      • 톨루엔 폭로 작업자에서 Aldehyde dehydrogenase2(ALDH2)의 유전자형과 요중 마뇨산

        This study was conducted to evaluate the distribution and the effect to urinary hippuric acid excretion according to the polymorphism of aldehyde dehydrogenase2(ALDH2) which is major role in oxidaton of benzaldehyde to benzoic acid. For 39 workers exposed to toluene and 42 white collar workers, we extracted the DNA from venouse blood and confirmed the genotype of aldehyde dehydrogenase2 using the polymerase chain reaction(PCR) and artificial created restriction site method. In workers exposed to toluene, normal (NN) type were 34 (88%) and inactive(ND) type were (12%), but in white collar workers NN type were 27(65%) and ND type were 14(33%), DD type were 1(2%). It showed the significant lower rate of inactive type of ALDH2 in workers exposed to toluene than white collar workers. The results of workplace toluene measurement were below 20ppm. For urinary hippuric acid there was not significant differnce between normal and inactive type of ALDH2. It suggested that further evaluation should be conducted in high concentration of toluene exposure group.

      • 培養鷄胚筋細胞分化에 미치는 細胞融合促進物質의 영향

        培養鷄胚筋細胸 分化에 미치는 促進物質의 영향을 알아보고자 2가양이온 (Ca2+, Mg2+) 胸胚抽出物 (EE), polyethylene glycol (PEG), 再生中인 도마뱀 (Lacertilia)꼬리 추출물 (BE)을 처리하여 細胸融合指數를 비교 조사하였다. 정상배양시의 細胸融合指數는 배양시작 70 시간에 최대에 달했으며 더이상 증가하지 않았고, Ca2+와 Mg2+ 처리할 경우에는 농도가 증가할수록 細胸融合指數는 저하되고, 근세포의 굵기도 정상세포보다 가늘고 핵이 모여있었다. Polyethylene Glycol (PEG)와 Blastemata Extract (BE) 처리군에서는 대조구와 큰 차이가 없는 것으로 나타났다. 鷄胚抽出物 (EE) 을 筋細胞分化에 필수적이나 과량으로 添加시에는 細胞融合은 억제되고 세포수는 증가되는것으로 나타났다. To study the effects of promoting factors on the differentiation of muscle cells, chick embryonic myoblasts were cultured with divalent cations (Ca2+, Mg2+) Chick embryo extract (EE), Polyethylene Glycol (PEG) and Blastema extract of Lacertilia (BE). Fusion index and Growth ratio were measured at various culture time. The fusion index of normal myoblast culture was reached maximum at 70 hours and the divalent cations (Ca2+, Mg2+) treated groups were decreased at higher concentration. The cellular formation of myoblasts treated with divalent cations was thiner and more aggregated between nuclei than that of normal myoblasts calture was. When polyethylene glycol (PEG) and Blastema extract of Lacertilia (BE) were treated to the myoblasts, tha fusion indices were similar to the control. Embryo Extract (EE) was seemed to be necessary for the fusion of chick embryonic myoblasts in culture, however, when excess of EE was added the fusion index was decreased and the number of cells was increased.

      • Fuzzy Difuntional Relation의 특성화

        본 눈문에서는 fuzzy difunctional relation 동치조건들을 소개하였고 이를 이용하여 fuzzy difunctional relation과 관련된 일부 결과를 제시하였으며 또한 반군에서의 퍼지합동관계를 α-cut로 특성화 하였다. In this paper, we shall introduce equivalent conditions of the fuzzy difunctional relation, give some results in connection with fuzzy difunctional relations and characterize the fuzzy congruence relation on a semigroup in terms of its α-cut. Also, closed relationship between fuzzy difunctional relations and fuzzy congruence relations will be revealed.

      • 대구지역 성인 남성의 혈청 지질 및 비타민 E 상태와 생활습관에 관한 연구

        Serum lipid and vitamin E levels were determined along with anthropometric measurements and smoking, drinking and exercise habits were asked in 357 healty male subjects with mean age of 49.4±6.7 years from Taegu area in Korea. Body mass index(BMI) and waist/hip(W/H) ratio, tricept skinfold thickness of the subjects were 23.01±23.0, 0.88±0.04 and 9.48±3.50mm, respectively. Their systolic and distolic blood pressures were 127.8±15.5 and 83.9±10. mmHg. Seventeen percent of the subject had BML over 25. Average serum levels of total cholesterol(TChol) and HDL-cholesterol and triglyceride(TG) were 189±49, 42.0±12.5 and 136±7mg/dl, respectively. Serum level of lipid peroxide measured as thiobarbituric acid reactive substances(TBARS) of the subjects was 2.01±0.78 MDA nmoles/ml and that of-tocopherol was 9.53±3.14㎍/ml. Exercise group had lower TG values than non-exercise one. Both smokers and alcohol drinkers had higher levels of TG and TBARS than non-smokers and non-drinkers. Vitamin E levels also appeared to be higher in the latter groups. It is concluded from the present study that vitamin E and other antioxidant nutrition have to be strengthened as well as modification of lifestyle factors both for those who are at risk of ischemic heart disease(IHD) and general public to retard and prevent IHD in Korea.

        Immunogenicity of a bivalent killed thimerosal-free oral cholera vaccine, Euvichol, in an animal model

        <P><B>Purpose</B></P><P>An oral cholera vaccine (OCV), Euvichol, with thimerosal (TM) as preservative, was prequalified by the World Health Organization (WHO) in 2015. In recent years, public health services and regulatory bodies recommended to eliminate TM in vaccines due to theoretical safety concerns. In this study, we examined whether TM-free Euvichol induces comparable immunogenicity to its TM-containing formulation in animal model.</P><P><B>Materials and Methods</B></P><P>To evaluate and compare the immunogenicity of the two variations of OCV, mice were immunized with TM-free or TM-containing Euvichol twice at 2-week interval by intranasal or oral route. One week after the last immunization, mice were challenged with <I>Vibrio cholerae</I> O1 and daily monitored to examine the protective immunity against cholera infection. In addition, serum samples were obtained from mice to measure vibriocidal activity and vaccine-specific IgG, IgM, and IgA antibodies using vibriocidal assay and enzyme-linked immunosorbent assay, respectively.</P><P><B>Results</B></P><P>No significant difference in immunogenicity, including vibriocidal activity and vaccine-specific IgG, IgM, and IgA in serum, was observed between mice groups administered with TM-free and -containing Euvichol, regardless of immunization route. However, intranasally immunized mice elicited higher levels of serum antibodies than those immunized via oral route. Moreover, intranasal immunization completely protected mice against <I>V. cholerae</I> challenge but not oral immunization. There was no significant difference in protection between two Euvichol variations.</P><P><B>Conclusion</B></P><P>These results suggested that TM-free Euvichol could provide comparable immunogenicity to the WHO prequalified Euvichol containing TM as it was later confirmed in a clinical study. The pulmonary mouse cholera model can be considered useful to examine <I>in vivo</I> the potency of OCVs.</P>

      • Inhibitory Effects of <i>Chrysanthemum boreale</i> Essential Oil on Biofilm Formation and Virulence Factor Expression of <i>Streptococcus mutans</i>

        <P>The aim of the study was to evaluate the antibacterial activity of essential oil extracted from <I>Chrysanthemum boreale</I> (<I>C. boreale</I>) on <I>Streptococcus mutans</I> (<I>S. mutans</I>). To investigate anticariogenic properties, and bacterial growth, acid production, biofilm formation, bacterial adherence of <I>S. mutans</I> were evaluated. Then gene expression of several virulence factors was also evaluated. <I>C. boreale</I> essential oil exhibited significant inhibition of bacterial growth, adherence capacity, and acid production of <I>S. mutans</I> at concentrations 0.1–0.5 mg/mL and 0.25–0.5 mg/mL, respectively. The safranin staining and scanning electron microscopy results showed that the biofilm formation was also inhibited. The result of live/dead staining showed the bactericidal effect. Furthermore, real-time PCR analysis showed that the gene expression of some virulence factors such as <I>gtf</I>B, <I>gtf</I>C, <I>gtf</I>D, <I>gbp</I>B, <I>spa</I>P, <I>brp</I>A, <I>rel</I>A, and <I>vic</I>R of <I>S. mutans</I> was significantly decreased in a dose dependent manner. In GC and GC-MS analysis, seventy-two compounds were identified in the oil, representing 85.42% of the total oil. The major components were camphor (20.89%), <I>β</I>-caryophyllene (5.71%), <I>α</I>-thujone (5.46%), piperitone (5.27%), <I>epi</I>-sesquiphellandrene (5.16%), <I>α</I>-pinene (4.97%), 1,8-cineole (4.52%), <I>β</I>-pinene (4.45%), and camphene (4.19%). These results suggest that <I>C. boreale</I> essential oil may inhibit growth, adhesion, acid tolerance, and biofilm formation of <I>S. mutans</I> through the partial inhibition of several of these virulence factors.</P>

      • Two distinct mechanisms of transcriptional regulation by the redox sensor YodB

        <P>For bacteria, cysteine thiol groups in proteins are commonly used as thiol-based switches for redox sensing to activate specific detoxification pathways and restore the redox balance. Among the known thiol-based regulatory systems, the MarR/DUF24 family regulators have been reported to sense and respond to reactive electrophilic species, including diamide, quinones, and aldehydes, with high specificity. Here, we report that the prototypical regulator YodB of the MarR/DUF24 family from Bacillus subtilis uses two distinct pathways to regulate transcription in response to two reactive electrophilic species (diamide or methyl-p-benzoquinone), as revealed by X-ray crystallography, NMR spectroscopy, and biochemical experiments. Diamide induces structural changes in the YodB dimer by promoting the formation of disulfide bonds, whereas methyl-p-benzoquinone allows the YodB dimer to be dissociated from DNA, with little effect on the YodB dimer. The results indicate that B. subtilis may discriminate toxic quinones, such as methyl-p-benzoquinone, from diamide to efficiently manage multiple oxidative signals. These results also provide evidence that different thiol-reactive compounds induce dissimilar conformational changes in the regulator to trigger the separate regulation of target DNA. This specific control of YodB is dependent upon the type of thiol-reactive compound present, is linked to its direct transcriptional activity, and is important for the survival of B. subtilis. This study of B. subtilis YodB also provides a structural basis for the relationship that exists between the ligand-induced conformational changes adopted by the protein and its functional switch.</P>

        Background: It has been well known that genetic mutations and the epidemiology of inflammatory bowel disease (IBD) differ between Asia and the West. However, TNF superfamily member 15 (TNFSF15) gene has been identified as a common susceptibility gene in Asian and Western IBD patients, suggesting that TNFSF15 may play an important role in the pathogenesis of IBD. TNFSF15/TNF-like cytokine 1A (TL1A) is a proinflammatory cytokine and a member of the TNFa superfamily, mainly expressed on activated T cells. We investigated mucosal TL1A expression by immunohistochemistry (IHC) in Korean IBD patients. Methods: TL1A expression was investigated in resected ileal specimens from 8 Crohn`s disease (CD) patients and 8 non-IBD controls by IHC using an affinity-purified monoclonal Antibody against TL1A. TL1A expression was also studied in endoscopically biopsied colonic specimens from 8 ulcerative colitis patients and 8 non-IBD controls. In addition, the mucosal expressions of CD3, CD4, and CD8, T cell markers, were examined in ileal and colonic specimens of IBD patients and non-IBD controls. Results: The expression of TL1A at the protein level was absent or minimal in ileal and colonic tissues from controls. On the contrary, intense expression of TL1A was identified in ileal and colonic specimens from IBD patients. Staining for TL1A was signifi - cantly increased in inflammed area of ileal tissues compared to non-inflammed area from CD patients. CD3, CD4, and CD8 expressions were also significantly increased in ileal and colonic specimens from IBD patients compared to those from controls. Conclusions: Mucosal TL1A is upregulated in IBD patients, suggesting an important role of TL1A in the pathogenesis of IBD. Further functional studies of TL1A will provide a better understanding of the pathogenesis of IBD.
