        黃菊과 黑薔薇를 이용한 기능성 花茶 개발 Ⅱ.花茶의 품질특성

        본 연구는 화차의 비율별 제조공정에 따른 화차의 관능적 품질을 평가하기 위해서 수행되었다. 녹차와 꽃의 시료에 대한 색도중 L, a, b값은 녹차가 29.93, -8.24, 9.27로 녹색을, 황국은 47.20, -1.69, 29.12로 밝은 황색을, 흑장미는 37.18, 4.47, 4.12로 흑적색을 띠웠다. 색상과 채도는 황국이 -17.23과 29.17로 가장 높아 색상이 가장 밝고 선명하였으며, 흑장미는 0.92와 6.08로 색상이 어둡고 둔탁한 적색을 띠워 녹차에 꽃을 혼합할 경우 시각적으로 황국은 쉽게 구별이 되었으며 흑장미는 쉽게 구별되지 않았다. 황국 화차는 꽃의 비율이 증가할수록 국화향, 상쾌하고 개운한 단맛의 증가와 더불어 떫은 맛이 감소하였으나, 혼합비율이 9:2 이상일 경우에는 쑥냄새와 같은 강한 국화향이 증가하여 녹차 고유의 향은 거의 상쇄되는 경향이었다. 장미 화차는 장미꽃이 증가할수록 시큼한 장미향이 강하게 증가하였고, 탄닌성분이 많아 떫고 텁텁한 맛과 시큼한 맛이 강한 것으로 평가되었다. 녹차에 알맞은 꽃의 혼합율은 9:1이하의 비율로 혼합했을 때 황국과 흑장미의 꽃잎색이 녹차와 어울려 전체적인 조화성과 풍미, 시각적 아름다움을 증가시켰다. 녹차에 꽃을 혼합한 화차는 녹차 단일의 차보다는 시각적 즐거움과 독특한 방향성이 부가되어 맛과 향이 풍부한 기능성 꽃차 개발에 좋은 재료로 평가되었다. This experiment was carried out to investigate the effect of mixed ratio on the sensory evaluation in manufacture of functional flower tea. Among the chromaticity about green-tea and material flower, the amount of L, a, b of green tea was 29.93, -8.24, 9.27 with green, the amount of a yellow chrysanthemum(C.indicum) was 47.20, -1.69, 29.12 with bright yellow, the amount of a black rose(Rosa hybrida)is 37.18, 4.47, 4.12 with dark red. The yellow chrysanthemum has brightest and most distinct color with -17.23 hue and 29.17 chrome value, but the black rose has dark dead red color with 0.29 hue 6.08 chrome value. When the yellow chrysanthemum mixed with green tea, it was easily distinguished with naked eye. But the black rose was not distinguishable easily. As the retio increase, the yellow chrysanthemum tea has more chrysanthemum scent, increase of refreshing and relieved sweetness, and less astringent taste. If the mixed rate was over 9:2, original scent of green tea was almost offset with strong chrysanthemum scent like mugwort's. As the ratio increase, the rose tea has strongly increased sourish, and because of the tannin it was appraised strong astringent, unpleasant, sourish taste. The best-mixed ratio of green tea and flower is 9:1. When it mixed at this ratio, the yellow chrysanthemum and petal color of black rose was well matched with green tea, and the tea has general harmony and flavor and it is visually beautiful. Flower tea, green tea from flower, has added visually joy and unique fragrance, it was valued as fine material for rich taste and scent technical tea development.

      • 토양물리성 및 엽내 무기성분과 적과수준이 감 대봉 품종의 낙과에 미치는 영향

        This experiment was conducted on persimmon (Diospyros kaki Thunb) to study the cultural method reducing preharvest drop through analysis of the character of dropped fruit, thinning degree and the nutrient state of tree. Fifteen 10-year-old 'Hachiya' persimmom trees were used. Thinning were applied to one fruit per one twig (A treatment), one fruit per two twigs (B treatment) and one fruit per three twigs (C treatment) which had about six-leaf on 20 June. The number of fruit seed was investigated on dropped and harvested fruit on 20 June, 11 July and 1 Oct. Leaf analysis was conducted on both tree with heavy fruits drop and tree without fruit drop on 20 July, 1999. Preharvest drop rate with thinning degree tended to more decrease in B treatment and C treatment than in A treatment and harvest quantity of fruit was higher on B treatment than on C treatment. Preharvest drop rate seemed to be higher on the upper part of tree than on the lower part. The number of seed in fruit was about 2.1 per dropped fruit and 3.9 per harvested fruit, which showed the tendency that Preharvest drop was decreased in the fruit with many seeds. The tree with high preharvest drop rate has lower N, Ca, Mg concentration and higher Mn concentration within leaves than that with low preharvest drop rate. Thinning degree, setting point and the number of seed in fruit, the concentration of mineral nutrient in leaf had an effect on preharvest drop. Therefore we guessed that movement and supply of nutrient could affect preharvest drop.

      • Assessment of microbial community in paddy soils cultivated with Bt and Nakdong rice

        The cultivation of genetically modified (GM) crops has increased due to their economic and agronomic advantages. Before commercialization of GM crops, however, we must assess the potential risks of GM crops on human health and environment. The aim of this study was to investigate the possible impact of Bt rice on the soil microbial community. Microbial communities were isolated from the rhizosphere soil cultivated with Bt rice and Nakdong, parental cultivar and were subjected to be analyzed using both culture-dependent and molecular methods. The total counts of bacteria, fungi, and actinomycetes in the rhizosphere of transgenic and conventional rice were not significantly different. Denaturing gradient gel electrophoresis (DGGE) analysis of PCR-amplified 16S rRNA genes revealed that the bacterial community structures during cultural periods were very similar each other. Analysis of dominant isolates in the rhizosphere cultivated with Bt and Nakdong rice showed that the dominant isolates from the soil of Bt rice and Nakdong belonged to the Proteobacteria, Cloroflexi, Actinobacteria, Firmicutes, and Acidobacteria. These results indicate that the Bt rice has no significant impact on the soil microbial communities during cultivation period. Further study remains to be investigated whether the residue of Bt rice effect on the soil environment.

        해피백 운동기구를 이용한 체간안정화 운동이 20대 대학생의 정적균형지수에 미치는 영향

        The purpose of this study was to investigate the effects of trunk stability exercise used Happyback on static balance ability in college students in 20s. Thirty subjects participated in the experiment. Males and females were divided into 15 subjects in the experimental group and 15 subjects in the control group by random same ball selection. The experimental group applied the trunk stabilization exercise using the Happyback, and the control group applied the trunk stabilization exercise without the Happyback. The exercise program consisted of a total of 5 sets of balance support posture, left side plank posture, right side plank posture, bridge posture, and quadruped posture in a sitting state, and was performed 3 times a week for 4 weeks. The static balance index was measured with Tetrax, a posture measuring device. The subjects measured the static balance index before and after the experiment. The experimental group decreased in SS and WDI, but was not significant, increased significantly in force plate AB and force plate BD, and increased but not significant in force plate CD, force plate AC, force plate AD, and force plate BC. It is considered to prove that the static balance index improved when normal people performed trunk stabilization exercise using Happyback. In conclusion, Trunk stabilization exercise using Happy Bag is highly recommended as a new exercise method for static balance at home.

        Evaluation of in vitro anti-oxidant and anti-inflammatory activities of Korean and Chinese Lonicera caerulea

        BACKGROUND/OBJECTIVE: The honeysuckle berry (HB) contains ascorbic acid and phenolic components, especially anthocyanins, flavonoids, and low-molecular-weight phenolic acids. In order to examine the potential of HB as a hepatoprotective medicinal food, we evaluated the in vitro anti-oxidant and anti-inflammatory activities of Korean HB (HBK) and Chinese HB (HBC). MATERIALS/METHODS: Antioxidant and anti-inflammatory effects of the extracts were examined in HepG2 and RAW 264.7 cells, respectively. The anti-oxidant capacity was determined by DPPH, SOD, CAT, and ARE luciferase activities. The production of nitric oxide (NO) as an inflammatory marker was also evaluated. The Nrf2-mediated mRNA levels of heme oxygenase-1 (HO-1), NAD(P)H dehydrogenase [quinone] 1 (Nqo1), and glutamate-cysteine ligase catalytic subunit (Gclc) were measured. The concentrations of HB extracts used were 3, 10, 30, 100, and 300 μg/mL. RESULTS: The radical scavenging activity of all HB extracts increased in a concentration-dependent manner (P < 0.01 or P < 0.05). SOD (P < 0.05) and CAT (P < 0.01) activities were increased by treatment with 300 μg/mL of each HB extract, when compared to those in the control. NO production was observed in cells pretreated with 100 or 300 μg/mL of HBC and HBK (P < 0.01). Treatment with 300 μg/mL of HBC significantly increased Nqo1 (P < 0.01) and Gclc (P < 0.05) mRNA levels compared to those in the control. Treatment with 300 μg/mL of HBK (P < 0.05) and HBC (P < 0.01) also significantly increased the HO-1 mRNA level compared to that in the control. CONCLUSIONS: Thus, the Korean and Chinese HBs were found to possess favorable in vitro anti-oxidant and anti-inflammatory activities. Nrf2 and its related anti-oxidant genes were associated with both anti-oxidant and anti-inflammatory activities in HB-treated cells. Further studies are needed to confirm these in vivo effects.

        Expression Profiling of Calcium Induced Genes in Cultured Human Keratinocytes

        <P>Terminal differentiation of skin keratinocytes is a vertically directed multi-step process that is tightly controlled by the sequential expression of a variety of genes. To examine the gene expression profile in calcium-induced keratinocyte differentiation, we constructed a normalized cDNA library using mRNA isolated from these calcium-treated keratinocytes. After sequencing about 10,000 clones, we were able to obtain 4,104 independent genes. They consisted of 3,699 annotated genes and 405 expressed sequence tags (ESTs). Some were the genes involved in constituting epidermal structures and others were unknown genes that are probably associated with keratinocytes. In particular, we were able to identify genes located at the chromosome 1q21, the locus for the epidermal differentiation complex, and 19q13.1, another probable locus for epidermal differentiation-related gene clusters. One EST located at the chromosome 19q13.1 showed increased expression by calcium treatment, suggesting a novel candidate gene relevant to keratinocyte differentiation. These results demonstrate the complexity of the transcriptional profile of keratinocytes, providing important clues on which to base further investigations of the molecular events underlying keratinocyte differentiation.</P>
