        생쥐 수정란의 양분에 의한 일란성 쌍태의 생산

        These experiments were carried out to produce monozygotic twin by bisection of mouse morula. The morula was bisected by microglass needle without any aid of microinstrument or by lateral incision using sharp blade attached to micromanipulator. Bisected dermi-embryos were cultured and transferred to pseudopregnant recipient mouse. The results obtained in these experiments were summarized as follows: 1. Of 285 decompacted morulae bisected by microglass needle, 180 embryos were divided into two demi-embryos, respectively, without any visible damage. 2. Total 180 pairs of demi-embryos separated from decompacted morula and subjected to in vitro culture were resulted in 85(47.2%) pairs of eu-blastocyst, 40(22.2%) pairs of eu-blastocyst and pseudo-blastocyst, 35(19.4%) pairs of pseudo-blastocyst and 20(11.1%) pairs of trophectodermal vesicle and degeneration. 3. Of 255 intact morulae bisected by microblade, 100(39.2%) embryos were divided into two demi-embryos, respectively, without damage. However, 95(37.3%) embryos were bisected into one normal and one damaged demi-embryos, respectively. The percentage of demi-embryos developed to blastocyst after in vitro culture with and without zona pellucida were 71.4 and 64.9% respectively. 4. Total 15 twins were produced following transfer of the 38 pairs of eu-blastocysts developed from demi-embryos to 25 recipients. However, none of pseudo-blastocyst gave birth to young, 5. The percentage of eu-blastocyst cultured with or without zona pellucida and developed to live young following transfer were 11.1 and 9.3% respectively.

      • 제주 이호 해수욕장의 계절별 지형변동의 현지관측

        이호 해수욕장의 계절별 지형변동의 현지관측을 통하여 해수욕장의 침식과 퇴적이 어떻게 진행되는가를 알아보려고 시도한 것이다. 현지관측은 계절별로 1회씩 수행하였다. 그리고 현지 측량으로 얻어진 결과를 이용하여 해안선 변화와 각 관측 지점에서의 침식과 퇴적의 이력을 통하여 연간 지형변동을 확인할 수가 있었다. 현지관측에 의한 제주 이호 해수욕장의 해안선은 겨울철에 전진하고 여름철에는 후퇴하는 경향을 해안선의 계절별 변화를 통하여 알 수 있었다. 지형변동 이력을 보다 정확하게 이해하기 위해서는 월별로 현지 관측을 수행하는 것이 좋은 듯하다. This field survey is tried to know how to arise the erosion and accretion of sand beach through the field survey of seasonal topography process in Iho beach. Field survey is carried out once a season. And, using the results obtained by field survey, annual topography process can be confirmed through the shoreline changes and the records of erosion and accretion at each surveying point. By the field survey, the shoreline of Iho beach has a tendency both advance in the winter and recession in the summer. In order to more understand about the records of topography process, field survey is good to be conducted every month.

        돼지 난포란의 성숙과 수정과정에서의 Cortical Granules 의 분포와 기능

        Recent advances with in vitro maturation and in vitro fertilization have contributed to increase the availability of mammalian embryos for the production of clone or transgenic animal. In livestock animals, however, the incidence of polyspermy during in vitro fertilization is much higher than in vivo fertilization. It has been observed that cortical granule reaction, which is a major factor for the block to polyspermy, is take place abnormally in the in vitro matured and fertilized livestock animals. Cortical granules are membrane-bound organelles and are situated at the oocyte cortex. Following sperm penetration, cortical granules extrude their contents into the perivitelline space. Cortical granule contents released into the perivitelline space are thought to be responsible for block to polyspermy at the vitelline membraneand the zona pellucida. Cortical granule contents of these material are probably glycosylated materials, but the exact function is not known. In immatured oocytes, cortical granules are dispersed through the cytoplasm of oocytes and moved next to the plasma membrane during maturation. Recent studies showed that microfilaments, one of the major cytoskeletons, have responsibility for the centrifugal movement of cortical granules during maturation. It was also suggested that in the pig the abnormal cortical reaction in vitro fertilization maybe related to the factors) from oviductal fluid. In this review paper, we addressed the distribution and the role of cortical granules during maturation and fertilization in mammalian oocytes.

        NUMERICAL ANALYSIS OF WAVE CHARACTERISTICS AROUND PERMEABLE SUBMERGED BREAKWATER ON THE POROUS SEABED

        In this study, wave characteristics coming with oblique incident angle to permeable trapezoidal submerged breakwater on the porous seabed are calculated by using boundary element method. This numerical analysis, based on the wave pressure function, is analyzing the continuity in the analytical region including fluid and structure. From the comparison of the reflection coefficients and damping coefficient, the results of this study are in good agreement with the existing results. The peak values of reflection coefficient obtained by permeable trapezoidal submerged breakwater on the porous seabed are smaller than those of permeable trapezoidal submerged breakwater on the non-porous seabed. The velocity vector in front of permeable trapezoidal submerged breakwater on the porous seabed is smaller than that in front of permeable trapezoidal submerged breakwater on the non-porous seabed with out the energy loss.

        In Vitro Developments of Porcine Embryos Produced by In Vitro Fertilization and Parthenogenetic Activation

        본 연구는 단위발생과 체외수정으로 생산된 돼지 난자의 발달양상과 inner cell mass(ICM) 그리고 trophectoderm (TE)의 세포배열을 조사하기 위하여 실시하였다. 단위발생은 ethanol 단독처리(haploid) 혹은 ethanol과 cytochalasin B을 공동처리(diploid)하였던바, 단위발생란은 체외수정란에 비하여 배반포까지의 발달이 저조하였지만, 단위발생에 있어서 ethanol과 cytochalasin B을 공동처리한 군이 ethanol 단독처리한 군보다 배반포까지 발달이 촉진되었다. 또한 단위발생란의 경우 total 세포수와 ICM 수에 있어서 체외수정란에 비하여 현저하게 감소되었지만, ethanol과 cytochalasin B을 공동처리한 단위발생란이 ethanol 처리된 단위발생란보다는 현저하게 높은 total 세포수와 ICM 수가 조사되었다. 이상의 결과로, 돼지의 착상전 배발달 양상과 ICM와 TE의 세포배열에 있어서 ploidy가 영향을 미친다는 것을 알수 있었다. The objective of this study is to determine developmental pattern and cell allocation to the ICM and TE in haploid and diploid of embryos following parthenogenetic activation and in vitro fertilization. The incidence of development to blastocyst was lower in the combined treatments of ethanol stimulation and cytochalasin B as compared to the control. However, the combined ethanol stimulation and cytochalasin B treatment (diploid) accelerated development to the blastocyst as compared to the ethanol treatment alone (haploid). Significantly reduction in the average number of total cells and ICM was observed in the parthenotes alone as compared to fetilized embryos, but those of combined ethanol stimulation and cytochalasin B treatment embryos were significantly increased as compared to ethanol alone embryos. These results suggested that the ploidy affects preimplantation developmental patters and cell allocation to the ICM and TE in the porcine.

        Fertilization and in vitro Development of Bovine Oocytes Following Round Spermatid Injection

        본 연구에서는 소 난자 세포질내 원형정자 및 원형정자 핵을 미세주입 후 수정과정과 체외 배 발달을 조사하였다. 원형정자 주입 3시간 전에 인위적 자극을 주고 원형정자를 주입했을 때 난자들의 수정률이 주입 직후에 자극을 준 것들과 자극을 주지 않은 것들 보다 높았다. 원형정자 주입과 원형 정자핵을 주입한 후 전핵 형성률과 전핵 이동률을 조사하였으나 유의차를 발견할 수 없었다. 전핵의 형성과 이동중에 미세소관의 움직임을 간접형광면역법 및 공촛점 현미경을 사용하여 조사해 본 결과, 난활성 직후 난자의 표층에서 미세소관이 발생해서 이것에 의해 웅성 및 자성전핵이 난자 중심부로 이전됨을 볼 수 있었다. 수정 후 원형정자의 세포질내 Mitochondria의 분포상태를 알아보기 위해 MitoTracker에 노출시킨 원형정자를 주입한 후 형광현미경에서 관찰해 보았다. 그 결과 원형정자의 Mitochondria가 주입 후 4-cell 이 전까지 는 관찰되었으나 그 이 후에는 발견 할 수 없었다. 원형정자를 미세주입한 후 소 난자를 체외에서 배양했을 때 2.5%의 난자가 배반포로 발달하였다. 이상의 결과는 체외 배양한 소 난자 내에 원형정자 혹은 원형정자 핵을 미세 주입하여 정상적인 수정과 배발달이 가능한 것을 보여 주는 것이다. In this study we determined fertilization processes and developmental ability of porcine oocytes following injection of round spermatid in the presence of artificial activation. Electrical stimulation at 3 h before spermatid injection significantly increased the incidence of normal fertilization as compared to those following injection without stimulation or with stimulation immediately after injection. The incidences of two pronuclear formation and apposition were not different in oocytes between following intracytoplasmic spermatid and spermatid nucleus injection. Indirect immunocytochemistry and laser scanning confocal microscopy study revealed that micro tubules were organized from the oocyte cortex following round spermatid injection, and this seemed to move both male and female pronuclei into the oocyte center. Paternal mitochondria which are introduced with spermatid have been observed up to 4-cell. Our study indicated that either round spermatid or it's nucleus can be used to produce viable bovine embryos by injection into unfertilized oocytes.

        Developmental Capacity of Bovine Follicular Oocytes after Ultra-Rapid Freezing by Electron Microscope Grid - I. Cryopreservation of Bovine Immature Oocytes

        This study was carried out to examine whether the developmental capacity of bovine immature oocytes frozen ultra-rapidly using electron microscope (EM) grids and EFS30 can be obtained. As freezing solution, we used EFS30 which consisted of 30% ethylene glycol, 0.5 M sucrose, 18% ficoll and 10% FBS added in D-PBS. As criterior of oocyte viability, the rates of maturation, fertilization and embryonic development were determined. The results obtained in this experiment were summarized as follows: When ultra-rapidly frozen immature oocytes were thawed, 43.2% of them were survived. The rates of maturation (84.1%) and normal 2 pronuclei formation (57.5%) of frozen immature oocytes were not significantly different when compared to those of control (92.5, 65.0%). In addition, the rates of $\geq2$-cell (65.0%) and blastocyst formation (30.8%) of freezing group were not significantly different when compared to those of control (73.7, 35.7%). These results demonstrate that developmental capacity of frozen-thawed bovine immature oocytes can be successfully obtained when survived from the ultra-rapid freezing method using EM grid and EFS30. 본 연구는 소 미성숙난자를 electron microscope (EM) grid와 동해제인 EFS30을 이용하여 초급속 동결하였을 때 정상적인 배 발달의 유도가능성 여부를 조사하고자 실시하였다. 동해제는 30% ethylene glycol, 18% ficoll, 0.5 M sucrose와 10% FBS 등이 PBS에 첨가되어 제작된 EFS30을 사용하였다. 난자 생존의 평가기준으로는 성숙, 수정 및 배발달을 조사하였다. 본 연구에서 얻어진 결과는 다음과 같다. 초급속동결-융해 후, 소 미성숙란의 생존율은 43.2%을 나타내었다. 동결-융해군의 체외 성숙 (84.1%)과 정상 자웅전핵 형성율 (57.5%)은 대조군의 결과 (92.5, 65.0%)와 비교하여 볼 때 유의한 차이는 없었다. 또한, 동결군의 체외수정 이후의 $\geq2$-세포기 형성 (65.0%)과 배반포형성율 (30.8%)도 대조군(73.7, 35.7%)의 결과와 유의한 차이를 나타내지 않았다. 따라서 소 미성숙난자는 EM grid와 EFS30 동결액을 이용한 초급속 동결방법에 의해 정상적인 배발달이 유도될 수 있다는 것을 알 수 있었다.
