This study was conducted to have basic and applied informations to establish the cultivation method and to improve the cultivation techniques of Angelica gigas Nakai through investigating the rates of germination and seedling establishment with treatment of plant growth regulator and seed priming. The germination rates of round shape seed were about 0.3~12% higher than those of parallel shape. The germination rates with different storage durations were increased at the level of 25oC and 4oC with the storage days, while they were clearly higher at -20oC compared to those at 4oC and 25oC without the storage durations. With the results of germination rates with treating of plant growth regulator and seed priming, their rates with the round shape seeds were comparatively higher about 6% than those with parallel shape seed and they were increased with increasing of the storage durations. The germination rates were generally appeared higher with GA3 than did those with seed priming. The highest germination rate was appeared at 50 ppm GA3 of plant growth regulator and at -0.5 MPa PEG6000 of seed priming. The germination rates?ㅤ ?ㅤ of Angelica gigas were different with different temperature, storage durations and the treatments of plant growth regulator and seed priming. These results could be used according to different cultivating conditions.
Research Notes : Detection of Extracellular Enzyme Activities in Ganoderma neo-japonicum

The ability of Ganoderma to produce extracellular enzymes, including β-g1ucosidase, cellulase, avicelase, pectinase, xylanase, protease, amylase, and ligninase was tested in chromogenic media, β-glucosidase showed the highest activity, among the eight tested enzymes. In particular, Ganoderma neo-japonicum showed significantly stronger activity for β-glucosidase than that of the other enzymes. Two Ganoderma lucidum isolates showed moderate activity for avicelase; however, Ganoderma neo-japonicum showed the strongest activity. Moderate ligninase activity was only observed in Ganoderma neo-japonicum. In contrast, pectinase, amylase, protease, and cellulase were not present in Ganoderma. The results show that the degree of activity of the tested enzymes varied depending on the Ganoderma species tested.

To determine the optimal media conditions for the detection of the cellulolytic activity in Ganoderma lucidum, we varied three media conditions: dye reagent, pH, and temperature. First, we evaluated the use of four dyes, Congo Red, Phenol Red, Remazol Brilliant Blue, and Trypan Blue. To observe the effect of pH on the chromogenic reaction, we also made and tested various media spanning acidic and alkaline pHs, ranging from 4.5 to 8.0. Furthermore, in order to research the effect of temperature on the clear zone and the fungus growing zone, we tested temperatures ranging from 15 to 35℃. On the whole, the best protocol called for Ganoderma lucidum transfer onto media containing Congo red with pH adjusted to 7.0, followed by incubation at 25℃ for 5 days. Our results will be useful to researchers who aim to study extracellular enzyme activity in Ganoderma lucidum

Ganoderma applantaum belonged to Polyporaceae of Basidiomycetes and has been known as one of the popular medicinal mushrooms due to high antitumor and anti-diabetes activity. This study was carried out to obtain the basic information for mycelial culture conditions of Ganoderma applantaum. According to colony diameter and mycelial density, the media for suitable mycelial growth of them were shown in PDA and MEA. The optimum temperature for mycelial growth was 25~30℃The colonies are white, flocculose and underside of the plates is whitish to cream. Carbon and nitrogen sources were mannose and malt extract, respectively. The optimum C/N ratio was 10:1 to 5:1with 2% glucose concentration, vitamin was thiamine- HCl, organic acid was succinic acid, and mineral salt was MgSO4ㆍ7H2O. Present experiments were conducted to determine the possibility of artificial culture with sawdusts of G. applantaum. The pH value which was 6.0 of oak sawdust and6.4 of ricebran. Mycelial density on oak sawdust of G.applantaum spp. was higher than Ganoderma lucidium (a control). Wt. of dried fruiting body (g) of G. applantaum. (origin culture: GBEA-01, strain name: SamJung(三淨)) showed that 16.0 g on oak sawdust bottle cultivation (850㎖), 7.8 g on oak sawdust and G. lucidum. This present study was undertaken to investigate comparative anti-tumor activity of water extracts of G. lucidum ASI 7125 (GLE), G. applantaum SamJung (GAE) in vitro. The anti-tumor activity in the present study was evaluated by sulforhodamine B (SRB) and microtetrazolium (MTT) assay in terms of cell survival level. The results showed that DW, GLE and GAE inhibited proliferation showing both tumor cells.

To elucidate structure of group "a" epitope, mouse antibodies that express idiotype monoclonal antibody and anti-idiotype monoclonal antibody against the group specific "a" determinant were purified by hydroxyapatite column. To obtain hepatitis B surface antigens (HBsAg). HBsAg positive blood was sequencially purified by ammonium sulfate precipitation, hydroxyapatite, sepharose 4B column chromatography and ultracentrifugation. The major protein (p25) and glycoprotein (gp30) of HBsAg were isolated by concanavalin-A-sepharose 4B. The ability of p25-gp30 among the HBsAg to inhibit the idiotype-anti-idiotype reaction was dependent on conformation, since reduced and alkylated p25-gp30 virtualy lost their inhibitory capacity when compared to native HBsAg. The data suggest that hepatitis B antigen is a conformational antigen critically dependent upon the disulfide bonds of p25-gp30.
