The influence of cooling rates on the mechanical properties of a Zr-based bulk metallic glass prepared with high rheologicalrate forming (HRRF) was investigated and compared with traditional suction cast methods. Amorphous samples of Zr57Cu-20Ni8Al10Ag5 were prepared in copper molds with different sizes in order to obtain different cooling rates for both HRRFand traditional cast methods. These specimens were subjected to compression experiments, including microhardness testing,X-ray diffraction testing and differential scanning calorimetry analysis. The results indicate that the plasticity of the samplesformed by HRRF are higher than that of the as-cast ones at the same cooling rates, while the microhardness manifests theopposite principle. As the cooling rate increases further, the difference in plasticity further increases between two methods,indicating that the plasticity of metallic glasses is more sensitive to cooling rates during the HRRF process. At the core ofthis phenomenon is the fact that HRRF methods can introduce more free volume into glasses than traditional cast methodswith an elevated cooling rate are able to.

The protein encoded by dynein axonemal heavy chain 1 (DNAH1) is a part of dynein, which regulates the function of cilia and sperm flagella. The mutant of DNAH1 causes the deletion of inner dynein arm 3 in the flagellum, leading to multiple morphological abnormalities of the sperm flagella (MMAF) and severe asthenozoospermia. However, instead of asthenozoospermia and MMAF, the result caused by the mutation of DNAH1 remains unknown. Here we report a male infertility patient with severe asthenozoospermia and teratozoospermia. We found two heterozygous mutations in DNAH1 (c.6912C>A and c.7076G>T) and which were reported to be associated with MMAF for the first time. We next collected and analyzed 65 cases of DNAH1 mutation and found that the proportion of short flagella is the largest, while the bent flagella account for the smallest, and the incidence of head deformity is not high in the sperm of these patients. Finally, we also analyzed 31 DNAH1 mutation patients who were treated with intracytoplasmic sperm injection (ICSI) and achieved beneficial outcomes. We hope our research will be helpful in the diagnosis and treatment of male infertility caused by DNAH1 mutation.
ODV-Associated Proteins of the <i>Pieris rapae</i> Granulovirus

<P><I>Alphabaculovirus</I> (lepidopteran-specific nucleopolyhedroviruses, NPV) and <I>Betabaculovirus</I> (granuloviruses, GV) are two main genera of the family Baculoviridae. The virion proteomes of <I>Alphabaculovirus</I> have been well studied; however, the <I>Betabaculovirus</I> virion compositions remain unclear. <I>Pieris rapae</I> granulovirus (PrGV) can kill larvae of <I>P. rapae</I>, a worldwide and important pest of mustard family crops. In this study, the occlusion-derived virus (ODV)-associated proteins of PrGV were identified using three mass spectrometry (MS) approaches. The MS analyses demonstrated that 47 proteins were present in PrGV-ODV. Of the 47 PrGV-ODV proteins, 33 have homologues identified previously in other baculovirus ODV/BVs, whereas 14 (P10, Pr21, Pr29, Pr35, Pr42, Pr54, P45/48, Pr83, Pr84, Pr89, Pr92, Pr111, Pr114 and FGF3) were newly identified ODV proteins. Seven of the 14 newly identified ODV proteins are specific to <I>Betabaculovirus</I>, including Pr35, Pr42, Pr54, Pr83, Pr84, Pr111 and Pr114. Furthermore, the data derived from these MS approaches were validated by immunoblotting analysis using antisera prepared from 11 randomly selected recombinant PrGV-ODV proteins (including 5 <I>Betabaculovirus</I>-unique proteins). Comparison analyses revealed the similar and different compositions between <I>Betabaculovirus</I> and <I>Alphabaculovirus</I> virions, which deepen our understanding of the baculovirus virion structure and provide helpful information on <I>Betabaculovirus</I>–host interaction studies.</P><P>We used three mass spectrometry (MS) approaches to identify the occlusion-derived virus (ODV)-associated proteins of the <I>Pieris rapae</I> Granulovirus. In this work, we investigate the existence of the extremal solutions for a class of fractional partial differential equations with or- der 1 < α < 2 by upper and lower solution method. Using the theory of Hausdorff measure of noncompactness, a series of results about the solutions to such differential equations is obtained.

To determine heat-shock protein (Hsp)90 expression is connected with cellular apoptotic response to heat stress and its mechanism, chicken (Gallus gallus) primary myocardial cells were treated with the Hsp90 promoter, aspirin, and its inhibitor, geldanamycin (GA), before heat stress. Cellular viability, heat-stressed apoptosis and reactive oxygen species level under different treatments were measured, and the expression of key proteins of the signaling pathway related to Hsp90 and their colocalization with Hsp90 were detected. The results showed that aspirin treatment increased the expression of protein kinase B (Akt), the signal transducer and activator of transcription (STAT)-3 and p-IKKa/b and the colocalization of Akt and STAT-3 with Hsp90 during heat stress, which was accompanied by improved viability and low apoptosis. GA significantly inhibited Akt expression and p-IKKa/b level, but not STAT-3 quantity, while the colocalization of Akt and STAT-3 with Hsp90 was weakened, followed by lower cell viability and higher apoptosis. Aspirin after GA treatment partially improved the stress response and apoptosis rate of tested cells caused by the recovery of Akt expression and colocalization, rather than the level of STAT-3 (including its co-localization with Hsp90) and p-IKKa/b. Therefore, Hsp90 expression has a positive effect on cellular capacity to resist heat-stressed injury and apoptosis. Moreover, inhibition of Hsp90 before stress partially attenuated its positive effects.
Modeling of ion diffusion coefficient in saturated concrete

This paper utilizes the modified Davis model and the mode coupling theory, as parts of the electrolyte solution theory, to investigate the diffusivity of the ion in concrete. Firstly, a computational model of the ion diffusion coefficient, which is associated with ion species, pore solution concentration, concrete mix parameters including water-cement ratio and cement volume fraction, and microstructure parameters such as the porosity and tortuosity, is proposed in the saturated concrete. Secondly, the experiments, on which the chloride diffusion coefficient is measured by the rapid chloride penetration test,have been carried out to investigate the validity of the proposed model. The results indicate that the chloride diffusion coefficient obtained by the proposed model is in agreement with the experimental result. Finally, numerical simulation has been completed to investigate the effects of the porosity, tortuosity, water-cement ratio, cement volume fraction and ion concentration in the pore solution on the ion diffusion coefficients. The results show that the ion diffusion coefficient in concrete increases with the porosity, water-cement ratio and cement volume fraction, while we see a decrease with the increasing of tortuosity. Meanwhile, the ion concentration produces more obvious effects on the diffusivity itself, but has almost no effects on the other ions.
Enhanced Lovastatin Production by Solid State Fermentation of Monascus ruber

The purpose of this study was to optimize the solid state cultivation of Monascus ruber on sterile rice. A single-level-multiple-factor and a single-factor-multiple-level experimental design were employed to determine the optimal medium constituents and to optimize carbon and nitrogen source concentrations for lovastatin production. Simultaneous quantitative analyses of the ${\beta}$-hydroxyacid form and ${\beta}$-hydroxylactone for of lovastatin were performed by the high performance liquid chromatography (HPLC) method with a UV photodiode-array (PDA) detector. The total lovastatin yield ($4{\sim}6\;mg/g$, average of five repeats) was achieved by adding soybean powder, glycerol, sodium nitrate, and acetic acid at optimized levels after 14 days of fermentation. The maximal yield of lovastatin under the optimal composition of the medium increased by almost 2 times the yield observed prior to optimization. The experimental results also indicated that the ${\beta}$-hydroxylactone form of lovastatin (LFL) and the ${\beta}$-hydroxyacid form of lovastatin (AFL) simultaneously existed in solid state cultures of Monascus ruber. while the latter was the dominant form in the middle-late stage of continued fermentation. These results indicate that optimized culture conditions can be used for industrial production of lovastatin to obtain high yields.

A psychrotrophic bacterium, Arthrobacter sp. ON14, isolated from Antarctica, was shown to exhibit a high β-galactosidase activity at a low temperature. A genomic library of ON14 was constructed and screened for β-galactosidase genes on functional plates containing 5-bromo-4-chloro-3-indolyl-β-D-galactopyranoside (X-gal) as the substrate. Two different β-galactosidase genes, named as galA, galB, were found in ON14. Computational analyses of the genes revealed that the encoded protein GalA belongs to family 2 of glycosyl hydrolysases and is a cold-active protein, whereas GalB belongs to family 42 of glycosyl hydrolysases and is a mesophilic protein. Reverse transcription analyses revealed that the expression of galA is highly induced at a low temperature (4℃) and repressed at a high temperature (28℃) when lactose is used as the sole carbon source. Conversely, the expression of galB is inhibited at a low temperature and induced at a high temperature. The purified GalA showed its peak activity at 15℃ and pH 8. The mineral ions Na+, K+, Mg2+, and Mn2+ were identified as enzyme activators, whereas Ca2+ had no influence on the enzyme activity. An enzyme stability assay revealed that the activity of GalA is significantly decreased when it is incubated at 45℃ for 2 h, and all its activity is lost when it is incubated at 50℃.
