Whole-genome resequencing analysis of 20 Micro-pigs

Background Miniature pigs have been increasingly used as mammalian model animals for biomedical research because of their similarity to human beings in terms of their metabolic features and proportional organ sizes. However, despite their importance, there is a severe lack of genome-wide studies on miniature pigs. Objective In this study, we performed whole-genome sequencing analysis of 20 Micro-pigs obtained from Medi Kinetics to elucidate their genomic characteristics. Results Approximately 595 gigabase pairs (Gb) of sequence reads were generated to be mapped to the swine reference genome assembly (Sus scrofa 10.2); on average, the sequence reads covered 99.15% of the reference genome at an average of 9.6-fold coverage. We detected a total of 19,518,548 SNPs, of which 8.7% were found to be novel. With further annotation of all of the SNPs, we retrieved 144,507 nonsynonymous SNPs (nsSNPs); of these, 5968 were found in all 20 individuals used in this study. SIFT prediction for these SNPs identified that 812 nsSNPs in 402 genes were deleterious. Among these 402 genes, we identified some genes that could potentially affect traits of interest in Micro-pigs, such as RHEB and FRAS1. Furthermore, we performed runs of homozygosity analysis to locate potential selection signatures in the genome, detecting several loci that might be involved in phenotypic characteristics in Micro-pigs, such as MSTN, GDF5, and GDF11. Conclusion In this study, we identified numerous nsSNPs that could be used as candidate genetic markers with involvement in traits of interest. Furthermore, we detected putative selection footprints that might be associated with recent selection applied to miniature pigs.

Backgrounds: Steatosis can also be a co-factor in many chronic liver diseases that can lead to fibrosis and cirrhosis. Although an important parameter, until now, evaluation of steatosis by non-invasive methods has remained challenging. Recently, controlled attenuation parameter (CAP) being implemented on FibroScan(®) (Echosens, Paris, France), both steatosis and fibrosis can be evaluated simultaneously, is reported to be efficient to detect even low grade steatosis noninvasively. Backgrounds and Aim: We analyzed the CAP value in health checkup subjects and the correlation between CAP value and other clinical parameters. Methods: CAP was retrospectively collected with other data including demographics, blood test results, and finding of abdominal ultrasonography. Steatosis grade was decided by cutoffs of CAP according to a previous report (Liver Int. 2012;32:911-8): 215 dB/m for S ≥ 1, 252 dB/m for S ≥ 2, and 296 dB/m for S3. Results: Total 179 subjects (M:F=95:84) were included. According to CAP value, S0:S1:S2:S3 patients were 49:52:51:27. Mean CAP values were 188.8 dB/m for S0, 233.4 dB/m for S1, 269.4 dB/m for S2, and 323.3 dB/m for S3. Steatosis grade according to CAP was significantly correlated with body weight (r=0.455, P<0.001), BMI (r=0.484, P<0.001), waist to hip ratio (r=0.409, P<0.001), systolic BP (r=0.273, P<0.001), diastolic BP (r=0.292, P<0.001), fasting glucose (r=0.271, P<0.001), triglycerides level (r=0.301, P<0.001), HDL cholesterol (r=- 0.241, P=0.001), LDL cholesterol (r=0.206, P=0.006), total cholesterol (r=0.201, P=0.007), and ultrasonographic fatty liver grade (r=0.505, P<0.001), but not with GGT, ALT and AST levels. However, ultrasonographic fatty liver grade was significantly correlated with GGT, ALT and AST as well as factors correlated with steatosis grade by CAP. Conclusion: In health check-up subjects, CAP sensitively detects steatosis by non-invasive measure. Histologic validation is warranted to use cut-offs of CAP for steatosis grade in nonchronic live disease subjects.

This paper has the objective to suggest specific specialized learning activities in Korean language education necessary for children of multi-cultural families. Korean language education for children with multi-cultural backgrounds should consider BICS(Basic Interpersonal Communicative Skill) and CALP(Cognitive Academic Language Proficiency) because Korean language education is conducted in the public education system. This paper focuses on the essence of an actual classroom the “learning activities.” In this paper, We first discuss the special characteristics of children and characteristics of KSL education. Taking this as a basis, this research is structured as “Teaching-Understanding-Application-Everyday life”, and the effective Korean language learning activities on each step are suggested. This paper presents a case that takes learning activities that are effective in understanding the content learned as a foundation and focuses on learning activities that can be applied to everyday and school life.

저자들은 약물치료 또는 케톤식이요법으로 조절되지 않는 난치성 간질을 가진 결절성 경화증 환아에서 간질 수술 후 발작이 없어지고 인지기능과 행동 발달을 보인 3례를 경험하였기에 문헌 고찰과 함께 보고하는 바이다. Tuberous sclerosis(TS) is a disorder of aberrant neuronal differentiation and prolifera-tion manifesting as multiple central nervous system(CNS) hamartomas. Seizures in pa-tients with TS are often intractable to anticonvulsant therapy. Although TS shows vari-ous lesions including multiple tubers, many patients with well localized epileptic focus to some tubers may have a benefit from epileptic surgery. We experienced three cases who had clinical improvement after epileptic surgery in intractable epilepsy complicated by TS. We report three cases and the brief review of literatures.
폴리(글리시딜 메타크릴레이트)가 그래프트된 다중벽 탄소나노튜브에 다양한 아민 그룹의 도입과 바이오센서 지지체로서의 응용

다양한 아민 그룹으로 개질된 다중벽 탄소나노튜브(이하 MWNT) 지지체를 기반으로 하여 페놀 화합물을 검 출하기 위한 티로시나아제가 고정된 바이오센서를 개발하였다. 방사선 중합법을 이용하여 MWNT에 글리시딜메타 크릴레이트를 중합한 후 중합 사슬의 아미노화 반응을 통해 MWNT에 다양한 아민 그룹을 도입시켰다. 이렇게 제조된 물질의 물리적, 화학적 특성은 SEM, XPS 그리고 TGA에 의해 평가되었다. 그리고 제조된 물질을 기반으로 제작된 티로시나아제가 고정화된 바이오센서의 전기화학적 특성도 평가하였다. 본 효소 바이오센서는 0.1-0.9mM의 페놀을 검출할 수 있다. 결합효과, pH, 온도 그리고 다양한 페놀화합물에 대한 반응과 같은 여러 가지 변수에 대하여도 최적화하였고 상용 레드와인에서의 페놀화합물 검출도 연구하였다. A tyrosinase-immobilized biosensor was developed based on various amine-modified multi-walled carbon nanotube (MWNT) supports for the detection of phenolic compounds. MWNTs with various amine groups were prepared by radiation-induced graft polymerization of glycidyl methacrylate (GMA) onto MWNT supports and the subsequent amination of poly (GMA) graft chains. The physical and chemical properties of the poly (GMA)-grafted MWNT supports and the aminated MWNT supports were investigated by SEM, XPS, and TGA. Furthermore, the electrochemical properties of the prepared tyrosinase-modified biosensor based on MWNT supports with amine groups were also investigated. The response of the enzymatic biosensor was in the range of 0.1-0.9mM for the concentration of phenol in a phosphate buffer solution. Various parameters influencing biosensor performance have been optimized: binder effects, pH, temperature, and the response to various phenolic compounds. The biosensor was tested on phenolic compounds contained in two different commercial red wines.
유연체를 고려한 블록 리프팅 동역학 해석 및 사용자 인터페이스 개발

본 연구에서는 블록 리프팅 작업시 사용되는 장비의 형상 모델링 작업을 CATIA 프로그램을 이용하여 수행한 후, 5 대의 골리앗 크레인, 8 대의 이퀄라이저(equalizer), 2 대의 리프팅 빔(lifting beam)을 상용 동역학 해석 시스템인 ADAMS 에 적용하였다. 골리앗 크레인 구현시 상부 트롤리(trolley)와 크레인의 체결부분은 병진 관절(slide joint)을 적용하여 실제와 동일한 구동이 가능하게 구현하였고, 와이어(wire)도 실제와 동일한 탄성효과를 나타내도록 하였으며, 이퀄라이저는 하중 균일화 과정을 물리적으로 재해석하여 구현함으로써 계산 시간을 대폭 단축하였다. 모델링 작업 이외에도 라이브러리에서 필요한 장비를 선택하여 적용하는 기능, 다양한 유연체 블록을 선택하여 적용하는 기능, 러그(lug) 위치 ID 입력만으로 러그와 와이어를 체결 또는 해제하는 기능, 러그 체결 위치 및 초기 와이어의 길이를 파악하여 적정 위치에 블록 로더(block loader)를 이동시키는 기능, 시나리오를 이용한 시뮬레이션을 제어 기능 등이 포함된 사용자 인터페이스를 개발하였다. 시나리오 제어 기능의 경우 블록을 들어올리거나 내리는 기능, 블록 거동의 안정화를 위하여 대기시간을 주는 기능, 트롤리를 움직이는 기능, 시뮬레이션 중 러그를 체결하거나 해제하는 기능이 적용되었다. 와이어, 이퀄라이저, 시나리오 구현 시 C++ 프로그래밍 언어를 이용하여 자체적으로 서브루틴으로 개발하였다. 또한 ADAMS 를 이용한 해석을 통해 블록 리프팅 작업 중 발생하는 와이어의 장력, 블록에 작용하는 응력, 변형량, 최대 응력 발생 부위 등을 구하고 이를 정적 해석결과와 비교 분석하여 해석 결과의 타당성을 검증하였다.
