        Chemical Constituents from Leaves of Pileostegia viburnoides Hook.f.et Thoms

        Phytochemical investigation on the leaves of Pileostegia viburnoides Hook.f.et Thoms led to the isolation of twenty-five compounds, and their structures were identified as n-dotriacontane (1), taraxeryl acetate (2), friedelin (3), epifriedelinol (4), canophyllal (5), stigmast-4-en-3-one (6), stigmasterol (7), (24R)-5A-stigmastane- 3,6-dione (8), ursolic acid (9), pomolic acid (10), umbelliferone (11), 4-epifriedelin (12), n-octatriacontanol (13), b-amyrin (14), a-amyrin (15), taraxerol (16), nonadecanol (17), friedelane (18), arachic acid (19), protocatechuic acid (20), n-pentatriacontanol (21), hexadecanoic acid (22), vincosamide (23), daucosterol (24), and skimming (25), respectively. To our best knowledge, compounds 1, 2, 12, 13, 17 - 19 and 21-23 were new within Saxifragaceae family. Compounds 15, 16, and 20 were produced from this genus for the first time. Compounds 4, 14 and 25 were first obtained from species P. viburnoides and compounds 3, 5 - 11, and 24 were achieved from the leaves of P. viburnoides for the first time. Furthermore, the anti-neuroinflammatory activity of these isolates was evaluated.

        Mechanisms of Extracellular NO and Ca2+ Regulating the Growth of Wheat Seedling Roots

        Our previous studies suggested the cross talk of nitric oxide (NO) with Ca2+ in regulating stomatal movement. However, its mechanism of action is not well defined in plant roots. In this study, sodium nitroprusside (SNP, a NO donor) showed an inhibitory effect on the growth of wheat seedling roots in a dose-dependent manner, which was alleviated through reducing extracellular Ca2+ concentration. Analyzing the content of Ca2+ and K+ in wheat seedling roots showed that SNP significantly promoted Ca2+ accumulation and inhibited K+ accumulation at a higher concentration of extracellular Ca2+, but SNP promoted K+ accumulation in the absence of extracellular Ca2+. To gain further insights into Ca2+ function in the NOregulated growth of wheat seedling roots, we conducted the patch-clamped protoplasts of wheat seedling roots in a whole cell configuration. In the absence of extracellular Ca2+,NO activated inward-rectifying K+ channels, but had little effects on outward-rectifying K+ channels. In the presence of 2 mmol L−1 CaCl2 in the bath solution, NO significantly activated outward-rectifying K+ channels, which was partially alleviated by LaCl3 (a Ca2+ channel inhibitor). In contrast,2 mmol L−1 CaCl2 alone had little effect on inward or outward-rectifying K+ channels. Thus, NO inhibits the growth of wheat seedling roots likely by promoting extracellular Ca2+ influx excessively. The increase in cytosolic Ca2+ appears to inhibit K+ influx, promotes K+outflux across the plasma membrane, and finally reduces the content of K+ in root cells.

        Default Prediction for Real Estate Companies with Imbalanced Dataset

        When analyzing default predictions in real estate companies, the number of non-defaulted cases always greatly exceeds the defaulted ones, which creates the twoclass imbalance problem. This lowers the ability of prediction models to distinguish the default sample. In order to avoid this sample selection bias and to improve the prediction model, this paper applies a minority sample generation approach to create new minority samples. The logistic regression, support vector machine (SVM) classification, and neural network (NN) classification use an imbalanced dataset. They were used as benchmarks with a single prediction model that used a balanced dataset corrected by the minority samples generation approach. Instead of using prediction oriented tests and the overall accuracy, the true positive rate (TPR), the true negative rate (TNR), G-mean, and F-score are used to measure the performance of default prediction models for imbalanced dataset. In this paper, we describe an empirical experiment that used a sampling of 14 default and 315 non-default listed real estate companies in China and report that most results using single prediction models with a balanced dataset generated better results than an imbalanced dataset.

      • Numerical simulation on the coupled chemo-mechanical damage of underground concrete pipe

        Long-termly used in water supply, an underground concrete pipe is easily subjected to the coupled action of pressure loading and flowing water, which can cause the chemo-mechanical damage of the pipe, resulting in its premature failure and lifetime reduction. Based on the leaching characteristics and damage mechanism of concrete pipe, this paper proposes a coupled chemo-mechanical damage and failure model of underground concrete pipe for water supply, including a calcium leaching model, mechanical damage equation and a failure criterion. By using the model, a numerical simulation is performed to analyze the failure process of underground concrete pipe, such as the time-varying calcium concentration in concrete, the thickness variation of pipe wall, the evolution of chemo-mechanical damage, the distribution of concrete stress on the pipe and the lifetime of the pipe. Results show that, the failure of the pipe is a coupled chemo-mechanical damage process companied with calcium leaching. During its damage and failure, the concentrations of calcium phase in concrete decrease obviously with the time, and it can cause an increase in the chemo-mechanical damage of the pipe, while the leaching and abrasion induced by flowing water can lead to the boundary movement and wall thickness reduction of the pipe, and it results in the stress redistribution on the pipe section, a premature failure and lifetime reduction of the pipe.

        Phase II Study of Preoperative Intra-Arterial Epirubicin, Etoposide, and Oxaliplatin Combined with Oral S-1 Chemotherapy for the Treatment of Borrmann Type 4 Gastric Cancer

        Purpose: A phase II study was conducted to evaluate the safety and efficacy of preoperative, intra-arterial perfusion of epirubicin, etoposide, and oxaliplatin combined with oral chemotherapy S-1 (SEEOX) for the treatment of type 4 gastric cancer. Materials and Methods: A single-center, single-arm phase II trial was conducted on 36 patients with histologically proven type 4 gastric cancer without distant peritoneal or organ metastasis. Patients received 3, 21-day courses of SEEOX preoperative chemotherapy. The primary endpoint was overall survival (OS) and the secondary outcomes assessed were chemotherapeutic response, radical resection rate, pathological regression, toxicities, postoperative morbidity, and mortality. Results: All patients were at an advanced stage of cancer (stage III or IV) and completed the entire course of treatment. Based on changes in tumor volume and peritoneal metastasis, the objective response rate was 55.6% (20/36; 95% confidence interval [CI], 38.5%-72.6%) and the disease control rate was 69.4% (25/36; 95% CI, 53.6%-85.3%). The radical resection rate was 75% (27/36; 95% CI, 60.1%-89.9%) and the proportion of R0 resections was 66.7% (21/36; 95% CI, 50.5%-82.8%). The pathological response rate was 33.3%, of which 13.9% showed complete pathological regression. The median survival was 27.1 months (95% CI, 22.24-31.97 months), and the 2-year OS was 48.5% (95% CI, 30.86%-66.1%). Conclusions: Preoperative SEEOX is a safe and effective treatment for type 4 gastric cancer. Based on these preliminary data, a phase III study will be conducted to confirm the superiority of this regimen over standard treatment.

        Knockdown of the olfactory co-receptor Orco impairsmate recognition in Tenebrio molitor (Coleoptera: Tenebrionidae)

        The yellow mealworm beetle, Tenebrio molitor (Coleoptera: Tenebrionidae), is a common insect pest that damages stored grain. The insect olfactory co-receptor (Orco) is a transmembrane protein localized on the surface of the dendrites of olfactory sensory neurons and plays an essential role in the olfactory perception. In this study, an Orco ortholog (named as Tmol\Orco) inT.molitorwas characterized. The deduced amino acid sequence of Tmol\Orco shared significant high identities with Orcos in other insect species from distinct Orders. Reverse transcription quantitative PCR (RT-qPCR) analysis showed that Tmol\Orco was predominantly expressed in the antennae of both male and female adults, and the difference between male antennae and female antennae was not significant. Double-stranded RNA (dsRNA) directly against the Tmol\Orco mRNA was synthesized and injected into the T. molitor male adults, RT-qPCR results demonstrated that the transcription level of Tmol\Orco was significantly reduced at 6 d after the injection. The mate-seeking behavior of Tmol\Orco-silenced beetles was significantly inhibited. Compare to the control individuals, the percentage of Tmol\Orco-silenced beetles successfully find mateswas significantly decreased and the average time they spent for searching were significantly increased. These results provided first direct evidence that suppression of Tmol\Orco transcription contributes to the decline in mate recognition of T. molitor

        Anti-inflammatory Activities of Lupane-triterpenoids In Vitro and Their Phytochemical Fingerprinting from Leaves of Acanthopanax gracilistylus

        The activities on the inhibition of NO on LPS-induced RAW 264.7 macrophages were investigated in this work. A simple and sensitive method has been developed and validated for fingerprinting analysis of leaves of Acanthopanax gracilistylus W.W. Smith (AGS). The cytotoxicity and inhibition of NO on LPS-induced RAW 264.7 cells of the extract and triterpenoids were determined. Optimal conditions of HPLC analysis were established as follows. The separation was performed with an ODS-C18 column at 30 oC, the detected wavelength was 210 nm, the flow rate was 1 mL/min, and the mobile phase consisted of acetonitrile (0.05% phosphoric acid) -0.05% phosphoric acid solution with gradient elution. Our results showed that impressic acid and acankoreaogenin was more effective on the inhibition of NO than the methanol extract and other compounds. There were seventeen peaks coexisted with similarities above 0.95 and nine lupane-triterpenoids including acankoreaogenin and impressic acid detected and identified. The result of anti-inflammatory activities provides a potential explanation for the use of AGS leaves as a herbal medicine in the treatment of inflammatory diseases. Our results also show that acankoreanogenin and impressic acid may be potentially useful in developing new anti-inflammatory agents. In addition, the fingerprint chromatography clearly illustrated and confirmed the material basis for the anti-inflammatory activities of this plant.

        Molecular cloning and characterization of two novel DREB genes encoding dehydration-responsive element binding proteins in halophyte Suaeda salsa

        The dehydration-responsive element-binding(DREB) proteins play an important role in regulatingexpression of stress-inducible genes under abiotic stresses. In this study, two genes encoding putative DREB proteins,named SsDREBa and SsDREBb, were cloned from halophyteSuaeda salsa L. using RACE method. The deducedSsDREBa and SsDREBb proteins contain a typical AP2/ERF domain. Multiple sequence alignments and phylogeneticanalysis revealed that the two SsDREB genes of S. salsa were highly similar in AP2/ERF domains at thenucleotide and amino acid levels and belong to the A-6subgroup of the DREB transcription factor subfamily. Asubcellular localization assay showed that both SsDREBslocalized to the nucleus. Yeast one-hybrid experimentstestified that both proteins were able to specifically bind tothe DRE sequence and activate the expression of the downstreamHIS reporter gene in yeast. Quantitative real-timePCR analysis demonstrated that under normal conditions,the expression level of SsDREBa was the most high in theroots and no SsDREBa mRNAs were detected in the stems;SsDREBb expressed at relatively higher levels in the leavesthan in the roots and stems. The expression of SsDREa andSsDREBb genes in S. salsa roots and leaves was remarkablyinduced by high-salt and dehydration treatments, butnot by cold and ABA, and exhibited stronger induction inroots and leaves, respectively. These results indicate thatthe SsDREBa and SsDREBb are novel stress-responsivetranscription factors, which are involved in the drought andhigh-salt stress responses through ABA-independent pathwaysand could be used for production of stress-toleranttransgenic crops.

        Molecular Characterization of Cold-Inducible β-Galactosidase from Arthrobacter sp. ON14 Isolated from Antarctica

        A psychrotrophic bacterium, Arthrobacter sp. ON14, isolated from Antarctica, was shown to exhibit a high β-galactosidase activity at a low temperature. A genomic library of ON14 was constructed and screened for β-galactosidase genes on functional plates containing 5-bromo-4-chloro-3-indolyl-β-D-galactopyranoside (X-gal) as the substrate. Two different β-galactosidase genes, named as galA, galB, were found in ON14. Computational analyses of the genes revealed that the encoded protein GalA belongs to family 2 of glycosyl hydrolysases and is a cold-active protein, whereas GalB belongs to family 42 of glycosyl hydrolysases and is a mesophilic protein. Reverse transcription analyses revealed that the expression of galA is highly induced at a low temperature (4℃) and repressed at a high temperature (28℃) when lactose is used as the sole carbon source. Conversely, the expression of galB is inhibited at a low temperature and induced at a high temperature. The purified GalA showed its peak activity at 15℃ and pH 8. The mineral ions Na+, K+, Mg2+, and Mn2+ were identified as enzyme activators, whereas Ca2+ had no influence on the enzyme activity. An enzyme stability assay revealed that the activity of GalA is significantly decreased when it is incubated at 45℃ for 2 h, and all its activity is lost when it is incubated at 50℃.
