한약재 혼합추출물이 생쥐 탈모 모델의 모발성장 촉진에 미치는 영향

Objective:This study was performed to examine the effect of mixed medicinal herbal extract including Lespedeza bicolor Turcz. and 18 others (CS-10-H) on hair growth in an alopecia model of C57BL/6 mice. Methods:Male mice were divided into 3 experimental groups for treatment of CS-10-H or 5% minoxidil or vehicle solution, respectively. Dorsal skin of animals were shaved and topically applied with an amount of 200 μL of test materials once a day for 3 weeks. The apparent hair growth was monitored at 5, 10, 15 and 21 days after topical application. The changes of hair density and hair thickness were evaluated by phototrichogram using folliscope. Protein expression levels of hair growth-related factors were measured by western blotting. Results:Application of minoxidil or CS-10-H stimulated the hair growth compared to vehicle treatment. Hair density of minoxidil and CS-10-H group was increased about 206% and 208% on 14 day compared to that of vehicle group, respectively. Hair thickness of minoxidil and CS-10-H group was also increased about 220 % and 200% on 14 day compared to that of vehicle group. The protein expression of hair growth factors, IGF-1 and VEGF was significantly up-regulated in CS-10-H and minoxidil group than in vehicle treated group. Conclusion:These data suggest that CS-10-H shows potential usefulness as ingredients of hair tonic and hair-restorer for development of cosmetics and quasi-drugs, as result of its potent stimulating activity on hair growth in C57BL/6 alopecia model.
콘택트렌즈 보존 용기 유래 Acanthamoeba lugdunensis KA/L5주의 내공생세균

콘택트렌즈 보존 용기 유래 가시아메바 KA/L5주의 세포질 내에 존재하는 bacterial endosymbiont(내공생세균)를 투과전자현미경으로 관찰하여 확인하였다. 숙주인 가시아메바 KA/L5주는 형태학적으로 제2군에 속하였고, rDNA PCR-RFLP 결과 A. lugdunensis로 동정되었다. 미토콘드리아 DNA RFLP와 동위효소 분석상 이 충주는 국내 콘택트렌즈 보존용기에서 가장흔히 분리되는 type인 KA/Ll주, 국내 임상 분리주 중 하나인 KA/E2주, 내공생세균을 가지는 것으로 보고된 병원 냉각수 유래 KA/W4주 및 L3a주와 동일하거나 매우 유사한 성적을 보였다. 내공생세균은 약 1.38× 0.507㎛의 크기였고, 아메바 세포질 내에 불규칙하게 분포하고 있었으며. 그 표면에 아메바의 ribosome이 부착되어 있었다. 내공생세균을 둘러싼 lacunae나 막과 같은 구조는 관찰되지 않았다. Legionella 특이 primer를 이용한 효소중합반응(PCR)에서 내공생세균의 염색체 DNA는 증폭되지 않았다. A. lugdunensis의 우리말 이름을 담수가시아메바로 제안한다. Transmission electron microscopy of an Acanthamieba isolate (KA/L5) from a contact lens case revealed bacterial endosymbionts within cytoplasm of the amoebae, The Acanthamoeba isolate belonged to the morphological group II. Based on the polymerase chain reaction (PCR) - restriction fragment length Polymorphism (RFLP) of 18S ribosomal RNA coding DNA (rDNA), the Isolate was identified as A. lugdunensis. Strain typing by isoenzyme analysis using isoelectric focusing (IEF) and mitochondrial (Mt) DNA RFLP revealed that the isolate was closely related with KA/Ll , the most predominant type of isolates from contact lens storage cases, KA/E2, a clinical isolate, KA/W4, preciously reported to host endosymbionts, and L3a strains of A. lugdunensis. The endosymbionts were similar to those of KA/W4 In aspects that they were randomly distributed in both trophozoites and cysts, and were rod-shaped bacteria measuring approximately 1.38x 0.50㎛. But the number of endosymbionts per amoeba was significantly lower than that of KA/W4. They were neither limited by phagosomal membranes nor included In lacunae- like structure.
경옥고가미방 추출물이 생쥐의 양모 및 발모 관련 단백 발현에 미치는 영향

Objective: Present study was carried out to investigate the effect of Gyungohkgo-gamibang extract on hair growth and protein expression in an alopecia model of C57BL/6 mice. Methods: Mice were divided into 3 experimental groups including normal (vehicle), Gyungohkgo-gamibang extract (YNS-10) and 5% minoxidil-treated group. The test materials were daily applied with 0.1 ml per mouse on shaved dorsal skin for 3 weeks. The hair growth was monitored by photograph at 5, 10, 15, 21 days after topical application. Then the changes of hair density and hair thickness in the hair-removed area were evaluated by phototrichogram using folliscope. Also the expression level of growth factors related to hair growth was measured by western blotting. Results: Application of minoxidil or YNS-10 stimulated the hair growth compared to vehicle treatment. Therefore hair density of minoxidil or YNS-10 application was increased about 200% and 210% more than in vehicle application on 14 day, respectively. And hair thickness of both minoxidil group and YNS-10 group was increased about 220% and 210 % more than in vehicle spreading on 14 day, respectively. Futhermore the protein expression of IGF-1 and VEGF were significantly up-regulated on 7 day in YNS-10 and minoxidil-spreaded group compared to vehicle-applied group. Conclusion: These data suggest that YNS-10 has potent stimulating activity on hair growth in C57BL/6 mice and potential usefulness as ingredients of hair tonic and hairrestore.

비타민 C는 신진대사에 연관되어 있으며 특히 항산화 기능을 가지고 있다. 본 연구에서는 생체에서 비타민 C를 합성할 수 없는 SMP 30 녹아웃 마우스에 간 절제술을 시행하여 간 재생에서 비타민 C의 역할을 관찰하였다. 간 절제술은 마우스 중간엽 및 좌엽을 제거한 부분절제술을 수행하였다. 마우스는 간 절제술 후 비타민 C를 투여한 군(KV)와 비타민 C를 투여하지 않는 군(KO)로 나누어서 비타민 C의 효과를 관찰하였다. 결과 비타민 C를 투여한 KV 마우스의 간 회복이 투여하지 않는 KO 마우스에 비해 촉진되었다. KV 마우스의 혈액에서 관찰된 간소상 지표인 아스파르타산 아미노전달효소 및 간 손상 정도가 KO 마우스에 비해 낮게 관찰되었다. KV 마우스에서는 HGF와 c-Met에 의해서 TGF-베타 수용체 신호전달계가 활성화되고 세포주기 조절인자인 cyclin D1과 PCNA의 발현이 빠르게 증가되었다. 반면 KO 마우스에서는 활성화 되지 않았다. 또한 ERK와 GSK-3β 단백질의 활성화가 관찰되었으며 세포분열 간세포들의 유의적인 증가가 관찰되었다. 그리고 KV 마우스에서는 혈중 알부민의 농도가 높은 것으로 확인되었다. 따라서 본 실험결과는 SMP 30 결핍 마우스에서 비타민 C 투여는 간 재생시스템의 활성화와 이에 따른 빠른 회복을 초래한다. The capacity for liver regeneration involves a variety of nutritional factors. Vitamin C has multiple metabolic and antioxidant functions. In this study, we investigated the role of vitamin C in liver regeneration following hepatectomy in senescence marker protein (SMP)-30 knockout (KO) mice. Partial hepatectomy was performed by resecting the median and left lateral lobes of mice. Vitamin C accelerated liver recovery in SMP30 KO mice treated with vitamin C (KV). The livers of the KV mice exhibited lower levels of aspartate aminotransferase and lower injury than those of the KO mice. Increased type II transforming growth factor-β receptor (TGF-βRII)-mediated regeneration signaling was accompanied by HGF and cMet in the KV but not the KO mice. Consistent with this, the expression of cell cycle regulatory proteins, including cyclin D1 and proliferating cell nuclear antigen (PCNA), increased rapidly in the KV mice. Enhanced activation of ERK and GSK-3β proteins and a significantly increased number of binuclear hepatocytes were also detected in the livers of the KV mice. Moreover, the KV mice synthesized the highest levels of albumin. These data suggest that treating SMP30 knockout mice with vitamin C resulted in earlier recovery and liver regeneration by activation of the regeneration system.
Adrenomedullin Deficiency Increases the Susceptibility of Liver Fibrosis Induced by CCl<SUB>4</SUB>

아드레노메둘린은 모든 조직에서 발현되는 펩타이드로 이 펩타이드 수용체가 간경화 유발 시 증가된다. 이번 연구에서는 사염화탄소(CCl4) 투여로 유도되는 간경화에서 아드레노메둘린의 효과를 아드레노메둘린 유전자 한쪽이 결핍된 생쥐에서 관찰하였다. 사염화탄소를 일회투여 받은 급성 군과 6주 동안 지속적으로 투여 받은 만성군으로 나누어 효과를 관찰하였다. 간경화 정도는 병리조직적 검사와 웨스턴 블롯 방법을 사용하여 측정하였다. 급성에서는 결핍된 생쥐와 야생형 생쥐에서 별다른 차이점이 없었다. 만성의 경우 아드레노메둘린이 결핍된 생쥐에서 심각한 간경화가 관찰되었다. 아스파르테이트 아미노전이요소(AST)와 알라닌 아미노전이요소(ALT) 수치가 아드레노메둘린이 결핍된 생쥐 군에서 높게 관찰되었다. 간경화 마커 단백질인 콜라젠, 알파-SMA과 TGF-beta가 아드레노메둘린이 결핍된 생쥐 군에서 높게 관찰되었다. 또한 아드레노메둘린 결핍 생쥐 군은 심각한 지질 과산화 반응이 확인되었고 이는 글루타시오 펄옥시다아제(GSH)의 감소가 원인이었다. 따라서 이 연구는 아드레노메둘린의 결핍은 사염화탄소로 유도된 간경화 감수성을 증가시키는 것으로 결론할 수 있다. 이로써 아드레노메둘린은 간경화 새로운 치료 타겟으로 사용될 수 있음을 나타낸다. Adrenomedullin (AM) is a peptide expressed in all body tissues, and its related receptors are increased in liver fibrosis. In this study, we evaluated the effect of AM deficiency on liver fibrogenesis induced by CCl4 using AM heterozygous (HT) mice. The animals received a single injection of CCl4 or olive oil for the acute experiment, and received CCl4 or olive oil three times a week for 6 weeks for the chronic experiment. Fibrosis was accessed using histopathological analysis and the western blot. The AM HT mice showed mild pericentrilobular degeneration when compared to the AM wild type (WT) mice. In the acute experiment, there was no significant difference between the AM WT and AM HT mice. However, in the chronic experiment, the CCl4-treated AM HT mice showed more severe liver fibrosis than that of the CCl4-treated AM WT mice. The AST and ALT levels of the AM HT CCl4 group were higher than those of the AM WT CCl4 group. Additionally, the collagen deposition, α-SMA protein and TGF-β protein were increased in the AM HT CCl4 group when compared to the AM WT CCl4 group. The AM HT mice also exhibited severe lipid peroxidation through the GSH decrement. Taken together, our data suggest that AM deficiency increases the susceptibility to liver fibrosis induced by CCl4, indicating a novel therapeutic target for patients with liver fibrosis.
