      • 당뇨병 백서의 간세포에서 Glucokinase 활성도 및 유전자 발현에 대한 인슐린의 영향

        목적 당대사의 조절 상태에 따른 생체 변화를 분자 수준에서 이해하고자 식이 조건을 달리한 정상 백서와 화학적으로 유도된 당뇨병 백서의 간조직에서 혈당수준과 인슐린치료 정도에 따라 나타나는 글루코키나제 활성도 및 유전자 발현을 분석하였다. 방법 스트렙토조토신 정맥투여 후 당뇨병의 유발을 확인하고, 인슐린을 1일 3회 3일간 복강내로 투여하여 상태를 안정시킨 후, 인슐린 투여군은 인슐린 투여 6시간이내에 그리고 인슐린 비투여군은 24시간 후 단두하여 채혈하고 복강을 열어 간조직을 채취하였다. 채취한 간조직에서 글루코키나제 활성도는 인산화된 포도당에서 NADH의 형성을 형광분광계로 측정하였으며, 글루코키나제 유전자 mRNA발현은 Northern 분석법을 이용하였다. 성적 정상 백서에서 공복상태와 식이를 섭취한 경우에 간조직의 글루코키나제 효소의 활성은 차이가 없었으나, 글루코키나제 유전자 mRNA 발현은 증가되었다. 당뇨병이 유발된 백서의 간조직에서 글루코키나제 효소의 활성 및 글루코키나제 유전자의 mRNA 발현은 정상 백서에 비하여 낮았다. 인슐린 투쳐 후 글루코키나제 효소의 활성 및 글루코키나제 유전자의 mRNA 발현이 증가되었고, 특히 혈당이 정상화된 경우에서 글루코키나제 유전자의 mRNA 발현이 증가도었다. 결론 인슐린에 의한 간조직에서 글루코키나제 효소의 활성 및 글루코키나제 유전자의 mRNA을 증가를 볼수 있었다. 당뇨병 백서에서 인슐린 투여 후에 혈당조절이 안된 경우 간조직의 글루코키나제 유전자의 mRNA 발현이 증가가 없는 것으로 보아 글루코키나제 mRNA의 발현에는 인슐린 이외의 다른 요소가 관여할 것으로 생각된다. The liver-specific hexokinase isoenzyme, referred to as glucokinase, is thought to play a key reglulatory role in hepatic glucose metabolism. The glucokinase gene is, therefore, of interest both because of its tissue-specific expression and because of the several regulatory processes that can be analyzed. The level of hepatic glucokinase activity appears to be determined essentially by regulation of the rate of enzyme synthesis, with insulin playing a leading role as an inducer. We investigated the role of insulin for the induction of glucokinase in the liver of diabetic rats. Experimental diabetes was induced by injection of streptozotocin 7 days before the experiment. Regular insulin was given by three days intraperitoneal injection at 8-h interval. The glucokinase mRNA in the liver was estimated by Nothern blot assay, as well as by fluorometric enzyme activity assay. Glucokinase activity was not reduced in the liver of normal fasting rats as compared to normal fed rats. And glucokinase activity was reduced in the liver of diabetic rats as compared to normal rats. In diabetic rats treated with insulin, glucokinase enzyme activity were increased. But glucokinase mRNA expression was only increased in normoglycemic diabetic rat with treated with insulin as compared to hyperglycemic rat. These data indicate that insulin stimulates hepatic glucokinase enzyme activity and mRNA expression. But other hormonal or metabolic factors may be contribute to regulation of glucokinase mRNA expression.

        Vasomotor Symptoms: More Than Temporary Menopausal Symptoms

        Vasomotor symptoms (VMS), such as hot flashes and night sweating, are classic menopausal symptoms experienced by a majority of perimenopausal and postmenopausal women. VMS have received a great deal of attention due to their relationship with cardiometabolic risk. Further, accumulating evidence indicates that VMS are associated with an increased risk of several chronic diseases, including metabolic syndrome, type 2 diabetes mellitus, nonalcoholic fatty liver diseases, and osteoporosis in perimenopausal and postmenopausal women. These findings suggest VMS as biomarkers of impaired cardiometabolic conditions rather than just temporary symptoms in menopausal women, warranting further studies to confirm the casual relationship of VMS with these diseases and the exact underlying mechanism in this context.

        The risk factors for herpes zoster in bortezomib treatment in patients with multiple myeloma

        <P><B>Background</B></P><P>Bortezomib has significant activity in treating multiple myeloma (MM). The risk of herpes zoster (HZ) has been reported to increase significantly with bortezomib treatment, but the predisposing factors for HZ are not clear. This study is a retrospective analysis of the relevant risk factors for HZ in Korean MM patients treated with bortezomib.</P><P><B>Methods</B></P><P>Sixty-six patients with refractory or relapsed MM who underwent chemotherapy with bortezomib were included in the study. Prophylactic antiviral drugs were not used for treatment. The following parameters were reviewed: age, gender, stage and type of MM, extent of previous treatment, history of HZ, duration from the time of diagnosis to the time of bortezomib treatment initiation, and absolute lymphocyte counts (ALC) at the time of bortezomib treatment initiation.</P><P><B>Results</B></P><P>The incidence of HZ was 16.7%. There were no intergroup differences between the HZ-positive and the HZ-negative groups with regard to a history of HZ, number of previous treatments, and exposure to steroids before bortezomib treatment. The median duration from the time of MM diagnosis to the time of bortezomib treatment initiation in the HZ-positive group was significantly shorter than that in the HZ-negative group. The median ALC at the time of bortezomib initiation in the HZ-positive group was significantly lower than that in the HZ-negative group.</P><P><B>Conclusion</B></P><P>Bortezomib itself might act as a risk factor for HZ by inhibiting cell-mediated immunity, and patients with low ALC at the time of bortezomib treatment initiation were at greater risk of HZ during bortezomib treatment.</P>

        이어도 종합해양과학기지의 진동 계측자료를 이용한 구조물 동특성 분석

        To study the ocean and meteorological issues related to global climate change and global warming, Korea is operating several ocean research stations (ORS), including Ieodo ORS, Gageocho ORS, and Sochengcho ORS in remote ocean area. In 2003, the Ieodo ORS was first constructed and in operation, and a lot of scientific studies were carried out using the observation data at Ieodo ORS. After the Gageodo ORS was attacked by Typhoon Muifa and was severely damaged to its structural members, ORSs were equipped with vibration measurement systems to monitor their structural integrities. In the case of Ieodo ORS, the vibration measurement system was instrumented in 2016. In this study, the structural dynamic characteristics of the Ieodo ORS were investigated using the vibration measurement data, including acceleration and tilt. As a result, three natural frequencies of two bending modes and torsional mode were estimated as 1.315 Hz, 1.390 Hz, and 1.590 Hz, respectively, and these natural frequencies were compared with those estimated using measurement data in 2005 and 2006 by Kim et al. (2006). It was found that the natural frequencies were decreased as about 2-3.8% due to the combined effect of structural degradation and the increase of mass of equipment. Long-term measurement data were also analyzed to obtain the variation of dynamic characteristics according to the environmental effects such as temperature and excitation force. It was also found that the dynamic characteristics of Ieodo ORS are not affected by temperature and tidal level while they are significantly affected by the level of excitation force, i.e., the amplitude of vibration responses can be used to account for the effect of excitation force indirectly.

        인슐린 비의존형 당뇨병 및 비만증 환자에서의 PPARγ2 유전자 다형성

        Background : Peroxisome proliferator activated receptor-gamma (PPAR-γ) is a nuclear receptor that regulate adipocyte differentiation and modulate intracellular insulin-signaling events. As such, PPARγ is a candidate gene for several human disorders including obesity and type 2 diabetes mellitus. The objective of our study was to examine the relationship between genetic variation of PPARγ2 and diabetes and obesity in Korean subjects. Methods : We studied 99 subjects with type 2 diabetes mellitus, 128 obesity patients and 97 controls. Screening for mutation at codon 12 and 115 of PPARγ2 were carried out by PCR-RFLP analyses. Statistical significance was evaluated by Chi-square test. Results : The allele frequency of the Pro12Ala PPARγ2 variant were 0.05 in controls, 0.06 in type 2 diabetes group, and 0.07 in obesity group (p=0.47). Pro115Gln variant were only proline homozygote in all groups. Genotype frequencies were also similar and conformed to expectations of the Hardy-Weinberg rule. The presence of P gene variant was no associated with concentrations of total cholesterol, triglyceride, HDL-cholesterol, and also with fasting glucose. Conclusion : We concluded that the Pro12Ala and Pro115Gln PPARγ2 missense mutation may not be associated with type 2 diabetes mellitus and obesity in Korean patients.(Korean J Med 59:132-141, 2000)

        성선자극 호르몬 저하성 성선기능 저하증 환자에서 Pulsatile Infusion Pump 를 이용한 성선자극 호르몬 방출 호르몬 ( GnRH ) 치료

        Hypogonadotropic hypogonadism result in absent or incomplete pubertal development, characterised by low plasma gonadotropin and gonadal steroid concentrations, and appears to be due to a deficiency of gonadotropin releasing hormone. We report a case of treatment of delayed puberty after pusatile administration of GnRH in a patient with hypogonadism. A 23 year old man was admitted with complaint of the absent of secondary sex characteristics. Diagnosis of hypogonadotropic hypogonadism was made by combined pituitary stimulation test, chromosomal study and abdominal sonography. GnRH (Luterelef , Ferring Co.) was injected subcutaneously at the interval of 90 min. by infusion pump in doses of 20 g/pulse. Gradual increases of serum gonadotropins, testosteron and sperm count. On fist day of therapy, serum testosteron level and sperm count was 0.6 ng/ml, one million per high power field respectively, and on 20 weeks of therapy, 4.5 ng/ml and 5 million per high power field respectively. GnRH pulse therapy is useful for delayed puberty and infertile man who diagnosed hypogonadotropic hypogonadism (J Kor Soc Endocrinol 5:318 322, 1990).

        한국인 뇌경색증환자에서 위험인자로서의 Lipoprotein (a)

        To evaluate the role of Lipoprotein (a) in patients with cerebral infarction in Korea, Lipid parameters were compared with control group. The control group was made up of 45 healthy persons and the patients group of cerebral infarction included 56 patients confirmed by computerized cerebral axial tomography. We tested 111 serum samples using Pharmacia 2-site IRMA kit. The results were as follows: 1) The concentrations of serum Lipoprotein (a) was significantly higher in patients group of cerebral infarction as compared to normal control (55.0±40.7 mg/dl). 2) In patients with cerebral infarction. 66.7% of male and 57.1% of female has serum Lipoprotein (a) concentration above mean normal level (≥29 mg/dl). 3) In female patients with low serum cholesterol group, the mean serum Lipoprotein (a) was higher than high serum cholesterol group (66.3±87.1 mg/dl vs. 51.3±44.0 mg/dl). We concluded that the high serum Lipoprotein (a) may be another risk factor for the cerebral infarction, especially in those who has normarum cholesterol level in Korea.         Blue Phosphorescence with High Quantum Efficiency Engaging the Trifluoromethylsulfonyl Group to Iridium Phenylpyridine Complexes

        <P>Incorporation of an electron-withdrawing −SO<SUB>2</SUB>CF<SUB>3</SUB> substituent to cyclometalating C^N-phenylpyridine (ppy) ligand resulted in an expected blue-shifted phosphorescence in the corresponding homoleptic <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>3</B></SUB> complex, showing the emission of λ<SUB>em</SUB> = 464 nm at 300 K. One of its heteroleptic derivatives, modified by a pyrazolyl borate LX ligand, <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>2</B></SUB><B>(bor)</B>, exhibited further blue-shifted phosphorescence of λ<SUB>em</SUB> = 460 nm at 300 K. Cyclic voltammograms (CVs) and density-functional theory (DFT) calculations supported the efficacy of the electron-withdrawing capability of the SO<SUB>2</SUB>CF<SUB>3</SUB> substituent lowering HOMO energy and obtained widened bandgaps and resumed blue emissions for all of the iridium complexes studied. The homoleptic complexes of both substituents, <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>3</B></SUB> and <B>Ir(ppySF)</B><SUB><B>3</B></SUB>, reached the higher quantum yields (<I>Φ</I><SUB>PL</SUB>) of (0.89 and 0.72), respectively. Similarly, emission quantum yields (<I>Φ</I><SUB>PL</SUB>) of the heteroleptic derivatives were reported to be (0.75, 0.83, and 0.87) for <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>2</B></SUB><B>(acac)</B>, <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>2</B></SUB><B>(bor)</B>, and <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>2</B></SUB><B>(pic)</B>, respectively. Emission kinetics support the enhanced quantum efficiency when <I>k</I><SUB>r</SUB> and <I>k</I><SUB>nr</SUB> values are compared between <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>3</B></SUB> and <B>Ir(ppySF)</B><SUB><B>3</B></SUB>, and both values favorably contribute to attaining a higher quantum efficiency for <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>3</B></SUB>. Among solution-processed multilayered devices having an ITO/PEDOT:PSS/TCTA:Ir dopant (10:1, <I>w</I>/<I>w</I>)/TmPyPB/Liq/Al structure, a heteroleptic dopant, <B>Ir(ppySCF</B><SUB><B>3</B></SUB><B>)</B><SUB><B>2</B></SUB><B>(bor)</B>, exhibited better device performance, reporting an external quantum efficiency (EQE) of 1.14%, current efficiency (CE) of 2.31 cd A<SUP>-1</SUP>, and power efficiency (PE) of 1.21 lm W<SUP>-1</SUP>, together with blue chromaticity of CIE<SUB>x,y</SUB> = (0.16, 0.32).</P><P>Blue phosphorescence of Ir(III) complexes can be tuned with the introducing of strong electron withdrawing substituents and the variation in the ancillary ligands.</P> [FIG OMISSION]</BR>
