        A study on efficient gate system based RFID at the container terminal

        It is a world trend to construct large terminal and develop automated container terminal to attract Super Post-Panamax and provide service which is based on differentiation In fact, there is insufficient research for automatization of terminal gate since the automatization of current constructed container terminal is only focused on increasing productivity and unmanned system through the automatization of quay, yard, etc. In this paper, we have investigated advantage/disadvantage of existing gate operation systems and compared each gate operation system in the aspect of raising terminal image and the productivity. For the specific study, we have used data from actual terminal gate operation and RFID model business sponsored by MOMAF (Ministry of Maritime Affairs and Fisheries). As a result, this paper carried out an efficient gate operation system and it has been expected that it will be performed as groundwork of automated gate operation system which is for design of container terminal and improvement of gate operation system.

      • 악성흑색종과 양성멜라닌세포성모반에서 bcl-2 단백발현에 관한 면역조직화학적 연구

        Background: The bcl-2 protein, contrast to p53, has been shown to be suppress the apoptosis, and overexpression of the bcl-2 protein has been reported in several malignant tumors. Although how the bcl-2 protein takes part in tumorigenesis has not been proved, the bcl-2 positive tumors appeared to have good prognosis in some malignant tumors. The expression pattern of the bcl-2 in melanocytic nevi and malignant melanoma have not been well known. The authors analyzed immunohistochemically expression of the bcl-2 protein to investigate the role of the bcl-2 protein in developing malignant melanoma. Method: 10 malignant melanoma and 10 benign melanocytic nevi were immunostained with anti-bcl-2 monoclonal antibody. Results: In malignant melanoma, 1 case was 2+ positive, 9 cases were 3+ positive. In benign melanocytic nevi, 2 cases were 2+ positive, 8 cases were 3+ positive. Conclusion: Bcl-2 protein is expressed by benign melanocytic nevi and malignant melanoma. There is no difference of bcl-2 protein expression in melanocytic nevi and malignant melanoma.

      • DSR을 기반으로 라우팅 테이블을 적용한 개선된 Ad hoc라우팅에 관한 연구

        In this paper, an improved source routing protocol is designed for mobile ad hoc network with unidirectional link. In the Mobile ad hoc network, nodes communicates with each other by usingmulti-hop method without any base station. In order for a source mobile node to send its data to a destination node through other intermediate nodes, mobile ad hoc routing protocol is required. DSR and AODV are the representative routing protocols for Mobile ad hoc network. However, unfortunately, current DSR and AODV routing protocol versions support mobile ad hoc network only with bidirectional links. We proposed and simulated a source routing protocol supporting mobile ad hoc network with unidirectional links which was based on DSR and AODV routing protocol. The result of simulation show average route setting time of 0.004~0.007 second and average date receiving rate of 97~97%.

      • 자전거 에르고미터운동시 유ㆍ무산소성 역치수준의 근전도 변화

        The aim of present study is investigate the change of IEMG and PEMG in the aerobic, anaerobic threshold by ventilation an blood locate concentration. Subjects of this study is adult sedentary men of age 32.18±4.28yrs. Subjects performed an incremental test on the bicycle ergometer from an initial work load of 0.5KP(60 rpm) to exhaustion by 3-min steps of 0.5KP. The results of this study was as follows: 1. The levels of relative exercises load in the ventilation, lactate, PEMG and IEMG by aerobic threshold were appeared 39.88±15.62%, 39.66±13.05%, 34.31±13.74% and 36.69±13.05%, respectively. It was not significant difference between variables. 2. The levels of relative exercise load in the ventilation, lactate, PEMG and IEMG by aerobic threshold were appeared 70.02±7.72%, 65.69±8.82%, 64.61±8.65% and 69.64±9.76%, respectively. It was not significant difference between variables. In this results above, It is considered that analysis methods by non-linear increase point of PEMG and IEMG related to both respiratory gas variables, blood lactate accumulation type and muscle function, muscle fiber recruitment type for determination of aerobic and anaerobic threshold.

      • 인체 흑색종 세포군에서의 Vascular Endothelial Growth Factor(VEGF) 분비

        Backgroung : Vascular endothelial growth factor(VEGF) is an angiogenic cytokine expressed by many human and animal tumors. Ultraviolet radiation(UVR) has been causally linked to melanoma initiation and progression. UVR is a known melanocyte mutagen and induces specific mutations in cellular oncogenes such as p53 and ras. Activated H-ras activates tumor angiogenesis in other cell types, and angiogenic switching is accompanied by up-regulation of VEGF. Objective : The purpose of this study was to determine whether VEGF release from human melanoma cell lines was altered by activating ras mutations or by inactivation of p53 induction and whether VEGF release of these transfected melanoma cell lines was changed after UVR radiation. Results : 1. Release of VEGF from menolayer culture of melanoma cells was significantly higher in cells with activating ras mutations and lower in cells with expression of dominant negative p53. 2. In spheroid culture, dominant negative p53 decreases VEGF release in WM35 cells, but activating ras did not affect VEGF release significantly. 3. The control cell line(WM35Neo) had higher VEGF release in the spheroid culture than in the monolayer culture, and dominant negative p53 or activating ras did not affect VEGF release significantly between these two culture systems. 4. Higher cell density increases VEGF release in all tested melanoma cell lines was shown in both spheroid and monolayer culture. This cell density effect was strongest in cells with activated ras in monolayer culture. 5. UVR inhibits VEGF release in all tested melanoma cell lines, and this effect was stronger in monolayer culture than in spheroid culture. The effect of UVR on VEGF release was significantly lower in the DNp53 transfectants. 6. UVR decreased the cell viability in a dose dependent manner regardless of the cell density or cell culture system. Conclusion : VEGF release in human melanoma was inhibited by ultraviolet radiation at cytotoxic doses, and this effect is stronger in monolayer culture than in spheroid culture. VEGF release from melanoma cell lines was also influenced by p53, ras and cell growth conditions in a complex manner.

      • 렙틴 수용체가 형질 도입된 신경세포에서 ER Stress로 유도된 렙틴저항성에 대한 일산화탄소의 억제작용

        연구 배경 및 목적: 렙틴이 음식의 섭취 양과 체중의 증가를 억제하는 중요한 호르몬 역할을 한다는 사실은 잘 알려졌다. 또한 렙틴에 대한 저항성이 비만의 중요한 원인이라는 연구가 많이 보고되고 있다. 그러나 렙틴 저항성에 대한 구체적인 기전은 아직 밝혀져 있지 않았다. 최근 endoplasmic reticulum (ER) 스트레스에 의한 unfolded protein response (UPR) 가 렙틴에 대한 저항성을 유발한 다는 보고가 있었다. 연구 대상 및 방법: 일산화탄소(carbon monoxide, CO)는 heme oxygenase (HO)-1이 헴(heme)을 산화적 분해로 만들어내는 무색, 무미, 무취의 기체로 항산화, 항염증, 항세포분열 및 항세포고사 작용을 나타내며 조직의 손상에 대한 세포보호작용을 한다. 최근 CO는 ER stress sensor 중의 하나인 protein kinase R-like endoplasmic reticulum kinase (PERK) 를 선택적으로 활성화 시키며 인산화된 PERK 는 HO-1의 전사조절인자인 Nrf2를 인산화로 활성화시켜 결국 HO-1의 유전자 발현을 촉진함이 보고 되였다. 또한 CO로 유도된 PERK의 활성화는 eukaryotic translation initiation factor (eIF) 2a를 인산화로 활성화 시키고 인산화된 eIF2a는 전사인자인 activating transcription factor (ATF)-4의 발현을 유도하지만 CO는 ER stress의 다른 두 개 sensors인 inositol requiring enzyme (IRE) 1a와 하위신호전단물질인 X-box binding protein (xbp)-1이나 ATF6의 활성화에는 영향을 미치지 아니한다. 그러나 ER stress 유발인자들인 thasigargin (TG), tunicamycin (TM), homocysteine 등으로 Xbp-1이나 ATF6가 활성화되는 것을 억제한다. 연구 결과: 본 연구에서는 ER stress 에 의해 활성화되는 UPR에 의해 유발되는 렙틴 저항성이 CO에 의하여 차단될 것이라는 가정을 세우고 다음과 같은 실험결과를 얻었다. TG나 TM에 의해 유도된 ER stress반응 즉 UPR은 렙틴의 신호전달로 야기되는 phospho-signal transducer and activation of transcription (STAT) 3로 인산화되는것을 억제시켰다. 그러나 ER stress를 유도시킬 시 CO를 CO releasing molecule (CORM)으로 처리하였을 때 UPR에 의한 렙틴의 신호전달로 말미암는 STAT3 인산화의 차단이 CO의 양과 노출 시간에 비례하여 제거 되었다. 결론: 이러한 실험결과는 CO에 의한 PERK 인산화는 ER stress 유도체에 의한 IRE1a의 하위 신호전달물질인 Xbp-1 이나 ATF6의 활성화를 억제하여 ER stress 반응인 UPR에 의한 렙틴 저항성을 차단함을 시사한다. 결론적으로 이러한 실험결과는 CO유도체를 이용하여 병인적인 렙틴 저항에 의한 여러 질병을 치료할 수 있음을 시사한다. Background: It is well known that leptin is an important circulating signal for the inhibition of food intake and the gain of body weight. Accumulating data suggest that leptin resistance has been considered to be one of the main causes of obesity. However, the detailed mechanism of leptin resistance is not known yet. Recently, endoplasmic reticulum (ER) stress-induced unfolded protein response (UPR) is reported to be responsible for the leptin resistance. Materials & methods: Carbon monoxide (CO), a reaction product of the cytoprotective heme oxygenase (HO)-1, is anti-apoptotic in a variety of models of cellular injury. Exogenous CO activated Nrf2 through the phosphorylation of protein kinase R-like endoplasmic reticulum kinase (PERK), resulting in HO-1 expression. CO-induced activation of PERK was followed by the phosphorylation of eukaryotic translation initiation factor 2a and the expression of activating transcription factor 4. However, CO fails to induce X-box binding protein-1 expression and activating transcription factor 6 cleavage. CO prevented X-box binding protein-1 expression and activating transcription factor 6 cleavage induced by ER-stress inducer such as thasigargin (TG), tunicamycin (TM) and homocysteine. Results: In the present study, we hypothesized that exogenous CO might block the leptin resistance which is induced by UPR during ER stress. Thasigargin or tunicamycin was used to induce ER stress. The activation status of leptin signals were measured by western blotting analysis using a phospho-signal transducer and activation of transcription3 (STAT3) antibody. In this study, ER stress markedly inhibited leptin-induced STAT3 phosphorylation. Conculusion: These results suggest that ER stress induces leptin resistance and exogenous CO-induced phosphorylation of PERK branch reversed ER stress-induced leptin resistance. Moreover, CO releasing molecule (CORM) blocks the inhibition of leptin-induced STAT3 phosphorylation. Together, the pathological mechanism of leptin resistance could be ameliorated by the use of exogenous CO..

      • 우수여자 유도선수들의 연간 트레이닝과 디트레이닝에 따른 무산소성 역치의 변화

        The seasonal variance for estimate of exercise intensity, energy expenditure, maximal oxygen uptake and anaerobic threshold of elite women judo players(Group Ⅰ:4,Group Ⅱ: 4) was measured during 3,5,7,9,10month. The results of this study is as follows. 1. Exercise intensity by %HRmax during seasonal training was shown range of 56.45-73.62, 65.56-82.15, 80.03-86.62, 62.06-68.83%HRmax, respectiveley, from 3 to 9month in the warm up, additional exercise, main exercise and cooling down. 2. Energy expenditure per min during seasonal training was shown range of 4.09-7.96, 5.41-8.66, 8.95-9.90 and 5.05-6.41Kcal/min, respectiveley, from 3 to 9month in the warn up, additional exercise, main exercise, main exercsie and cooling down. 3. Changes of maximal oxygen uptake(VO₂max)during seasonal training and detraining not significant difference between both group, and in the seasonal variance. However, maximal heart rate during detraining expressed significant difference(p<0.05) between both group. 4. Anaerobic threshold levels during seasonal training and detraining not significant difference between both group, and in the seasonal variance. However, AT-VE(1/min) and AT- work time(min) of group Ⅱ expressed significant difference(p<0.01) in the seasonal variance.

      • 콘택트렌즈 보존 용기 유래 Acanthamoeba lugdunensis KA/L5주의 내공생세균

        콘택트렌즈 보존 용기 유래 가시아메바 KA/L5주의 세포질 내에 존재하는 bacterial endosymbiont(내공생세균)를 투과전자현미경으로 관찰하여 확인하였다. 숙주인 가시아메바 KA/L5주는 형태학적으로 제2군에 속하였고, rDNA PCR-RFLP 결과 A. lugdunensis로 동정되었다. 미토콘드리아 DNA RFLP와 동위효소 분석상 이 충주는 국내 콘택트렌즈 보존용기에서 가장흔히 분리되는 type인 KA/Ll주, 국내 임상 분리주 중 하나인 KA/E2주, 내공생세균을 가지는 것으로 보고된 병원 냉각수 유래 KA/W4주 및 L3a주와 동일하거나 매우 유사한 성적을 보였다. 내공생세균은 약 1.38× 0.507㎛의 크기였고, 아메바 세포질 내에 불규칙하게 분포하고 있었으며. 그 표면에 아메바의 ribosome이 부착되어 있었다. 내공생세균을 둘러싼 lacunae나 막과 같은 구조는 관찰되지 않았다. Legionella 특이 primer를 이용한 효소중합반응(PCR)에서 내공생세균의 염색체 DNA는 증폭되지 않았다. A. lugdunensis의 우리말 이름을 담수가시아메바로 제안한다. Transmission electron microscopy of an Acanthamieba isolate (KA/L5) from a contact lens case revealed bacterial endosymbionts within cytoplasm of the amoebae, The Acanthamoeba isolate belonged to the morphological group II. Based on the polymerase chain reaction (PCR) - restriction fragment length Polymorphism (RFLP) of 18S ribosomal RNA coding DNA (rDNA), the Isolate was identified as A. lugdunensis. Strain typing by isoenzyme analysis using isoelectric focusing (IEF) and mitochondrial (Mt) DNA RFLP revealed that the isolate was closely related with KA/Ll , the most predominant type of isolates from contact lens storage cases, KA/E2, a clinical isolate, KA/W4, preciously reported to host endosymbionts, and L3a strains of A. lugdunensis. The endosymbionts were similar to those of KA/W4 In aspects that they were randomly distributed in both trophozoites and cysts, and were rod-shaped bacteria measuring approximately 1.38x 0.50㎛. But the number of endosymbionts per amoeba was significantly lower than that of KA/W4. They were neither limited by phagosomal membranes nor included In lacunae- like structure.

        We investigated the effect of aqueous extract of Ammomum xanthioides(AXAE) on immediate hypersensitivity. AXAE inhibited systemic anaphylaxis induced by compound 48/80 in mice. AXAE inhibited serum histamine levels induced by compound 48/80 in mice. Moreover, AXAE dose-dependently inhibited histamine release in peritoneal mast cells activated by compound 48/80. These results indicate that the AXAE may be benificial in the regulation of immediate type allergic reaction.

      • 피로를 유발하는 등속성 수축시와 회복기 혈중젖산 및 외측광근의 EMG Power Specturm 변화

        To examine the change of blood lactate concentration, EMG power spectrum, and Torque during fatiguing isokinetic contraction and recovery phrase, 6 sprinters performed maximal isokinetic knee extension at angular velocity of 3.14 rad.sec??(180°sec??) to 75% decreasing point (25%PT) using CybexⅡ system. In exercise phase, there was significant correlation between the decrement of peak EMG and median frequency and that of torque, and decrement of torque was greater than that of EMG . In recovery phase, EMG was restituded as normal value rapidly in a minute, but torque was restituded more slowly, and blood lactate concentration increased gradually from 50%PT to 3 minute after recovery. As a result of this study, it is suggested that muscle fatique due to decreasing of metabolic substrate and accumulation of metabolic products in peripheral site of muscle, and that inhibitory factors act neuromuscular junction in the end of exercise from rapid restitution of EMG in recovery phase.
