      • Vinblastine이 백서 배양 Type-1 별아교세포에 미치는 세포독성에 관한 연구

        It has been reported that vinblastine, anticancer drug, is a neuroteratogen inducing various neural malformations such as microcephaly and neural tube defect(NTD), but the study on the toxicity of neuroglial cells, especially type-1 astrocytes using culture system is not well established. In order to evaluate the cytotoxicity of vinblastine on cultured type-1 astrocytes of neonatal rat brain in vitro. MTT assay and also light and electron microscopic studies were carried out. The results were as follows: 1. MTT_90 and MTT_50 in cultured type-1 astrocytes were 1×10 exp (-1) μM and 1×10 exp (2)μM of vinblastine, respectively. 2. Vinblastine was highly toxic in cultured type-1 astrocytes(MTT_50 ≤ 100 μM). 3. In a light microscopy, cultured type-1 astrocytes showed decrease of cells in number, cytoplasmic perforation, vacuolization and cytoplasmic granulation after cultured type-1 astrocytes were treated with 1×10 exp (2)μM of vinblastine for 24 hours. Cultured type-1 astrocytes damaged by vinblastine showed degenerative changes of cells morphologically. 4. In an electron microscopy, increment of glial fibrillary acidic protein(GFAP), increased free-ribosomes, cisternal dilatation of rough endoplasmic reticulum (rER). few dense bodies and vacuoles were shown in cultured type-1 astrocytes treated with 1×10 exp (2)μM of vinblastine for 24 hours. The results indicate that vinblastine has markedly cytotoxic effect on the type-1 astrocytes of neonatal rat brain in vitro study.

      • 수열반응에 의한 비정질 알루미나의 상전이 및 물성평가

        수열반응에 의한 비정질 활성알루미나[Al_2O_3·XH_2O(0.2<X<1.0)]의 상전이 및 물성을 알아보기 위해 수열 처리하였다. 비정질 알루미나를 일축가압 성형하여 시편을 제작한 후에 오토클레이브에서 110℃∼300℃로 각각 수열처리하였다. 수열처리로 만들어진 각각의 시편들을 400℃∼800℃로 열처리하여 밀도, 비표면적, 결정상 등의 물성을 측정하였다. 비표면적은 9∼250㎡/g, 밀도는 2.88∼3.02g/㎤으로 나타났고 110℃에서 수열처리 한 시편에서는 베마이트와 비정질상이 공존하였으나 150℃ 이상에서는 잘 발달된 베마이트상만 존재하였다. 수열처리 한 시편을 열처리 한 경우 비표면적이 30∼230㎡/g이고 3.03∼3.36g/㎤인 γ-알루미나로 전이되었음을 확인하였다. The phase transitions and properties of amorphous alumina were investigated after hydrothermal treatment was done. The specimens were prepared by press, hydrothermally treated in autoclave at 110℃∼300℃, and then the specimens were heat treated at 400℃∼800℃ range. Density, specific surface area, phase, and microstructure were investigated. The specific surface area and density were 9∼250㎡/g, and 2.88∼3.02g/㎤, respectively. The specimen hydrothermally treated at 150℃ was found to be boehmite and amorphous phase, but the specimens above 150℃ was boemite phase only. The specimens heat treated at 400℃∼800℃ were found to be γ-alumina with specific surface area and density of 30∼230㎡/g and 3.03∼3.36g/㎤, respectively.

        항정신병약물 사용 중인 정신분열병 환자에서 올란자판으로의 교체 방법에 관한 연구(II) : Comparison of Safety 안전성 비교

        연구목적: 이 다기관 공동임상연구는 사용 중인 항정신병약물을 ’직접 교체 방법’또는 ’시작-감량 교체 방법’중 한 가지 방법으로 올란자핀으로 교체한 후, 안정성 측면에서 두 교체 방법 간의 비교와 교체후의 변화를 관찰하기 위한 것이다. 방법: 국내 13개 병원의 입원 및 외래에 내원한 환자들 중 ICD-10 지단기준으로 정신분열병에 해당되며, 임상적으로 항정신병약물 교체가 필요한 환자를 대상으로 하였다. 두 가지 교체 방법 중 한 가지를 무작위로 피험자에 적용하였으며, ’직접 교체 방법’에 배정된 경우에는 사용중인 항정신병약물을 일시에 중단하고 10㎎의 올란자핀을 바로 투여하였고, ’시작-감량 교체 방법’에 배정된 경우는 10㎎의 올란자핀 투여하고 2주에 걸쳐서 기존 약물을 감량하여 중단하였다. 올란자핀 사용기간은 총 6주이며, 용량은 5∼20㎎ 범위로 제한하였다. 한정성 평가를 위해서 체중, 생명징후, 자발적인 이상반응 복, 실험실 검사 그리고 Simpson-Angus Scale(SAS), Barnes akathisia rating scale(BARS), Abnormal involuntary movement scale(AIMS). Liverpool University neuroleptic side effect rating scale(LUNSERS)등을 이용하였다. 결과: 총 103명의 정신분열병 환자를 대상으로 하였다. 사용한 올란자핀의 용량, 벤조디아제핀의 병용률, 탈락률과 탈락 사유, 자발적인 이상반응 보고, 생명징후, 실험실 검사 그리고 대부분의 부작용 척도 상에서 임상적으로 의미 있는 차이를 두 교체 방법간에 발견하지 못하였다. 다만 AIMS의 감소는 ’직접 교체 방법’군에서 보다 적었고, 항콜린제의 병용률은 ’시작-감량 교체 방법’군에서 보다 많았다. 기저 상태에서 전체 피험자의 SAS와 BARS 점수는 각각 3.5점과 1.8점이었으며 70% 이상의 피험자가 고프로락틴 혈증을 보였다. 올란자핀으로 교체한 후, SAS, BARS, AIMS 점수의 유의한 감소가 있었으며 고프로락틴 혈증을 보인 피험자 분율도 약 30%이하로 감소하였다. 그러나 교체 방법과 상관없이 올란자핀 교체 후 유의한 체중 증가가 있었다. 결론: 이 연구를 통해 교체 방법에 관계없이 비교적 안전하고 용이하게 올란자핀으로 교체 할 수 있음을 알 수 있었다. 그리고 기존 항정신병약물을 올란자핀으로 교체함으로써 일부 부작용들을 줄일 수 있음을 간접적으로 관찰할 수 있었다. 하지만 이 연구는 여러 제한점과 문제점을 지니고 있기 때문에 보다 체계적인 연구를 통해 검정이 필요하리라 생각된다. Objectives: This multicenter clinical trial involving 13 hospital sites compared the safely of switching to olanzapine between ’direct switching method’ and ’start-tapering switching method’. Method: This study included both inpatients and outpatients who fulfilled the criteria for schizophrenia as defined in the ICD-10, and were in need to be appropriate for switching antipsychotics. Subjects were randomly assigned to one of the two switching methods. For ’direct switching method’group, previous antipsychotics were abruptly discontinued and 10㎎ of olanzapine was administered, and previous antipsychotics was gradually tapered for 2 weeks. Olanzapine was used for 6 weeks and the dose was adjusted within the range of 5-20㎎. The safety of switching to olanzapine was measured with vital sings including body weight, adverse events reported spontaneously, laboratory tests, and various scales such as Simpson-Angus Scale(SAS), Barnes Akathisia Rating Scale(BARS). Abnormal Involuntary Movement Scale(AIMS), and Liverpool University Neuroleptic Side Effect Rating Scale(LUNSERS). Results: 103 patients were switched to olanzapine in this study. The comparison between two switching methods did not show any significant difference in the dosage of olanzapine used, the concomitant use of benzodiazepine, the rate and reasons of drop-out, the adverse events, vital signs, laboratory tests, and most scales for measuring side-effects. However, the decrease in AIMS scores was significantly lower in ’direct switching method’ group, and the concomitant use of anticholinergics was comparatively greater in ’start-tapering switching method’ group. At baseline, SAS and BARS scores were 3.5 and 1.8 points respectively, and more than 70% of the subjects showed hyperprolactinemia. After switching to olanzapine, SAS, BARS, and AIMS scores were significantly decreased and the proportion of the patients with hyperprolactinemia was also decreased to less than 30%. However significant weight gain after the treatment of olanzapine was observed regardless of switching method. Conclusion: This study may suggest that switching to olanzapine can be done with relatively high safety regardless of switching methods and olanzapine can significantly decrease some side-effects induced by other antipsychotics.

      • Electric Characterization of Piezoelectric Actuators Joined by Ceramic Slurry

        <P>Complicate shaped multilayer ceramic devices, especially piezoelectric actuated devices, are difficult to be produced due to non-easy fabrication processes such as such as cutting, lamination, and co-firing. These issues related to the fabrication can be solved through a specially designed bonding method of green body joining, which offers several advantages such as high density, structural matches to matrix materials, and improvement of mechanical properties owing to similar shrinkage rate with matrix materials. In this study, the bonding procedure and electrical properties of piezoelectric actuators fabricated by green body joining method with ceramic slurry is reported. While electric properties of actuators with interlayer (green body/ceramic slurry) displayed similar electric properties to those of actuators without the interlayer, actuators bonded by epoxy showed electrical properties different than those of actuators with and without the interlayer. By employing a binder with similar composition to that of the matrix material, the piezoelectric and dielectric properties of the interlayer have been improved after the sintering process.</P>

      • Highly sensitive and selective multidimensional resistive strain sensors based on a stiffness-variant stretchable substrate

        <P>Highly stretchable strain sensors that are capable of collecting complex multi-axial, multidimensional strain information in real time are crucial in practical applications for human motion detection. Here we present a highly sensitive and selective multidimensional resistive strain sensor based on a monolithic integration of a stiffness-variant stretchable substrate and sensing film comprising a cross-shaped silver nanowire percolation network in a single device. The multidimensional strain sensor efficiently distinguishes strains in various directions with a large gauge factor (GF) of >20 and a wide strain-detectable range of up to 60%. The sensor also features a maximum difference in GF between the <I>x</I>- and <I>y</I>-axes of >20 and long-term performance stability for up to 500 strain cycles. The practicality of the sensor as a human motion detector is demonstrated by attaching it directly to a part of the human body and measuring the multidimensional strains that occur during motions in real time.</P>

        Analysis of Post-Weld Deformation at the Heat-Affected Zone Using External Forces Based on the Inherent Strain

        An analytical method to predict the post-weld deformation at the heat-affected zone (HAZ) is presented in this paper. The method was based on the assumption that the post-weld deformation is caused by external forces resulting from the inherent strain, which is defined as the irrecoverable strain after removing structural restraints and loadings. In general, the equivalent loading method can be used to analyze distortions in welding areas because it is efficient and effective. However, if additional loads are applied after welding, it is difficult to determine the final strain on a welded structure. To determine the final strain of a welded structure at the HAZ more accurately, we developed a modified equivalent loading method based on the inherent strain that incorporated hardening effects. The proposed method was applied to calculate the residual stress at the HAZ. Experiments were also conducted on welded plates to evaluate the validity of the proposed method.

        Characteristics of a New Oyster Mushroom Variety 『Samkang』 for the Bottle and P.P vinyl Bag Culture

        ‘Samkang’ a new variety of oyster mushroom, fitting for the bottle culture, was bred and by mating between monokaryons isolated from MT05433 and AE1-1. In the major characteristics of fruit body, the pilei were thick and dark-gray and the pileistripes were thick and long with softness. It was great in elasticity and cohesivness of tissue as compare to chunchu-1ho. The optimum temperature for the mycelial growth was around 26~28℃ and that for the pinheading and growth of fruitbody was around 14~16℃. In the bottle culture, it was required around 26 days in incubation period and 4 days in primordia formation. The fruit body was grew vital and uniform. The yield were shown by 136.5g/bottle.
