Molecular characterization of chemosensory protein genes in Bactrocera minax (Diptera: Tephritidae)
Wang Zhao‐Xiang,Qi Zhen‐hua,Chen Jian,Wang Fu‐Lian,Gui Lian‐You,Zhang Guo‐Hui 한국곤충학회 2021 Entomological Research Vol.51 No.7
Chemoreception is of great importance for survival of insects. Chemosensory proteins (CSPs) are believed to be involved in the perireceptor events of chemosensory system in many insect species, but this has not been clarified in the citrus fruit fly, Bactrocera minax, a serious dipteran pest of citrus crops. Previous studies indicated that four CSP genes were identified in many fly speices in Diptera. In the present study, we also identified four CSP genes in B. minax, namely BminCSP1–4, from a transcriptome database. All CSP proteins encoded by these genes bear the typical hallmarks of the CSP family: an N-terminal signal peptide and the four highly conserved cysteine residues. Phylogenetic analysis comparing with other dipteran CSPs indicated that dipteran CSPs may evolved from three ancestral CSP genes, and revealed the sequence diversities of BminCSPs and showed that BminCSP1–4 are clustered in separate groups, indicating the possibility of their contrasting function in B. minax. Furthermore, the tissue distribution of the four CSP genes in adult B. minax was analyzed by real-time quantitiative PCR. The results demonstrated that BminCSP3 are significantly transcriptionally enriched in antennae; BminCSP4 expressed primarily in heads; BminCSP1 and BminCSP2 showed high expressions in different tissues, such as antennae, abdomens and wings. Based on these findings, the different implications for the functions of BminCSPs are discussed. This study will offer a significant indication for further functional studies of the CSPs in B. minax

A psychrotrophic bacterium, Arthrobacter sp. ON14, isolated from Antarctica, was shown to exhibit a high β-galactosidase activity at a low temperature. A genomic library of ON14 was constructed and screened for β-galactosidase genes on functional plates containing 5-bromo-4-chloro-3-indolyl-β-D-galactopyranoside (X-gal) as the substrate. Two different β-galactosidase genes, named as galA, galB, were found in ON14. Computational analyses of the genes revealed that the encoded protein GalA belongs to family 2 of glycosyl hydrolysases and is a cold-active protein, whereas GalB belongs to family 42 of glycosyl hydrolysases and is a mesophilic protein. Reverse transcription analyses revealed that the expression of galA is highly induced at a low temperature (4℃) and repressed at a high temperature (28℃) when lactose is used as the sole carbon source. Conversely, the expression of galB is inhibited at a low temperature and induced at a high temperature. The purified GalA showed its peak activity at 15℃ and pH 8. The mineral ions Na+, K+, Mg2+, and Mn2+ were identified as enzyme activators, whereas Ca2+ had no influence on the enzyme activity. An enzyme stability assay revealed that the activity of GalA is significantly decreased when it is incubated at 45℃ for 2 h, and all its activity is lost when it is incubated at 50℃.

Background: A number of studies have reported the association of X-ray repair cross-complementing group 1 (XRCC1) Arg399Gln polymorphism with susceptibility to hepatocellular carcinoma (HCC). However, the results were inconsistent and inconclusive. The aim of this study was to comprehensively explore the association of XRCC1 Arg399Gln variant with HCC risk. Materials and Methods: Systematic searches of PubMed, Elsevier, Science Direct, CNKI and Chinese Biomedical Literature Database were performed. Pooled odds ratio (OR) with 95% confidence intervals (CI) was calculated to estimate the strength of association. Results: Overall, we observed an increased HCC risk among subjects carrying XRCC1 codon 399 Gln/Gln, Arg/Gln and Gln/Gln+Arg/Gln genotypes (OR=1.20, 95%CI: 1.05-1.38, OR=1.16, 95%CI: 1.05-1.28, and OR=1.14, 95%CI: 1.04-1.24, respectively) based on 20 studies including 3374 cases and 4633 controls. In subgroup analysis, we observed an increased risk of XRCC1 codon 399 Gln/Gln, Arg/Gln and Gln/Gln+Arg/Gln polymorphisms for HCC in hospital-based study (OR=1.25, 95%CI: 1.03-1.51, OR=1.21, 95%CI: 1.07-1.36 and OR=1.18, 95%CI: 1.06-1.31, respectively) and in Asian population (OR=1.19, 95%CI: 1.03-1.38, OR=1.17, 95%CI: 1.04-1.30 and OR=1.14, 95%CI: 1.04-1.25, respectively). Limiting the analysis to the studies with controls in agreement with Hardy-Weinberg equilibrium (HWE), we observed an increased HCC risk among Gln/Gln, Arg/Gln and Gln/ Gln+Arg/Gln genotype carriers (OR=1.17, 95%CI: 1.05-1.29, OR=1.12, 95%CI: 1.00-1.25 and OR=1.11, 95%CI: 1.02-1.21, respectively). Conclusions: This updated meta-analysis results suggest that XRCC1 Arg399Gln variants may contribute to HCC risk. Well-designed studies with larger sample size were required to further verify our findings.

Mussels are major fouling organisms causing serious technical and economic problems. In this study, antifouling activity towards mussel was found in three compounds isolated from a marine bacterium associated with the sea anemone Haliplanella sp. This bacterial strain, called PE2, was identified as Vibrio alginolyticus using morphology, biochemical tests, and phylogenetic analysis based on sequences of 16S rRNA and four housekeeping genes (rpoD, gyrB, rctB, and toxR). Three small-molecule compounds (indole, 3-formylindole, and cyclo (Pro-Leu)) were purified from the ethyl acetate extract of V. alginolyticus PE2 using column chromatography techniques. They all significantly inhibited byssal thread production of the green mussel Perna viridis, with EC50 values of 24.45 μg/ml for indole, 50.07 μg/ml for 3-formylindole, and 49.24 μg/ml for cyclo (Pro-Leu). Previous research on the antifouling activity of metabolites from marine bacteria towards mussels is scarce. Indole, 3-formylindole and cyclo (Pro-Leu) also exhibited antifouling activity against settlement of the barnacle Balanus albicostatus (EC50 values of 8.84, 0.43, and 11.35 μg/ml, respectively) and the marine bacterium Pseudomonas sp. (EC50 values of 42.68, 69.68, and 39.05 μg/ml, respectively). These results suggested that the three compounds are potentially useful for environmentally friendly mussel control and/or the development of new antifouling additives that are effective against several biofoulers.
Collision characteristics of droplet pairs with the presence of arriving distance differences
Xiang Wang,Zhaomiao Liu,Yan Pang 한국공업화학회 2019 Journal of Industrial and Engineering Chemistry Vol.69 No.-
Four collision behaviors are observed for droplet pairs at a T-junction and phase diagrams are constructed by considering the arriving distance difference, the droplet length and the capillary number. Critical conditions for both coalescence and splitting are shown. The main coalescence regime is varied for asynchronous droplet pairs and the cause of the coalescence which occurs while two droplets are pulled apart is confirmed. Movements of the front caps of various droplet pairs are exhibited to show their different degrees of competitions to penetrate into the downstream channel and the advantage that favors coalescence is acquired by comparison.

This paper presents a compression-friendly low power test scheme in EDT environment. The proposed approach exploits scan slices reusing to reduce the switching activity during shifting for test scheme based on linear decompressor. To avoid the impact on encoding efficiency from resulting control data, a counter is utilized to generate control signals. Experimental results obtained for some larger ISCAS’89 and ITC’99 benchmark circuits illustrate that the proposed test application scheme can improve significantly the encoding efficiency of linear decompressor.

Xiang Wang,Xiaoming Zhang,Mei Li 보안공학연구지원센터 2015 International Journal of u- and e- Service, Scienc Vol.8 No.10
Sensor Network has been widely used in all fields, and a large number of heterogeneous data is produced. In order to make these data shared and integrated semantically, semantic web technology is introduced in the sensor network. In this paper, we make a comprehensive survey about Semantic Sensor Web (SSW) from the perspective of ontology, mapping and query respectively. Firstly, we present state of the art of SSW from three aspects: ontologies before SSN ontology, SSN ontology and the extensions of SSN ontology, in which the extensions of SSN ontology are introduced emphatically. Secondly, we discuss the mapping from the stream data to ontology and focus on the comparison of the mapping languages in order to find out the best mapping method of SSW. Thirdly, we describe the research status of stream data query languages and analyze the characteristics, the application fields, the related researches and tools of the query languages in detail. Finally, the future research directions of SSW are proposed.
