 시험 결과, 배추의 생체수량은 자갈의 함량이 증가할수록 감소하는 경향이었고, 특히 질소시비량 240 kg/ha 구에서는 현저한 수량감소를 보였다. 질소시비량 60 kg/ha 구에서는 자갈함량 30%, 질소시비량 120 kg/ha 구에서는 자갈함량 50%를 변환점으로 하여 배추수량이 급격히 감소하였다. 배추의 생육장해는 질소시비량이 240 kg/ha인 구에서는 대부분 위조현상이 나타났으며 결구 초기인 8월 초순에는 자갈함량이 50% 이상이 되면 질소시비량을 ha당 120 kg만 시용해도 50% 이상이 위조현상을 보였다. 침출수의 암모늄태 질소는 자갈함량과 질소시비량이 많을수록 침출수의 농도가 높아지는 경향을 보였는데, 특히 배추의 고사가 발생한 30% 이상의 자갈함량과 질소시비량 240 kg/ha인 처리구의 농도는 139-339 mg/L이었다. 침출수의 질소용탈량은 자갈함량에 따라 정의 상관관계를 보였고, 자갈함량이 10% 증가될 때마다 pot당 0.14g씩 증가하여 자갈함량이 10%일 때에 비하여 자갈함량이 30%가 되면 약 3.8배 증가하였다. 배추재배 시험 후 토양의 화학성은 NH₄-H 농도와 pH는 처리간에 일정한 경향을 보이지 않았고, NO₃-N 농도는 자갈함량 30%까지는 증가하다가 50%부터는 감소하는 경향을 보였다. 사양토에서 자갈의 함량이 30%일 때 질소비료 240 kg/ha, 자갈함량이 50%일 때 질소비료 120 kg/ha을 시용하면 배추의 생육장해를 가져올 수 있다. 따라서 이러한 토양에 시비를 하게 되면 염류장해와 위조현상을 초래하므로 토양유실이 심한 고랭지는 특수환경을 고려한 시비기준이 설정되어야 할 것으로 판단된다. Continuous monocropping of Chinese cabbage in Gangwon highland increased gravel and sand contents due to surface soil erosion. Nutrient leaching and Chinese cabbage growth were investigated with different treatments of gravel contents and nitrogen application levels by using 0.5 m²Wagner pots. Gravel contents were 0, 10, 30, 50, 70, and 90%(w/w), nitrogen application levels were 60, 120, and 240 kg/ha, and manure compost application rate was 15 ton per hectare, respectively. Wagner pots were filled with loamy sand soil mixed with 5 cm-sized gravels. Fresh weight of Chinese cabbage was decreased as gravel contents in soil increased, and particularly severely decreased at 240 kg-N/ha. Yields of Chinese cabbage were remark-ably decreased at the rate of 60 kg-N/ha with 30% gravel content and 120 kg-N/ha with 50% gravel content. Most of Chinese cabbages were severely wilted by heavy N application at the rate of 240 kg-N/ha in the middle of growth stages regardless of gravel contents, while about 50% of Chinese cabbage showed wilting symptom in the treatment of more than 50% of gravel contents and 120 kg-N/ha. N content in leachate in-creased as gravel content and N application increased. The relationship between gravel content and N con-tents showed linear regression: N in leachate = 0.014(gravel content) -0.039 (r = 0.961). Particularly, NH₄-N contents in leachates with more than 30% gravel content and 240 kg-N/ha ranged from 139~339 mg/L. Chinese cabbage growth in loamy sand soil containing 30%, and 50% gravel contents could be adversely affected by N application at the rate of 240, and 120 kg-N/ha, respectively.

        Congenital Non-Neural Granular Cell Tumor Mimicking Nevus Lipomatosus Superficialis

        A 4-month-old infant presented with asymptomatic soft nodules on his right forearm, which had developed since birth. On the suspicion of nevus lipomatosus superficialis (NLS), biopsy was performed. Histopathologic findings showed monomorphic polygonal cells with abundant granular cytoplasm. Immunohistochemical stains for CD68 and vimentin were strongly positive, but were negative for S-100 protein. Based on the pathologic findings, the patient was diagnosed as non-neural granular cell tumor (NN-GCT). GCT can be divided into conventional and non-neural GCT by immunoreactivity for S-100 protein. NN-GCT is typically manifested as a well-circumscribed, papulo-nodular dermal mass, and is known to occur in a younger group than does in conventional GCT, but is rare among children. To our knowledge, there have been no case reports of NN-GCT which appeared at birth and presented as grouped nodules. Therefore, we report this interesting case of congenital NN-GCT clinically mimicking NLS. (Ann Dermatol 29(6) 776∼778, 2017)

        지방공기업의 재정성과에 영향을 미치는 외부환경 요인에 관한 연구 -도시개발공사를 중심으로

        현재 지방재정 악화의 숨은 원인으로서 지방공기업의 막대한 부채가 심각한 우려를 야기하고 있는 가운데, 지방공기업의 재정성과 제고를 위한 실효성 높은 정책대안의 모색이 더욱 요구된다. 이와 같은 문제의식을 바탕으로 본 연구에서는 광역자치단체 산하 16개 도시개발공사의 2006년부터 2015년까지의 패널데이터를 가지고서 이들 지방공기업의 재정성과에 영향을 미치는 외부환경 요인들을 탐색하였다. 본 연구에서의 분석결과, 재정적 요인인 관할 지방자치단체의 재정자주도와 정치적 요인인 단체장의 지지율 및 정치성향이 도시개발공사의 재정성과에 유의미한 영향을 주는 것으로 나타났다. 또한 정당성 확보 요인으로서 자신을 제외한 나머지 도시개발공사의 전년도 재정성과 역시 도시개발 공사의 재정성과에 유의미한 영향을 미치는 것으로 나타났다. 이와 같은 분석결과를 바탕으로 도시개발공사의 재정성과는 조직의 내부적 특성에 못지않게 관할 지방자치단체와의 관계를 비롯한 외부환경 요인에 의해 영향을 받는다는 것을 확인할 수 있다. 따라서 본 연구에서 지방공기업 재정성과의 향상을 위해 제시하고자 하는 정책적 시사점은 다음과 같다. 먼저 단기적 관점에서, 자치단체장의 합리적인 정책결정능력 제고를 위한 제도적 장치가 구비되어야 하고, 이를 위해 지방의회의 역량은 더욱 강화되어야 한다. 보다 장기적 관점에서는, 지방 공기업의 온전한 자율경영을 가능케 해주는 제도적 여건을 마련하는 것이 필요하며, 특히 정치적 독립성과 정책적 합리성을 바탕으로 지방공기업이 성과에 대한 책임을 오롯이 짐으로써 당해 기업의 실질적인 주인으로 거듭나야 할 것이다. With local public enterprises` growing debts causing concerns for local finances, there is an increasing need for effective measures to improve the financial performance of local public enterprises. With this in mind, the present study has explored environmental factors influencing the financial performance of local public enterprises using panel data from 2006 to 2015 involving 16 Urban Development Corporations under Regional Governments in Korea. The study has found that the financial autonomy of local governments as a financial factor and the approval rating and political ideology of chief executive officers as a political factor have a statistically significant effect on the financial performance of Urban Development Corporations. In addition, the financial performance of other Urban Development Corporations as a legitimacy factor was also found to have a significant effect. Taken together, these results suggest that the financial performance of local public enterprises is affected not only by their internal organizational characteristics but also by their external environmental factors including their interaction with the host governments. The policy implications made by this study are clear: First of all, in the short term, institutional arrangements need to be in place to improve the decision-making capability of local chief executive officers and also strengthen legislative control over the government and its public enterprises. In the long term, it is necessary for local governments to provide local public enterprises with managerial discretion and political independence and instead make sure that they assume responsibility for their financial performance.

      • Clinical efficacy of diphenylcyclopropenone immunotherapy as monotherapy for multiple viral warts: A retrospective review of 250 patients

        Background: Diphenylcyclopropenone (DPCP) immunotherapy for viral warts has been used for many years, but no studies have analyzed the treatment effects of DPCP alone, without the use of conventional therapy before immunotherapy. Objectives: We evaluated clinical efficacy of DPCP monotherapy compared to cryotherapy and pulsed dye laser therapy. We also assess the impact of initial sensitization on clinical response. Methods: We retrospectively analyzed the medical records of 250 patients with viral warts between January 2008 and December 2015. Results: DPCP-only group (n=43) showed a lower clinical response (75.6%) than cryotherapy-only (n=171, 89.8%, p<.0.01) and PDL-only group (n=36, 90.3%, p<.0.01). The positive clinical response in the successful sensitization group (n=21) was 76.3% (119/156), while 74.4% in the sensitization fail group (p=0.870). Conclusion: DPCP monotherapy has a lower clinical response than conventional therapy. Initial sensitization to DPCP does not predict a failed response with continued immunotherapy for viral wart.

      • [P148] A case of allergic contact dermatitis due to ciprofloxacin otic drop

        A 51-year-old man presented with erythematous scaly papules and patches with oozing on the right ear lobe, face, and neck for two months. He had a history of canal wall up mastoidectomy with canaloplasty due to chronic otitis media with tympanic membrane rupture about a year ago. Ciprofloxacin otic drop had been used 1 month after the surgery because of continuous otorrhea, which was considered as surgical complication. Pruritic papules and patches with oozing and crust formation spread from right ear lobe to cheek and neck after using ciprofloxacin otic drop. As patch test showed positive results for otic drop, patient was diagnosed as allergic contact dermatitis (ACD) due to ciprofloxacin ear drop. ACD caused by topical antibiotics is rare, but well documented side effect. Aminoglycoside, polypeptides, β-lactam, macrolide, benzoyl peroxide, clindamycin, mupirocin, fusidic acid are well-known antibiotic sensitizer. However, fluoroquinolone as a potent contact allergen had not been reported to our knowledge. Indeed our case presented strong positivity in patch test, which can be an obvious evidence for fluroquinolone as a potent skin sensitizer. We herein report this interesting case of ACD caused by ciprofloxacin otic drop.

        Knockdown of Archvillin by siRNA Inhibits Myofibril Assembly in Cultured Skeletal Myoblast

        A myofiber of skeletal muscle is composed of myofibrils, sarcolemma (plasma membrane), and constameres, which anchor the myofibrils to the sarcolemma. Achvillin is a recently identified F-actin binding muscle protein, co-isolates with dystrophin and caveolin-3 in low-density sarcolemma of striated muscle, and colocalizes with dystrophin at costameres, the specialized adhesion sites in muscle. Archvillin also binds to nebulin and localizes at myofibrillar Z-discs, the lateral boundaries of the sarcomere in muscle. However other roles of archvillin on the dynamics of myofibrillogenesis remain to be defined. The goal of this study is, by using siRNA-mediated gene silencing technique, to investigate the effect of archvillin on the dynamics of myofibrillogenesis in cell culture of a mouse skeletal myogenic cell line (C2C12), where presumptive myoblasts withdraw from the cell cycle, fuse, undergo de novo myofibrillogenesis, and differentiate into mature myotubes. The roles of archvillin in the assembly and maintenance of myofibril and during the progression of myofibrillogenesis induced in skeletal myoblast following gene silencing in the cell culture were investigated. Fluorescence microscopy demonstrated that the distribution of archvillin was changed along the course of myofibril assembly with nebulin, vinculin and F-actin and then located at Z-lines with nebulin. Fluorescence microscopy demonstrated that knockdown of mouse archvillin expression led to an impaired assembly of new myofibrillar clusters and delayed fusion and myofibrillogenesis although the mouse archvillin siRNA did not affect those expressions of archvillin binding proteins, such as nebulin and F-actin. This result is corresponded with that of RT-PCR and western blots. When the perturbed archvillin was rescued by co-transfection with GFP or Red tagged human archvillin construct, the inhibited cell fusion and myotube formation was recovered. By using siRNA technique, archvillin was found to be involved in early stage of myofibrillogenesis. Therefore, the current data suggest the idea that archvillin plays critical roles on cell fusion and dynamic myofibril assembly.
