        도토리 추출물의 3T3-L1 세포 분화억제 효과

        본 연구에서는 만성질환의 주요 위험인자인 비만을 예방하기 위한 식품소재로서 3T3-L1세포를 이용하여 탈피한 도토리의 항비만 효과를 알아보고자 하였다. 3T3-L1 세포에서 생존율(MTT assay)을 측정한 결과, AE와 AW 시료 모두 $500{\mu}g/mL$ 농도에서는 다소 생존율의 감소를 보여, $300{\mu}g/mL$를 최종 농도로 정하였다. 3T3-L1 세포의 지질축적 억제 효과를 측정한 결과, 농도 $100{\mu}g/mL$로 처리하였을 때 두 시료 모두 지질축적량의 증가를 보였으나, $200{\mu}g/mL$ 처리농도에서 AE 시료는 82%로, AW 시료는 74%로 감소되다가 $300 {\mu}g/mL$ 농도에서는 두 시료 모두 약 53% 수준까지 지질축적이 억제되었다. 3T3-L1 세포에서 중성지방 억제 효과를 확인한 결과, AE 시료의 경우 $200{\mu}g/mL$ 농도에서 11%의 감소율, $300{\mu}g/mL$ 농도에서 42% 수준의 감소율을 보였다. AW 시료도 $200{\mu}g/mL$ 농도에서 5%의 감소율과 $300{\mu}g/mL$에서 41%의 감소율을 보였다. 3T3-L1 세포의 ROS 생성량을 측정한 결과, 시료 농도 $200{\mu}g/mL$에서 AE는 42%, AW는 33%로 $300{\mu}g/mL$에서는 AE는 58%, AW는 52%로 ROS 생성량의 억제를 보였다. 3T3-L1 세포에서 mRNA 발현에 미치는 영향을 대조군과 비교하였을 때 두 시료(AE와 AW) 모두 $300{\mu}g/mL$ 농도에서 $PPAR-{\gamma}$은 54%와 38%, aP2는 40%와 18% 수준의 발현을 억제시키는 것으로 나타났다. 이상의 결과로 볼 때 탈피한 도토리는 3T3-L1 세포의 분화를 억제함으로써 새로운 항비만 소재로의 가능성이 있는 것으로 판단된다. This study aimed to investigate the suppressive effect of acorn extracts, by evaluating 70% ethanol extract (AE) and hot water extract (AW) using 3T3-L1 preadipocytes. We applied various levels (0, 100, 200, 300 and $500 {\mu}g/mL$) of AE and AW to 3T3-L1 preadipocytes. The cell viability of the 3T3-L1 preadipocytes was not affected by up $300 {\mu}g/mL$ of extracts, but was suppressed by level $500{\mu}g/mL$ of both AE and AW by 20% and 9% respectively. The accumulation of lipid droplets in differentiated 3T3-L1 preadipocytes was dose-dependently suppressed by AE and AW. Especially, at high concentrations ($300{\mu}g/mL$), AE (42%) was more effective than AW (41%). Reactive oxygen species (ROS) was also dose-dependently suppressed by treatment with AE (58%) and AW (52%). With regard to the mRNA related to differentiated 3T3-L1 preadipocytes, $PPAR-{\gamma}$ and aP2 were suppressed by treatment with AE (54 and 40%) and AW (38 and 18%). From our results, acorn extract (AE) has more suppressive effects than AW in differentiated 3T3-L1 preadipocytes. We therefore concluded that acorn has suppressive effects against obesity in differentiated 3T3-L1 cells due to antioxidation.

      • 베체트병 환자에서 Azathioprine으로 유발된 급성 췌장염 1례

        Although many drug are thought capable of initiating acute pancreatitis, following azathioprine administration represents the 5% incidence by far the highest recorded incidence to date. We report a 35-year-old male who developed acute pancreatitis in the course of treatment with azathioprine for Behcet's disease. He was admitted due to abdominal pain, diarrhea, and fever for 10 days. Serum amylase and lipase levels were 510U/L and 3702U/L respectively. The abdominal CT scan revealed diffuse enlargement of the pancreas. There was no history of alcohol use or gall bladder disease. We considered drugs, especially azathioprine, as the cause of acute pancreatitis. We diagnosed it as acute pancreatitis. The patient was treated conservatively with hydration and analgesics. The symptom subsided within four days. Five days later, azathioprine was resumed. And the patient complained of abdominal pain. The elevated levels of serum amylase and lipase confirmed the recurrence of pancreatitis. His symptom subsided when azathioprine intake the stopped, and the serum amylase and lipase levels returned to normal level within five days.

      • Acute exacerbation of interstitial lung disease; subgroup analysis for causes, risk factors for mortality rate, and steroid responsiveness

        Background: Interstitial Lung Disease (ILD) has known as a poor prognosis disease, particularly idiopathic pulmonary fibrosis (IPF). Corticosteroid is widely used for treatment of acute exacerbation (AE)-ILD. This study aims to clarify efficacy of corticosteroid use, and compare the mortality rate depending on the subgroups of ILD. Materials and Methods: We collected the data from the patients who have diagnosed ILD and visited emergency room (ER) with the respiratory symptoms from January 1, 2016 to December 31, 2018. We classified AE-ILD patients into 3 groups depending on methylprednisolone dose per kilogram. Also, we compared the mortality of IPF patients to that of non-IPF patients. Results: A total case of AE-ILD visited ER from 2016 to 2018 was 209 cases, including IPF cases (n=122) and non-IPF cases (n=87). We divided total patients (n=209) into three groups depending on steroid use, and these include group 1 of 0-0.49 mg/kg (n=78), group 2 of 0.5-0.99 mg/kg (n=58), and above 1.0 mg/kg (n=41), each. We found poor prognosis from the cases of IPF compared to non-IPF cases (p=0.029, OR 2.463). Furthermore, there was significant differences of mortality rate according to steroid dose (p=0.036, OR 1.712), CRP elevation (p=0.016, OR 1.004), and intubated case (p=0.000, OR 8.152). Also, we categorized IPF (n=122) to non-IPF cases (n=87) and analyzed the mortality rate. Most of the patients were death from ILD itself (n=23, 63.9%), Pneumonia(n=8, 22.2%), and Lung cancer (n=5, 13.9%). Bacterial infection (n=26) was dominant cause of AE, followed by fungal infection (n=15), and viral infection (n=10). Conclusions: This retrospective analysis of corticosteroid treatment shows more fatal outcome when we use high dose steroid in AE-ILD cases, and there was significant difference in mortality rate depending on the subgroup of ILD and medical background, such as intubation status, higher CRP level, and IPF cases.

        The level of urinary aflatoxin M1 in Korean adults

        Competitive ELISA methods were used to measure the level of aflatoxin M1 (AFM1) from urine in 1008 Korean adults. Subjects were selected by random sampling in all areas of Korea, except Cheju-do. The recovery rate of AFM1 using this method was 105% (73-124%). The geometric mean of urinary AFM1 in all subjects was 3.43 pg/mL (3.67 ng/g creatinine). The level of AFM1 in males was statistically higher, compared with female subjects. However, the levels of AFM1 did not differ according to age. Subjects in Chungbuk-do showed the highest urinary AFM1 concentration, whereas subjects in Kyeongnam-do showed the lowest concentration. Assuming an excretion rate of 5%, this AFM1 excretion corresponds to approximately 0.1 microgram/day in Korean adults.

        Idiopathic hypereosinophilic syndrome is characterized by marked elevation of eosinophil count ( >1,500/㎣) in peripheral blood without explaining etiology. It shows tissue involvement in many organs such as liver, G-I tract, myocar dium, lung, skin, kidney and nerve. Some case of hypereosinophilic syndrome with hepatic involvement have been reported, but not much. We have recently experienced a case with idiopathic hypereosinophilic syndrome of a 40 year old man who presented with RUQ pain. By abdominal CT, ill- defined subcapsular hematoma was detected. Liver biopsy showed eosinophilic infiltration within the portal tract and sinusoid. Drain procedure and corticosteroid adminiiatbrought marked improvement of clinical manifestation, hematologic abnormality and radiologic abnormality.

        항암화학요법 관련 오심과 구토 완화를 위한 비약물요법에 관한 연구동향

        Purpose: Purposes of this study were to understand the current trends on complementary therapy in relieving chemotherapy-induced nausea and vomiting and to suggest the future research direction. Method: Subjects were selected on CINAHL, MEDLINE, Korean Academy Data Base from 1980 to 2001 which used nausea, vomiting, chemotherapy and complementary therapy as key words in experimental studies. Eight korean articles and twenty-one international articles were analyzed in terms of general characteristics, research methods, and types of complementary therapy. Data were analyzed by using descriptive statistical methods. Result: Since 2000, researchers have more actively used complementary therapy. In subject characteristics, mean age was 35.5 years old, 45% of the researchers were performed with high level of incidence of chemotherapy induced vomiting, 14% of them set limit of consecutive cycle during research, and 65% of them did not comment the selecting criteria of sample. About 60% of them were designed post-test only control group: 35% used INV by Rhodes, 31% used Likert scale, and 24% used VAS for dependent variable. Muscle relaxation therapy was mostly applied for relief of nausea and vomiting. Conclusion: Further studies will be needed to control extrinsic variables affecting nausea and vomiting in research design and to accumulate evidence with studies applying various complementary therapies.

        피질이형성증과 동반된 난치성 간질의 외과적 치료

        The advent of neuroimaging technique has enabled the accurate diagnosis of structural abnormalities associated with intractable epilepsy. Among these, neuronal migration disorders(NMDs) or cortical dysplasias(CDs) involving cerebral cortex are increasingly recognized as a major cause of intractable epilepsy. CDs are expressed as variety of forms depending upon the time of injury to the developing nervous system. CDs are highly and intrinsically epileptogenic. It is crucial to completely resect the epileptogenic zone in focal CDs for better seizure outcome. Hemispherectomy or multilobar resection is preferred for hemimegalencephaly. Callosotomy can be chosen in the diffuse CDs who have intractable drop attacks. However, resective surgery can benefit the selected patients with diffuse CDs if localizing information persists.

      • 정수공정에서 이취미물질 제거에 관한 연구

        이취미물질중에서 페놀, Geosmin, 2-MIB를 선정하여 응집제(PACS)와 흡착제(PAC) 및 산화제(Cl2, ClO2)로 처리하였을 때 제거율을 알아보았다. 페놀의 경우 이산화염소(ClO2)와 분말활성탄(PAC)으로 처리하였을 때 제거효율이 높았고, Geosmin과 2-MIB는 분말활성탄(PAC)으로 처리하였을 때 제거효율이 높았다. 그러나 페놀, Geosmin, 2-MIB 모두 응집제(PACS), 염소(Cl2)로 처리하였을 경우에는 제거효율이 거의 없음을 알 수 있었다. The removal ratio of phenol, geosmin, and 2-MIB selected in odor and taste causing materials was investigated, when water was treated with coagulant(PACS), adsorbent(PAC), and oxidants(Cl2, ClO2). Phenol was removed at a relatively higher rate by the treatment with CIO2 and PAC(Powder Activated Carbon). The other side, geosmin and 2-MIB were removed at a higher rate by the treatment with PAC. But it was found that the treatment with coagulant(PACS) and chlorine(Cl2) for the removal of phenol, geosmin and 2-MIB was little removal efficiency.

        We introduced mechanical stimuli and micro patterned substrate with micro fibers to investigate the effects of those on neurite outgrowth along with nerve growth factor (NGF) in vitro. Micro fiber substrates were fabricated using an electros pinning process. And PC-12 cells cultured on substrates were simulated with newer growth factor and laminar flow shear stress in a fluid flow system. The results suggest that micro fiber substrates and fluid-induced shear stress are promising for simulating neuronal regeneration in a desired direction.

        자소엽 추출물이 식중독 유발세균의 증식에 미치는 영향

        In this study, Perilla frutescens which has been used as a folk remedy was investigated its antimicrobial activity. First, the Perilla frutescens was extracted with methanol at room temperatures, and fractionation of the methanol extracts from Perilla frutescens was carried out by using petroleum ether, chloroform, and ethyl acetate, respectively. The antimicrobial activity of the Perilla frutescens extracts was determined using a paper disc method and the growth inhibition curve was determined using methanol extracts of Perilla frutescens against Salmonella enteritidis and Bacillus cereus. The methanol extract of Perilla frutescens showed strong antimicrobial activity against Bacillus cereus at the concentration of 1.000 ppm. The 4,000 ppm of methanol extract from Perilla frutescens. retarded the growth S. enteritidis and Bacillus cereus up to 72 hours.
