        Evaluation of the Physiological Quality of Wheat Seed as Influenced by High Parent Plant Growth Temperature

        To examine the effect of high temperature during seed development and maturation on seed quality of wheat, seeds of four heat-tolerant and two heat-sensitive wheat genotypes obtained normal and high temperature growing conditions were tested for quality. High parent plant growth temperature during seed development and maturation resulted in poor seed quality. The effect of parent plant growth temperature on seed germination and speed of germination was inconsistent between heat tolerant and heat-sensitive wheat genotypes. Seed vigor as indicated by seed density, conductivity of seed leachates, seedling dry weight, production of normal seedlings, seed reserve utilization efficiency, and seedling emergence were reduced in all wheat genotypes due to higher parent plant growth temperature but heat-sensitive wheat genotypes were affected more than the heat-tolerant genotypes.

        Isolation and identification of infectious bursal disease virus from broiler and layer chickens during the outbreak year 2007 in Bangladesh

        The objective of the present study was to isolate and identify infectious bursal disease viruses (IBDVs) from broiler and layer chickens of outbreaks of infectious bursal disease (IBD) in three districts of Bangladesh. A total of 70 bursal samples were collected from dead broiler (n=40) and layer (n=30) chickens showing specific lesions of IBD from seven commercial poultry farms of three different districts (Mymensingh, Chittagong and Tangail) of Bangladesh during the year 2007. Five representative bursal samples from each farm were used for the isolation of IBDVs using 9-day-old embryonated eggs of seronegative flock of layer birds and for identification the samples were subjected to agar gel immunodiffusion test (AGIDT), immunohistochemistry (IHC) and reverse transcription-polymerase chain reaction (RT-PCR). Out of 35 bursal samples, IBDVs were successfully isolated from 28 (80%) samples. By AGIDT, 32 (91.4%) samples were found positive for IBDV antigen. Results of AGIDT clearly indicated that IBDVs detected in 29 bursal samples of six affected farms were identical to each other but not to IBDVs present in the remaining three samples of another farm. Indirect immunoperoxidase staining of the bursal sections revealed the presence of IBDV antigen in 32 (91.4%) samples and the IBDV antigen was detected mainly in the cortex of the lymphoid follicles of the bursal tissues. In histopathology, cell depletion, atrophy and necrosis were observed in many bursal follicles with severe edema of interfollicular septa. Of the 35 bursal samples, 34 (97.1%) samples generated 254 bp product by RT-PCR. In conclusion, the results of virus isolation and identification by AGIDT, IHC and the analysis of viral genome by RT-PCR confirmed the outbreaks of acute IBD in commercial poultry of Bangladesh. Moreover, histopathological findings and results of AGIDT gave a clear indication that the isolates from six outbreaks were different from classical strain and it seems to be of very virulent strain. On the other hand, the isolates from the other outbreak were similar to the classical strain.

        Effect of a Probiotic Strain, Enterococcus faecium, on the Immune Responses of Olive Flounder (Paralichthys olivaceus)

        The present study was aimed to investigate the effect of a probiotic, Enterococcus faecium, on the immune responses against infection with the marine fish pathogen Lactococcus garvieae in olive flounder (Paralichthys olivaceus). The immune responses were assessed by lysozyme activity, complement activity, protease activity, and expression of proinflammatory cytokines by RT-PCR. The lysozyme and complement activities were increased between 9 to 15 and 9 to 13 days, respectively, and antiprotease activity was slightly elevated after 5 days of probiotic treatment. The TNF-α and IL-1β expressions were observed from kidney and spleen. The results of this study reveal that E. faecium induces immune-responsible materials and protects olive flounder from lactococcosis.

        Species-Specific Duplex PCR for Detecting the Important Fish Pathogens Vibrio anguillarum and Edwardsiella tarda

        Vibriosis caused by Vibrio anguillarum and edwardsiellosis caused by Edwardsiella tarda are septicemic diseases of many commercially important freshwater and marine fishes, and threaten the aquaculture industry in Korea. Early diagnosis and accurate identification of these two bacterial species could help to prevent these diseases and minimize the damage to cultured marine species. This study designed a duplex polymerase chain reaction (PCR) method for the simultaneous detection of two major fish pathogens: V. anguillarum and E. tarda. Each pair of oligonucleotide primers exclusively amplified the target groEL gene of the specific microorganism. Twenty-two Vibrio and ten non-Vibrio enteric species were used to check the specificity of the primers, which were found to be highly specific for the target species, even among closely related species. The detection limit was 400 pg for V. anguillarum and 4 ng for E. tarda when mixed purified DNA was used as the template. This assay showed high specificity and sensitivity in the simultaneous detection of V. anguillarum and E. tarda from artificially inoculated seawater and fish.

        Alleviation of Drought Stress in Maize by Exogenous Application of Gibberellic Acid and Cytokinin

        The study was conducted to investigate the ameliorative roles of $GA_3$ and CK on adverse effects of drought in maize. Drought stressed maize plants were applied with $GA_3$ and CK at 50, 100, and $150mg\;L^{-l}$ as foliar spray at the vegetative and the reproductive stages. Plant height, internode length, stem diameter, leaf chlorophyll index, and dry matter production were significantly affected by drought. In most cases, $GA_3$ and CK significantly improved the depressed plant traits, but in varying degrees depending on the growth stage encountering hormones, and their types and concentrations. Both $GA_3$ and CK were found to be very effective in alleviating drought-imposed adverse effects on maize at the vegetative phase. Such alleviating effects varied depending on the concentration of the hormones. Application of CK at $150mg\;L^{-l}$ was excellent resulting in a 106% yield advantage compared to drought stress and 79.9% increase relative to well-watered controls. Conversely, $GA_3$ at $50mg\;L^{-l}$ performed well showing 78.8% increase in grain yield. However, both $GA_3$ and CK had very little effect on improving the depressed growth and yield attributes in maize at the reproductive phase. The relative yield advantages for the hormones were mainly attributed to improving the cob and seed-bearing capacity of drought-stressed maize plants.
