        Effects of four Bacillus spp. of soil origin on the Colorado potato beetle Leptinotarsa decemlineata (Say)

        Four species of Bacillus were isolated from soil in an effort to find safe, effective and alternative biological control agents against plant pests. These bacteria were identified as Bacillus pumilus, Bacillus sphaericus,Bacillus megaterium and Bacillus cereus on the basis of fatty acid methyl ester analysis and carbon utilization profiles by using Microbial Identification and Biolog Microplate Systems. Laboratory experiments carried out to determine the insecticidal activities of these isolates showed that B. pumilus caused 95.7 and 26.7% mortality and B. sphaericus caused 74.5 and 23.3% mortality of Leptinotarsa decemlineata larvae and adults, respectively. B. cereus and B. megaterium showed 51.1 and 29.7%, respectively, of L. decemlineatalarvae. This study presents at least two Turkish isolates from the genus Bacillus showing high insecticidal activity against L. decemlineata.

        Synthesis of Thiadiazole Derivatives Bearing Hydrazone Moieties and Evaluation of Their Pharmacological Effects on Anxiety, Depression, and Nociception Parameters in Mice

        Novel thiadiazole derivatives bearing hydrazone moieties were synthesized through the reaction of 2-[(5-methyl-1,3,4-thiadiazol-2-yl)thio)]acetohydrazide with aldehydes/ketones. The chemical structures of the compounds were elucidated by 1H-NMR, 13C-NMR, MS-FAB spectral data, and elemental analyses. Behavioral effects of the test compounds in mice were examined by hole-board, activity cage, tail suspension and modified forced swimming tests (MFST). Antinociceptive activities were evaluated using the hot-plate and tail-clip methods. Results of the experiments indicated that the test compounds did not significantly change the exploratory behaviors or locomotor activities of animals in the hole-board and activity cage tests, respectively. Administration of the reference drug fluoxetine (10 mg/kg) and compounds 3a, 3b, 3c, 3j, 3k, and 3l significantly shortened the immobility times of animals in the tail suspension and MFST tests, indicating the antidepressant-like effects of these derivatives. Morphine (10 mg/kg) and compounds 3a, 3b, 3c, 3d, 3e, 3j, 3k, and 3l increased the reaction times of mice in both the hot-plate and tail-clip tests, indicating the antinociceptive effects of these compounds. To the best of our knowledge, this is the first study of central nervous system activities of chemical compounds carrying thiadiazole and hydrazone moieties together on their structures.

        Synthesis of new 1-phenyl-2-(4-substituted-piperazin-1-yl)- propanol derivatives and evaluation of their antidepressant-like effects

        In this study, we synthesized eight novel1-phenyl-2-(4-substituted-piperazin-1-yl)-propanol derivativesand evaluated their antidepressant-like activities. Thechemical structures of the synthesised compounds wereelucidated by spectroscopy and elemental analyses. Potential antidepressant-like effects of the test compounds(20 mg kg-1) were investigated using the tail-suspensiontest and modified forced swimming test (MFST) in mice. Additionally, the spontaneous locomotor activity of themice was assessed using the activity cage apparatus. Boththe reference drug fluoxetine (20 mg kg-1) and the testcompounds 3a–3e and 3g significantly shortened theimmobility time of the mice in both the behavioural tests. These test compounds also increased the swimming time inMFST without any change in the climbing duration. Compounds 3c–3e and 3g were significantly more potent ininducing these effects than 3a and 3b. None of the compoundschanged the locomotor activities of the animals,thus antidepressant-like effects of test compounds werespecific. The findings support those of previous studies thatreported antidepressant-like activities of aryl alkanolpiperazine derivatives.

        An Analysis of the Potential Relationship between Crowe Type and Lower Extremity Morphology in Patients with Developmental Dysplasia of the Hip

        Purpose: To test whether Crowe type is related to femoral alignment and leg length discrepancy by evaluating the preoperative lengths and coronal alignment of femurs, pelvic parameters and hip morphology of patients who underwent primary hip arthroplasty due to coxarthrosis secondary to developmental dysplasia of the hip (DDH). Materials and Methods: Medical records of patients with coxarthrosis secondary to DDH who were treated with total hip arthroplasty at Department of Orthopaedics and Traumatology, Istanbul University Faculty of Medicine between 2008 and 2017 were reviewed. The mechanical axis of lower limbs was analyzed; pelvic height and femoral and tibial lengths were measured. All femurs were classified according to the Dorr classification. Results: A total of 97 patients were eligible for analysis and were diagnosed with unilateral DDH (n=51) or bilateral DDH (n=46). In those diagnosed with unilateral DDH, the affected pelvis, femur, and tibia were often shorter than the unaffected side. In those diagnosed with bilateral DDH, femoral and pelvic lengths were unpredictable. In the femoral coronal alignment test, data varied widely but were within normal limits. The difference in the Dorr types of femurs was significant between dysplastic and normal sides of patients with unilateral DDH (P=0.001) but not those with bilateral DDH. Conclusion: Especially in patients with unilateral DDH, pelvic heights and femoral and tibial lengths on the affected side may be shorter compared with unaffected side regardless of the Crowe type. Femoral coronal alignment is unpredictable for both groups. Careful preoperative analyses of femoral coronal alignment and pelvic length are advised.

        Differential antioxidative and biochemical responses to aluminium stress in Brassica juncea cultivars

        Aluminium (Al) toxicity in acidic soils limits crop production worldwide. We evaluated eleven genotypes of Brassica juncea (Mustard) under Al stress on basis of their growth and shortlisted two best among them for further comparative analysis. Our objective was to elucidate individual and differential oxidative damage and defence response elicited by Al treatment in selected mustard genotypes, ‘Pusa Tarak’ and ‘Pusa Vijay’. Thirty-day-old plants of both genotypes were subjected to Al stress for a period of 24 h and 72 h. Concentration of superoxides was visible much higher in leaves of ‘Pusa Vijay’ both at 24 h and 72 h, also confirmed by oxidative stress marker thiobarbituric acid reactive substances (TBARS). The activities of the enzymatic antioxidants superoxide dismutase (SOD), ascorbate peroxidase (APX), glutathione reductase (GR), glutathione S-transferase (GST), and catalase (CAT) were significantly higher in ‘Pusa Tarak’ compared to control and ‘Pusa Vijay’ at both time stages. Levels of non-enzymatic antioxidants glutathiones and ascorbates were already higher in ‘Pusa Vijay’; however, Al treatment increased their levels in both genotypes with non-significant changes on inter-genotypes basis. More and significant decline by Al in chlorophylls was observed in ‘Pusa Vijay’. Interestingly, increase in proline content by Al was much prominent in ‘Pusa Tarak’ compared to ‘Pusa Vijay’. The in vitro antioxidant capacity estimation of mustard genotypes, evaluated by measuring 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and hydroxyl radical scavenging (HRSA) activities proved that extract of ‘Pusa Tarak’ can detoxify more radicals than ‘Pusa Vijay’. We conclude that ‘Pusa Tarak’ can upregulate cellular antioxidants and osmoregulation, and quench more radicals compared to ‘Pusa Vijay’. The results will contribute for selection of better Brassica genus to be grown in Al rich acidic soils, and help in designing strategies for crop breeding and cultivation.

        3D finite element modelling of drilling process of Al7075-T6 alloy and experimental validation

        In this study, the performances of twist and 3-flute drills in the drilling process of Al7075-T6 aluminium alloy were investigated experimentally and numerically. In the experimental phase, the drilling processes were carried out with the twist and 3-flute drills at three different feed rates (0.05, 0.1, 0.2 mm/rev) and four different cutting speeds (60, 90, 120, 150 m/min). The thrust forces occurring during the drilling processes were measured with a Kistler 9257b dynamometer. In the numerical phase, a 3D finite element model of the drilling process was carried out with Deform 3D V6.1 software. During the finite element analyses of the drilling processes, the thrust forces were numerically obtained and compared with the experimental thrust forces. Also, the torque and tool stress occurring in the drilling process were determined numerically. At the end of the study, the results showed that there is a good agreement between the experimental and numerical results. Additionally, it is shown that the thrust force, torque and tool stress obtained with the twist drill were less than those of the 3-flute drill.

        The Early Histological Effects of Intravesical Instillation of Platelet- Rich Plasma in Cystitis Models

        Purpose: To evaluate the early histological effects of the intravesical instillation of platelet-rich plasma (PRP) in rabbit models of interstitial and hemorrhagic cystitis. Methods: Thirty-six rabbits were classified into 6 groups: saline (S), S+PRP, hydrochloric acid (HCl), HCl+PRP, cyclophosphamide (CyP), and CyP+PRP. At 48 hours after induction, PRP was prepared and intravesically administered to the S+PRP, HCl+PRP, and CyP+PRP groups. Bladder sections were stained with toluidine blue for mast cell counting and with hematoxylin and eosin for histopathology and mitotic index determination. The proliferation index was determined by proliferating cell nuclear antigen (PCNA) immunolabeling. The nonparametric Mann-Whitney U-test was used for statistical analysis. Results: No abnormalities were observed in the S group, whereas increased interstitial edema and increased average mitotic and proliferation indices were observed in the S+PRP group (P=0.023, P=0.004, and P=0.009, respectively). Intense epithelial loss, hemorrhage, and leukocyte infiltration were detected in the HCl and HCl+PRP groups, whereas a significantly increased average mitotic index was observed in the HCl+PRP group (P=0.002). When compared with its CyP counterpart, a significant reduction in hemorrhage and an increase in leukocyte infiltration and mitotic index were observed in the CyP+PRP group (P=0.006, P=0.038, and P=0.002, respectively). In addition, PCNA staining revealed a significantly increased proliferation index in the HCl+PRP and CyP+PRP groups (P=0.032 and P=0.015, respectively). Conclusions: The intravesical instillation of PRP increased the mitotic index in the saline and cyclophosphamide groups while decreasing macroscopic bleeding.

        Determining the Optimum Process Parameters of Selective Laser Melting via Particle Swarm Optimization Based on the Response Surface Method

        Manufacturing high-quality and desired products from additive manufacturing necessitate careful adjustment of the processparameters. Various methods can be utilised to determine optimum process parameters, such as the Taguchi method, Designof Experiments (DoE). Rather than evaluating limited information obtained from statistical analysis of the experiments, optimisationmethods can help find the best possible combination for the process parameters. Therefore, an optimisation approachbased on Particle Swarm Optimization (PSO) was utilised to find the optimum process parameters. The most importantprocess parameters of Selective Laser Melting (SLM) such as laser power, layer thickness, scan speed, and build orientationwere selected as input parameters, and their effects on the tensile properties of the manufactured part were investigated to findout the optimal operating conditions for the SLM process. Since there is not any explicit mathematical expression relatingthese process parameters to the tensile strength, the Response Surface Method (RSM) was used to obtain a meta-model sothat it can be used as an objective function in the optimisation formulation. This approach enabled us to predict the optimumprocess parameters to maximise the tensile strength without conducting an excessive number of experiments. Moreover, themathematical model can also predict tensile strength corresponding to the parameter values that are not tested according tothe DoE chosen for such studies. Furthermore, it was also shown that the PSO outperforms the Genetic Algorithm (GA),which is widely employed to find out the optimum process parameters, in terms of less number of iteration.

        Relationships of the Vitamin D and Platelet Indices in Sjögren’s Syndrome

        Primer Sjögren’s Syndrome (pSS) is an autoimmune/inflammatory illness. The platelet indices (PIs) indicate the inflammatory response and activity/severity of many diseases. A vitamin D deficiency is accompanied by the increased tendency of autoimmune diseases. This study investigated whether the vitamin D levels are related to the altered platelet indices in pSS. A total of 261 individuals were included in this analytical cross-sectional study. The laboratory data of pSS patients were evaluated and the relationship between the PIs and vitamin D status was examined. According to these findings, in patients with pSS, the vitamin D levels were lower than the healthy control group (P<0.05). The vitamin D levels were negatively associated with PDW (P=0.012), but positively correlated with PCT (P<0.001). The cut-off point was obtained with receiver operating characteristics (ROC) curves for PDW: 12.53 (AUC 0.921, sensitivity 90%, specificity 85%), for PCT; 0.29 (AUC 0.660, sensitivity 68%, specificity 55%). In multivariate linear regression analysis, the most significant parameters for the effects of PDW are the following: vitamin D (=−0.373; t=−2.626; sig.=0.013) and plateletcrit (=−0.308; t=−2.13; sig.=0.040). A vitamin D deficiency may be accompanied by changes in PIs in pSS. A higher PDW and lower PCT supports the underlying inflammation, which may be vitamin D related useful parameters to consider in approaching to pSS.

        Anti-fibrotic effect of tocotrienols for bladder dysfunction due to partial bladder outlet obstruction

        Purpose: To investigate potential beneficial effects of tocotrienols which have been suggested to inhibit hypoxia-inducible factor (HIF) pathway, on partial bladder outlet obstruction (PBOO)-induced bladder pathology. Materials and Methods: PBOO was surgically created in juvenile male mice. Sham-operated mice were used as controls. Animals received daily oral administration of either tocotrienols (T3) or soybean oil (SBO, vehicle) from day 0 to 13 post-surgery. Bladder function was examined in vivo by void spot assay. At 2 weeks post-surgery, the bladders were subjected to physiological evaluation of detrusor contractility in vitro using bladder strips, histology by H&E staining and collagen imaging, and gene expression analyses by quantitative PCR. Results: A significant increase in the number of small voids was observed after 1 week of PBOO compared to the control groups. At 2 weeks post-surgery, PBOO+SBO mice showed a further increase in the number of small voids, which was not observed in PBOO+T3 group. PBOO-induced decrease in detrusor contractility was similar between two treatments. PBOO induced bladder hypertrophy to the same degree in both SBO and T3 treatment groups, however, fibrosis in the bladder was significantly less prominent in the T3 group than the SBO group following PBOO (1.8- vs. 3.0-fold increase in collagen content compared to the control). Enhanced levels of HIF target genes in the bladders were observed in PBOO+SBO group, but not in PBOO+T3 group compared to the control. Conclusions: Oral tocotrienol treatment reduced the progression of urinary frequency and bladder fibrosis by suppressing HIF pathways triggered by PBOO.
