Background: Shared decision making using patient decision aids (PtDAs) was established over a decade ago, but few studies have evaluated its efficacy in Asian countries. We therefore evaluated the application of PtDAs in a decision conflict between two muscle relaxant reversal agents, neostigmine and sugammadex, and sequentially analyzed the regional differences and operating room turnover rates.Methods: This multicenter, outcome-assessor-blind, randomized controlled trial included 3,132 surgical patients from two medical centers admitted between March 2020 and August 2020. The patients were randomly divided into the classical and PtDA groups for pre-anesthesia consultations. Their clinicodemographic characteristics were analyzed to identify variables influencing the choice of reversal agent. On the day of the pre-anesthesia consultation, the patients completed the four SURE scale (sure of myself, understand information, risk-benefit ratio, encouragement) screening items. The operating turnover rates were also evaluated using anesthesia records. Results: Compared with the classical group, the PtDA group felt more confident about receiving sufficient medical information (P < 0.001), felt better informed about the advantages and disadvantages of the medications (P < 0.001), exhibited a superior understanding of the benefits and risks of their options (P < 0.001), and felt surer about their choice (P < 0.001). Moreover, the PtDA group had a significantly greater tendency to choose sugammadex over neostigmine (P < 0.001). Conclusions: PtDA interventions in pre-anesthesia consultations provided surgical patients with clear knowledge and better support. PtDAs should be made available in other medical fields to enhance shared clinical decision-making.
Removal of the Glycosylation of Prion Protein Provokes Apoptosis in SF126

Although the function of cellular prion protein (PrP$^C$) and the pathogenesis of prion diseases have been widely described, the mechanisms are not fully clarified. In this study, increases of the portion of non-glycosylated prion protein deposited in the hamster brains infected with scrapie strain 263K were described. To elucidate the pathological role of glycosylation profile of PrP, wild type human PrP (HuPrP) and two genetic engineering generated non-glycosylated PrP mutants (N181Q/N197Q and T183A/T199A) were transiently expressed in human astrocytoma cell line SF126. The results revealed that expressions of non-glycosylated PrP induced significantly more apoptosis cells than that of wild type PrP. It illustrated that Bcl-2 proteins might be involved in the apoptosis pathway of non-glycosylated PrPs. Our data highlights that removal of glycosylation of prion protein provokes cells apoptosis.

Glucagon-like peptide-1 (GLP-1) was a potential therapeutic drug for type II diabetes, mainly because of the stimulatory effect on insulin secretion under condition of high blood glucose. We used PCR to obtain a recombination gene, GGH, in which two GLP-1 (GLP-1A2G) mutants were connected in series and then fused to the N terminal of human serum albumin. The fusion gene was inserted into pGAPZaA plasmid with Saccharomyces cerevisiae α- factor secretion signal sequence, and was expressed by the glyceraldehyde-3-phosphate dehydrogenase (GAP) promoter. The engineered strain was constructed by integrating the recombinant plasmid pGAPZαA/GGH into the genome of Pichia pastoris GS115. Genome PCR and western blot showed that the recombinant P. pastoris successfully expressed the fusion protein GGH. The yield of GGH reached 78 mg/L after 72 h fermentation in a flask, using glucose as the optimal carbon source. Fed-batch fermentation was investigated in a 5 L bioreactor, and the expression level of GGH reached 246 mg/L in 52 h. The fusion protein GGH was purified in four steps, and the final purity was 96.1%. The in vitro bioactivity of GGH was the same as that expressed in P. pastoris by the AOX1 promoter. This study described an efficient way to express GGH fusion protein in P. pastoris using GAP promoter, fermentation was easier to control without carbon source change and fermentation time was 20 h less than AOX1 promotercontrolled GGH fermentation.

A new chromone glycoside, 3,5,7-trihydroxylchromone-3-O-$\alpha$-L-arabinopyranoside (1), together with quercetin (2), (+)-catechin (3), (-)-epi-catechin (4) were isolated from the aerial parts of Rhododendron spinuliferum. The structure of 1 was elucidated on the basis of spectroscopic and 2D-NMR spectral analysis. In addition, 1 exhibited mild inhibitory effect on NO production in LPS-stimulated RAW264.7 cells.

Hierarchically porous carbons are obtained by a facile process of carbonizing the precursor of diblock copolymer xPS-b-polymethylmethacrylate (PMMA). In the diblock copolymer, there is a carbon source of cross-linked polystyrene (xPS) and a sacrificial block of PMMA. The atom transfer radical polymerization was used to synthesize the diblock copolymer with controlled molecular weight and narrow polydispersity in dimethylformamide at 120 ℃. The obtained hierarchically porous carbons (HPCs) exhibit remarkable electrochemical properties, such as higher specific capacitance of 204 Fg -1 at the current density of 0.5Ag -1 in 6 mol L -1 aqueous KOH electrolyte. In addition, it also presents an excellent rate performance of 64% capacitance retention as the current density increases to 10Ag -1 and a superior cycling stability of 98% capacitance retention after 5000 cycles at the current density of 1Ag -1.
Osteopontin enhances sperm capacitation and in vitro fertilization efficiency in boars

In this study, we used more reliable experimental materials and methods to detect the effects of osteopontin (OPN) on boar sperm in vitro capacitation, acrosome reaction, and fertilization efficiency. We reorganized and obtained the OPN protein of the porcine source. Immunofluorescence and Western blot show the localization and expression of the OPN protein before and after sperm capacitation. To determine whether OPN can affect sperm during sperm capacitation, we examined cyclic adenosine monophosphate (cAMP) concentrations after sperm capacitation, and the results showed that OPN significantly increased the cAMP concentration in sperm (p < 0.05). Flow cytometry showed that 0.1 μg/mL OPN-treated sperm had better acrosome reaction ability. In vitro fertilization (IVF) showed that 0.1 μg/mL OPN significantly increased the rate of embryo division. In conclusion, this study found that 0.1 μg/mL porcine OPN protein can significantly improve porcine capacitated sperm motility, cAMP concentration after capacitation sperm, acrosome reaction ability, and embryo division during IVF and provides new clues to explore the mechanism of OPN’s function on sperm.
Eclosion hormone functions in larva-pupa-adult ecdysis in Leptinotarsa decemlineata

In Drosophila melanogaster, ecdysis-triggering hormone (ETH) and eclosion hormone (EH) mutually enhance the release of each other through a positive feedback loop. Both are involved in the stimulation of ecdysis behavior series and molting-related physiological preparations. However, whether 20-hydroxyecdysone (20E) controls eh expression and whether EH plays a similar role in non-Drosophilid insects remain to be determined. In Lep tinotarsa decemlineata, we found that Ldeh mRNA levels were high in the early and/or late stages and low in the intermediate period within each larval instar, indicating that the mRNA peaks positively correlate with 20E pulses. Consistent with the indication, we confirmed that 20E signaling stimulated the transcription of Ldeh. RNA interference (RNAi)-aided knockdown of Ldeh at the final instar stage slightly impaired pupation and signifi cantly affected wing expansion. Around 20% of the Ldeh RNAi larvae remained as prepupae, completely wrapped in the old larval cuticles. These prepupae gradually darkened, dried and withered, and finally died. The remaining (about 80%) Ldeh hypomorphs became pupae and emerged as abnormal adults, bearing smaller and wrinkle elytra and hindwings. The adult tracheae in the Ldeh hypomorphs were full of liquid. In addition, the low pupation rate and high deformed adult proportion cannot be rescued by ingestion of either 20E or juvenile hormone. Thus, EH is critical for ecdysis and trachea clearance in L. decemlineata. The failure of trachea clearance may disenable air-swallowing during larva-pupa-adult transition and thus impacts wing growth and expansion.

Gynandromorphic ticks are extremely rare, and often attract parasitologists’ attention. During our examination of tick specimens, an engorged gynandromorph of Hyalomma asiaticum was noticed. This is the first record of gynandromorphic ticks from China. In this study, several important morphological structures of normal and gynandromorphic H. asiaticum were analyzed. Comparing to the normal H. asiaticum, the gynandromorphic specimen was a typical bipartite protogynander. Its right side showed normal female characteristics, whereas the left side had normal male traits. Different from other gynandromorphic ticks containing 1 anus, this tick reported here had 2 complete anuses, and the anus of the male part had a single adanal plate.
