        In 2019, the daily generation of food waste in Korea was 14,314 tons, which is a steady increase compared to the past. To efficiently use food waste as an effective resource, composting and feeding are carried out. In Korea, attention is gradually focused on a sustainable resource circulation society. Compost made of food waste has problems with foreign substances, heavy metals, and salt content, and solid fuelization research is being actively conducted to compensate for this. In this study, infrared spectroscopy(FT-IR), organic content measurement(TGA), and chemical composition were measured to compare the fuel characteristics with wood pellets, which are major solid fuels, in order to examine the physical and chemical properties of food waste compost.

        Comparative Analysis of Pigment Biosynthesis Gene Expression Related to Enhanced Skin Color in ‘Mishima’ Apples during Fruit Development

        In apple (Malus domestica), red coloration is an attractive trait for consumers. The color of fruit skin is determined by the distribution of carotenoids and anthocyanins in the skin tissue. ‘Fuji’ apples have red stripes over a yellow-green background, and ‘Mishima’, a ‘Fuji’ bud sport variety, has superior coloration compared to its parent. To investigate the genetic mechanism underlying the enhanced pigmentation in ‘Mishima’ fruit skin, we characterized the expression of pigment biosynthesis genes in ‘Fuji’ and ‘Mishima’ apples during fruit development. CIELAB data showed lower L* (relative lightness) and higher a*/b* ratio (relative redness) values in ‘Mishima’ apples, suggesting increased red pigmentation, which correlated with the levels of pigment biosynthesis gene expression. In ‘Mishima’, two carotenoid biosynthesis genes (MdZISO and MdLCY-ε) were upregulated at many stages of fruit development. Furthermore, enhanced red pigmentation was resulted from higher expression levels of three anthocyanin regulatory (MdMYB10, MdWD40, and MdGST) and three structural (MdCHS, MdCHI, and MdF3’H) genes, suggesting their positive interaction. These data further our understanding of the molecular basis of the enhanced skin color of ‘Fuji’ apple varieties.

      • NEDD4L limits cAMP signaling through ubiquitination of CREB‐regulated transcription coactivator 3

        <P>The transcription factor cAMP-responsive element-binding protein (CREB) is involved in a variety of physiologic processes. Although its activity appears to be largely correlated with its phosphorylation status, cAMP-mediated dephosphorylation and the subsequent nuclear migration of the CREB-regulated transcription factors (CRTCs) are required to stimulate CREB transcriptional activity. Among the 3 identified mammalian homologs of CRTCs, CRTC3 has been shown to be expressed predominantly in adipose tissues in response to catecholamine signals that regulate lipid metabolism. Here, we show that prolonged cAMP signaling down-regulates CRTC3 in a proteasome-dependent manner and that neural precursor cell-expressed developmentally down-regulated gene 4-like (NEDD4L), a specific ubiquitin ligase for CRTC3, is responsible for this process. By recognizing the PY motif of CRTC3, NEDD4L interacts with CRTC3 and promotes its polyubiquitination. Interaction between NEDD4L and CRTC3 is further boosted by cAMP signaling, and this enhanced interaction appears to be dependent on the cAMP-mediated phosphorylation of NEDD4L at the Ser448 site. Furthermore, we show that food withdrawal stimulates NEDD4L phosphorylation in mice, which then show a decrease of adipose tissue CRTC3 protein levels.         국내산 아로니아 두 품종(네로, 바이킹)의 영양성분 및 생리활성 분석

        본 연구에서는 국내 충북에서 재배되는 아로니아의 활용성 제고를 위해 두 품종(네로, 바이킹)의 영양성분과 생리활성을 분석하였다. 수분 함량, 조회분, 조지방, 조단백, 조섬유, 탄수화물의 함량을 분석한 결과, 네로, 바이킹 각각 수분 함량은 13.21%, 13.94%, 조지방은 0.73%, 0.33%, 조단백 5.47%, 6.51%, 조섬유 15.65%, 16.15%, 조회분 0.58%, 0.64%였으며, 탄수화물은 100에서 위의 값을 뺀 값으로 64.36%, 62.43%로 검출되었다. 네로, 바이킹의 생리활성은 안토시아닌이 각각 233.23 mg%, 235.24 mg%이었으며, 총 폴리페놀 함량은 393.73 mg%, 510.60 mg%, DPPH 전자공여능은 71.31%, 74.48%, ${\alpha}$-glucosidase 저해활성은 77.93%, 88.64%로 측정되었다. 결과적으로 아로니아는 생리기능성 물질인 폴리페놀 화합물과 안토시아닌의 함량 및 항당뇨 활성이 높은 것으로 나타났다. 충북에서 재배된 아로니아의 두 품종인 네로와 바이킹을 비교하였을 때, 총 폴리페놀과 혈당강하능 면에서 네로보다 바이킹이 우수한 것으로 보아, 이를 바탕으로 바이킹을 이용한 다양한 가공품이 개발된다면 아로니아의 이용성이 증진될 것이라고 생각된다. The nutritional compositions and physicochemical properties of two aronia varieties (Viking and Nero) cultivated in Chungbuk were investigated. The proximate compositions of Nero and Viking powder were:13% moisture, 0.58% and 0.6% ash, 0.73% and 0.33% crude lipid, 5.47% and 6.51% crude protein, 15.65% and 16.15% crude dietary fiber, and 64.36 and 64.43% crude carbohydrate, respectively. The proximate contents of the aronia samples were not significantly different. Among the physicochemical properties, there was also no significant change in total anthocyanin and antioxidant activity. However, Viking had higher total polyphenol contents and ${\alpha}$-glucosidase inhibition activity than Nero. The mineral contents of Al, Fe, Mn, Mg, Ca, Na, K, Cu and Zn in the two aronias were determined. Cu and Zn were not detected in both varieties. With regard to Al, Na and K, Nero was higher than Viking, while Ca was higher in Viking than Nero. From the results, Viking could be suggested to be beneficial for food processing.

        컨조인트 분석을 이용한 싱글 남성의 도시락 선택속성에 대한 연구

        본 연구는 싱글 남성 252명을 대상으로 2020년 7월부터 9월까지 도시락 이용실태 및 도시락 선택속성 및 수준을 조사하고 최적의 도시락 속성 및 수준을 도출하여 싱글 남성들이 가장 선호하는 도시락 속성을 제시하고자 하였다. 조사대상자들의 가장 큰 도시락 구매동기는 ‘원하는 시간대 구매가능’이며, 30대와 40대 이상의 조사대상자에게는 ‘맛’, ‘메뉴 다양성’, ‘광고 및 전단지’ 및 ‘배달 가능’이 20대 조사대상자들보다 유의적으로 높은 구매동기로 나타났다. 도시락 섭취장소는 20대는 주로 ‘매장’, ‘집’, 30대와 40대 이상은 ‘직장’, ‘차 안에서’도 섭취하는 것으로 나타났다. 편의점과 도시락 전문점에서 도시락을 구매하는 이유는 집 또는 직장과의 근접성 때문이며, 도시락 전문점에서 구매 할 경우는 ‘제품의 만족도’도 구매에 영향을 주고 있었다. 이는 도시락전문점의 도시락이 편의점의 도시락보다 가격대에 따른 메뉴의 다양성 측면에서 영향을 준 것으로 보인다. Focus group interview 결과, 도시락의 선택속성은 ‘구매장소’, ‘배달 유무’, ‘메뉴 구성’, ‘가격’으로 구성하였다. ‘구매장소’의 속성 수준은 ‘편의점’과 ‘도시락 전문점’의 2가지, ‘배달유무’는 배달의 ‘유’와 ‘무’, ‘메뉴 구성’은 ‘메뉴 1’, ‘메뉴 2’, ‘메뉴 3’의 3가지, ‘가격’은 ‘3,500원’, ‘5,500원’, ‘8,500원’의 3가지로 구성하였다. 도시락 속성의 상대적 중요도 분석결과, ‘가격’, ‘메뉴 구성’, ‘배달 유무’, ‘구매장소’의 순으로 나타났다. 선택속성의 수준에 대한 효용값 분석 결과, ‘가격’은 ‘3,500원’, ‘5,500원’, ‘8,500원’의 순으로 저렴한 가격의 도시락을 선호하며, 메뉴 구성은’ ‘메뉴 3’, ‘메뉴 2’, ‘메뉴1’의 순으로 육류 메뉴 구성의 도시락을 선호하였고, ‘배달유무’는 ‘유’를, ‘구매장소’는 ‘도시락 전문점’을 더 선호하였다. 각 속성과 속성 수준들의 효용값을 이용하여 싱글 남성들이 가장 선호하는 가상의 도시락 수준을 도출한 결과, ‘도시락 전문점’, ‘배달 이용’, ‘메뉴 3’, ‘3,500원’으로 도시락 선호예측 점수는 26.629점이었다. BTL 모형을 바탕으로 예측된 시장 점유율은 57.6%로 나타났다. 속성의 부분가치를 이용한 시장세분화 결과, 2 집단으로 구분되었으며, 군집1은 ‘도시락 전문점’, ‘메뉴 2’, ‘8,500원’, ‘배달 유’로 조사대상자의 31.3%가 해당되었다. 군집 2는 ‘도시락 전문점’, ‘메뉴 3’, ‘3,500원’, ‘배달 무’로 조사대상자의 68.7%가 해당되었다. 이상의 결과, 조사대상 싱글 남성들의 소득 수준 및 직업 등의 다양한 특성들이 도시락 선택속성에 영향을 주고 있었다. 조사대상자들은 가격은 저렴하지만 메뉴 구성이 다양한 도시락을 희망하고 있으며, 배달을 통한 구매가 점점 확대 될 것으로 보인다. 따라서 본 연구에서 실시한 속성의 중요도 및 시장세분화 결과는 향후 도시락 시장에서 소비자의 메뉴 요구도와 가격을 고려한 세분화된 도시락 제품 개발 및 마케팅 전략 구축을 제시한 것에 의의가 있겠다. 그러나 본 연구는 일부 편의점 및 도시락 전문점의 메뉴 및 가격조사를 바탕으로 연구가 진행되었기 때문에 도시락 업체 전체로 일반화하는 데 제한이 있으므로 향후 도시락 업체를 확대하여 후속 연구가 진행되어야 하겠다. This study analyzed the comparative importance of the selection attributes of a lunch box (dosirak) in single men and suggests a solution for the ideal type of lunch box that would appeal to single men using conjoint analysis techniques. A total of 252 questionnaires were distributed to single men who had previously purchased a lunch box in convenience stores or franchise dosirak stores. The conjoint design was applied to evaluate a hypothetical lunch box for single men. As a result, the order of importance of the attributes of the lunch box was found to be the price (41.66%), menu composition (24.62%), delivery status (17.45%), and place of purchase (16.27%). The ideal combination of the selection attributes of lunch box was a purchase in a franchise dosirak store, having a delivery service, a type 3 menu (Rice・Kimchi+fried pork cutlet, chicken, fish cutlet), and a low price of 3,500 won. The most preferred lunch box showed an incremental 57.6% potential market share using choice simulation compared to the lunch box being currently sold. Using the utility of attributes, the lunch box market for single men was classified into two segments. As a result of market segmentation, single men of cluster 1 preferred a lunch box with the menu type 2 and a price of 8,500 won while single men of cluster 2 preferred a lunch box with the menu type 3 and a price of 3,500 won. These results will help establish a database that contributes to the development of differentiated products and building marketing strategies that can satisfy the needs of single men.

        Role of IL-17A in the development of colitis-associated cancer.

        <P>A close relationship between inflammation and colon cancer has been widely accepted, and interleukin (IL)-17A plays an important role in controlling colonic inflammation. However, the role of IL-17A has not yet been validated in colitis-associated cancer (CAC). This study aims to identify the effects of IL-17A in tumorigenesis utilizing IL-17A-deficient mice in an experimental CAC model. CAC was induced in both the IL-17A-deficient and the C57BL/6 (wild-type, WT) mice by injection of 12.5 mg/kg azoxymethane followed by three rounds of 1.7% dextran sodium sulfate exposure to elicit colitis. On day 63 after the start of the study, mice were sacrificed. Colonic inflammation, proliferation and tumorigenesis were evaluated. Tumor numbers per mouse (1.43 versus 5.80; P = 0.02) and mean tumor size (1.17 versus 3.58 mm; P = 0.01) were significantly decreased in IL-17A-deficient mice compared with WT mice. Furthermore, the inflammation and the proliferation scores of IL-17A-deficient mice were significantly lower than WT mice. In the analysis of inflammatory mediators, IL-6, interferon-γ, tumor necrosis factor-α and IL-17A were markedly decreased in IL-17A-deficient mice compared with WT mice. In the western blot analysis, p-STAT3, cyclin D1, cyclin-dependent kinase 2, cyclin E, Glycogen synthase kinase 3-β and p-Akt were downregulated in IL-17A-deficient mice. Immunohistochemical staining with p-STAT3, Ki-67 and β-catenin revealed lower number of stained cells in IL-17A-deficient mice compared with WT mice. IL-17A ablation significantly decreases CAC tumorigenesis and thus may play an important role associated with chronic colitis.</P>

        Outcomes of a Modified CALGB 19802 Regimen in Adult Acute Lymphoblastic Leukemia

        We analyzed the efficacy and toxicity of a modified Cancer and Leukemia Group B (CALGB) 19802 regimen in adult acute lymphoblastic leukemia (ALL). From February 2002 to August 2005, 25 adults with untreated ALL were enrolled in the study. Compared to the original regimen, the modified CALGB 19802 regimen consisted of a 4-drug induction (cyclophosphamide, daunorubicin, vincristine, and prednisone) instead of a 5-drug induction (L-asparaginase was added to the previous regimen). This was followed by high-dose methotrexate (1,000 mg/m2×3 days) and cytarabine (2,000 mg/m2×4 days) for the consolidation cycles. High-dose systemic and intrathecal methotrexate was given for central nervous system prophylaxis. Twentythree patients (92%) achieved a complete remission (CR), and two patients (8%) had refractory disease. With a median follow-up of 21.5 months, 10 patients (40%) were alive and continued to be in CR. The 3-yr probability of an event-free survival and the overall survival were 39.0% and 47.4%, respectively. Treatment related mortality and major grade 3 to 4 neurotoxicity occurred in 1 patient and 3 patients, respectively. The modified CALGB 19802 regimen demonstrated a high remission rate and a favorable survival rate.

      • Expression and antigenicity of recombinant human respiratory syncytial virus glycoproteins having different affinity tags

        <P><B>Abstract</B></P> <P>Human respiratory syncytial virus (HRSV) is a main cause of lower respiratory tract infections in infants and the elderly. Glycoprotein (G) is major antigen on the viral surface, and plays a key role for virus entry. Therefore, purification of the glycoprotein of HRSV is critical for the development of HRSV vaccine and serological diagnosis. In this study, we report the design and characterization of glycoprotein engineered rationally to enhance the protein solubility and to facilitate efficient purification. We permuted HRSV glycoproteins with two tags: (i) an immunoglobulin (Ig) M signal peptide and a protein A B domain tag to render HRSV glycoprotein secret into the culture media and (ii) a foldon and 6 × histidine tag with or without transmembrane domain. Three recombinant baculoviruses were constructed: (i) transmembrane-truncated HRSV glycoprotein (amino acid positions 66–298) inserted with the N-terminal IgM signal peptide and protein A B domain (MG-GΔTM), (ii) truncated HRSV glycoprotein (amino acid positions 66–298) fused with a C-terminal foldon and 6 × histidine tag (GΔTM-FH), and (iii) full-length HRSV glycoprotein (amino acid positions 1–298) fused with a C-terminal foldon and 6 × histidine tag (G-FH). Highly soluble recombinant MG-GΔTM protein was clearly purified using one-step affinity chromatography with IgG-sepharose resin, whereas the recombinant G-FH protein and truncated GΔTM-FH were purified partially using nickel-resin. Although, the antigenicity of GΔTM-FH was stronger than highly mannose-rich MG-GΔTM protein, MG-GΔTM induced neutralizing antibodies efficiently in the mice to protect from infectious HRSV.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Three HRSV-G proteins were expressed with different tagging proteins. </LI> <LI> MG tag enhanced the production and the solubility of HRSV-G protein successfully. </LI> <LI> MG-GΔTM is glycosylated abundantly and induces the neutralizing antibodies. </LI> </UL> </P>

      • New Constitutively Active Phytochromes Exhibit Light-Independent Signaling Activity

        <P>Plant phytochromes are photoreceptors that mediate a variety of photomorphogenic responses. There are two spectral photoisomers, the red light-absorbing Pr and far-red light-absorbing Pfr forms, and the photoreversible transformation between the two forms is important for the functioning of phytochromes. In this study, we isolated a Tyr-268-to-Val mutant of Avena sativa phytochrome A (AsYVA) that displayed little photoconversion. Interestingly, transgenic plants of AsYVA showed light-independent phytochrome signaling with a constitutive photomorphogenic (cop) phenotype that is characterized by shortened hypocotyls and open cotyledons in the dark. In addition, the corresponding Tyr-303-to-Val mutant of Arabidopsis (Arabidopsis thaliana) phytochrome B (AtYVB) exhibited nuclear localization and interaction with phytochrome-interacting factor 3 (PIF3) independently of light, conferring a constitutive photomorphogenic development to its transgenic plants, which is comparable to the first constitutively active version of phytochrome B (YHB; Tyr-276-to-His mutant). We also found that chromophore ligation was required for the light-independent interaction of AtYVB with PIF3. Moreover, we demonstrated that AtYVB did not exhibit phytochrome B activity when it was localized in the cytosol by fusion with the nuclear export signal and that AsYVA exhibited the full activity of phytochrome A when localized in the nucleus by fusion with the nuclear localization signal. Furthermore, the corresponding Tyr-269-to-Val mutant of Arabidopsis phytochrome A (AtYVA) exhibited similar cop phenotypes in transgenic plants to AsYVA. Collectively, these results suggest that the conserved Tyr residues in the chromophore-binding pocket play an important role during the Pr-to-Pfr photoconversion of phytochromes, providing new constitutively active alleles of phytochromes by the Tyr-to-Val mutation.</P>

        Effect of porous polycaprolactone beads on bone regeneration: preliminary in vitro and in vivo studies

        Background: For the effective bone regeneration with appropriate pathological/physiological properties, a variety of bone fillers have been adapted as a therapeutic treatment. However, the development of ideal bone fillers is still remained as a big challenge in clinical practice. The main aims of this study are i) fabrication of a highly porous PCL beads; and ii) the estimation of the potential use of the porous PCL beads as a bone filler through preliminary animal study. Results: The porous PCL beads with size range of 53 ~ 600 μm (425 ~ 500 μm dominantly) are fabricated by a spray/precipitation method using a double nozzle spray and PCL solution (in tetraglycol). The PCL beads show highly porous inner pore structure and the pores are interconnected with outer surface pores. For the preliminary animal study, we recognize that the porous PCL bead can induce the new bone formation from the outer surface of bone defect toward the bone marrow cavity through the bead matrix. Conclusions: From the preliminary results, we can suggest that the highly porous PCL beads may be a promising candidate as a bone filler (scaffolding matrix) for the effective bone regeneration.
