        Sialoglycoprotein from Gadous morhua eggs improve high bone turnover activity via down-regulating BMP-2/Smads and Wnt/b-catenin signal pathways

        The effect of sialoglycoprotein isolated from Gadous morhua eggs (Gm-SGP) on ovariectomized (OVX) induced osteoporosis, which is characterized by high bone turnover activity was investigated. Results revealed that Gm-SGP significantly increased bone mineral density, enhanced bone biomechanical properties and repaired the microstructure of the trabecular bone. Also, the treatment with Gm-SGP remarkably decreased biochemical marker contents or activities, such as serum BALP, PICP, BMP-2, TrACP, Cath-K, urine Ca and P, leading to the reduction in bone turnover. The elevation in the rate of bone formative process contributed in the increase of bone turnover. Both BMP-2/Smads and Wnt/b-catenin signaling pathways played an important role in osteogenesis. Gm-SGP suppressed the key factors expression in these two pathways such as BMP-2, Smad1, Smad4, Lrp-5b, Runx2, Osx, ALP, Col1, OCN and b-catenin. These findings might provide some theoretical basis for the application of Gm-SGP as a potential anti-osteoporotic drug or as functional food.

      • Clinical Application of Transcatheter Arterial Chemoembolization Combined with Synchronous C-arm Cone-Beam CT Guided Radiofrequency Ablation in treatment of Large Hepatocellular Carcinoma

        Objective: This work aimed to evaluate the safety and clinical efficacy of transcatheter arterial chemoembolization (TACE) combined with c-arm cone-beam CT guided synchronous radiofrequency ablation (RFA) in treatment of large hepatocellular carcinoma (HCC). Methods: 21 patients with large HCC were studied from January 2010 to March 2012. TACE combined with synchronous C-arm cone-beam CT guided RFA were performed on a total of 25 lesions. Conventional imaging examination (CEUS, enhanced CT or MRI) and AFP detection were regularly conducted to evaluate the technical success rate of combined treatment, complications, treatment response, time without disease recurrence and survival rate. Results: The technical success rate of combined treatment was 100%, without any significant complication. After 1 month, there were 19 cases with complete response and 2 cases with partial response, with an complete response rate of 90.4% (19/21) and a clinical effective rate of 100% (21/21). The complete response rates of single nodular lesions (100%, 17/17) was significantly higher than that of multiple nodular lesions (50%, 2/4) (P<0. 05). During 2 to 28 months of follow-up, in 19 cases with complete response, the average time without disease recurrence was $10.8{\pm}6$ months. The total survival rates of 6, 12 and 18 months in 21 patients were 100%, respectively. Conclusion: TACE combined with synchronous C-arm CT guided RFA is safe and effective for treatment of large HCC. The treatment efficacy for single nodular lesion is better than that for multiple nodular lesions.

      • Geometric Optimization of a Mathematical Model of Radiofrequency Ablation in Hepatic Carcinoma

        Radio frequency ablation (RFA) is an effective means of achieving local control of liver cancer. It is a particularly suitable mode of therapy for small and favorably located tumors. However, local progression rates are substantially higher for large tumors (>3.0 cm). In the current study, we report on a mathematical model based on geometric optimization to treat large liver tumors. A database of mathematical models relevant to the configuration of liver cancer was also established. The specific placement of electrodes and the frequency of ablation were also optimized. In addition, three types of liver cancer lesion were simulated by computer guidance incorporating mathematical models. This approach can be expected to provide a more effective and rationale mechanism for employing RFA in the therapy of hepatic carcinoma.

      • Prognostic Value of MGMT Promoter Methylation and TP53 Mutation in Glioblastomas Depends on IDH1 Mutation

        Several molecular markers have been proposed as predictors of outcome in patients with glioblastomas. We investigated the prognostic significance of $O^6$-methylguanine-DNA methyltransferase (MGMT) promoter methylation and TP53 mutation status dependent on isocitrate dehydrogenase 1 (IDH1) mutation in glioblastoma patients. A cohort of 78 patients with histologically confirmed glioblastomas treated with radiation therapy and chemotherapy were reviewed retrospectively. We evaluated the prognostic value of MGMT promoter methylation and TP53 mutation status with regard to progression-free survival (PFS) and overall survival (OS). It was revealed that mutations in IDH1, promoter methylation of MGMT, TP53 mutation, age, Karnofsky performance status (KFS), and extension of resection were independent prognostic factors. In patients with an IDH1 mutation, those with an MGMT methylation were associated with longer PFS (p=0.016) and OS (p=0.013). Nevertheless, the presence of TP53 mutation could stratify the PFS and OS of patients with IDH1 wild type (p=0.003 and 0.029 respectively, log-rank). The MGMT promoter methylation and TP53 mutation were associated with a favorable outcome of patients with and without mutant IDH1, respectively. The results indicate that glioblastomas with MGMT methylation or TP53 mutations have improved survival that may be influenced by IDH1 mutation status.

        Evaluating and Mitigating Malicious Data Aggregates in Named Data Networking

        Named Data Networking (NDN) has emerged and become one of the most promising architectures for future Internet. However, like traditional IP-based networking paradigm, NDN may not evade some typical network threats such as malicious data aggregates (MDA), which may lead to bandwidth exhaustion, traffic congestion and router overload. This paper firstly analyzes the damage effect of MDA using realistic simulations in large-scale network topology, showing that it is not just theoretical, and then designs a fine-grained MDA mitigation mechanism (MDAM) based on the cooperation between routers via alert messages. Simulations results show that MDAM can significantly reduce the Pending Interest Table overload in involved routers, and bring in normal data-returning rate and data-retrieval delay.

        Flutter suppression of long-span suspension bridge with truss girder

        Section model wind tunnel test is currently the main technique to investigate the flutter performance of long-span bridges. Further study about applying the wind tunnel test results to the aerodynamic optimization is still needed. Systematical parameters and test principle of the bridge section model are determined by using three long-span steel truss suspension bridges. The flutter critical wind at different attack angles is obtained through section model flutter test. Under the most unfavorable working condition, tests to investigate the effects that upper central stabilized plate, lower central stabilized plate and horizontal stabilized plate have on the flutter performance of the main beam were conducted. According to the test results, the optimal aerodynamic measure was chosen to meet the requirements of the bridge wind resistance in consideration of safety, economy and aesthetics. At last the credibility of the results is confirmed by full bridge aerodynamic elastic model test. That the flutter reduced wind speed of long-span steel truss suspension bridges stays approximately between 4 to 5 is concluded as a reference for the investigation of the flutter performance of future similar steel truss girder suspension bridges.

      • Predictive and Prognostic Biomarkers for Patients Treated with Anti-EGFR Agents in Lung Cancer: A Systemic Review and Meta-Analysis

        Background: Several studies have investigated predictive and prognostic biomarkers for patients treated with anti-epidermal growth factor receptor (EGFR) agents in lung cancer. However, the conclusion is controversial. Materials and Methods: A meta-analysis was conducted to evaluate the associations of mutant K-ras, PIK3CA and PTEN deficiency with the efficacy of anti-EGFR agents in lung cancer. The primary endpoint was objective response rate (ORR). The secondary endpoints were overall survival (OS) and progression-free survival (PFS). Results: A total of 61 studies were included in the final meta-analysis. The result showed that K-ras mutation was a good predictor for ORR (RR=0.42, 95%CI, 0.33-0.55, p=0.000) and an effective prognostic marker for OS (HR=1.37, 95%CI, 1.15-1.65, p=0.001) and PFS (HR=1.33, 95%CI, 1.05-1.69, p=0.019). However, PTEN deficiency or PIK3CA mutation did not show any significance predictive value for ORR (PTEN, RR=0.82, 95%CI, 0.56-1.19, p=0.286; PIK3CA, RR=1.08, 95%CI, 0.17-6.66, P=0.938). And PTEN deficiency or expression of PIK3CA did not show significance prognostic value for OS (PTEN, HR=0.88, 95%CI, 0.31-2.46,P=0.805; PIK3CA, HR=0.79, 95%CI: 0.23-2.68, P=0.706). Conclusions: Our meta-analysis showed that K-ras mutation may be an effective predictor in lung cancer patients treated with anti-EGFR agents. Whereas, the predictive and prognostic value of PTEN deficiency and PIK3CA mutation need to be further investigated.

      • Taxol Produced from Endophytic Fungi Induces Apoptosis in Human Breast, Cervical and Ovarian Cancer Cells

        Currently, taxol is mainly extracted from the bark of yews; however, this method can not meet its increasing demand on the market because yews grow very slowly and are a rare and endangered species belonging to first-level conservation plants. Recently, increasing efforts have been made to develop alternative means of taxol production; microbe fermentation would be a very promising method to increase the production scale of taxol. To determine the activities of the taxol extracted from endophytic fungus N. sylviforme HDFS4-26 in inhibiting the growth and causing the apoptosis of cancer cells, on comparison with the taxol extracted from the bark of yew, we used cellular morphology, cell counting kit (CCK-8) assay, staining (HO33258/PI and Giemsa), DNA agarose gel electrophoresis and flow cytometry (FCM) analyses to determine the apoptosis status of breast cancer MCF-7 cells, cervical cancer HeLa cells and ovarian cancer HO8910 cells. Our results showed that the fungal taxol inhibited the growth of MCF-7, HeLa and HO8910 cells in a dose-and time-dependent manner. IC50 values of fungal taxol for HeLa, MCF-7 and HO8910 cells were $0.1-1.0{\mu}g/ml$, $0.001-0.01{\mu}g/ml$ and $0.01-0.1{\mu}g/ml$, respectively. The fungal taxol induced these tumor cells to undergo apoptosis with typical apoptotic characteristics, including morphological changes for chromatin condensation, chromatin crescent formation, nucleus fragmentation, apoptotic body formation and G2/M cell cycle arrest. The fungal taxol at the $0.01-1.0{\mu}g/ml$ had significant effects of inducing apoptosis between 24-48 h, which was the same as that of taxol extracted from yews. This study offers important information and a new resource for the production of an important anticancer drug by endofungus fermentation.

      • <i>ITGB6</i> loss-of-function mutations cause autosomal recessive amelogenesis imperfecta

        <P>Integrins are cell-surface adhesion receptors that bind to extracellular matrices (ECM) and mediate cell–ECM interactions. Some integrins are known to play critical roles in dental enamel formation. We recruited two Hispanic families with generalized hypoplastic amelogenesis imperfecta (AI). Analysis of whole-exome sequences identified three <I>integrin beta 6</I> (<I>ITGB6</I>) mutations responsible for their enamel malformations. The female proband of Family 1 was a compound heterozygote with an <I>ITGB6</I> transition mutation in Exon 4 (g.4545G > A c.427G > A p.Ala143Thr) and an <I>ITGB6</I> transversion mutation in Exon 6 (g.27415T > A c.825T > A p.His275Gln). The male proband of Family 2 was homozygous for an <I>ITGB6</I> transition mutation in Exon 11 (g.73664C > T c.1846C > T p.Arg616*) and hemizygous for a transition mutation in Exon 6 of <I>Nance–Horan Syndrome</I> (<I>NHS</I> Xp22.13; g.355444T > C c.1697T > C p.Met566Thr). These are the first disease-causing <I>ITGB6</I> mutations to be reported. Immunohistochemistry of mouse mandibular incisors localized ITGB6 to the distal membrane of differentiating ameloblasts and pre-ameloblasts, and then ITGB6 appeared to be internalized by secretory stage ameloblasts. ITGB6 expression was strongest in the maturation stage and its localization was associated with ameloblast modulation. Our findings demonstrate that early and late amelogenesis depend upon cell–matrix interactions. Our approach (from knockout mouse phenotype to human disease) demonstrates the power of mouse reverse genetics in mutational analysis of human genetic disorders and attests to the need for a careful dental phenotyping in large-scale knockout mouse projects.</P>
