        Specific Expression of Interferon-γ Induced by Synergistic Activation Mediator-Derived Systems Activates Innate Immunity and Inhibits Tumorigenesis

        The synergistic activation mediator (SAM) system can robustly activate endogenous gene expression by a single-guide RNA. This transcriptional modulation has been shown to enhance gene promoter activity and leads to epigenetic changes. Human interferon-γ is a common natural glycoprotein involved in antiviral effects and inhibition of cancer cell growth. Large quantities of high-purity interferon-γ are important for medical research and clinical therapy. To investigate the possibility of employing the SAM system to enhance endogenous human interferon-γ with normal function in innate immunity, we designed 10 single-guide RNAs that target 200 bp upstream of the transcription start sites of the interferon-γ genome, which could significantly activate the interferon-γ promoter reporter. We confirmed that the system can effectively and highly activate interferon-γ expression in several humanized cell lines. Moreover, we found that the interferon-γ induced by the SAM system could inhibit tumorigenesis. Taken together, our results reveal that the SAM system can modulate epigenetic traits of non-immune cells through activating interferon-γ expression and triggering JAK-STAT signaling pathways. Thus, this strategy could offer a novel approach to inhibit tumorigenesis without using exogenous interferon-γ.

        회사범죄는 중국 단위(單位)범죄의 중대한 유형으로 실무에서 가장 흔하게 볼 수 있는 단위범죄의 형식이다. 회사범죄에 대한 정의는 회사주체, 회사의지와 범죄행위 3가지 기본요소를 포함하여야 한다. 또한 고의와 과실 두 가지 죄과(罪過)형식도 포함하여야 한다. 회사가 범죄를 성립하는데 있어서는 자연인 범죄와는 다른 성립조건이 있어야 한다. 회사범죄의 귀책근거를 확립하여 회사의 독립적인 범죄주체의 지위를 중시하며 회사제도 등에서 회사범죄의 특징적인 요인 및 형벌목적을 구현하는 데서 출발하여 회사범죄 귀책근거의 정당성과 합리성을 고찰해야 한다. 일부 고의범죄의 법정범죄 종류 외에 회사도 과실 범죄와 일부 자연범죄의 종류를 실시할 수 있는데 회사과실범죄는 업무과실과 감독관리과실 두 가지 형식을 지닌다. 회사범죄의 책임자에 대한 판단은 신분 특징과 행위특징 두 가진 측면을 근거로 하여야 하며 또한 다른 죄과형식과 회사제도를 고려하여야 한다. 단지 한 사람의 자연인을 주주로 하는 1인 회사가 실시한 범죄행위는 회사범죄가 아닌 자연인 범죄를 성립하여야 한다. 민법의 법인인격 부인제도를 이용하여 1인의 회사범죄 주체자격을 부인하는 것은 부적절하며 응당 형법 중의 회사범죄 성립조건으로 그 자격을 부인하여야 한다. 한 명의 자연인 주주인 1인회사는 “회사 의지 지배 하에서 실시한 범죄행위”라는 이 조건에 부합하지 않기 때문이다. The corporation crime is an important and common type of the unit crime in China. The concept of corporation crime should include the corporation subject,the corporation will and the corporation action. And the concept should include intentional crime and negligent crime. How can a corporation`s action can be judged as a crime that depends on the 30 article in criminal law of China and the details about different charges. The theory basis of the corporation`s criminal responsibility should regard corporation as a relatively independent subject. In additions, different company systems and the expected aim of penalty for company crime are also the essential elements should be considered. The corporation can commit both intentional crimes and negligent crimes. And the latter include the vocational negligence and the supervision management negligence. What kind of natural person should take criminal responsibility of their corporation`s crime actions? That depends on the natural person`s status trait and action trait. The intentional crime and negligence crime, corporations of different forms also have different natural persons to take criminal responsibility. According to Chinese Company Law, one-man corporation is a kind of new corporation form according to Chinese Company law. If the one-man corporation which has only a natural stockholder commit a crime, the stockholder should take the criminal responsibility instead of The one-man corporation`s crime act is the natural stockholder`s crime act actually.

      • MiR-99a Inhibits Cell Proliferation and Tumorigenesis through Targeting mTOR in Human Anaplastic Thyroid Cancer

        MicroRNAs (miRNAs) are emerging as critical regulators in carcinogenesis and tumor progression. Recently, miR-99a has been reported as a tumor suppressor gene in various human cancers, but its functions in the context of anaplastic thyroid cancer (ATC) remain unknown. In this study, we reported that miR-99a was commonly downregulated in ATC tissue specimens and cell lines with important functional consequences. Overexpression of miR-99a not only dramatically reduced ATC cell viability by inducing cell apoptosis and accumulation of cells at G1 phase, but also inhibited tumorigenicity in vivo. We then screened and identified a novel miR-99a target, mammalian target of rapamycin (mTOR), and it was further confirmed by luciferase assay. Up-regulation of miR-99a would markedly reduce the expression of mTOR and its downstream phosphorylated proteins (p-4E-BP1 and p-S6K1). Similar to restoring miR-99a expression, mTOR down-regulation suppressed cell viability and increased cell apoptosis, whereas restoration of mTOR expression significantly reversed the miR-99a antitumor activity and the inhibition of mTOR/p-4E-BP1/p-S6K1 signal pathway profile. In clinical specimens and cell lines, mTOR was commonly overexpressed and its protein levels were statistically inversely correlated with miR-99a expression. Taken together, our results demonstrated for the first time that miR-99a functions as a tumor suppressor and plays an important role in inhibiting the tumorigenesis through targeting the mTOR/p-4E-BP1/p-S6K1 pathway in ATC cells. Given these, miR-99a may serve as a novel prognostic/diagnostic and therapeutic target for treating ATC.

        Automatic carrier phase delay synchronization of PGC demodulation algorithm in fiber-optic interferometric sensors

        Phase-generated carrier (PGC) demodulation algorithm is the main demodulation methods in Fiber-optic interferometric sensors (FOISs). The conventional PGC demodulation algorithms are influenced by the carrier phase delay between the interference signal and the carrier signal. In this paper, an automatic carrier phase delay synchronization (CPDS) algorithm based on orthogonal phase-locked technique is proposed. The proposed algorithm can calculate the carrier phase delay value. Then the carrier phase delay can be compensated by adjusting the initial phase of the fundamental carrier and the second-harmonic carrier. The simulation results demonstrate the influence of the carrier phase delay on the demodulation performance. PGC-Arctan demodulation system based on CPDS algorithm is implemented on SoC. The experimental results show that the proposed algorithm is able to obtain and eliminate the carrier phase delay. In comparison to the conventional demodulation algorithm, the signal-to-noise and distortion ratio (SINAD) of the proposed algorithm increases 55.99dB.

        Simple Joule-heating pyrolysis in air boosts capacitive performance of commercial carbon fiber cloth

        Commercial carbon fiber cloth (CFC) is treated by the Joule-heating pyrolysis method in air to boost its capacitive performance on the premise of energy- and time-saving considerations. A thermoelectric coupling model suitable for the Joule-heating pyrolysis is successfully established based on the comparisons between the simulated temperatures and actually measured ones. The temperature field on CFC surface induced by the Joule heat presents a concentric-ellipse shape that the temperature in the core is the maximal and gradually decays outward. Increasing the direct current (DC) voltage which is applied to the CFC from 1.0 to 6.0 V, the core temperature on the CFC surface can be raised from 31 to 519 °C. The specific surface area and hydrophilicity of the as-prepared porous CFC are greatly improved compared with the pristine one. Electrochemical test shows that the optimal Joule-heating pyrolysis parameters falls at 5.0 V and 12.5 min, and the areal specific capacitance of as-obtained CFC-5.0-12.5 is about 80 folds that of the pristine CFC. In addition to the much shorter preparation time, all the characteristics including areal specific capacitance, rate performance, and electrical conductivity of the Joule-heating pyrolyzed CFC are superior to those of the electrical furnace pyrolyzed counterpart. The aqueous symmetrical supercapacitor made of CFC-5.0-12.5 electrodes exhibits considerable power and energy densities with respect to the previously reported carbon electrode-based supercapacitors. For conductive precursors, the Joule-heating pyrolysis can be an ideal substitute for the traditional electric furnace pyrolysis.

        Improved Work Function of Preferentially Oriented Indium Oxide Films Induced by the Plasma Exposure Technique

        The preferentially oriented In2O3 thin films were prepared on glass substrates by conventional magnetron sputtering with Ar+ plasma exposure at room temperature. Based on the x-ray diffraction, x-ray photoelectron spectroscopy, and UV photoelectron spectroscopy results, it was found that the Ar+ plasma exposure not only enhanced the low-temperature crystallization of In2O3 thin films, but also led to a dramatic improvement in the work function. Furthermore, it demonstrated that the shift mechanism of the work function in In2O3 thin films mainly combined with theelimination of oxygen defects and the change of the preferential orientation of In2O3 film surface.

      • Atomistic Numerical Simulation on Nanoupsetting Process of Copper Brick

        Nanofabrication techniques such as nanoforging have been used to fabricate nanosize components in recent years. As a basic form of nanoforging process, nanoupsetting was studied in this paper via atomistic simulation. First a simplified computation model for upsetting process of single crystalline copper was created, and then the deformation mechanism was investigated at the atomic scale. The results show that the plastic deformation was initiated by the highlevel strain at the corners of the brick; partial dislocations nucleated at the corners and propagated along the [101] and [011] directions in the ( ? ? 1) plane; after unloading, the defects did not disappear or move out to the surface due to lattice friction; the computed load during nanoupsetting agrees well with the theoretical value. At last, the FEM simulation results for this single crystal were also shown, but no load drop was found in its load-displacement curve, which indicates that atomistic simulation is more elaborate than crystal plasticity FEM for nanoscale forming problems.
