        PAHs 오염 유해 토양 부지의 생물학적 교정 : 동시 산화

        Bioremediation of PAHs(Polynuclear Aromatic Hydrocarbons) contaminated soils was evaluated using cooxidation process. Bioremediation potential for hazardous PAHs constituents was substantial in the presence of growth substrate phenanthrene. Mean half-life values of pyrene and benz(a)anthracene with phenanthrene substrate were 27.7% and 33.3%, respectively, of the half lives for each pyrene and benz(a)anthracene without phenanthrene. Soil enzyme activities were significantly enhanced by the addition of phenanthrene substrate through catalyzing oxidation of pyrene and benz(a)anthracene.

      • Hepatotoxicity를 유도하는 thioacetamide의 in vivo와 in vitro간에 발현 profiling 비교

        Microarray 기술은 단일 hybridization에서 매우 광대하게 유전자의 발현을 profiling할수 있다, 그리고 Microarray 기술은 독성과 관련하여 생물학적 systems을 위한 강력한 분자유전적 기술로서 사용되어지고 있다, 우리가 중점을 두고 있는 부분은 rat간과 cell line(WB-F344)에서 hepatotoxicity와 관련이 있는 유전자 발현의 결정을 위해서 독성학의 사용이다. Thioacetamide는 hepatotoxic를 사용하여 thioacetamide에 의해서 in vivo와 in vitro에서 특이적으로 발현되는 유전자를 profiling했다, 5~6주된 남성 rat(Sprague-Dawely VAF+albino)3마리와 WB-F344 cell line에 24시간 동안 thioacetamide(500㎎/㎏ and 5mM and 10mM)를 처리하였다, 전체 RNA는 thioacetamide가 처리되어진 rat의 간에서 추출하였고 cDNA microarray 기술을 사용하기 위해서 형광물질과 합성하였다, 직접 만든 4.8KcDNA microarrays는 유전자 발현을 profiling하는데 사용되어졌다. 게다가 in vitro에서는 in vivo에서 관찰되어진 유전자 발현과 유사한 성향을 가지고 있음을 보여주었다. Thioacetamide is a hepatotoxic compound that affects liver metabolism inhibits mRNA rtansport and induces immune suppression We carried out DNA microarray analyses to assess thioacetamide-specific expression profiles in vivo and in vitro. Male Sprague-Dawelt VAF+ albino 3 rats of 5-6weeks old and WB-F344 cell line were treated with a dose (500㎎/㎏ and 5mM and 10mM) of thioacetamide for 24h Total RNA isolated from Rat liver that treated thioacetamide was used to synthesis of fluorescence for cDNA microarray. 4.8K cDNA microarray in house has been used for gene expression profiling. Furthermore in vitro systems appeared similar in their ability to reproduce thioacetamide-induced changes in gene expression observed in vivo

        日本團塊世代의 服飾文化

        團塊世代とは、日本のベビ一ブ─マ─として多い人口數を背景に、これまで日本の社會で多樣な文化と社會的な樣相を作ってきた。特に音樂、ファッション、文化などの分野に與えた影響は大きい。そこで本硏究は、團塊世代の服飾に焦点を合わせ、その特性を分析した。硏究結果は、次のようである。團塊世代の大きな特徵は、マスマ一ケティングを支援し、マスコミや企業と共に消費を創り出し續いてきたという点である。そして今まで靑年文化(1960年代半ば-1970年代)、ニュ一ファミリ一(1970年後半-1980年代)、アクティブ·シニア─層(2007年の以後)を形成してきた。その過程の中で團塊世代は、①アイビ一ルックで服飾文化にTPOの槪念をもたらすと共にコダワリの感覺を共有した。②ジ一ンズ·カジュアルルックで、單品とアクセサリ一で自分の個性を表現し、自然體の感覺を共有した。③センスティブ·カジュアルルックで服飾にライフスタイルとセンスという意味を持ってきた。④今、團塊世代を指すアクティブ·シニア層とは、自分なりのコダワリと價値觀を持って、仕事と趣味に意欲のある健康なシニア─世代を意味する。服飾の特徵としては、靑年期から長年期までの樣タな經驗が蓄積されており、自然體、コダワリ、ライフスタイル、センス、エ一ジレスなどの感覺を共有しているのが分かった。 The Dankai Generation is the baby boomer generation in Japan born between 1947 and 1949. They have piloted many cultural and social changes, especially in the fields of music, fashion and culture. This study focuses on the costume culture of the Dankai generation and analyzes the flow and characteristics.The contents of the study are as follows. One characteristic of the Dankai generation is that they have continuously supported mass marketing and created consumption together with the mass media and corporations. Also, they have formed the Youth Culture from mid-1960’s to 1970’s (first wave), the New Family late 1970’s to 1980’s (second wave) and the Active Seniors after 2007 (third wave); and during the courses, they have shared, first, the concept of TPO and sensation of kakkoii and kodawari in ivy look, second, the sensation of ko and sizentai in jean casual look, and third, assigned meaning of lifestyle and sense in a sensitive casual look. The current designation, Active Seniors, refers to a healthy generation of seniors who are zealous in occupations and hobbies with their own kodawari and sense. In analyzing the characteristics of their costume, it was evident that various experiences had accumulated from their youth like jeans. In essence, they are sharing of sense, emphasis on private lifestyle, sense and ageless, sizentai .

      • 응급 내시경적 역행성 담췌관 조영술(Endoscopic Retrograde CholangioPancreatography)은 모든 담석 췌장염 환자에서 필요한가?

        목적:내시경적 역행성 담췌관 조영술(Endoscopic Retrograde Cholangiopancreatography, 이하 ERCP)과 내시경적 유두 괄약근 절개술(Endoscopic Sphincterotomy,이하 ES)은 급성 담석 췌장염 환자의 진단과 치료에 유용하다. 이 연구에서는 담석 췌장염 환자에서 응급ERCP의 유용성에 관하여 알아보고자 하였다. 방법:1994년 7월부터 2002년 12월까지 본원에서 급성 담석 췌장염을 진단받고 ERCP를 시행 받은 환자 66명을 대상으로 후향적인 연구를 하였다. 담석으로 인한 폐쇄성 황달은 응급 ERCP의 명백한 적응증이 되므로 이 연구에서 제외하였다. 대상 환자를 A군(응급 ERCP군 ; 내원 72시 간 내에 ERCP를 시행 받은 군)과 B군(정규 ERCP군 ;내원 72시간 후에 ERCP를 시행 받은 군)으로 나누고 두 군 사이의 임상양상과 부작용 발생 빈도를 비교하였다. 결 과:두 군에서 생화학적 검사 결과, 췌장염의 정도, 췌장염의 부작용, 췌장 유두부의 특성. 입원 기간 사이에는 유의한 차이가 없었다. A군이(40.9%) B군에(24.2%) 비해 ERCP에서 육안적으로 담석이 확인된 경우가 많았다. 결 론:ERCP가 급성 담석 췌장염의 진단과 치료에서 매우 유용하지만. 명백한 폐쇄성 황달이 없을 경우에는 응급 ERCP가 반드시 필요하지는 않다. Objectives : Endoscopic Retrograde Cholangiopancreatogrphy (ERCP) and endoscopic sphincterotomy are useful for both diagnosis and treatment of patients with acute gallstone pancreatitis. In this study, we assessed the feasibility of emergency ERCP in patients with all gallstone pancreatitis. Methods : We retrospectively reviewed the medical records of 66 patients, who underwent ERCP with a diagnosis of acute gallstone pancreatitis between July 1994 and December 2002. Obstructive jaundice from gallstones is excluded, because it is a distinct indication of emergency ERCP. Patients were divided into the group A (emergency ERCP group : ERCP was performed within 72 hours after hospitalization, mean 37.0±16.4 hours, range 6-70 hours) and group B (elective ERCP group ; ERCP, over 72 hours after hospitalization, mean 124.0±49.3 hours, range 76-288 hours). Comparisons of the clinical characteristics and incidence of complications were made between these two groups. Results : There was no significant difference for biochemical measurements, severity of pancreatitis, complications of pancreatitis, characteristics of ampulla, and length of hospital stay between the two groups, Group A had more cases (40.9%) with macroscopic stones on ERCP than group B (24.2%). Conclusion : Although ERCP was a very useful modality for the diagnosis and treatment of patients having acute gallstone pancreatitis, an emergency ERCP would not be necessary unless there is definite obstructive jaundice.

        Human Beings as Co-Creators with God:The Search for Workers’ Dignity in Capitalist Society

        This paper begins with a question raised by a Christian Asian migrant worker at a sushi restaurant in Toronto, “Can the God of the employer and the God of the employee be the same God?” The question contains his perplexity about the sharply divided reality between employers and employees in capitalist society, and about which side God is standing on. In order to engage in the worker’s experience theologically, I will explore the meaning of human beings as workers and human labor through a theological lens. To discuss this, I will critically analyze Laborem exercens (On Labour), the encyclical letter of Pope John Paul II and To Work and to Love by Dorothee Sölle. The authors of both documents are engaged in a theology of creation in order to understand the human being and the nature of work. Both also engage in a critical dialogue with Marxism in order to address the distortion of labor and the laboring human in capitalism, while maintaining their own understanding of the human and work. Through the dialogue with two authors’ works, I will maintain that human beings are created as working beings in the image of God the Creator and called to participate in co-creation with God by seeking solidarity in resisting a dominant ideology which deprives workers of their dignity.

        Adenovirus-mediated heme oxygenase-1 gene transfer to neonatal porcine islet-like cluster cells: the effects on gene expression and protection from cell stress

        Porcine islet xenotransplantation is a promising strategy for the treatment of diabetes that overcomes donor shortages. However, islet xenografts are susceptible to oxidative stress and apoptosis. Heme oxygenase-1 (HO-1) has been shown to protect cells from oxidative stress, apoptosis and inflammation. Here, we investigated whether introduction of human HO-1 (hHO-1) into neonatal porcine islet-like cell clusters (NPCCs) can induce beneficial transcriptional changes in NPCCs against cellular stress. NPCCs were transduced with either adenovirus-HO-1 (Ad-HO-1) or control adenovirus-GFP (Ad-GFP). After treatment with hydrogen peroxide (H2O2) for 24 hours, nitrite oxide (NO) production assays were performed to detect oxidative stress. Microarray analysis was performed using a pig oligonucleotide 44 K gene chip. We profiled transcriptional changes to apoptosis, oxidant and inflammatory genes, and real-time PCR analysis was also performed to confirm the microarray results. Survival of NPCCs after treatment with H2O2 was significantly higher in the Ad-HO-1 group (p⁄0.001), and NO production also decreased in the Ad-HO-1 group (p⁄0.01). The microarray results showed that the expression of pro-apoptosis genes such as CASP3, CASP7, CASP10, CIDE-B and CIDE-C was significantly decreased in the Ad-HO-1 virus group (CASP10; p⁄0.05, CASP3, CIDE-C; p⁄0.01, CASP7, CIDE-B; p⁄0.001). We also found that the expression of oxidative stresses genes including COX1, COX2, CYB5A, SDHD and NOS2 was decreased, and that the anti-oxidant genes Gpx1 and SOD2 were increased in the Ad-HO-1 group (NOS2; p⁄0.05, COXI, COX2, CYB5A, SDHD, SOD2, GPX1; p⁄0.001). However, inflammatory gene expression was not significantly changed. Realtime PCR analysis confirmed the results of the microarray analysis. These results shed light on the underlying mechanisms of the protective effects of hHO-1 on porcine islets from cellular stresses and suggest that hHO-1 could be a promising target gene for the production of transgenic pigs that confer improved islet xenograft survival.

Total RNA isolated from Rat liver that treated toxicants compounds was used to synthesis of fluroescene for cDNA microarray. 4.8K CDNA microarray in house has been used for gene expression profiling. We found that the level of gene expression related necrosis and Apotosis.
