      • 1-Di-butylaminonaphthalene-5-Sulfonyl Chloride와 Pyridine 置換體와의 反應

        Nucleophilic substitution reaction of bansyl chloride with mathyl substituted pyridines have been studied kinetically at 0℃ in methanol-acetonitrile binary solvent mixtures. The order of magnitude of reactivity is γ-picoline>β-picoline>pyridine to react with bansyl chloride. The Bronsted βand Hammett ??values indicated that the bond formation is advanced more than the bond breaking at the transition state. The reactivity of nucleophiles is in accord with charge controlled and the reactivity of substrate has shown an orbital controlled reaction.

      • 자궁경부암에서 Glutathione S Transferase-π의 분포에 관한 연구

        Uterine cervical carcinoma is the most common malignant tumor of woman in korea. GST-π is multifunctional and is implicated to detoxication of drug and carcinogenesis. This enzyme is expressed in many tumors of human. Relationship between GST-π and drug resistence on uterine cervical carcinoma is not known, but condyloma accuminatum, CIN and invasive carcinoma express this isoenzyme, and it is regarded as a useful marker in preneoplasia and neoplasia. To observe the expression of GST-π in CIN and invasive carcinoma, we analyzed CIN 26 cases and invasive squamous cell carcinoma 61 cases by immunohistochemical stain using monoclonal mouse anti-human GST-π. The results are as followings: 1) The GST-π showed positive reactionin normal and metaplastic epthelium adjacent to CIN and invasive carcinoma in forty-three cases(62.3%) of sixty-nine cases. 2) The GST-π showed positive reactionin CIN in eighteen cases(69.2%) of twenty-six cases. Positive rates of CIN Ⅰ, CIN Ⅱ and CIN Ⅲ are 66.7%, 71.5%, 68.8%, respectively. 3) The GST-π showed positive reaction in invasive squamous cell carcinoma in fifty-one cases(83.6%) of sixty-one cases. Positive rates of microinvasive and invasive squamous cell carcinoma are 84.6% and 83.3%, respectively. According to cell type, keratinizing and nonkeratinizing type are 81.8% and 84.6%, respectively. Above results revealed that GST-π may be significance as a immuohistochemical marker in CIN and invasive SCC, and may be implicated in intrinsic resistance for anticancer drug.

      • 대장선암에서 p53과 c-erbB-2의 표출 및 상관관계에 관한 연구

        Colorectal adenocarcinoma is relatively common malignant tumor in korea and various oncogenes has found in tumor cells. Colorectal adenocarcinoma show overexpression of p53 and c-erbB-2. The relation between expression of p53 and c-erbB-2 and prognosis of patients in colorectal adenocarcinoma has studied recently, but that is controversy. To evaluate relation between expression of p53 and c-erbB-2 and histopathologic finding prognostic factors, we have performed immunohistochemical staining, using mouse anti-p53 and anti-human c-erbB-2, for 52 cases of colorectal adenocarcinoma. The results were as follows; 1) Expression rate of p53 and c-erbB-2 in colorectal adenocarcinoma were 71.2% and 51.9%, respectively. 2) Expression rate of p53 in left side(70.4%), differentiated(75.6%) and invasion to serosa and surrounding adipose tissue(72.5%) were higher than in right side(37.5%), poorly differnetiated(42.9%) and invasion to muscle(50%). 3) Expression rate of c-erbB-2 in left side(51.9%), differentiated(57.8%) and invasion to serosa and surrounding adipose tissue(54.5%) were higher than in right side(37.5%), poorly differnetiated(14.3%) and invasion to muscle(37.5%). 4) The expression of p53 and c-erbB-2 showed coexpression in colorectal adenocarcinoma. 5) The expression of p53 and c-erbB-2 showed no differenece according to presence or absence of lymphatic metastasis. Above result revealed that p53 and c-erbB-2 are coexpression in colorectal adenocarcinoma. The expression of each p53 and c-erbB-2 and the simultaneous expression of p53 and c-erbB-2 may be related to left side and differentiated cases. Our studies for relation with prognosis of patients in colorectal adenocarcinoma showed different results according to depth of invation and metastasis to lymph node and we should be followed for them.

      • Dabsyl Chlorides의 親核性 置換反應

        Kinetic studies of nuclsophilic substitution reaction of dabsyl chloride with pyridines have been investigated at 30℃ in a range of methanol-acetonitrile bianary solvent mixtures. The order of magnitude fo reactivity is β-picoline > pyridine > 1.5-N D A > α-picoline to react with dabsyl chloride, especially in case of α-picoline, the reactivity revealed lower than others, it is suggestd that nitrogen atom of pyfidine ring has a steric hinderance by the neightboring methyl group. The value of ρ(-0.96∼4.59) and β(0.36∼0.67) associated with a change of substituent in the nucleophile are large and indicate a realtively advanced bond formation in the transition state. Solvatochraomic correlations were predicted the importance of bond formation transition state, showing a greater contribution of polaritypolarizability (π*) lone paired to hydro-gen bond donar acidity (α). We conclude that the reaction of dabsyl chloride with pyridine proceed via S2 type reaction mechanism, as well as the reaction of dansyl chloride pyridine.

      • 위선암에서 p53, c-erbB-2, 그리고 DNA ploidy의 상관관계에 관한 연구

        Gastric carcinoma is the most common malignant tumor in korea and many oncogenes have found in gastric carcinoma. The relation between the expression of p53 and c-erbB-2 and the prognosis of patient in gastric adenocarcinoma have studied recently, but it is controversy. To evaluate correlation between the pathologic findings including prognostic factors and the expression of p53 and c-erbB-2, and DNA ploidy, we have performed immunohistochemical staining using 83 cases adenocarcinoma and flow cytometry with 56 cases. The results were as follows; 1) The expression rate of p53 was 45.8% in gastric adenocarcinoma. Differentiated, intestinal, advanced, and lymph node metastatic cases showed higher expression rates than poorly differentiated, diffuse, and non-metastatic cases, without statistically significance. 2) The expression rate of c-erbB-2 was 62.6% in gastric adenocainoma. c-erbB-2 was related to differentiated and intestinal types. Advanced and metastatic cases showed higher expression rates of c-erbB-2 than early and non-metastatic cases, but there was not statistic significance 3) Gastric adenocarcinoma showed correlation of DNA diploidy with poorly differentiated type and aneuploidy with differentiated. 4) DNA diploidy and aneuploidy showed correlation with c-erbB-2 negative and postive cases respectively, but correlation between DNA ploidy with p53 expression was not proved. Above results showed that p53 and c-erbB-2 may be related poor prognosis but there was no statistic significance. DNA aneuploidy and diploidy showed correlation with c-erbB-2 expressed and non-expressed cases, respectively, but not with p53 expression.

      • Paraquat lung에서 Fibronectin 분포에 관한 연구

        To investigate the process of the Paraquat induced pulmonary fibrosis, 10 mg/Kg and 20mg/kg of paraquat was administered intraperitoneally each of 10 rats respectively. Three, five, seven, ten and fifteen days after this injection, the rats were sacrificed. Immunohistochemical studies for fibronectin and histologic studies were carried out. Results were as follows. 1. Toxic effect of the paraquat seems to be depend on the dose of administered paraquat since the groups of the rate treated with 10 mg/kg revaled no significant tissue damage after 7 days. 2. The pulmonary changes seen in 20 mg/Kg treated group could be classified into two stages: the early changes such as hemorrhage, edema and hyaline membrane formation which are similar to the findings observed in adult respiratory distress syndrome and the late change characterized by fibrosis which observed in mild degree even after 2 weeks, so the fibrosis seen to need more time for full blown. 3. The positivity of fibronectin were observed in macrophage and exudate in the early stage of tissue destruction that the fibronectin productin is preceded fibrosis and plays an important role for the collection of inflammatory cells such as macrophaques and fiboblasts. The more tissue desturction is severe, the more strong positivity of fibronectin was observed in the early stage of tissue destruciton.

      • 자궁 경부 질환에서 p53과 bcl-2 발현 양상

        Background: The function of p53 protein was known to accelerate the apoptosis while bcl-2 protein was known to suppress the apoptosis. The patterns of expression of mutant p53 and bcl-2 protein has been reported in various malignant tumors. Although the mechanisms of the p53 and bcl-2 protein in tumorgenesis has not been completely proved, the bcl-2 positive tumors appeared to have good prognosis than tumors with mutant p53 expression. The authors analyzed immunohistochemical expression of mutant p53 and bcl-2 protein in epithelial lesions of uterine cervix. Method: Nine cases of squamous metaplasia, 9 cases of mild dysplasia(CIN Ⅰ), 11 cases of moderate dysplasia(CIN Ⅱ), 16 cases of carcinoina in situ(CIS) and 13 cases invasive squamous cell carcinoma(SCC) were immunohistochemically stained with p53 and bcl-2 monoclonal antibodies. Result: 1. The p53 was weakly positive in 3 out of 9 cases(33%) of squamous metaplasia, and the positive cells were observed only in the basal layer. 3 out of 9 cases(33%) in mild dysplasia and 4 out of 11 cases(36%) in moderate dysplasia showed weakly positive reaction. 2. The p53 showed weakly positive reaction in 7 out of 16(44%) CIS cases and in 9 out of 23(38%) cases the invasive SCC and the positive cells were observed in the tumor cells. 3. The bcl-2 protein showed positive reaction of the reserve cells and along the basal layer of the uterine cervix. However, the positive reactions disappeared in reserve cells and basal cells in 3 out of 9 cases(33%) in squamous metaplasia, 3 out of 9 cases in mild dysplasia(33%), 5 out of 9 cases(56%) in moderate dysplasia, 9 out of 16 cases(56%) in CIS and 14 out of 23 cases(54%) in invasive SCC. This meaned that the positive rate of the bcl-2 protein appeared to decrease according to the progression of the lesion. Above result suggested that the expression of the mutant p53 in the epithelial lesions uterine cervix could be different from those of the other organs in view of tumorgenesis and prognosis; the expression of p53 in the epithelial lesion of the uterine cervix have no significant meaning in progression and prognosis of the lesions. The disappearance of the bcl-2 of the basal cells and reserve cells in the epithelial lesions of uterine cervix suggested a relationship with the prognosis of the lesios, however, not statistically significant.

      • 위선암에서 p53과 P-glycoprotein의 분포에 관한 연구

        Gastric adenocarcinoma express many oncogenes, such as p53, c-erbB, k-sam, etc. The p53 especially acts as tumor suppressor in normal tissue and various mutational changes. of p53 are known as doing important role in carcinogenesis of gastric adenocarcinoma. P-glycoprotein, as a pleotrophic membrane transport protein, shows specific distribution in many normal tissues and malignant tumors. This protein, produced by MDR gene, shows multidrug resistance to anticancer drug by pumping out intracellular drug. resulting in resistance to chemotherapeutics. The expression of p53 and P-glycoprotein according to clincopathologic findings in gastric adenocarcinoma has been studied, even though their results are controversial. To observe the relationship between both antigens and clinicopathologic findings of gastric adenocarcinoma. we were analysesed 40 cases gastric adenocarcinoma then processed for immunohistochemical stain by SLAB method using p53 and JSB-1 monoclonal antibodies. The results were as followings: 1) The p53 and P-glycoprotein showed positive in 20 cases(50%) and 21 cases(52.5%) of 40 cases respectively, and 10 cases(25%) showed positive reaction to both antigens. 2) p53 revealed higher positive rate in intestinal type(54.6%), differentiated form(57.1%), advanced form(58.1%) compared to diffuse type(28.6%), poorly differentiated form(42.1%), early cancer(22.2%), but no difference according to age, sex, lymphatic metastasis and signet ring cell within tumor. 3) P-glycoprotein revealed higher positive rate in interstinal type(54.6%), differentiated form(61.9%), advanced form(58.1%), absence of signet ring cell (63.3%) compared to diffuse type (28.6%), poorly differentiated form (36.8%), early cancer(33.3%), presence of signet ring cell(20.0%), but no difference according to sex, age, lymphatic metastasis. 4) Positive rate in intestinal type, differentiated form, advanced form, and presence of signet ring cells in gastric adenocarcinoma showing both positive reaction of p53 and P-glycoprotein, are higher, but no difference according to age, sex, lymphatic metastasis. 5) In noneoplastic mucosa adjacent to cancer, p53 revealed negative reaction in all cases. P-glycoprotein showed positive reaction in all metaplastic glands 33 cases and parietal cells 17 cases, and in 16 cases(40%) of 40 cases of luminal surface and gastric glandular epthelium, and showed negative reaction in all 12 cases of chief cell. Above results revealed that positive rates of p53 and P-glycoprotein in gastric adenocarcinoma are almost same and there is no relationships between both antigens. Both antigens showed simillar high positive rate in intestinal type, differentiated form and advanced form, indicating association with these clinicopathologic findings.
