        6-(3.4-Dichlorophenyl)amino-7-chloro-5.8-quinolinedione (RCK50) was tested for antifungal activities in mice systemically infected with Candida albicans. The therapeutic potential of RCK50 was also assessed in comparison with ketoconazole. RCK50 had ED_50 0.22±0.01 ㎎/㎏. Ketoconazole as a positive control had ED_50 6.00±1.70 ㎎/㎏. Intraperitoneally administered RCK50 at the ED_50 for 7 days and 14 days reduced Candida albicans colony count in the kidneys and liver. And administered RCK50 at the ED_50 for 14 days improved survival rates. The genotoxicities of RCK50 had been evaluated. RCK50 was negative in Ames test with Salmonella typhimurium and chromosomal aberration test in CHL cells. RCK50 did not show any clastogenic effect in mouse peripheral blood and was negative in mouse micronucleus assay. These results indicate that RCK50 has no genotoxic potential under these experimental conditions. Acute oral toxicity studies of RCK50 were carried out in ICR mice of both sexes. RCK50 did not show acute oral toxicities and LD_50 values were over 2.850 ㎎/㎏ in ICR mice.

      • A rare case of extensive papillary muscle calcification in senile cardiac calcification syndrome

        광범위 유두근 석회화는 흔하지 앉으며, 원인과 임상적 중요성에 관한 문헌만 드물게 찾아볼 수 있다. 반면에 작은 석회 침착은 노인에서 흔하며 심장 첨부에 주로 나타난다. 유두근 석회화는 관상동맥질환, 확장성 심근병증, 승모관 질환, 고칼슘혈증, 그리고 말기신부전에서 인산칼슘의 중가와 연관성이 있다. 저자는 심장초옴파와 MDCT를 통해 진단된 전외측 유두근의 광범위 석회화의 드문 사례가 있어 보고한다.

        Proficiency test of radioactivity measurement of gamma-ray emitting radionuclides in Korea

        The Korea Research Institute of Standards and Science (KRISS) has been conducting profciency tests of the radioactivity measurement of gamma-ray emitting radionuclides for domestic institutes in Korea. The test scheme used the z′-score with a robust analysis based on the International Standard Organization (ISO) 17043 and ISO 13528 standards for evaluating the measurement performance. The z′-score employed a robust standard deviation by Algorithm-A in the ISO 13528 standard instead of using a traditional standard deviation of participant results. The profciency test materials (PTMs) were prepared by spiking a diluted standard solution of radionuclides of interest into acidifed water with 1 M HCl. Combinations of 134Cs, 137Cs, 57Co, 60Co, 88Y, 113Sn, and 51Cr were used as PTMs. KRISS satisfes profciency testing requirements, and the test is accredited by the Korea Laboratory Accreditation Scheme (KOLAS). KRISS has conducted profciency tests annually since 2016, and more than 70 laboratories have tested over 650 test items so far. This study presents the results provided by the participants and the observed trends.

        속단(續斷) 추출물의 C57BL/6 마우스를 이용한 2주 경구투여 독성시험

        Objectives : A root of Dipsacus asperoides C. Y. Cheng et T. M. Ai (D. asperoides) has been traditionally used as a medicinal resource in several Asian countries, including Korean and traditional Chinese medicine that has been traditionally used for treating several medical conditions including pain, arthritis, and bone fractures in Korea. In the present study, we investigated potential subacute toxicities of D. asperoides extract. Methods : C57BL/6 mice (male, 7weeks) were randomly divided into 4 groups of 5 mice. Except for the control group, the mice were orally administrated D. asperoides extract at doses of 50, 150, or 450 mg/kg/day for 2 weeks. At the end of the treatment period, all mice were euthanized, and the following parameters were examined: mortality, body weight, clinical signs, gross findings, hematology, serum biochemistry, organ weight, and histopathology. Results : There were no abnormalities in mortality, clinical signs, body weight, gross findings, or organ weight after repeated administration of D. asperoides extract for 2 weeks, compared with the control group. In addition, there were no significant changes in hematological, serum biochemical, and histopathological parameters between the control group and D. asperoides extract administrated groups with doses of up to 450 mg/kg/day. Conclusion : In this study, D. asperoides extract showed no significant toxicities at a dose of up to 450 mg/kg/day in mice. Although we could not confirm the toxic dose of D. asperoides extract, it can be considered safe for further pharmacological use.

        소절개 유두괄약근절개술과 함께 내시경적 큰풍선확장술을 시행한 후 총담관담석의 재발과 내시경적 치료

        Background/Aims: Recent studies have reported the potentials of endoscopic papillary large balloon dilatation (EPLBD) with minor endoscopic sphincterotomy (EST) for the complete removal of common bile duct (CBD) stone in the high risk groups. However, there have been no reports about the recurrence of the CBD stone after EPLBD with minor EST. The aim of this study was to evlauate the recurrence of CBD stone after EPLBD with minor EST. Methods: A total of 1,036 patients who underwent endoscopic treatment due to CBD stones at Pusan University Hospital were enrolled. The patients were classified into two groups: those who underwent EPLBD with minor EST (group 1) and those who underwent EST treatment (group 2). We investigated clinical factors and recurrence rate between two groups. Results: The recurrence of CBD stone occurred in total of 74 patients (7%), and the recurrence rates of CBD stone were 21/321 (6.5%) in Group 1 and 53/715 (7.4%) in Group 2. There were no difference in the presence of diverticulum and the number and size of recurrent CBD stone between the two groups. In case of diverticulum existence, recurrence rates were 12/158 (7.6%) in Group 1 and 21/101 (20.8%) in Group 2. When compared to the case of no diverticulum existence (Group 1: 9/163 [5.5%], Group 2: 32/614 [5.2%]), the recurrence rate of CBD stone was significantly lower if treated after EPLBD with minor EST (p<0.01). Conclusions: CBD stone that recurs after going through EPLBD with minor EST can be successfully removed with an endoscopic treatment. The recurrence of CBD stone was especially lower in cases with periampullary diverticulum and treated with EPLBD with minor EST. Our results will be helpful in endoscopic retreatment and preventing the recurrence of CBD stone. (Korean J Gastroenterol 2011;57:352-357)

        Hypocholesterolemic Effect of Amaranth Squalene (Amaranth esculantus) in Rats Fed a High Cholesterol Diet

        In experiment 1, rats (n=6) fed diet containing 10 g/kg cholesterol for 4 wk (control) with either no amaranth (control), amaranth grain (300 g/kg, AG) or amaranth oil (90 g/kg, AO). Both the AG and AO groups had lower concentration of serum and hepatic cholesterol and triglyceride than the controls (p<0.05). Fecal excretions of cholesterol and bile acid in AO group increased about 4 fold and 2 fold, respectively, while AG affected only bile acid excretion (p<0.05). In experiment 2, rats (n=6) were fed the cholesterol diet for 4 wk and injected intraperitoneally with saline (control) or amaranth squalene (AS) for 7 d. The hypolipidemic effect of AS was evident in both serum and liver. Fecal excretions of cholesterol and bile acid were greater (p<0.05) in AS than control. HMG - CoA (3 - hydroxy - 3 - methylglutaryl coenzyme A) reductase activity was reduced in AS group (11.6%, p=0.13). This study suggests that the cholesterol - lowering effect of AS is mediated by greater fecal elimination of steroids through interference with cholesterol absorption.

        Background: In a previous study, we reported short term effectiveness and safety of dupilumab in Korean atopic patients Objectives: To report the long-term effectiveness and safety of dupilumab in Korea Methods: Ninety-nine patients with moderate to severe AD treated with dupilumab for 52 weeks were included and analyzed. They were evaluated using Eczema Area and Severity Index (EASI), Numerical Rating Scale (NRS), Patient Oriented Eczema Measure (POEM), and Dermatology Quality of Life Index (DLQI) at baseline, week 16, 32 and 52. Laboratory assessments were also performed at same visits. Results: Efficacy outcomes showed higher improvement at 52 weeks compared with 16 weeks; with high percentual reductions in EASI (88.1 %), peak pruritus NRS (65.6 %), POEM (67.2 %), and DLQI (69.0 %) compared to baseline. Proportion of patients achieving EASI 75 and 90 were 90.2 % and 53.7 %. There was a significant decrease in immunoglobulin E (IgE), total eosinophil count and Lactate dehydrogenase (LDH) levels. POEM and DLQI had a high correlation with clinical measured outcomes. In the analysis for the factors affecting the response of treatment about achieving EASI 90, female gender (OR 2.5), eosinophilia (OR 0.2) and elevated LDH (OR 0.07) were significantly associated. Most frequent adverse events included facial erythema (19.2 %) and conjunctivitis (17.2 %). Conclusion: Dupilumab treatment for 52 weeks in Korean patients with moderate-to-severe AD confirmed long term effectiveness and safety in real-world.

        Daidzein Activates Choline Acetyltransferase from MC-IXC Cells and Improves Drug-Induced Amnesia

        <P>The choline acetyltransferase (ChAT) activator, which enhances cholinergic transmission <I>via</I> an augmentation of the enzymatic production of acetylcholine (ACh), is an important factor in the treatment of Alzheimer’s disease (AD). Methanolic extracts from <I>Pueraria thunbergiana</I> exhibited an activation effect (46%) on ChAT <I>in vitro</I>. <I>Via</I> the sequential isolation of <I>Pueraria thunbergiana</I>, the active component was ultimately identified as daidzein (4′,7-dihydroxy-isoflavone). In order to investigate the effects of daidzein from <I>Pueraria thunbergiana</I> on scopolamine-induced impairments of learning and memory, we conducted a series of <I>in vivo</I> tests. Administration of daidzein (4.5 mg/kg body weight) to mice was shown significantly to reverse scopolamine-induced amnesia, according to the results of a Y-maze test. Injections of scopolamine into mice resulted in impaired performance on Y-maze tests (a 37% decreases in alternation behavior). By way of contrast, mice treated with daidzein prior to the scopolamine injections were noticeably protected from this performance impairment (an approximately 12%–21% decrease in alternation behavior). These results indicate that daidzein might play a role in acetylcholine biosynthesis as a ChAT activator, and that it also ameliorates scopolamine-induced amnesia.</P>

        Synthesis of Lysophosphatidylcholine Analogues Using D-Mannitol as a Chiral Template and Their Biological Activity for Sepsis

        LPC analogues including natural and unnatural LPC, 3-L-2-PC, acetylated LPC and ethylene glycol derivative are prepared from D-mannitol using in convenient procedures by only changing the synthetic sequences, and their protective activities against cecal ligation and puncture (CLP)-induced severe sepsis are compared. The chirality at C2 position in LPC is found to be required as (S)-configuration for sepsis inhibition, comparing from the protection activity between LPC 6 and unnatural LPC 8. The hydroxyl functionality is also very important and required at C2 or C3 position as shown in the protection activities of ethylene glycol analogue 11 and 3-L-2-PC 9.

      • Stable semi-transparent CH<sub>3</sub>NH<sub>3</sub>PbI<sub>3</sub> planar sandwich solar cells

        <P>Semi-transparent CH<SUB>3</SUB>NH<SUB>3</SUB>PbI<SUB>3</SUB> (MAPbI<SUB>3</SUB>) planar sandwich solar cells could be fabricated by simply laminating an FTO (F doped tin oxide)/TiO<SUB>2</SUB>/MAPbI<SUB>3</SUB>/wet hole transporting material (HTM) with additives and PEDOT:PSS (poly(3,4-ethylenedioxythiophene):poly(styrenesulfonic acid))/indium tin oxide (ITO). The best FTO/TiO<SUB>2</SUB>/MAPbI<SUB>3</SUB>/P3HT with additives/PEDOT:PSS/ITO planar sandwich structured solar cells exhibited a 12.8% (deviation: 11.7% ± 0.74%) average power conversion efficiency (<I>η</I><SUB>avg</SUB>) but poor visible transmittance due to strong absorption by P3HT. Meanwhile, the semi-transparent FTO/TiO<SUB>2</SUB>/MAPbI<SUB>3</SUB>/PTAA with additives/PEDOT:PSS/ITO planar sandwich solar cells exhibited a 15.8% (deviation: 14.45% ± 0.76%) <I>η</I><SUB>avg</SUB> without significant <I>J</I>–<I>V</I> hysteresis with respect to the forward and reverse scan directions. The average visible transmittance (AVT) was controlled from 17.3% to 6.3% and the corresponding <I>η</I><SUB>avg</SUB> changed from 12.55% to 15.8%.
