        Realgar transforming solution-induced differentiation of NB4 cell by the degradation of PML/RARa partially through the ubiquitin–proteasome pathway

        PML/retinoic acid receptor alpha (RARa), as ahallmark of acute promyeloid leukemia (APL), is directlyrelated to the outcome of clinical APL remedy. It isreported that arsenicals can effectively degrade PML/RARa, such as arsenic trioxide and realgar. However, thehigh toxicity or insolubility have hampered their clinicalapplications. Realgar transforming solution (RTS) wasproduced from realgar by bioleaching process in our lab. Previous studies demonstrated that RTS had a significantanti-cancer ability on chronic myeloid leukemia throughoncoprotein degradation. The capacity of RTS on treatingAPL is what is focused on in this study. The results showedthat RTS had a noticeable sensitivity in NB4 cell, and RTSremarkably down-regulated PML/RARa expression andinduced cell differentiation. Further, RTS could accumulatePML/RARa into the nuclear bodies and then executedegradation, which could be reversed by proteasomeinhibitor MG132. The results also exhibited that thereduction of RTS-induced PML/RARa expression accompaniedby the elevation of ubiquitin and SUMO-1 proteinexpression. Finally, PML and SUMO-1 had been demonstratedto be co-localized after RTS treatment byimmunofluorescence co-localization assay and immunoprecipitationassay. In conclusion, these results suggestedthat RTS-induced cell differentiation may attribute to thePML/RARa degradation partially through the ubiquitin–proteasome pathway.

        Dynamic experimental study on single and double beam-column joints in steel traditional-style buildings

        In order to study the failure mode and seismic behavior of the interior-joint in steel traditional-style buildings, a single beam-column joint and a double beam-column joint were produced according to the relevant building criterion of ancient architectural buildings and the engineering instances, and the dynamic horizontal loading test was conducted by controlling the displacement of the column top and the peak acceleration of the actuator. The failure process of the specimens was observed, the bearing capacity, ductility, energy dissipation capacity, strength and stiffness degradation of the specimens were analyzed by the load-displacement hysteresis curve and backbone curve. The results show that the beam end plastic hinge area deformed obviously during the loading process, and tearing fracture of the base metal at top and bottom flange of beam occurred. The hysteresis curves of the specimens are both spindle-shaped and plump. The ultimate loads of the single beam-column joint and double beam-column joint are 48.65 kN and 70.60 kN respectively, and the equivalent viscous damping coefficients are more than 0.2 when destroyed, which shows the two specimens have great energy dissipation capacity. In addition, the stiffness, bearing capacity and energy dissipation capacity of the double beam-column joint are significantly better than that of the single beam-column joint. The ductility coefficients of the single beam-column joint and double beam-column joint are 1.81 and 1.92, respectively. The cracks grow fast when subjected to dynamic loading, and the strength and stiffness degradation is also degenerated quickly.

      • Maltol Inhibits Apoptosis of Human Neuroblastoma Cells Induced by Hydrogen Peroxide

        To analyze the effect of Maltol on the apoptosis of Human Neuroblastoma Cells (SH-SY5Y) treated by free radical which was generated from Hydrogen Peroxide ($H_2O_2$), flow cytometry analysis on Phosphatidylserine (PS) inverting percentage was applied to determine the apoptosis. MTT (3-(4,5-dimethythiazol-2-yl)-2,5-diphenyl-tetrazolium bromide) assay was employed to analyze the cell viability. DNA electrophoresis was used to detect DNA fragmentation. Moreover intracellular calcium of concentration ($[Ca^{2+}]_i$) was measured by fluorescence emission. Flow cytometry analysis on the function of mitochondria and Western blto analysis of NF-${\kappa}B$. The results showed that the pretreatment with maltol for 2 hours could prevent the $H_2O_2$-induced apoptosis. Maltol could reduce the inverting percentage of PS, DNA fragmentation and $[Ca^{2+}]_i$, and enhance the cellular function of mitochondria. NF-${\kappa}B$ activated by $H_2O_2$ is reduced. The experiments suggest that maltol could effectively inhibit the apoptosis induced by $H_2O_2$. As a novel anti-oxidant, maltol is a new promising drug in protecting the neurological cells from the damage by free radical.

      • Is Hepatectomy for Huge Hepatocellular Carcinoma (≥10cm in Diameter) Safe and Effective? A Single-center Experience

        Background: This retrospective study aimed to validate the safety and effectiveness of hepatectomy for huge hepatocellular carcinoma (HCC). Materials and Methods: Data of patients who underwent hepatectomy for HCC between January 2006 and December 2012 were reviewed. The patients were divided into three groups: huge HCC(${\geq}10cm$ in diameter), large HCC(${\geq}5$ but<10 cm in diameter) and small HCC(<5cm in diameter). Results: Characteristics of pre-operative patients in all three groups were homogeneously distributed except for alpha fetal protein (AFP)(p<0.001).The 30, 60, 90-day post-operative mortality rates were not different among the three groups (p=0.785, p=0.560, and p=0.549). Laboratory data at 1, 3, and 7 days after surgery also did not vary. The 5-year overall survival (OS) and 5-year disease-free survival (DFS) rates in the huge and large HCC groups were lower than that of the small HCC group (OS: 32.5% vs 36.3% vs 71.2%, p=0.000; DFS: 20.0% vs 24.8% vs 40.7%, p=0.039), but there was no difference between the huge and large HCC groups (OS: 32.5% vs 36.3%, p=0.667; DFS: 20.0% vs 24.8%, p=0.540). In multivariate analysis, five independent poor prognostic factors that affected OS were significantly associated with worse survival (p<0.05), namely, AFP level, macrovascular invasion, Edmondsone Steiner grade, surgical margin and Ishak score. AFP level, macrovascular invasion, microvascular invasion, and surgical margin influenced disease-free survival independently (p<0.05). Conclusions: The safety of hepatectomy for huge HCC is similar to that for large and small HCC; and this approach for huge HCC may achieve similar long-term survival and disease-free survival as for large HCC.

        Power–Delay Tradeoff in Wireless Powered Communication Networks

        <P>In this paper, we investigate the power-delay tradeoff for the newly emerging wireless powered communication networks, in which wireless powered devices (WPDs) harvest energy from a power station via wireless energy transfer in the downlink and then communicate with an information receiving station in the uplink. Each WPD is equipped with an energy buffer and a data buffer to store the random harvested energy and the bursty data arrivals, respectively. To minimize the time-averaged power consumption, a stochastic optimization problem is formulated subject to both constraints of data queue stability and harvested energy availability. By employing Lyapunov optimization theory, we transform the stochastic optimization problem into a series of deterministic problems, where each is further proved to become the standard convex optimization problem and, hence, can be effectively and optimally solved by standard convex optimization techniques. Correspondingly, we propose an Online Power and TIMe Allocation (Optima) algorithm, which requires no a priori distribution knowledge of channel states and data arrivals. Most importantly, the proposed algorithm achieves the power-delay tradeoff as [O(1/V), O(V)], with V being a system control parameter, and provides a significant method to control the power-delay performance on demand in system design. Simulation results verify the theoretical analysis and validate the effectiveness of the proposed algorithm.</P>

      • Systematic Review of Single Large and/or Multinodular Hepatocellular Carcinoma: Surgical Resection Improves Survival

        Background: The role of surgical resection for patients with single large (${\geq}5cm$) and/or multinodular (${\geq}2$) hepatocellular carcinoma (HCC) is still controversial. This systematic review was performed to evaluate the safety and efficacy of resection for patients with single large and/or multinodular HCC. Materials and Methods: Databases (the PubMed, Web of Science, Embase, and Cochrane databases) were systematically searched to identify relevant studies exploring the safety and efficacy of resection for single large and/or multinodular HCC, published between January 2000 and December 2014. Perioperative morbidity and mortality, overall survival, and disease-free survival of the resection group were calculated. In addition, these outcome variables were also calculated for the control group in the included studies. Results: One randomized controlled trial and 42 nonrandomized studies involving 9,580 patients were eligible for analysis. Eight (1,594 patients) of the 43 studies also reported the outcomes of transarterial chemoembolization (TACE). Although 51.4% of patients featured cirrhosis, 90.7% of them demonstrated Child-Pugh A liver function in the resection group. The median rates of morbidity (24.5%) and mortality (2.5%) after resection were significantly higher than that of TACE (11.0%, P<0.001; 1.9%, P<0.001). However, patients who underwent resection had significantly higher median one-, three-, and five-year overall survival (76.1%, 51.7%, and 37.4%) than those who underwent TACE (68.3%, 31.5%, and 17.5%, all P<0.001). The median 1-, 3-, and 5-year DFS rates after resection were 58.3%, 34.6%, and 24.0%, respectively. Conclusions: Although tumor recurrence after resection for patients with single large and/ or multinodular HCC continues to be a major problem, resection should be considered as a strategy to achieve long-term survival.

        Mitochondrial citrate accumulation drives alveolar epithelial cell necroptosis in lipopolysaccharide-induced acute lung injury

        Necroptosis is the major cause of death in alveolar epithelial cells (AECs) during acute lung injury (ALI). Here, we report a previously unrecognized mechanism for necroptosis. We found an accumulation of mitochondrial citrate (citratemt) in lipopolysaccharide (LPS)-treated AECs because of the downregulation of Idh3α and citrate carrier (CIC, also known as Slc25a1). shRNA- or inhibitor–mediated inhibition of Idh3α and Slc25a1 induced citratemt accumulation and necroptosis in vitro. Mice with AEC-specific Idh3α and Slc25a1 deficiency exhibited exacerbated lung injury and AEC necroptosis. Interestingly, the overexpression of Idh3α and Slc25a1 decreased citratemt levels and rescued AECs from necroptosis. Mechanistically, citratemt accumulation induced mitochondrial fission and excessive mitophagy in AECs. Furthermore, citratemt directly interacted with FUN14 domain-containing protein 1 (FUNDC1) and promoted the interaction of FUNDC1 with dynamin-related protein 1 (DRP1), leading to excessive mitophagy-mediated necroptosis and thereby initiating and promoting ALI. Importantly, necroptosis induced by citratemt accumulation was inhibited in FUNDC1-knockout AECs. We show that citratemt accumulation is a novel target for protection against ALI involving necroptosis.

        Magnetoelectric characteristics around resonance frequency under magnetic field in Pb(Zr, Ti)O3/Terfenol-D laminate composite

        Dependence of magnetoelectric modulus and phasic difference on the bias magnetic field was studied in a magnetoelectric Pb(Zr, Ti)O3/Terfenol-D laminate composite. The shifts of both magnetoelectric resonant peak and phasic bounce under the bias magnetic field lead to a nonlinear relationship between them. The acoustic velocity of Terfenol-D has a nonlinear dependence on magnetic field, resulting in a similar relationship between the acoustic velocity of the composite and magnetic field. As a result, the magnetoelectric modulus and phase around the resonance frequency exhibit very sensitive dependences on the bias magnetic field.

        A case study on safety integrity level analysis for shale gas station

        Safety integrity level (SIL), as defined in IEC 61511, is a widely used safety performance measure for safety instrumented functions. Shale gas station is an important part of shale gas gathering and transportation system. To ensure the safety of its instrument system is of great significance to the safe and stable operation of the whole gathering and transportation system. This paper is motivated to conduct a case study on safety integrity analysis for shale gas station by using the layer of protection analysis method (LOPA). Firstly, the equipment under control (EUC) is identified. It is followed by the analysis of reaction reasons on safety instrumented function (SIF) from safety instrumented system (SIS). Finally, according to the identification of independent protection layer (IPL) and probability of failure on demand (PFD), the SIL analysis for every SIF is carried out. The result shows that the SIS of this shale gas station has reached the required of SIL and there is no need for further increase in SIS. Utilizing this method can accurately determine the reliability of the SIS in the station, and it can also be the reference of safe operation for this shale gas station.
