      • 출아효모에서 TAR 클론닝법을 이용한 고등동물의 게놈으로부터 특정 염색체 부위의 분리

        복잡한 게놈 분석에 용이하도록 효모의 인공 염색체(YAC) 클론닝 시스템은 발달되어왔다. YAC은 박테리아의 인공 염색체(BAC)보다 더 큰 단편을 클론닝할 수 있고, 또한 클론된 단편을 쉽게 변형시킬 수 있다. 형질전환과 연계된 재조합법(Transformation-Associated Recombination; TAR)은 게놈 라이브러리를 만들지 않고 직접 게놈 DNA로부터 분리하고자 하는 유전자나 특정 염색체 부분을 클론닝 할 수 있는 방법이다. 이 방법은 spheroplast transformation을 수행하는 동안, 목적으로 하는 유전자의 5' 그리고 3' 염기 배열(hooks)을 지닌 TAR 벡터와 게놈 DNA 사이에서 일어나는 상동성 재조합에 의해 이루어진다. 효모 내의 in vivo 재조합을 이용한 TAR 클론닝법은 복잡한 게놈으로부터 목적의 염색체 부분을 원형 YAC의 형태로서 선택적으로 분리할 수 있다. 그러므로 TAR 클론닝 법은 특정 염색체로부터 YACs을 만드는데 매우 유용하여, 전체 게놈으로부터 특이적 유전자나 유전자의 family를 분리하는데 효과적인 방법으로 사료된다. Yeast artificial chromosome (YAC) cloning systems have advanced the analysis of complex genomes considerably. They permit the cloning of larger fragments than do bacterial artificial chromosome systems, and the cloned material is more easily modified. Transformation-associated recombination (TAR) is a cloning technique that allows specific chromosomal regions or genes to be isolated directly from genomic DNA without prior construction of a genomic library. This technique involves homologous recombination during spheroplast transformation between genomic DNA and a TAR vector that has 5' and 3' gene targeting sequences (hooks). Using in vivo recombination in yeast, TAR cloning selectively isolates, as circular YACs, desired chromosome segments or entire genes from complex genomes. We propose that TAR cloning can provide an efficient means for generating YACs from specific chromosomes and that TAR cloning may be useful for isolating families of genes and specific genes from total genome DNA.

        The purpose of this study was to examine the application of storytelling and audio-visual aids video in particular, to determine their effect in children's achievement in the functional skills of listening and speaking, and the degree of interest of Korean pre-school children in learning English. One way te sustain the curiosity and interest of pre-school students in an English classroom is to use stories. Students can be given as much as possible to listen and practice speaking the foreign language in these stories. In conclusion, it is suggested that instruction using storytelling contributes to the learner's development of listening and speaking skills and the children took an interest and pleasure in listening to the stories. As well, storytelling motivates children to develop a better understanding and knowledge of literature. This study found that the use of storytelling, especially fur the early beginners in the English classroom created a lively and happy atmosphere, where children were allowed to use their creative imagination.

        Fibrothecoma are mesenchymal tumors deriving from the ovarian stroma and consisting of theca-like elements and fibrous tissues. The reported incidence is between 0.4 and 8.0% of all ovarian tumors and this side variability can be attributed to the difficulty in differentiation fibrothecoma from ovarian fibroma or hyperplastic stroma nodules. We experienced a rare case of ovarian fibrothecoma in woman at 38 weeks gestation with previous repeat cesarean section diagnosed after tertiary cesarean delivery and bilateral tubal ligation with a brief review of literature.

        In past decade, ectopic pregnancy developing in the previous cesarean section scar has been the rarest forms of ectopic pregnancy and very dangerous because of the risk of uterine rupture and hemorrhage. These pregnancies once carried a maternal mortality rate of 40% to 45%, and until recent years they usually resulted in abdominal hysterectomy as a life-saving procedure. However, increased awareness of the importance of preserving childbearing capacity has led to several successful attempts at conservative management that include intraamniotic and systemic methotrexate administration, suction curettage followed by insertion of an intracervical 30-mL Foley catheter ballooning and angiographic artery embolization. We present three cases of ectopic pregnancy developing in the previous cesarean section scar in which successfully treated with conservative management.

        Persistent tumor develops in approximately 20% of patients with a complete mole, and chemotherapy is required to achieve remission. Following evacuation of hydatidiform mole, careful hCG monitoring is mandatory since it is the most reliable and sensitive method for the early detection of gestational trophoblastic disease. In carefully selected patients in whom the risk of developing gestational trophoblastic disease is significant or when the availability of hCG testing is suboptimal, chemoprophylaxis has been shown to decrease the risk of gestational trophoblastic tumor. We report here two cases of patient, 16-year-old and 45-year-old women who experienced unusual course after the evacuation of a complete mole and markedly elevated serum β-hCG levels. The patient developed persistent gestational trophoblastic disease and were successfully treated with EMA-CO and hysterectomy.

        Variant of CHD1 gene resulting in a Korean case of Pilarowski-Bjornsson syndrome

        Many monogenic neurodevelopmental disorders have been newly identified in recent years owing to the rapid development of genetic sequencing technology. These include variants of the epigenetic machinery – up to 300 known epigenetic factors of which about 50 have been linked to specific clinical phenotypes. Chromodomain, helicase, DNA binding 1 (CHD1) is an ATP-dependent chromatin remodeler, known to be the causative gene of the autosomal dominant neurodevelopmental dis-order Pilarowski-Bjornsson syndrome. Patients exhibit various degrees of global developmental delay, autism, speech aprax-ia, seizures, growth retardation, and craniofacial dysmorphism. We report the first case of Pilarowski-Bjornsson syndrome in Korea, due to a de novo missense variant of the CHD1 gene (c.862A>G, p.Thr288Ala) in a previously undiagnosed 17-year-old male. His infantile onset of severe global developmental delay, intellectual disability, speech apraxia, and failure to thrive are compatible with Pilarowski-Bjornsson syndrome. We also noted some features not previously reported in this syndrome such as skeletal dysplasia and ichthyosis. Further studies are needed to discover the specific phenotypes and pathogenic mechanisms behind this rare disorder.

        The South Korean society is now in the middle of crisis caused by chronic low-growth, income polarization, low birth rate and aging population. These crisis factors are causing geographical imbalance and inequality, so some regions are known to be on the verge of extinction. The present study aims to present possible solutions for achieving sustainable regional growth by capitalizing on the transitional factors. The ‘4P (Private-Public-People-Partnership)’ framework was applied to analyze the regional innovation ecosystem. This tool employs both quantitative and qualitative approaches. It quantitatively analyzes private and people elements while diagnosing public and partnership elements qualitatively. The quantitative analysis was conducted in cooperation with the Korea Enterprise Data (KED) using its corporate transactions network database. For the qualitative diagnosis, the research team collaborated with the Korea Small Business Institute (KSBI) by referencing KSBI’s regional development policy analyses. In a transitional era, regional innovation policy requires a location-based innovation policy approach. Now, it has become important to redefine the roles between the central and regional players, so that regional players take the lead while the central government only plays a supporting role instead of trying to solve regional issues with the policy lead of the central government. Therefore, regional innovation policies need to be based on the mutually beneficial relations between the central and regional players and be implemented by 1) linking R&D and non-R&D, 2) balancing science and technology innovation and lifestyle, 3) taking a comprehensive and integrated approach to science and technology, environment, energy and industrial policies and 4) combining various policies into a policy package. Against this backdrop, the present study has identified the following 10 policy initiatives from the analysis of regional innovation ecosystem; 1) building a real-time monitoring system to assess business ecosystem by region, 2) encouraging changes in pivot companies’ roles for the transformation of existing companies and supporting their growth to create more pivot companies, 3) building regional brands based on the entrepreneurship of residents living in regional cities and provinces, 4) enhancing integrity between the central government’s balanced regional development plan and regional governments’ own development plans, 5) strengthening planning capacity of regional players, 6) building a quantitative statistical base to monitor policies for supporting regional SMEs, 7) establishing an inter-ministerial mechanism for coordinating laws and regulations relevant to regional issues, especially those for designating special locations, 8) providing support differentiated by designated location, 9) developing new growth engines and opportunities to overcome regional crises, and 10) devising and implementing a policy mix based on cooperative governance.

        A novel routing protocol for cognitive radio networks with cooperation process

        Cognitive radio networks (CRNs) are composed of mobile users who can use multiple spectrum bands for communication. CRNs allow unlicensed users (called cognitive users) to efficiently utilize unused licensed spectrums without interfering with communications of licensed users (called primary users). The main goals of CRNs are to mitigate spectrum saturation and to improve spectrum utilization. This paper introduces state-of- the-art routing protocols for CRNs and addresses some limitations of these protocols. To resolve the limitations, we suggest a new research direction for routing protocols in CRNs. We implement our protocol to compare with the existing routing protocols for multi-hop CRNs. Our protocol shows good performance compared to the existing routing protocols in terms of network performance and PU protection.

        DEVELOPMENT AND IMPLEMENTATION OF DISTRIBUTED HARDWARE-IN-THE-LOOP SIMULATOR FOR AUTOMOTIVE ENGINE CONTROL SYSTEMS

        A distributed hardware-in-the-loop simulation (HILS) platform is developed for designing an automotive engine control system. The HILS equipment consists of a widely used PC and commercial-off-the-shelf (COTS) I/O boards instead of a powerful computing system and custom-made I/O boards. The distributed structure of the HILS system supplements the lack of computing power. These features make the HILS equipment more cost-effective and flexible. The HILS uses an automatic code generation extension, REAL-TIME WORKSHOP$^{ (RTW$^{) of MATLAB$^{ tool-chain and RT-LAB$^{, which enables distributed simulation as well as the detection and generation of digital event between simulation time steps. The mean value engine model, which is used in control design phase, is imported into this HILS. The engine model is supplemented with some I/O subsystems and I/O boards to interface actual input and output signals in real-time. The I/O subsystems are designed to imitate real sensor signals with high fidelity as well as to convert the raw data of the I/O boards to the appropriate forms for proper interfaces. A lot of attention is paid to the generation of a precise crank/ earn signal which has the problem of quantization in a conventional fixed time step simulation. The detection of injection! command signal which occurs between simulation time steps are also successfully compensated. In order to prove the feasibility of the proposed environment, a simple PI controller for an air-to-fuel ratio (AFR) control is used. The proposed HILS environment and I/O systems are shown to be an efficient tool to develop various control functions and to validate the software and hardware of the engine control system.
