        Production and characterization of monoclonal antibodies against an avian influenza virus (H9N2)

        In this report, fifteen monoclonal antibodies (MAbs) against an avian influenza virus (H9N2 subtype) were newly produced and characterized. These MAbs proved to react to the epitopes of nucleocapsid protein (NP), hemagglutinin (HA), neuraminidase (NA) and non-structural protein 1 (NS1) of Korean H9N2 strain, respectively. Two HA-specific MAbs showed the ability to inhibit the hemagglutination activity of H9N2 subtype avian influenza virus when tested by hemagglutination inhibition (HI) assay. All MAbs did not cross-react with other avian-origin viruses (Newcastle disease virus, infectious bursal disease virus, infectious bronchitis virus and avian rotavirus) by immunofluorescence test or enzyme-linked immunosorbent assay. The MAbs produced in this study could be useful as the materials for diagnostics and therapeutics against Korean-lineage H9N2 virus infections.

        Identification of Korean native cattle persistently infected with BVDV using Ear-notch method

        Bovine viral diarrhea Virus (BVDV) infections cause respiratory, gastrointestinal, and reproductive problems, such as infertility, abortion, stillbirth, and sickly offspring. Many countries have reduced the economic damage through the application of different control programmes, and some have successfully eradicated BVD. Detection and elimination of cattle persistently infected (PI) with BVDV is important for BVD eradication because PI cattle are a main source of BVD transmission. In this study, the prevalence of Korean native cattle persistently infected (PI) with BVDV was investigated and determined in 49 farms with 3,050 cattle. The all samples were collected by ear notch sampling. Korean native cattle with initial positives on antigen-ELISA (Ag-ELISA) were sampled again after 3~4 weeks and cattle with second positives in both Ag-ELISA and immunohistochemistry (IHC) were identified as PI cattle. Among the 49 farms, 14 (28.6%) farms had at least more than one PI cow and 21 (0.69%) of 3,050 cattle were determined as PI cattle. As a result of this work, it is suggested that national BVD eradication program is required to reduce economic losses by BVDV infection in Korean cattle industries.

        Seroprevalence of specific Brucella infection of cattle in Bangladesh Agricultural University Veterinary Clinics and its surrounding areas

        A cross sectional survey was conducted to determine the seroprevalence of brucellosis in cattle in Bangladesh Agricultural University (BAU) Veterinary Clinics, in BAU Dairy Farm and Vabokhali from June 2008 to November 2008. A total of 200 serum samples were collected from BAU Veterinary Clinic, from BAU Dairy Farm and Vabokhali. Among the serum samples 143 sera samples were collected from BAU Veterinary Clinic, 42 serum samples from BAU Dairy Farm and 15 serum samples from Vabokhali. Sera were separated from blood samples and tested with specific Brucella abortus antigen (BAA) test and B. melitensis antigen (BMA) test. The overall seroprevalence of brucellosis in cattle was 5% in BAA and 0.5% in BMA. It was observed that, a significant higher prevalence of B. abortus was found in female than male. An insignificant higher prevalence of brucellosis was found in adult cattle (aged above 5 years), in cross breed cattle, in cattle with grazing, cattle breed by natural breeding, and in pregnant cows. Although insignificant but a higher prevalence of brucellosis was found in aged cattle than young cattle, cross bred cattle, pregnant cattle than non pregnant cattle, cattle with grazing. A higher prevalence of brucellosis was found in female cattle than male.

        Brucellosis outbreak of Korean indigenous cattle at Yeongwol and Pyeongchang county in Korea

        This study was attempted to investigate the properties of brucellosis in Korean indigenous cattle at the Yeongwol and Pyeongchang county. Brucella spp was differentiated and identified from cotyledons, amniotic fluids and supramammary lymph nodes which confirmed with clinical, serological, epidemiological evidences (69 cases) from January to June, 2004. Isolation frequency of this causative agent from supramammary lymph nodes, cotyledons and amniotic fluids from 38 pregnant Korean indigenous cattle were $39.1\%,\;87.5\%,\;and\;63.2\%$, respectively, and finally confirmed with Brucella abortus biotype 1 through biochemical and serological test. A Brucella specific DNA with 711bp band was detected by PCR assay using BCSP primer. The two cases were definite epidemiological evidences that infected Korean indigenous cattle acrossed the border to Yeongwol and Pyeongchang from near two provinces. Effective prevention programs are urgently needed for further spreading this epidemics.

        Prevalence of honeybee diseases in Incheon area in 2011

        This study investigated the occurrence of honeybee diseases in Incheon area, at the point of great widespread of sacbrood disease in the country. Sixteen resident beekeeping apiaries; 3 native honeybee and 13 European honeybee apiaries were selected for this research. Over 20 adult bees were evenly collected from the most colonies of each apiary three times (March, June, November) within a year. In this work, 13 honeybee diseases including 7 viral diseases, 2 bacterial diseases, 2 fungal diseases, and 2 parasitic diseases were detected by preliminary inspections and PCR. As a result, viral infections were confirmed at 34 among 48 apiaries (70.8%) over the entire examination period. Parasitic diseases showed the highest detection rate of 45.8%, which are detected in 44 among 96 cases. In the seasonal prevalence, 30 cases (15.6%) of 7 pathogens were detected from 14 apiaries in March, 50 cases (24.0%) of 9 pathogens and 56 cases (26.9%) of 9 pathogens were detected from all apiaries in June and November, respectively. Nosema was shown to be the most prevalent pathogen from March to November, followed by sacbrood virus (SBV) and stonebrood. The spread of SBV infection in Incheon would be under-estimated by the increasing of detection rate over the time. Especially, Chinese sacbrood virus was detected from 4 European honybee apiaries, but clinical symptoms were not found. No chalkbrood, acute bee paralysis virus, and chronic bee paralysis virus were detected in this study. The effective therapy and preventive measures should be prepared for beekeeping industry.

        Pathogenesis of infectious bronchitis virus with different routes of inoculation and the effect of in vivo serial passage in nephropathogenicity using cloacal infection

        In this study, we wanted to determine if the respirotropic JMK strain of infectious bronchitis virus(IBV), which has a spike glycoprotein gene that is 99% similar to the nephropathogenic Gray strain of IBV, could adapt and cause lesions in the kidney following intracloacal passage in chickens. Two day old specific pathogen free(SPF) cchickens were infected with Gray and JMK strains by the intraocular and cloacal route. Several tissue samples were collected at various times. Viruses were recovered from more tissues and earlier in the infection from chickens infected cloacally than chickens infected intraocularly. Virus was isolated from the kidney of chickens infected with Gray by the intraocular route and JMK by the intracloacal route, but not from chicken given JMK the intraocular route. Histopathologically, interstitial nephritis was observed in Gray infected chickens. However, viral RNA or antigen were not detected in the kidney by in situ hybridization and immunohistochemistry. We further passaged the JMK strain ten times in two day old SPF chickens using cloacal inoculation. We examined the virus titer and histopathological change in the kidney at each passage level. The amount of virus recovered from the kidney was stable throughout this serial passage and the passaged virus did not caused renal damage. Further, virus could not be isolated from the kidney when chickens were infected with the passaged virus by the intraocular route. We conclude that the JMK strain has a strict upper respiratory tract tropism since cloacal passage did not produce nephrotropism or nephropathogenicity.

        Development of serodiagnostic surface plasmon resonance imaging assay for the detection of antibodies to porcine circovirus type 2

        A surface plasmon resonance imaging (SPRI) assay was developed for measuring porcine circovirus type 2 (PCV2) antibody using a recombinant capsid protein as an antigen. The diagnostic potential of SPRI for detecting antibodies to the PCV2 capsid protein was compared with that of a conventional enzyme-linked immunosorbent assay (ELISA) using 70 pig serum samples taken from 6 pig farms. There was a strong positive correlation between the SPRI and ELISA (n = 70, r = 0.911, P<0.01). Therefore, this recombinant capsid protein can be used as an antigen for serological studies, and the SPRI, a label-free and high-throughput method, is expected to be a valuable tool in the serodiagnosis of PCV2 infection.

        A case of nonserotypable Escherichia coli infection in a Korean rabbit farm

        Rabbits are highly susceptible to colibacillosis, and no treatment is able to control the disease effectively. Rabbits raised in a farm in Chonbuk province presenting persistent diarrhea and death were submitted for diagnosis. Ninety percent of the infected animals died; weanlings suffered the most mortality. Necropsies showed prominent hemorrhagic foci along the intestinal and cecal serosae. In histopathological examination, rod-shaped bacteria were observed in the necrotic areas of the tips of villi in the small intestine and neutrophils infiltration was found around the necrotic villous areas. The affected animals consistently yielded Escherichia coli isolates from the intestines. The isolated organism was atypically indole-negative and was nonserotypable using 62 known O group-typing sera. Further microbiological and epidemiological works to recognize and control colibacillosis infection in farmed rabbits in Korea is therefore critical.

        Isolation and identification of infectious bursal disease virus from broiler and layer chickens during the outbreak year 2007 in Bangladesh

        The objective of the present study was to isolate and identify infectious bursal disease viruses (IBDVs) from broiler and layer chickens of outbreaks of infectious bursal disease (IBD) in three districts of Bangladesh. A total of 70 bursal samples were collected from dead broiler (n=40) and layer (n=30) chickens showing specific lesions of IBD from seven commercial poultry farms of three different districts (Mymensingh, Chittagong and Tangail) of Bangladesh during the year 2007. Five representative bursal samples from each farm were used for the isolation of IBDVs using 9-day-old embryonated eggs of seronegative flock of layer birds and for identification the samples were subjected to agar gel immunodiffusion test (AGIDT), immunohistochemistry (IHC) and reverse transcription-polymerase chain reaction (RT-PCR). Out of 35 bursal samples, IBDVs were successfully isolated from 28 (80%) samples. By AGIDT, 32 (91.4%) samples were found positive for IBDV antigen. Results of AGIDT clearly indicated that IBDVs detected in 29 bursal samples of six affected farms were identical to each other but not to IBDVs present in the remaining three samples of another farm. Indirect immunoperoxidase staining of the bursal sections revealed the presence of IBDV antigen in 32 (91.4%) samples and the IBDV antigen was detected mainly in the cortex of the lymphoid follicles of the bursal tissues. In histopathology, cell depletion, atrophy and necrosis were observed in many bursal follicles with severe edema of interfollicular septa. Of the 35 bursal samples, 34 (97.1%) samples generated 254 bp product by RT-PCR. In conclusion, the results of virus isolation and identification by AGIDT, IHC and the analysis of viral genome by RT-PCR confirmed the outbreaks of acute IBD in commercial poultry of Bangladesh. Moreover, histopathological findings and results of AGIDT gave a clear indication that the isolates from six outbreaks were different from classical strain and it seems to be of very virulent strain. On the other hand, the isolates from the other outbreak were similar to the classical strain.

        Generation of ovine recombinant prion protein (25-232): Characterisation via anti-PrP monoclonal antibodies and CD spectroscopy

        In prion pathogenesis, the structural conversion of the cellular prion protein $(PrP^c)$ to its abnormal isomer $(PrP^{Sc})$ is believed to be a major event. The susceptibility or resistance to natural sheep scrapie is associated with polymorphisms of host PrP gene (PRNP) at amino acid residues 136, to a lesser extent 154. The 112 residue in ovine PrP displays a natural polymorphism, Methionine to Threonine, which has not been thoroughly investigated. However the cell-free conversion assay showed that ARQ with Thr112 $(T_{112}ARQ)^{1)}$ presents lower convertibility to $PrP^{Sc}$than wild type ARQ $(M_{112}ARQ)$ [1] In this study we generated ovine recombinant PrPs of 112 allelic variants by metal chelate affinity chromatography and cation exchange chromatography. The final purity of the ovine PrP ARQ was more than $95\%$. These variants showed similar immunoreactivity against anti-PrP monoclonal antibodies in Western blot and ELISA. The refolded $M_{112}ARQ$ and $M_{112}ARQ$ presented the secondary structural content to similar extent via CD spectroscopy analysis. The inherited structural features of $M_{112}ARQ$ and $M_{112}ARQ$ under the different biophysical conditions are in the middle of investigation.
