        Adenovirus-mediated heme oxygenase-1 gene transfer to neonatal porcine islet-like cluster cells: the effects on gene expression and protection from cell stress

        Porcine islet xenotransplantation is a promising strategy for the treatment of diabetes that overcomes donor shortages. However, islet xenografts are susceptible to oxidative stress and apoptosis. Heme oxygenase-1 (HO-1) has been shown to protect cells from oxidative stress, apoptosis and inflammation. Here, we investigated whether introduction of human HO-1 (hHO-1) into neonatal porcine islet-like cell clusters (NPCCs) can induce beneficial transcriptional changes in NPCCs against cellular stress. NPCCs were transduced with either adenovirus-HO-1 (Ad-HO-1) or control adenovirus-GFP (Ad-GFP). After treatment with hydrogen peroxide (H2O2) for 24 hours, nitrite oxide (NO) production assays were performed to detect oxidative stress. Microarray analysis was performed using a pig oligonucleotide 44 K gene chip. We profiled transcriptional changes to apoptosis, oxidant and inflammatory genes, and real-time PCR analysis was also performed to confirm the microarray results. Survival of NPCCs after treatment with H2O2 was significantly higher in the Ad-HO-1 group (p⁄0.001), and NO production also decreased in the Ad-HO-1 group (p⁄0.01). The microarray results showed that the expression of pro-apoptosis genes such as CASP3, CASP7, CASP10, CIDE-B and CIDE-C was significantly decreased in the Ad-HO-1 virus group (CASP10; p⁄0.05, CASP3, CIDE-C; p⁄0.01, CASP7, CIDE-B; p⁄0.001). We also found that the expression of oxidative stresses genes including COX1, COX2, CYB5A, SDHD and NOS2 was decreased, and that the anti-oxidant genes Gpx1 and SOD2 were increased in the Ad-HO-1 group (NOS2; p⁄0.05, COXI, COX2, CYB5A, SDHD, SOD2, GPX1; p⁄0.001). However, inflammatory gene expression was not significantly changed. Realtime PCR analysis confirmed the results of the microarray analysis. These results shed light on the underlying mechanisms of the protective effects of hHO-1 on porcine islets from cellular stresses and suggest that hHO-1 could be a promising target gene for the production of transgenic pigs that confer improved islet xenograft survival.

      • Induction of heme oxygenase-1 protects against podocyte apoptosis under diabetic conditions

        Heme oxygenase-1 (HO-1) is an anti-oxidant enzyme normally upregulated in response to oxidant injury. Here we determined the role of HO-1 in podocyte apoptosis in glomeruli of streptozotocin-treated rats and in immortalized mouse podocytes cultured in media containing normal or high glucose. HO-1 expression, its activity, the ratio of Bax/Bcl-2 protein, and active caspase-3 fragments were all significantly higher in isolated glomeruli of diabetic rats and in high glucose–treated podocytes. These increases were inhibited by zinc protoporphyrin treatment of the rats or by HO-1 siRNA treatment of the podocytes in culture. The number of apoptotic cells was also significantly increased in the glomeruli of diabetic rats and in high glucose–treated podocytes. Inhibition of HO-1 accentuated the increase in apoptotic cells both in vivo and in vitro. Our findings suggest that HO-1 expression protects against podocyte apoptosis under diabetic conditions.

        Language Change : From Syntactic Incorporation to Lexical Compounding

        Han, Ho. 1998. Language Change: From Syntactic Incorporation to Lexical Compounding. Linguistics, 6-2, 67-81. This study explores ongoing structural change with respect to light verb ha- constructions (henceforth, LVC) in Korean. The ha- LVC generates through the process of combining a Sino-Korean(SK) noun with the LV ha-. In the literature this process is considered to be involved with some sort of incorporation in Baker's (1988) sense. In negating an LV complex as V, only one type of negatives is judged perfect. However, it will be shown that there is age variance in terms of acceptability of the other type of negatives. With language acquisition data and historical linguistic theories, I will argue that the structure of the LV ha- complex appears to be currently under change. (Ajou University)

      • 정지궤도 위성과 지구국간 광통신 링크의 전송속도 해석

        정지궤도 위성과 지구국간 광통신을 대기상태와 앙각의 함수로써 해석할 수 있는 한모델(Han's model)을 제시하였다. 한모델에서는 대기상태를 맑은날, 구름낀 날, 안개, 헤이즈, 비, 눈의 여섯가지 전형적인 상태로 구분한다. 비트오류을 $10^{-7}$을 만족하는 데이터 전송속도를 한모델을 이용하여 상향링크와 하향링크에 대해 해석하였다. 상향링크시의 포인팅손실이 하향링크시의 대기에 의한 공간 가간섭 저하보다 크기 때문에 데이터 전송속도는 하향링크 보다 오히려 상향링크시에 제한됨을 알 수 있었다. Han's model, which is able to analyze optical communication between earth station and geo-satellite as a function of atmospheric conditions and elevation angles. is presented. In Han's model, atmospheric conditions are roughly classified into six basic types; clear sky, cloud, haze, fog, rain and snow. Data rate satisfying for the BER below $10^{-7}$ is analyzed by Han's model in case of up-link and down-link, respectively. Data rate is more limited by up-link than by down-link because the pointing loss caused by atmosphere on the up-link is greater than the spatial coherence degradation caused by atmosphere on the down-link.

        Can Markedness Take Over Subset Principle?

        Han, Ho. 1999. Can markedness Take Over Subset Principle? Linguistics 7-2, 117-127. This study explores whether or not markedness can account for the acquisition data that the subset principle provide explanation for. Goodluck (1991) stipulates that logically the subset principle can be reduced to the notion of markedness. This stipulation is logically quite right, but it overlooks some cases of incongruity among markedness, the subset principle, and children's hypothesis testing. For the former issue, I will consider null subject utterances in child English, while for the latter issue, children's errors will be taken into account, which appear to result from hypothesis testing. I will conclude that markedness and the subset principle need to be considered as separate principles, countering Goodluck's (1991) stipulation. (Ajou University)

        Research Trends in Young Children’s Theory of Mind using Keyword Network Analysis

        This study aims to investigate the trends in research and find implications for future research to understand the characteristics of young children’s theory of mind from 1997 to 2021. Out of the research papers published in journals in the field of early childhood education, 175 articles that fit the purpose of this study were analyzed using network analysis methods. The summary of the analysis results was as follows: First, in the study of young children’s theory of mind, “theory of mind”, “false belief”, “pretend play”, “emotion”, “executive function”, “social ability”, “story”, “young children”, “peer competence”, and “picturebook” showed high frequency. Second, it can be seen that the network structure for each period gradually changes from the first to the third periods. This demonstrated that while the research of theory of mind was somewhat slow but steadily active, the keywords identified in the early childhood theory of mind study evolved into a broad and rich array. Based on the results of this study, this study suggests diversification of the theory of mind research to perspectives of social cognition development of young children.

        독도·울릉도 ‘가지’(강치)에 대한 인식의 변화와 그 의미

        Gaji(可之), which is sea lion, was one of the specialties in Ulleungdo(Ulleung Island) and Dokdo and is now widely called ‘Gangchi’. Japan illegally occupied Dokdo in order to secure the exclusive fishing righls there. Thus the stludy about Gaji provides important clues for refuting Japan’s claim on Dokdo and also helps to prove thal Dokdo is Korean territory. This paper analyzes the change of views on ‘Gaji’ in Ulleungdo and Dokdo from lhe early Chosun Dynasty to 1905, when Japan seized Dokdo, and the implication of lhe change. The first official record of Gaji in the early Chosun Dynasty shows that Kim In-woo dedicated its leather in 1416. With the increasing interests on Ulleungdo and Dokdo. Inspectors were dispatched regularly to the islands. Inspector Jang Han-sang, who recognized the existence of Dokdo and witnessed Gaji, dedicated its leather when his returned. Thereafter it was regularized that the Inspectors who visited Ulleungdo dedicate Gaji leather as tribute. Gaji was recognized as a symbol of Ulleungdo’s typical souvenirs and this customary practice lasted until December 1894. when Government Patrol and Inspection System (sutoje) was abolished. Ulleungdo Inspector Yi Gyu-won left valuable records of Gaji in 1882 with mentions on its habitat, capture, use, and the fishing activities of the captors. With the accumulation of information about Gaji by Inspectlors. it was mentioned in several writings including Yi Ik’s Seonghosaseol, Sin Gyeong-jun’s Ganggyego and Dongguk-munheonbigo. Yeojigo, and Yi Gyu-gyeong’s Ojuyeonmunjangjeon-sango. Especially it attracts our attention thal Yi Gyu-gyeong recordro the sea lion as ‘Gaji’ referring to the writings of Sin Gyeong-jun. He also comparro and analyzed the origins of the names and the characleristics of sea lion, sea horse and other sea creatures in order to identify Gaji clearly. Thus, the current word Dokdo ‘Gangchi’ should be replaced with ‘Gaji’ or ‘Gaje’, a dialect in Ulleungdo. In this context, it is very significanl lhat Sin Gyeong-jun reconfirmed Woosan and Ulleungdo as wwo different islands belonging to Chosun in Ganggyego and Dongguk-munheonbigo. With regular dispatch of Inspectors, Woosan(Dokdo) was recognized as our territory and it was important to have precise knowledge on the geography, topography, and local products of Ulleungdo and Dokdo like Gaji. After lhe opening of ports, both the government and media shared the information that Gaji was a local product of Ulleungdo and Dokdo. Especially Hwangseong-sinmun made strong claims on Dokdo’s dominium as Korean territory, indicating Yu Deuk-gong’s poem which suggested Gaji and Imperial Edict No. 41 as the supporting evidence for il even after the Japanese seizure of the island. These records show that Gaji was widely recognized as the symbol of the evidence for Dokdo’s dominium as Korean territory.

        동아시아 도성의 공간구조에 있어서 문화적 차이에 관한 연구

        도시는 인간이 어떠한 목적을 위해 2차원의 지도 위에 의도적으로 그려나간 인공적인 공간이다. 그러나 그 의도와 목적이 그려진 2차원의 지도가 3차원 공간인 지형 위에 가시적으로 구체화되어야 하므로 도시는 건설되는 장소와 분리하여 생각할 수 없다. 한 장소는 다른 장소와 같을 수 없고, 장소가 가지고 있는 지리 체계와 경관 그리고 그것이 결합하여 만들어내는 고유한 감각, 즉 장소성은 다른 장소에 복제되거나 이식될 수 없다. 따라서 장소는 경험과 인식에 의해 결정되는 것으로 이는 곧 문화적인 것임을 의미하며 장소 선택에 관한 문제와 장소의 해석에 관한 문제 역시 문화적 배경과 연관된 고유한 특성에 의해 결정되는 것이라 할 수 있다. 동아시아에서 도성이 들어설 장소를 획득하기 위하여 크게 중심축을 설정하는 방법과 중심점을 설정하는 방법이 확인된다. 전자의 방법은 대지에 중 심축을 설정함으로서 장소성을 부여하며, 후자의 방법은 장소성을 가진 대지를 점유하기 위한 방법으로 사용되었다. 또한 전자는 주로 중국의 도성에서 후자는 주로 한국의 도성에서 확인된다. 그러나 중심축을 설정하는 방법에는 문화적 차이가 있어, 한(漢)문화 중심의 한계(漢系)왕조의 도성과 비교하여 비한계(非漢系) 왕조의 도성에서는 방형(方形)의 평면형태와 대칭구성의 기준선으로서 중심축을 설정하는 경향이 강하게 나타나고 있음을 알 수 있다. 도시는 과거의 어느 지점으로부터 연속되어 내려오는 공간으로 역사 속의 도시를 탐구하는 작업은 현재를 이해하는 일임과 동시에 수많은 변화 속에서 변하지 말아야 할 장소의 가치 체계를 판별해 내는 작업일 것이다. City is artificial space drawn intentionally for specific purpose by human on the two dimensional map. However the map has to be realized on topology, the three dimensional space, and city cannot be thought without place.<br/> One place cannot be same with another place. Sense of place; specific sense combined with geographical conditions and landscapes; can't be duplicated to the other place. Therefore place defined by experience and perception means cultural dimension and the matter in terms of choosing and analyzing a place is also defined by a specific characteristic based on cultural background.<br/> In Asian Capital city plans, it was researched that two methods to specify the center in a city; one was making a central axis to define placeness and the other one was pointing a center to occupy meaningful place. the former was mainly checked among Chinese ancient cities and the latter was checked among Korean ancient cities. However the ways to set a central axis had cultural differences; comparing with Chinese Han, in non-Chinese Han's cities, a central axis was drawn as a baseline to symmetric organization in square-shaped city plan.<br/> City that we live in is from the past regardless of the depth of time. To study ancient city structure has the meaning for discover the unchangeable value of places.

        Methanol extract of Myelophycus caespitosus ameliorates oxidative stress‑induced cytotoxicity in C2C12 murine myoblasts via activation of heme oxygenase-1

        Myelophycus caespitosus, a brown alga belonging to genus Myelophycus, has been traditionally used as a food and medicinal resource in Northeastern Asia. However, few studies have been conducted on its pharmacological activity. In this study, we evaluated whether methanol extract of M. caespitosus (MEMC) could protect against oxidative damage caused by hydrogen peroxide (H2O2) in C2C12 murine myoblasts. Our results revealed that MEMC could suppress H2O2-induced growth inhibition and DNA damage while blocking the production of reactive oxygen species. In H2O2-treated cells, cell cycle progression was halted at the G2/M phase, accompanied by changes in expression of key cell cycle regulators. However, these effects were attenuated by MEMC. In addition, we found that MEMC protected cells from induction of apoptosis associated with mitochondrial impairment caused by H2O2 treatment. Furthermore, MEMC enhanced the phosphorylation of nuclear factor-erythroid-2 related factor 2 (Nrf2) and expression and activity of heme oxygenase-1 (HO-1) in H2O2-treaetd C2C12 myoblasts. However, such anti-apoptotic and cytoprotective effects of MEMC were greatly abolished by HO-1 inhibitor, suggesting that MEMC could increase Nrf2-mediated activity of HO-1 to protect C2C12 myoblasts from oxidative stress.
