        SIRT5-related desuccinylation modification of AIFM1 protects against compression-induced intervertebral disc degeneration by regulating mitochondrial homeostasis

        Mitochondrial dysfunction plays a major role in the development of intervertebral disc degeneration (IDD). Sirtuin 5 (SIRT5) participates in the maintenance of mitochondrial homeostasis through its desuccinylase activity. However, it is still unclear whether succinylation or SIRT5 is involved in the impairment of mitochondria and development of IDD induced by excessive mechanical stress. Our 4D label-free quantitative proteomic results showed decreased expression of the desuccinylase SIRT5 in rat nucleus pulposus (NP) tissues under mechanical loading. Overexpression of Sirt5 effectively alleviated, whereas knockdown of Sirt5 aggravated, the apoptosis and dysfunction of NP cells under mechanical stress, consistent with the more severe IDD phenotype of Sirt5 KO mice than wild-type mice that underwent lumbar spine instability (LSI) surgery. Moreover, immunoprecipitation-coupled mass spectrometry (IP-MS) results suggested that AIFM1 was a downstream target of SIRT5, which was verified by a Co-IP assay. We further demonstrated that reduced SIRT5 expression resulted in the increased succinylation of AIFM1, which in turn abolished the interaction between AIFM1 and CHCHD4 and thus led to the reduced electron transfer chain (ETC) complex subunits in NP cells. Reduced ETC complex subunits resulted in mitochondrial dysfunction and the subsequent occurrence of IDD under mechanical stress. Finally, we validated the efficacy of treatments targeting disrupted mitochondrial protein importation by upregulating SIRT5 expression or methylene blue (MB) administration in the compression-induced rat IDD model. In conclusion, our study provides new insights into the occurrence and development of IDD and offers promising therapeutic approaches for IDD.

        中国税收滞纳金制度现状分析及发展研究

        The collection of late tax filing charges is one means of increasing tax revenues. At present, due to the unclear start and end periods of late tax filing charges, its function has changed from encouraging taxpayers to repay their tax arrears as soon as possible to becoming the main source of conflict between tax authorities and taxpayers. This paper engages in a preliminary discussion on the improvement of the late tax filing charge system, including such aspects as sorting out the legal nature of late tax filing charges, setting up a clear collection method for late tax filing charges, establishing a system that’s integrated with other jurisdictions, and strengthening the advocacy of tax delay charges by tax authorities and administrative departments. This paper is structured as follows: The introduction provides a general overview of the topic’s background, clarifying questions and methods of research. The second section clarifies the difference between late tax filing charges and administrative late fees, defining the boundary between several related concepts and revealing some normative problems. The third section discusses the creation of the late tax filing charges collection system. In combining the first chapter’s question and the second chapter’s analysis of theory and practice, we put forward some suggestions to improving the tax delay charge collection system. Combining the perspective of comparative law, aimed at the settlement of disputes over the collection of late tax filing charges, this paper elaborates on how to strengthen the publicity of late tax filing charges by tax collection and administration departments as well as the creation of specialized tax courts. The conclusion traces the collection of late tax filing charges to the theoretical issues of tax collection and management, summarizing the significance of the late tax filing charge system in promoting the improvement of the tax system.

        Weak fault detection for wind turbine bearing based on ACYCBD and IESB

        To detect the rolling bearing fault of a wind turbine at incipient injury phase, a novel method combining adaptive cyclostationary blind deconvolution (ACYCBD) with instantaneous energy slice bispectrum (IESB) is proposed. Cuckoo search algorithm (CSA) is fused with cyclostationary blind deconvolution (CYCBD) to optimize cyclic frequency and filter length parameters, and the satisfied processing result is able to adaptively use this developed ACYCBD method. Besides, fusing the complementary superiorities of frequency weighted energy operator and slice bispectrum, a new frequency domain analysis method named IESB is put forward to identify the characteristic frequency components. During fault diagnosis, the deconvolution signal with higher signal to noise ratio is first separated from the collected signal by applying ACYCBD method. However, owing to the intense background noises, external interferences still remain in the deconvolution signal. Then the IESB is further used as a post-processing method, the residual noises are suppressed effectively, and the characteristic spectral lines are highlighted immensely. In the end, the incipient fault injury of wind turbine bearing can be diagnosed by analyzing the acquired instantaneous energy slice bispectrum. The analyzed results of wind turbine bearing fault signals from the experimental rig and the actual engineering testing field demonstrate the feasibility and the superiority of this novel detection method.

        중국인 한국어 학습자가 한자어를 쓸 때의 오류 분석 및 교육 방법

        외국어를 공부할 때 생기는 오류는 모국어의 영향으로 인한 경우가 적지 않다. 특히 한국어를 배우는 중국어권 학습자들은 한국어의 한자어 어휘를 습하는 과정에서 이러한 오류가 많이 발생한다. 한국어 한자어 어휘와 중국어 한자어는 뜻이 일대일로 대응하지 않은 것들이 많은데 많은 중국어권 학습자들은 모국어의 영향을 받아서 양국의 한자어가 뜻이 같을 뿐만 아니라 쓰임새도 같다고 생각하는 경향이 있다. 본고에서는 한국어를 배우는 중국어권 학습자들을 대상으로 문장 번역 테스트를 통해 한자어 어휘 사용실태를 파악하고 그 결과를 분석한 후, 이러한 오류를 줄일 수 있는 교육 방법에 대해 제시하고자 한다. When learning a language, it is hard for learners to avoid various kinds of mistakes. Because there are a lot of Chinese characters in Korean language, the Chinese learners of the Korean language believe that it is not difficult to learn the Korean. In fact, the meaning of Chinese characters is not one-to-one correspondent to the meaning of Chinese characters in Korean. However, there are still a lot of students who think that not only the meaning but also the usage of the characters in the two languages are the same. This leads to a lot of language errors in the two languages of the Korean language learners. This article tries to let Korean learners to translate Chinese articles into Korean and analyzes the translation of the sentences whether there really has an impact of Chinese characters. This article also proposes some strategies of how to reduce the language errors from the mother tongue influence, and it explains the reasons to these language errors.

        <P>Colloids with well-defined multicavities are synthesized through the hydrolytic removal of silica cluster templates from organo-silica hybrid patchy particles. The geometry of the cavities stems from the originally assembled cluster templates, displaying well-defined three-dimensional symmetries, ranging from spherical, linear, triangular, tetrahedral, trigonal dipyramidal, octahedral, to pentagonal dipyramidal. The concave surface of the cavities is smooth, and the cavity shallowness and size can be varied. These particles with multicavities can act as “lock” particles with multiple “key holes”.         Step-by-step identification of industrial robot dynamics model parameters and force-free control for robot teaching

        In order to solve the problem of lack of flexibility in direct teaching of industrial robots under complex working conditions, dynamic inertia term compensation is combined with the traditional force-free control algorithm to reduce the traction in the teaching process in this paper. Firstly, considering the multi-dimensional, strongly nonlinear and multi-parameter characteristics of the 6-DOF manipulator dynamic model, a step-by-step parameters identification method based on genetic algorithm is proposed. This method can effectively reduce the computational complexity and improve the optimization speed of industrial robot dynamic model identification. Secondly, based on the internal torque analysis of industrial robot joints, a forcefree compensation control algorithm based on the torque control is designed. Finally, the effectiveness of the step-by-step robot dynamic model parameter identification method is verified by a traction teaching experiment. In the verification experiment, the relative error rate between the predicted torque and the actual torque calculated from the identification results is 1.64 % to 5.99 %. After increasing the inertia term compensation, the traction force of the teaching process is reduced by 28.6 %. The experimental results show that the identification result is more accurate, and the proposed force-free control algorithm significantly improves the compliance of the teaching process.

        Design and Kinematic Modeling of a Notch Continuum Manipulator for Laryngeal Surgery

        A notch continuum manipulator with a large cavity is presented. The continuum manipulator which is designed for laryngeal surgery is composed of guidewire discs and a Nitinol skeleton. The deformation of the continuum manipulator is superimposed by the flexible units, therefore its posture and driving force are coupled. Based on the small deformation cantilever beam theory, the mechanical model from the microscopic to the macroscopic is used to establish the mapping from the driving space to the joint space. The manipulator is assumed to be a multi-joint series robot. Based on the mechanical model, the mapping from the joint space to the Cartesian space is established by the Denavit-Hartenberg (D-H) method. The kinematics is established from the driving space to the Cartesian space. The proposed kinematic model is verified by a prototype. The experimental results show the validity of the model.

        Transcriptome Analysis Reveals the Putative Polyketide Synthase Gene Involved in Hispidin Biosynthesis in Sanghuangporus sanghuang

        Hispidin is an important styrylpyrone produced by Sanghuangporus sanghuang. To analyzehispidin biosynthesis in S. sanghuang, the transcriptomes of hispidin-producing and non-producingS. sanghuang were determined by Illumina sequencing. Five PKSs were identifiedusing genome annotation. Comparative analysis with the reference transcriptome showedthat two PKSs (ShPKS3 and ShPKS4) had low expression levels in four types of media. Thegene expression pattern of only ShPKS1 was consistent with the yield variation of hispidin. The combined analyses of gene expression with qPCR and hispidin detection by liquid chromatography-mass spectrometry coupled with ion-trap and time-of-flight technologies(LCMS-IT-TOF) showed that ShPKS1 was involved in hispidin biosynthesis in S. sanghuang. ShPKS1 is a partially reducing PKS gene with extra AMP and ACP domains before the KSdomain. The domain architecture of ShPKS1 was AMP-ACP-KS-AT-DH-KR-ACP-ACP. Phylogenetic analysis shows that ShPKS1 and other PKS genes from Hymenochaetaceaeform a unique monophyletic clade closely related to the clade containing Agaricales hispidinsynthase. Taken together, our data indicate that ShPKS1 is a novel PKS of S. sanghuanginvolved in hispidin biosynthesis.

        A Study on BMPR-IB Genes of Bayanbulak Sheep

        The average twin lambing rate of Bayanbulak sheep is 2% to 3%. However, a flock of sheep with a close genetic relationship and an average of 2 to 3 lambs per birth has been found recently. To determine the major genes controlling the prolificacy of the flock in the present study, the flock was designated A while 100 normal Bayanbulak sheep were randomly selected to comprise the control flock B. Ligase detection reaction method was applied to detect and analyze the 10 mutational loci of the 3 candidate prolificacy genes including bone morphogenetic protein type I receptors, bone morphogenetic protein 15, and growth differentiation factor 9. The 10 mutational loci are as follows: FecB locus of the BMPR-IB gene; $FecX^I$, $FecX^B$, $FecX^L$, $FecX^H$, $FecX^G$, and $FecX^R$ of the BMP15 gene; and G1, G8, and FecTT of the GDF9 gene. Two mutations including BMPR-IB/FecB and GDF9/G1 were found in Bayanbulak sheep. Independence test results of the two flocks demonstrate that the FecB locus has a significant effect on the lambing number of Bayanbulak sheep. However, the mutation frequency of the G1 locus in GDF9 is very low. Independence test results demonstrate that the GDF9 locus does not have a significant impact on the lambing performance of Bayanbulak sheep. Among the 10 detected loci, BMPR-IB/FecB is the major gene that influences the high lambing rate of Bayanbulak sheep.
