        Modeling and optimization of cutinase production by recombinant Escherichia coli based on statistical experimental designs

        Statistics-based experiment designs were used to optimize the culture medium (glucose, yeast extract, IPTG,tween-60, and CaCl2) for cutinase production by recombinant Escherichia coli. A 25-1 fractional factorial design augmented with center points revealed that glucose, yeast extract, and IPTG were the most significant factors, whereas the other factors were not important within the levels tested. The method of steepest ascent was used to approach the proximity of optimum, followed by a central composite design to develop a response surface for culture condition optimization. The optimum culture medium for cutinase production was found to be: glucose 33. 92 g/L, yeast extract 30.92 g/L,and IPTG 0.76 g/L. A cutinase production of 145.27±1.5 U/mL, which was in agreement with the prediction, was observed in triplicate verification experiments. The results obtained here verified the effectiveness of the applied methodology and may be helpful for cutinase production on an industrial scale.

        Comparison between Craniospinal Irradiation and Limited-Field Radiation in Patients with Non-metastatic Bifocal Germinoma

        Purpose Whether craniospinal irradiation (CSI) could be replaced by limited-field radiation in non-metastatic bifocal germinoma remains controversial. We addressed the issue based on the data from our series and the literature. Materials and Methods Data from 49 patients diagnosed with non-metastatic bifocal germinoma at our hospital during the last 10 years were collected. The Pediatric Quality of Life Inventory 4.0 was used to evaluate health-related quality of life (HRQOL). Additionally, 81 patients identified from the literature were also analyzed independently. Results In our cohort, 34 patients had tumors in the sellar/suprasellar (S/SS) plus pineal gland (PG) regions and 15 in the S/SS plus basal ganglia/thalamus (BG/T) regions. The median follow-up period was 52 months (range, 10 to 134 months). Our survival analysis showed that patients treated with CSI (n=12) or whole-brain radiotherapy (WBRT; n=34) had comparable disease-free survival (DFS; p=0.540), but better DFS than those treated with focal radiotherapy (FR; n=3, p=0.016). All 81 patients from the literature had tumors in the S/SS+PG regions. Relapses were documented in 4/45 patients treated with FR, 2/17 treated with whole-ventricle irradiation, 0/4 treated with WBRT, and 1/15 treated with CSI. Survival analysis did not reveal DFS differences between the types of radiation field (p=0.785). HRQOL analysis (n=44) in our cohort found that, compared with S/SS+PG germinoma, patients with BG/T involvement had significantly lower scores in social and school domains. However, HRQOL difference between patients treated with CSI and those not treated with CSI was not significant. Conclusion In patients with non-metastatic bifocal germinoma, it is rational that CSI could be replaced by limited-field radiation. HRQOL in patients with BG/T involvement was poorer.

        Cytotoxic Constituents from Angelicae Sinensis Radix

        Cytotoxic bioassay-guided fractionation of methanol extract of Angelicae Sinensis Radix led to the isolation of a new dimeric Z-ligustilide, named neodiligustilide (1), together with three known compounds, Z-ligustilide (2), 11(S),16(R)-dihydroxy-octadeca-9Z,17-dien-12,14-diyn-1- yl acetate (3), and 3(R),8(S)-falcarindiol (4). Among them, 2 showed the strongest cytotoxicity against L1210 and K562 cell lines with IC50 values of 2.27 ± 0.10 and 4.78 ± 0.18 µM, respectively, while 1 showed moderate cytotoxicity with IC50 values of 5.45 ± 0.19 and 9.87 ± 0.14 µM. Two polyacetylenes, 3 and 4, showed cytotoxicity only against L1210 cell line with IC50 values of 2.60 ± 0.90 and 2.87 ± 0.49 µM, respectively.

        Different modes of bubble migration near a vertical wall in pure water

        The migration of deformable bubbles with different sizes near a vertical wall in pure water was experimentally studied by the dual-camera orthogonal shadow method. The key factors affecting the migration are discussed. It was found that the effect of size and initial distance on bubble migration is indirect. For bubbles of the same diameter (deq=3.10 mm), the same mode occurs when the initial distance (0.5≤L* ≤3.5) is changed. When the initial distance (L* =0.5) is constant, the same mode appears for bubbles of different sizes (2.27 mm≤deq≤3.40 mm). Furthermore, different modes are found in the same initial distance (L* =0.5) and size (deq=3.10 mm). In fact, the migration mode is determined by the initial distance, Reynolds number and Weber number. The size and initial distance can influence the frequency of different modes. Considering a variety of factors, four basic migration modes are summarized. New correlations are proposed to predict bubble behavior in different modes near the wall. The kinematic and dynamic parameters of different modes are discussed. It is expected that the results can be helpful to predict the gas-liquid flow near a wall and to improve related heat and mass transfer process.

        Dynamic behavior of an ellipsoidal bubble contaminated by surfactant near a vertical wall

        Adding a small amount of surfactant to gas-liquid two-phase flow can markedly change bubble behavior,which has crucial application value in nuclear energy, petrochemical, chemical, and environmental engineering. In thispaper, the dynamic behavior of a single ellipsoidal bubble (Re~800) contaminated by surfactant rising near a verticalwall in stagnant liquid is studied using the shadow method. The effects of different concentrations of sodium dodecylsulfate solution (100 ppm, 400 ppm, 800 ppm) and initial dimensionless distances on bubble dynamics were compared. The dynamic parameters, shape oscillation, force, and energy of the bubble were analyzed. The results show that thecritical initial dimensionless distance at which the collision occurs is decreased due to a dimensionless distance changefrom 3.3 to 0.23, accelerating the transition from zigzag to spiral movement. Transverse movement of the contaminatedbubble is restrained. Because of the Marangoni effect caused by the surfactant, the boundary condition changesfrom zero shear to non-zero shear, resulting in a decrease in velocity and an increase in the drag coefficient. As the surfactantconcentration increases, the lift coefficient does not significantly change with concentration variations. Theinfluences of the wall effect on velocity and drag gradually weaken. Comparing free-rising and collision conditions, theaspect ratio of the contaminated bubble is distinct from the regularity of a clean bubble. The surfactant also changes thewall-normal velocity frequency and symmetrical shape frequency and inhibits energy conversion during collision.

      • Cytotoxicity of Triterpenes Isolated from <i>Aceriphyllum rossii</i>

        <P>Bioassay-guided fractionation of a MeOH extract of the whole plant of <I>Aceriphyllum rossii</I> (Saxifragaceae) led to the isolation of two new triterpenes, 3α,23-isopropylidenedioxyolean-12-en-27-oic acid (1) and 23-hydroxy-3-oxoolean-12-en-27-oic acid (2), together with six known triterpenes, 3-oxoolean-12-en-27-oic acid (3), 3α-hydroxyolean-12-en-27-oic acid (4), β-peltoboykinolic acid (5), aceriphyllic acid A (6), oleanolic acid (7), and gypsogenic acid (8). The structures of these compounds were elucidated on the basis of physicochemical and spectroscopic analyses. These compounds were evaluated for <I>in vitro</I> cytotoxicity against the K562 and HL-60 cell lines. Olean-12-en-27-oic acid derivatives (1—6) exhibited considerable cytotoxicity against K562 and HL-60 cell lines with IC<SUB>50</SUB> values ranging from 12.2 to 28.7 μ<SMALL>M</SMALL> and from 12.1 to 25.8 μ<SMALL>M</SMALL>, respectively.</P>
