        Genetic Diversity in the Coat Protein Genes of Prune dwarf virus Isolates from Sweet Cherry Growing in Turkey

        Sweet cherry is an important fruit crop with increasing economical value in Turkey and the world. A number of viruses cause diseases and economical losses in sweet cherry. Prune dwarf virus (PDV), is one of the most common viruses of stone fruits including sweet cherry in the world. In this study, PDV was detected from 316 of 521 sweet cherry samples collected from 142 orchards in 10 districts of Isparta province of Turkey by double antibody sandwich-enzyme linked immunosorbent assay (DAS-ELISA). The presence of PDV in ELISA positive samples was confirmed in 37 isolates by reverse transcription- polymerase chain reaction (RT-PCR) method. A genomic region of 862 bp containing the coat protein (CP) gene of PDV was re-amplified from 21 selected isolates by RT-PCR. Amplified DNA fragments of these isolates were purified and sequenced for molecular characterization and determining genetic diversity of PDV. Sequence comparisons showed 84-99% to 81-100% sequence identity at nucleotide and amino acid level, respectively, of the CP genes of PDV isolates from Isparta and other parts of the world. Phylogenetic analyses of the CP genes of PDV isolates from different geographical origins and diverse hosts revealed that PDV isolates formed different phylogenetic groups. While isolates were not grouped solely based on their geographical origins or hosts, some association between phylogenetic groups and geographical origins or hosts were observed.

        Gallstone Disease Does Not Predict Liver Histology in Nonalcoholic Fatty Liver Disease

        Background/Aims: We sought to examine whether the presence of gallstone disease (GD) in patients with biopsy-proven nonalcoholic fatty liver disease (NAFLD) is associated with liver fibrosis and histological nonalcoholic steatohepatitis (NASH) score. Methods: We included 441 Turkish patients with biopsy-proven NAFLD. GD was diagnosed in the presence of sonographic evidence of gallstones, echogenic material within the gallbladder with constant shadowing and little or no visualization of the gallbladder or absence of gallbladder at ultrasonography, coupled with a history of cholecystectomy. Results: Fifty-four patients (12.2%) had GD (GD+ subjects). Compared with the GD- subjects, GD+ patients were older, had a higher body mass index and were more likely to be female and have metabolic syndrome. However, GD+ patients did not have a higher risk of advanced fibrosis or definite NASH on histology. After adjustment for potential confounding variables, the prevalence of GD in NAFLD patients was not associated with significant fibrosis (≥2) (odds ratio [OR], 1.06; 95% confidence interval [CI], 0.53 to 2.21; p=0.68) or definite NASH (OR, 1.03; 95% CI, 0.495 to 2.12; p=0.84). Conclusions: The presence of GD is not independently associated with advanced fibrosis and definite NASH in adult Turkish patients with biopsy-proven NAFLD.

        The Antioxidant Potency of Punica granatum L. Fruit Peel Reduces Cell Proliferation and Induces Apoptosis on Breast Cancer

        Pomegranate (Punica granatum L.) is known to possess pharmacological activities, such as antioxidant and anticancer. In this study, we evaluated the antioxidant potency of a methanolic pomegranate fruit peel extract (PPE) and the relation with its antiproliferative and apoptotic effects on MCF-7 human breast cancer cells. Total phenolic content and antioxidant activity of PPE were determined using the Folin–Ciocalteau and the 2,2-diphenyl-l-picrylhydrazyl free radical methods, respectively. Phenolic acids present in the extract were characterized by a reverse-phase high-performance liquid chromatography (HPLC) method. Cell proliferation was assessed by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide reduction assay. The apoptotic effects were determined by in situ Tdt-mediated dUTP nick end-labeling assay, and Bax/Bcl-2 mRNA expression levels were measured by reverse transcription–polymerase chain reaction. The extraction yield as a percentage of plant material was 37.97% (wt/wt), and total phenolic content was 331.28 mg of gallic acid equivalents/g of extract. According to HPLC analysis, the most abundant phenolic acid detected in the extract was ellagic acid. MCF-7 cell proliferation decreased depending on PPE concentration (25, 50, 100, 200, and 300 ㎍/mL) and incubation times (24, 48, and 72 hours). After 48 and 72 hours, the apoptotic cell numbers were significantly increased at 100, 200, and 300 ㎍/mL PPE concentrations. In addition, expression of the pro-apoptotic gene Bax was increased, and that of the anti-apoptotic gene Bcl-2was decreased after 200 and 300 ㎍/mL PPE treatment for 48 and 72 hours. Because PPE reduced cell proliferation and induced apoptosis on MCF-7 cancer cells, we believe that PPE has important antioxidant and apoptotic effects.

        Objective: Brucellosis is a characterized zoonotic disease by multisystem involvement. Although musculoskeletal involvement is the most frequent case, the pulmonary system is rarely involved. Nodular involment is one of the rarest amoung pulmonary involvements. This study presents a 21- year-old-male with pulmonary involvement without any musculoskeletal system involvement and he was received a diagnosis in the early days of hospitalization. Case: The patient had complaints of one -month-long cough and sputum production and he informed that he used different antibiotics. After the last center he went detected hypogonadism in the patient, he was referred to our hospital`s endocrinology clinic. Physical examination showed fever:37.2,nb:112 beats/min and fi ndings in accordance with the hypogonadism and also late inspiratory crackles had been heard by oscultation. The captured chest radiograph indicated suspicious nodular appearance and suspected infi ltrated area. The laboratory tests erythrocyte sedimentation rate:41mm/saat,C-Reactive Protein:14.1,WBC : 4.08 x103/ul. The patient stated that he was working as a village shephard 1 month ago who did not suffer from signifi - cant weight loss nor fever. Detailed medical history stated that the last examination indicated brucella rosebengal positive, and the antiglobulin (coombs) test in brucellosis 1/160 was also positive. Altough the sputum culture and tuberculosis acid fast stain were negative, the patient had a positive blood culture regarding brucella melitensis. The thorax CT image showed nodules in the lungs with the ground glass opacity and infi ltrative image has been detected in subpleural area. There was nothing in bones scintigraphy. The patient was treated rifampicin 1x600 mg and doxycycline 2x100 mg for 6 weeks. Along with the treatment the patient`s complaints disappeared, and post-treatment blood cultures was negative. Control thorax CT reveals disappearanceof lesions in the lung area. Conclusion: Anamnesis and disease suspicion are the most important things regarding diagnosis of Brucellosis.

        Conventional Galactography and MR Contrast Galactography for Diagnosing Nipple Discharge: Preliminary Results

        Objective: We wanted to compare the clinical usefulness of conventional galactography and MR contrast galactography for diagnosing patients with nipple discharge. Materials and Methods: Both conventional galactography and MR contrast galactography were performed prospectively in 16 patients. Gadopentate dimeglumine (0.1 ml) was mixed with non-ionic contrast medium (0.9 ml) to obtain a resultant volume of 1 ml and this was used for both examinations. Following conventional galactography, MR contrast galactography was performed after direct injection of contrast media into the duct. Results: Conventional galactography and MR contrast galactography were concordant in 13 (81%) of 16 patients; the results were normal in five, ductal dilatation was noted in four and intraductal filling defects were noted in four. The remaining three (19%) patients demonstrated discordant findings on the two examinations. While conventional galactography revealed filling defects, the MR contrast galactography results were normal in two patients. The third patient had kinks-stricture on conventional galactography and MR contrast galactography showed ductal dilatation. This suggested there were false positive results for the three patients’ conventional galactography, and all the three patients with discordant results underwent surgery and the histopathologic evaluation showed fibrocystic changes. Conclusion: MR contrast galactography may be used as an alternative imaging modality for making the diagnosis of pathologic nipple discharge. However, statistically supported studies with large pools of subjects for comparing the galactography and MR contrast galactography results are needed to confirm our findings.         Synthesis of new 1-phenyl-2-(4-substituted-piperazin-1-yl)- propanol derivatives and evaluation of their antidepressant-like effects

        In this study, we synthesized eight novel1-phenyl-2-(4-substituted-piperazin-1-yl)-propanol derivativesand evaluated their antidepressant-like activities. Thechemical structures of the synthesised compounds wereelucidated by spectroscopy and elemental analyses. Potential antidepressant-like effects of the test compounds(20 mg kg-1) were investigated using the tail-suspensiontest and modified forced swimming test (MFST) in mice. Additionally, the spontaneous locomotor activity of themice was assessed using the activity cage apparatus. Boththe reference drug fluoxetine (20 mg kg-1) and the testcompounds 3a–3e and 3g significantly shortened theimmobility time of the mice in both the behavioural tests. These test compounds also increased the swimming time inMFST without any change in the climbing duration. Compounds 3c–3e and 3g were significantly more potent ininducing these effects than 3a and 3b. None of the compoundschanged the locomotor activities of the animals,thus antidepressant-like effects of test compounds werespecific. The findings support those of previous studies thatreported antidepressant-like activities of aryl alkanolpiperazine derivatives.

        원격 의료 진단 시스템을 위한 사용자 기반 적응 대역폭 비디오 시스템

        Adaptive bitrate (ABR) streaming technology has become an important and prevalent feature in many multimedia delivery systems, with content providers such as Netflix and Amazon using ABR streaming to increase bandwidth efficiency and provide the maximum user experience when channel conditions are not ideal. Where such systems could see improvement is in the delivery of live video with a closed loop cognitive control of video encoding. In this paper, we present streaming camera system which provides spatially and temporally adaptive video streams, learning the user's preferences in order to make intelligent scaling decisions. The system employs a hardware based H.264/AVC encoder for video compression. The encoding parameters can be configured by the user or by the cognitive system on behalf of the user when the bandwidth changes. A cognitive video client developed in this study learns the user's preferences (i.e. video size over frame rate) over time and intelligently adapts encoding parameters when the channel conditions change. It has been demonstrated that the cognitive decision system developed has the ability to control video bandwidth by altering the spatial and temporal resolution, as well as the ability to make scaling decisions
