Sample Preparation and Stability of Human Serum and Urine Based on HPLC-DAD for Metabonomics Studies

Many literatures focus on the biological relevance and the identification of biomarkers for disease activity assessment while less attention has been paid to the development of standard procedures for sample preparation and storage based on liquid chromatography technique. The influencing factors including protein precipitation, storage temperature, storage time, and reconstitution by ultra pure water were analyzed employing HPLC-DAD. The effects were investigated from five participants over three months by principal components analysis (PCA) and the values of percent changes (PC). The samples with protein precipitation might slow the rate of bacterial enzymatic conversion. After protein precipitation, the average PC of urine samples ($0.136{\pm}0.013$, n = 5) is relatively less than that of the serum samples ($0.173{\pm}0.026$, n = 5) for three months. Minimal effects on metabolic profiles of serum and urine (PC < 0.15) are reasonable for metabolomic studies after protein precipitation and storage at $-20^{\circ}C$ for two months.
Triterpenoid saponins from Clinopodium chinense (Benth.) O. Kuntze and their biological activity

Four new ursane-type triterpenoid saponins, clinopoursaponins A–D (1–4), six new oleanane-type triterpenoid saponins, clinopodiside VII–XII (5–10), as well as eight known triterpene analogues (11–18), were isolated from the aerial parts of Clinopodium chinense (Benth.) O. Kuntze. The structures of the new compounds were determined based on extensive spectral analyses, including 1D (1H and 13C) and 2D NMR experiments (COSY, NOESY, HSQC, 2D TOCSY, HSQC-TOCSY and HMBC), HR-ESI-MS and chemical methods. Compounds 1–18 were evaluated for their protective effects against anoxia/reoxygenation-induced apoptosis in H9c2 cells and cytotoxicities against murine mammary carcinoma cell line 4T1. Compounds 8, 9 and 18 exhibited significant protective effects, while compound 1 exhibited cytotoxic activity with IC50 value of 7.4 μm compared to 7.6 μm for the positive control 10-hydroxycamptothecin.
중국 조손가정 초기 성인기 손자녀의 부모화 경험과 대인관계문제 간의 관계에 대한 연구 -불안 애착의 매개효과를 중심으로

본 연구는 부모화 경험, 불안 애착이 조손가정 초기 성인기 손자녀의 대인관계문제에 미치는 영향을 밝히고자 하는 목적으로 실시되었다. 중국에 거주하는 18~29세의 조부모와 단독적으로 3년 이상의 생활경험을 갖고 있는 209명을 대상으로 연구를 진행하였으며, SPSS 23.0와 Process Macro 3.3 프로그램을 사용하여 통계 분석을 실시하였다. 주요 결과, 첫째, 조손가정 초기 성인기 손자녀의 부모화 경험, 불안 애착, 대인관계문제는 중간보다 조금 높은 수준으로 나타났다. 둘째, 부모화 경험과 불안 애착이 대인관계문제에 유의미한 영향을 미치는 것으로 나타났으며, 부모화 경험과 대인관계문제 간의 관계에서 불안 애착의 부분 매개효과가 확인되었다. 따라서 본 연구는 조손가정 손자녀의 부모화 경험과 대인관계문제에 주목할 필요가 있다는 것을 보이고 조손가정을 위한 프로그램과 교육을 개발하는 필요성을 제시하였고 상담 및 치료 개입에 대해 논의하였다. Objectives: The purpose of this study is to identify the influence of parentification and anxiety attachment on interpersonal relationship problems among early adult grandchildren in multigenerational households. Methods: The study was conducted on 209 participants coming from multigenerational households aged 18 to 29 living in China, and the collected data were analyzed using SPSS 23.0 and SPSS PROCESS macro 3.3 Results: First, the grandchildren have experienced extensive parentification, have formed anxiety attachment, and have faced problems in interpersonal relationships. Second, parentification and anxiety attachment had significant effects on interpersonal problems, and a partial mediating effect of anxiety attachment was confirmed in the relationship between parentification and interpersonal problems. Conclusions: This study suggests that it is necessary to pay attention to the parentification and interpersonal relationships and to develop meaningful programs for multigenerational households.

The purpose of this study was to investigate the eating patterns of children with cerebral palsy having motor disturbances as well as stiffness. The food habits, nutritional status, and snack intakes of 1 to 7 year-old children with cerebral palsy were examined. The subjects were grouped into three categories according to their table-utensil handling skills: superior, normal, and inferior. The children in the superior group were significantly taller and heavier compared to children in the other two groups. The %EARs of folic acid and total calorie intake were insufficient in all three groups; however, their %EARs of other nutrients were fully sufficient. When comparing the children`s intake frequencies and preferences for snacks, the superior group showed a greater likelihood to consume various kinds of snacks than the inferior group. And the inferior group disliked more kinds of snacks than the other two groups. It was also shown that the inferior group had a significantly higher tendency for problems in chewing and swallowing. These results indicate that the development of table utensil-handling skills is very important for the food intake and growth of children with cerebral palsy, and the better their table utensil-handling skills the greater their physical development. Thus, considering their preference and intake frequency, it seems necessary that children in the inferior group be provided a greater variety of snacks and foods to receive more calories.

We design iterative methods based on the hybrid steepest descent algorithms for solving variational inclusions, equilibrium problems. Our results unify, extend and improve the corresponding results given by many others.

Purpose The purpose of this study was to investigate the survival trends and patterns of failure in patients with stage II nasopharyngeal carcinoma (NPC) treated with radiotherapy (RT) and chemotherapy over the last 20 years. Materials and Methods Thirty-eight hundred and eight patients diagnosed with stage II NPC between January 1990 and December 2012 were involved in this retrospective cohort study. All patients were treated with RT. According to the main imaging techniques and RT technology, we categorized these patients into four calendar periods: 1990-1996, 1997-2002, 2003-2007, and 2008-2012. Overall survival (OS), progression-free survival (PFS), locoregional relapse-free survival (LRFS), and distant metastasis–free survival (DMFS) were served as the clinical outcome. Results After a median follow-up period of 84.7 months, we observed increasing trends in survival and disease control. The 3- and 5-year OS rates increased from 87.1% and 78.7% in the first calendar period to 97.4% and 94.5% in the last calendar period, respectively (p < 0.001). Additionally, significant increasing trends could be seen in the PFS and LRFS during the four calendar periods. In the subgroup analysis, the LRFS in patients older than 50 years at diagnosis showed greater improvement than younger patients. However, the rate of distant metastasis was stable and relatively low, as the 5-year DMFS ranged from 90.5% to 94.7% among the four calendar periods. Conclusion The survival rates in patients with stage II NPC showed increasing trends from 1990 to 2012. The advance of RT provided excellent locoregional control and enhanced OS.

To investigate the expression of hPOT1 in the HeLa cell line and screen point mutations of hpot1 in different tumor tissues a two step osmotic method was used to extract nuclear proteins. EMSA was performed to determine the expression of hPOT1 in the HeLa cell line. PCR was also employed to amplify the exon14 sequence of the hpot1 gene in various of cancer tissues. A SV gel and PCR clean-up system was performed to enrich PCR products. DNAStar was used to analyse the exon14 sequence of the hpot1 gene. hPOT1 was expressed in the HeLa cell line and the signal was gradually enhanced as the amount of extracted nuclear proteins increased. The DNA fragment of exon14 of hpot1 was successfully amplified in the HeLa cell line and all cancer tissues, point mutations being observed in 2 out of 3 cases of endometrial cancer (66.7%) despite the hpot1 sequence being highly conserved. However, the sequence of hpot1 exon14 do not demonstrate point mutations in most cancer tissues. Since hPOT1 was expressed in HeLa cell and the probability of gene point variants was obviously higher in endometrial cancer than other cancers, it may be involved in the pathogenesis of gynecological cancers, especially in cervix and endometrium.

(Received 25 August 2008) The physical origin of the temperature dependence of the optical transmission of a magnetic fluid under an applied magnetic field is studied in this paper When a magnetic field is applied parallel to the plane of a magnetic fluid thin film, magnetic chains form in the same direction as the magnetic field, which suppresses the optical transmission. We observed that the optical transmission could be tuned by varying the ambient temperature of the magnetic fluid thin film. The design and the experimental results are also analyzed. Inhibitory Effects of Phenolic Alkaloids of Menispermum Dauricum on Gastric Cancer in Vivo

The present study was conducted to investigate effects and mechanisms of action of phenolic alkaloids of Menispermum dauricum (PAMD) on gastric cancer in vivo. In vitro, cell apoptosis of human gastric cancer cell line SGC-7901 was observed using fluorescence staining. In vivo, a mice model was constructed to observe tumor growth with different doses. Cell apoptosis was examined using flow cytometry and K-RAS protein expression using Western blotting. The mRNA expression of P53, BCL-2, BAX, CASPASE-3, K-RAS was examined by real-time PCR. PAMD significantly suppressed tumor growth in the xenograft model of gastric cancer in a dose-dependent manner (p<0.01). Functionally, PAMD promoted cell apoptosis of the SGC-7901 cells and significantly increased the rate of cell apoptosis of gastric tumor cells (p<0.05). Mechanically, PAMD inhibited the expression of oncogenic K-RAS both at the mRNA and protein levels. In addition, PAMD affected the mRNA expression of the cell apoptosis-related genes (P53, BCL-2, BAX, CASPASE-3). PAMD could suppress gastric tumor growth in vivo, possibly through inhibiting oncogenic K-RAS, and induce cell apoptosis possibly by targeting the cell apoptosis-related genes of P53, BCL-2, BAX, CASPASE-3.
