        Production and Characterization of Enzymes Involved in Chitin Catabolic Cascade from a Bacterial Strain Isolated from Soil

        The present study was aimed to isolate the bacteria producing chitinolytic enzymes including chitin deacetylase (CDA) from soil origin. About 24 bacteria were screened for their chitinolytic enzymes producing ability on the basis of colloidal chitin. Amongst one of the most potent isolates designated to JI-02 was further selected as the most prominent chitinolytic and CDAlike enzyme (CDA-X) producer based on the ability of deacetylation of an artificial substrate pNP-acetamilide. The maximum production of CDA-X in crude enzyme from JI-02 was observed in the presence of 1% colloidal chitin at 37°C and pH 6.4 after 120 h of incubation. Potent enzyme activity of CDA-X was determined toward pNP-acetamilide, demonstrating that optimal pH, temperature and ionic strength of the CDA-X were assessed to be 7.0, 52°C and 125 mM, respectively. Unexpectedly, Nacetylglucosamine (GlcNAc) was not fully converted by the action of CDA-X to glucosamine (GlcN) under the established conditions. Further study on enzyme activity toward chitin-oligosaccharides consisting of multi-N-acetylglucosamine (GlcNAc)n, n=2-5 may be necessary to elucidate the mode of action which requires the minimum size of (GlcNAc)n. However, our data suggest that CDA-X can convert chitin to chitosan with the maximum yield of approximately 0.08g/L during the fermentation of the strain in the presence of chitin. Furthermore, we found that crude enzyme showed the significant chitosanolytic activity. Taken together, our data suggest that the strain JI-02 may be a potential candidate as a particular strain for better understating of chitin catabolic pathway.

        The purpose of this study was to determine the maximal cardiopulmonary response of healthy male college students in treadmill forward walking Subjects were composed of fifteen healthy male college students. Maximal heart rate and VO₂max Were determined for each subjects by administering a Bruce treadmill exercise test of the before and after forward walking training for eight weeks, Wilcoxon signed rank test was used to evaluate the changes of the forward walk training, The study showed that maximal heart rate were 170.4 beat/min in pre-training, and 177.0 beats/min in post-training respectively resting heart rate was 71.7 beat/min in pretraining and 68.3beat in post. The maximal oxygen consumption were 2283.3㎖/min, 45.6% in pre-training, and 2736.9㎖/min, 57.3% in post-training. There were significant differences (P<0.05) in maximal heart rate, heart rate ratio of maximal heart rate(% HR), oxygen consumption and oxygen consumption ratio of maximal oxygen consumption(% VO₂max) between the pre and post-training, The total lipid and HDL cholesterol, Triglyceride level were 633.3㎎/㎗, 40.8㎎/㎗, 99.3㎎/㎗ in pre-training, and 510.1㎎/㎗, 57.8㎎/㎗, 85.6㎎/㎗ in post-training and there were significant increased of HDL cholesterol in post-training. We concluded that 7km/hr forward walk training for eight weeks was more effective exercise program to promote health in the college male students.

        Actinomycosis is chronic, granulomatous, suppurative and fibrosing disease caused by Actinomyces. Actinomyces are anaerobic, G(+), non-acid-fast, branched, filamentous bacteria. The most commonly found microorganism is Actinomyces israelii. Common site for isolation of actinomyces are dental plaque, dental caries, calculus, and tonsillar crypt. A breach in the integrity of the mucosa by direct trauma or following a fracture, tooth extraction, root canal therapy or some intraoral surgical procedure is thought to be the most likely portal of entry. This is a case report of 23 years old male with cervicofacial actinomycosis developed after extraction and treated with surgical excision and antibiotics.

        This study was carried out to evaluate the application possibility of UF Membrane System to papermaking industry in order to optimize the recycling of white water. In this study, for the efficiency test of UF modules, the MWCO(molecular weight cut-off) of the membrane was estimated by the COD measurement from UF filtrate water. From the rejection rate of membrane, three kinds of UF module were selected and evaluated in the factors of COD, SS, turbidity and flow rate.

        유치원 실내공간구성의 관계론적 고찰 -자유선택활동에서의 유아 간사회적 상호작용을 중심으로-

        본 연구는 유치원의 실내공간구성 원리를 고찰해 보고 다양한 프로그램의 공간구성에 따른 유아들의 자유선택활동 시 유아간의 활동 및 상호작용의 의미를 탐색하였다. 유치원의 교실공간과 유아의 사회적 관계와의 관련성 및 그 의미에 대해 알아보고자 하였으며 유치원 교실공간의 교육적 가치와 의미를 탐구하고자 하였다. 이를 위해 다양한 프로그램을 운영하는 유치원 4곳을 선정하여 유아들의 자유선택활동 시간을 중심으로 동영상 촬영과 각 기관의 유아 8명, 교사 4명과의 면담으로 실시되었다. 연구결과 유치원의 공간구성에 따라 유아들은 공간을 명확한 경계를 가진 공간과 느슨한 경계를 가진 공간, 성취지향적인 공간과 대인관계 지향적인 공간. 의식과 무의식적인 공간으로 인식하고 있었다. 공간은 다양한 이미지와 의미를 가지고 있으며 목적에 충실한 절대적인 공간의 의미뿐만 아니라 관계론적 의미를 가지고 있음을 알 수 있었다. 따라서 아동중심과 발달에 적합한 공간구성 원리는 하나의 전통적인 원리로만 이루어질 수 없으며, 유치원의 실내공간구성은 유아들의 소통이 가능한 장소로서의 교실 공간 의미로 재해석될 필요가 있음을 시사한다. This study was to consider the meaning of activities and interaction among young children according to space construction of various programs in free-choice activities. That is, the purpose of this study was to analyze the correlation and meaning between classroom` space and social relation of young children, and to study the educational value and meaning of classroom space in kindergarten. The research participants were 8 young children and 4 teachers of 4 kindergartens situated in Busan.. The data was analysed by in-depth interview of the young children and teachers and recordings of free-choice activities in the kindergarten carrying each program. The conclusions drawn from the study are as follows. First, young children recognized space according to space construction in kindergarten as the space with a well-defined or loose-defined boundary, and aim of achievement or personal relations, and conscious or unconscious influence. That is, space has various images and meaning, and not only meaning for purpose but also meaning for human relations. In conclusion, the developmentally appropriate principle of space construction is impossible to contain by a traditional that. 〔I don`t understand this sentence.〕Therefore, we suggest that space construction in kindergarten could do with reinterpretation as a meaningful of place of communication among young children.

        Background/Aims: Sustained virologic response (SVR) has been regarded as therapeutic end point in chronic hepatitis C (CHC). But, ideal follow-up time for these patients still remains undefined. In analyses of SVR durability, the incidence of late relapse is extremely low (<1%) and patients with SVR have significantly fewer liver related complications, less hepatocellular carcinoma (HCC), and liver-related deaths. We investigated the durability of SVR and development of HCC to assess the clinical outcome in chronic hepatitis C (CHC) patients who achieved SVR after combination therapy of pegylated interferon (PEG-IFN) and ribavirin. Methods: The study was conducted on 293 CHC patients who achieved an SVR after PEG-IFN and ribavirin combination treatement. Liver function tests, qualitative HCV RNA assays and imaging study (USG or CT) were performed every 6 or 12 months during the follow-up period. Results: Among the 293 patients, 285 (genotype 1, 136; genotype non-1, 149) were followed up for more than 6 months after SVR. These 285 patients were aged 51.2 ± 11.0 years (mean ± SD), and 169 (59.3%) of them were male. The median follow-up duration was 29.4 months (range 6-81 months). The reappearance rate of HCV RNA during follow-up was 2.1% (n=6). Among 6 relapsers, five patients were genotype 1 , three had dose reduction of PEG-IFN or ribavirin (1:2) during combination therapy and the rest of them were treated with shorter duration (24wks). The median follow up duration of relapse was 5 months (range 3 -9). Six patients (2.1%) developed HCC during the follow-up period and the median follow up duration was 23.3 months (range 3 -74). Conclusions: These results suggests that even though, the relapse rate is low, the relapse could occur after SVR achievement, and all cases of relapse occurred within one year after SVR. Furthermore, periodic surveillance for HCC should be done because risk of HCC development was same with rate of relapse and still exist even after SVR achievement.

        Enzymatic Characterization of α-L-Fucosidase

        The aim of the present study is to define the strain that degrade fucoidan as sole carbon source. A bacterium, designated as the strain HH-0518, utilizing fucose was isolated from soil. HH-0518, belonging to the Enterobacteriaceae family by 16S rDNA and physiological analyses, can hydrolyze p-nitropheny-α-L-fucoside (pNP-α-L-fucoside) used as a sole substrate. It was elucidated that α-L-fucosidase of HH-0518 which can hydrolyze pNP-α-L-fucoside was localized on the cell membrane, especially. The α-L-fucosidase appeared to have optimal pH and temperature at 8.0 and 37 o C. Additionally, kinetic parameters of α-L-fucosidase toward artificial substrates pNP-α-L-fucoside were determined to be Km = 1.05 mM and Vmax = 300 pmole/mg/ min, respectively. No significant enzyme activity was observed in the presence of variety of carbon sources including monosaccharides such as galactose, mannose, fructose, sucrose, xylose, glucosamine and N-acetylglucosamine, compared to the medium containing fucoidan only. To the best of our knowledge, we first report the characteristics of α-L-fucosidase from Enterobacter hormaechei strain.

      • Effects of matured silkworm hemolymph on suppressing melanin synthesis and skin flora

        In this study, matured silkworm hemolymph was studied for inhibitory effect on melanocyte formation, tyrosinase activities and antibacterial effect. A matured silkworm hemolymph mass purification method was developed and some of suppressing melanin formation effects were investigated, too. Antibacterial effects were tested by Escherichia coli, Staphylococcus aureus, Staphylococcus epidermidis and Candida albicans. As a result, the advanced purification method was higher in yield rate than existing one by 53%. A free radical scavenging activity of hemolymph was higher than sericin on the market(IC50, 202.9±33.1) and lower than vitamin C(0.57±0.02) by 6.4±0.3 ㎍/㎖. An inhibitory effect on melanin synthesis was higher in hemolymph compared to morus bark and arbutin by 9.15% and 11.56%, respectively. The antibacterial effects on Escherichia, Staphylococcus were detected at 2% hemolymph concentration, which was relatively good. Thus, these results suggest that hemolymph of matured silkworm may have beneficial properties as a material for cosmetics.
