<P>Brassinosteroids (BRs) are essential phytohormones regulating various developmental and physiological processes during normal growth and development. cog1-3D (cogwheel1-3D) was identified as an activation-tagged genetic modifier of bri1-5, an intermediate BR receptor mutant in Arabidopsis (Arabidopsis thaliana). COG1 encodes a Dof-type transcription factor found previously to act as a negative regulator of the phytochrome signaling pathway. cog1-3D single mutants show an elongated hypocotyl phenotype under light conditions. A loss-of-function mutant or inducible expression of a dominant negative form of COG1 in the wild type results in an opposite phenotype. A BR profile assay indicated that BR levels are elevated in cog1-3D seedlings. Quantitative reverse transcription-polymerase chain reaction analyses showed that several key BR biosynthetic genes are significantly up-regulated in cog1-3D compared with those of the wild type. Two basic helix-loop-helix transcription factors, PIF4 and PIF5, were found to be transcriptionally up-regulated in cog1-3D. Genetic analysis indicated that PIF4 and PIF5 were required for COG1 to promote BR biosynthesis and hypocotyl elongation. Chromatin immunoprecipitation and electrophoretic mobility shift assays indicated that COG1 binds to the promoter regions of PIF4 and PIF5, and PIF4 and PIF5 bind to the promoter regions of key BR biosynthetic genes, such as DWF4 and BR6ox2, to directly promote their expression. These results demonstrated that COG1 regulates BR biosynthesis via up-regulating the transcription of PIF4 and PIF5.</P>
Fyn promotes proliferation, differentiation, survival and function of osteoclast lineage cells

<P><B>Abstract</B></P><P>c‐Src and Lyn are the only Src family kinases (SFKs) with established activity in osteoclasts (OCs). c‐Src promotes function via cytoskeletal organization of the mature resorptive cell while Lyn is a negative regulator of osteoclastogenesis. We establish that Fyn, another SFK, also impacts the OC, but in a manner distinctly different than c‐Src and Lyn. Fyn deficiency principally alters cells throughout the osteoclastogenic process, resulting in diminished numbers of resorptive polykaryons. Arrested OC formation in the face of insufficient Fyn reflects reduced proliferation of precursors, in response to M‐CSF and retarded RANK ligand (RANKL)‐induced differentiation, attended by suppressed activation of the osteoclastogenic signaling molecules, c‐Jun, and NF‐κB. The anti‐apoptotic properties of RANKL are also compromised in cells deleted of Fyn, an event mediated by increased Bim expression and failed activation of Akt. The defective osteoclastogenesis of Fyn−/− OCs dampens bone resorption, in vitro. Finally, while Fyn deficiency does not regulate basal osteoclastogenesis, in vivo, it reduces that stimulated by RANKL by ∼2/3. Thus, Fyn is a pro‐resorptive SFK, which exerts its effects by prompting proliferation and differentiation while attenuating apoptosis of OC lineage cells. J. Cell. An Isolated Microlens Observed from <i>K2</i> , <i>Spitzer</i> , and Earth

<P>We present a combined analysis of the observations of the gravitational microlensing event OGLE-2015-BLG-0479 taken both from the ground and by the Spitzer Space Telescope. The light curves seen from the ground and from space exhibit a time offset of similar to 13 days between the caustic spikes, indicating that the relative lens-source positions seen from the two places are displaced by parallax effects. From modeling the light curves, we measure the space-based microlens parallax. Combined with the angular Einstein radius measured by analyzing the caustic crossings, we determine the mass and distance of the lens. We find that the lens is a binary composed of two G-type stars with masses of similar to 1.0 M-circle dot and similar to 0.9 M-circle dot located at a distance. of similar to 3 kpc. In addition, we are able to constrain the complete orbital parameters of the lens thanks to the precise measurement of the microlens parallax derived from the joint analysis. In contrast to the binary event OGLE-2014-BLG-1050, which was also observed by Spitzer, we find that the interpretation of OGLE-2015-BLG-0479 does not suffer from the degeneracy between (+/-, +/-) and (+/-, -/+) solutions, confirming that the four-fold parallax degeneracy in single-lens events collapses into the two-fold degeneracy for the general case of binary-lens events. The location of the blend in the color-magnitude diagram is consistent with the lens properties, suggesting that the blend is the lens itself. The blend is bright enough for spectroscopy and thus this possibility can be checked from future follow-up observations.</P>
Expression Analysis of the Mx Gene and Its Genome Structure in Chickens

Among the known interferon-induced antiviral mechanisms, the Mx pathway is one of the most powerful pathways. The Mx protein has direct antiviral activity and inhibits a wide range of viruses by blocking an early stage of the viral replication cycle. Cloning, characterization, and expression of Mx in vivo and in vitro have been conducted. The chicken Mx gene spans 21 kb and is made up of 14 exons and 13 introns, of which the promoter region was analyzed. The real-time PCR results showed that Mx expression was increased in chicken embryo fibroblasts (CEF) after 12- and 24-h induction with polyI: C. Induction of Mx expression by poly I: C in vivo revealed tissue-specific patterns among the chicken tissues tested. A trace expression of Mx was detected in healthy chicken liver tissues from adult chickens without inducement; the expression levels in the liver, heart, and gizzard were higher than in the muscle and kidney. This is the first report to demonstrate the expression of a glutathione-S-transferase-tagged-Mx fusion protein of 75 KDa, as well as the biological activity tested by SDS-PAGE and western blotting.

<P>In this paper, we present an analysis of the binary gravitational microlensing event OGLE-2015-BLG-0196. The event lasted for almost a year, and the light curve exhibited significant deviations from the lensing model based on the rectilinear lens-source relative motion, enabling us to measure the microlens parallax. The ground-based microlens parallax is confirmed by the data obtained from space-based microlens observations using the Spitzer telescope. By additionally measuring the angular Einstein radius from the analysis of the resolved caustic crossing, the physical parameters of the lens are determined up to the twofold degeneracy, u(0) < 0 and u(0) > 0, solutions caused by the well-known 'ecliptic' degeneracy. It is found that the binary lens is composed of two M dwarf stars with similar masses, M-1 = 0.38 +/- 0.04M(circle plus) (0.50 +/- 0.05M(circle plus)) and M-2 = 0.38 +/- 0.04M(circle plus) (0.55 +/- 0.06M(circle plus)), and the distance to the lens is D-L = 2.77. +/- 0.23 kpc (3.30 +/- 0.29 kpc). Here the physical parameters outside and inside the parentheses are for the u(0) < 0 and u(0) > 0 solutions, respectively.</P>

<P>We report the first mass and distance measurements of a caustic-crossing binary system OGLE-2014-BLG-1050 L using the space-based microlens parallax method. Spitzer captured the second caustic. crossing of the event, which occurred similar to 10 days before that seen from Earth. Due to the coincidence that the source-lens relative motion was almost parallel to the direction of the binary-lens axis, the fourfold degeneracy, which was known before only to occur in single-lens events, persists in this case, leading to either a lower-mass (0.2 and 0.07 M-circle dot) binary at similar to 1.1 kpc or a higher-mass (0.9 and 0.35 M-circle dot) binary at similar to 3.5 kpc. However, the latter solution is strongly preferred for reasons including blending and lensing probability. OGLE-2014-BLG-1050 L demonstrates the power of microlens parallax in probing stellar and substellar binaries.</P>
Fabrication of p-Type Nitrogen-Doped MgZnO by Depressing N-Related Donors

Wurtzite nitrogen-doped MgZnO (MgZnO:N) films were grown on c-plane sapphire by using plasma-assisted molecular beam epitaxy with radical NO as the oxygen source and nitrogen dopant. P-type conduction of MgZnO:N was obtained by decreasing the donor defects ((N2)O, VO, etc.) through annealing, revealing a hole concentration of 6.1 × 1017 cm−3 and a mobility of 6.42 cm2/Vs. Furthermore, as-grown p-type films with a hole concentration of 1 × 1017 cm−3 and a mobility of 3 cm2/Vs were obtained by decreasing the (N2)O double donor defect through control of the plasma conditions. ZnMgO:N/ZnO p-n junctions were obtained by using these p-type films. ode-like, rectifying I-V characteristics with a threshold voltage of about 5 V and a different reverse leakage current were observed at room temperature.

<P>The new tuberculosis (TB) lead ecumicin (<B>1</B>), a cyclic tridecapeptide, was isolated from <I>Nonomuraea sp.</I> MJM5123, following a high-throughput campaign for anti-TB activity. The large molecular weight of 1599 amu detected by LC-HR-MS precluded the initial inference of its molecular formula. The individual building blocks were identified by extensive NMR experiments. The resulting two possible planar structures were distinguished by LC-MS<SUP>2</SUP>. Effect of surface modification layer thickness on the gas sensitivity of SnO2 films

Nickel was used as a surface modification material to enhance the gas-sensing properties of SnO2 films. The thickness of the Ni modification layer was controlled by the sputtering time. The surface morphology and surface chemical composition of the surface modification layers of different thicknesses were analyzed by Field Emission Scanning Electron Microscopy (FESEM) and X-ray Photoelectron Spectroscopy (XPS), respectively. The gas sensitivity to clean air with a low hydrogen concentration of 1000 ppm of the modified tin dioxide thin films was investigated at temperatures from 20oC to 100oC. The results show that the thickness of the modification layer plays an important role in the enhancement of the gas-sensing properties and surface electric properties of SnO2 films. A Ni modification layer with a thickness of 50 nm on the surface of a SnO2 film can improve the hydrogen gas sensitivity of a SnO2 film up to 112%, and reduce the response time to 80s.
