        Optimization of Siderophore Production by Bacillus sp. PZ-1 and Its Potential Enhancement of Phytoextration of Pb from Soil

        In this study, the siderophore-producing characteristics and conditions of Bacillus sp. PZ-1 were investigated and the enhancement of siderophores on Pb uptake and translocation in Brassica juncea were determined. Results of single factor experiment showed that glucose, pH, and Pb(NO<sub>3</sub>)<sub>2</sub> could stimulate PZ-1 growth and siderophore production. The maximum siderophore production of 90.52% siderophore units was obtained by response surface methodology optimization at the glucose concentration of 21.84 g/l, pH 6.18, and Pb(NO<sub>3</sub>)<sub>2</sub> concentration of 245.04 μmol/l. The type of siderophore was hydroxamate and its concentration in the fermentation broth amounted to 32.24 μg/ml. Results of pot experiments indicated that the siderophores enhanced B. juncea to assimilate more Pb from soil with the uptake ratio from 1.04 to 2.74, and to translocate more Pb from underground to overground with the TF values from 1.21 to 1.48. The results revealed that Bacillus sp. PZ-1 could produce abundant siderophores and might be potentially used to augment the phytoextraction of Pb from soil.

        Characterization of siderophore produced by Pseudomonas syringae BAF.1 and its inhibitory effects on spore germination and mycelium morphology of Fusarium oxysporum

        In this study, an antagonistic bacterium against Fusarium oxysporum was identified and designated as Pseudomonas syringae strain BAF.1 on the basis of 16S rDNA sequence analysis and physiological-biochemical characteristics. It produced catechol-species siderophore at a molecular weight of 488.59 Da and a maximum amount of 55.27 μg/ml with glucose as a carbon source and asparagine as a nitrogen source at a C/N ratio of 10:1, 30°C and pH 7. The siderophore exhibited prominent antagonistic activity against Fusarium oxysporum with a maximum inhibition rate of 95.24% and had also suppressive effects on other kinds of 11 phytopathogenic fungi in the absence of FeCl3·6H2O. Spore germination was completely inhibited by 50 μl of the siderophorecontaining solution, and the ultrastructures of mycelia and spores were also considerably suppressed by siderophore treatment as established by electron microscopy observation. These results indicate that the siderophore produced by Pseudomonas syringae BAF.1 could be potentially used for biocontrol of pathogenic Fusarium oxysporum.

        Effects of LED Light Quality Combinations on Growth and Leaf Colour of Tissue Culture-Generated Plantlets in Sedum rubrotinctum

        Different plants have different requirements with regard to light quality, including combinations of R/B LED light on tissue-culture-generated plants. However, the effects of R/B LED light combinations on the growth and colour changes of leaves in succulent plants remains unknown. In this study, to identify the most beneficial light quality and R/B light ratios for the growth of the succulent plant species Sedum rubrotinctum and its production of attractive colours, the growth parameters, pigment levels and leaf colour phenotypes of tissue-culture-generated S. rubrotinctum plantlets were evaluated when the plantlets were exposed to R, 4R1B, 3R1B, 2R4B, B and W treatments for 90 days. The results showed that light quality obviously influenced the growth and leaf colour of tissue-culture-generated S. rubrotinctum plantlets. R light hastened the elongation of stems, and B light was more beneficial for biomass accumulation and leaf and root growth. However, compared with B light, R light, and W light, the R/B combination resulted in a greater amount of fresh biomass and higher values for the shoot/root FW, stem diameter, crown width, total root length, root surface area, and root volume. The R/B combination also markedly increased the contents of anthocyanins and the R/green chromaticity of the leaves. In addition, a principal component analysis (PCA) revealed the following comprehensive ranking: 4R1B > 2R4B > 3R1B > W > B > R. Therefore, the 4R1B combination is suggested as the best treatment for promoting the growth of tissue-culture-generated S. rubrotinctum plantlets and their production of attractive colours. This study provides a practically significant and optimal solution for growth regulation via light quality for the production of S. rubrotinctum as well as important implications for other succulent plant species in the Crassulaceae family.

        Characterization of P450 monooxygenase gene family in the cotton aphid, Aphis gossypii Glover

        Cytochrome P450 monooxygenases play an important role in the biosynthesis and detoxification of endogenous compounds. In this study, we analyzed a number of P450 genes through transcriptome-wide and genome analyses in Aphis gossypii Glover, the most notorious cotton pests. A total of 49 CYP-putative sequences were identified and classified into 13 families and 21 subfamilies. We identified a putative P450 motif (FXXGXXXCXG), and further analyzed other conserved sequences, which indicate that forty-seven of the fortynine genes seem to be functional, but one is likely non-functional and one is predicted pseudogenes. We found that P450-encoding genes are conserved in gene structure and organization (number, position, and phase of introns) in cotton aphids. The size, transcription orientation, and comparison with other insects of the cotton aphid P450 family suggested that it has evolutionary differentiation. The distribution across the genome and phylogenetic clustering analysis indicated past occurrence of gene duplication events. These results may provide valuable clues for understanding the evolution of the aphid genome, studying the functions of P450s in cotton aphid, and further selecting candidate genes for novel insect control methods.

        High-performance Zinc-Tin-Oxide thin film transistors based on environment friendly solution process

        Zinc-Tin-Oxide (ZTO) thin films were fabricated using a simple and eco-friendly sol-gel method and their application in thin film transistors (TFTs) was investigated. Annealing temperature has a crucial influence on the structure and electrical properties of sol-gel ZTO thin films. The ZTO thin films annealed at 300–600 °C revealed smooth and uniform surfaces with amorphous state, in addition, a high optical transparency over 90% of the ZTO films in the visible range was obtained. The electrical performance of ZTO TFTs showed obvious dependence on annealing temperature. The ZTO TFTs annealed at 500 °C showed a high carrier mobility of 5.9 cm2 /V, high on/off current ratio (Ion/off) of 106 -107 , and threshold voltage (Vth) of 1.03 V. To demonstrate the application of sol-gel ZTO films in low-power display fields, we also fabricated ZTO TFTs with a solution-processed high-permittivity (high-k) ZrTiOx dielectric layer. The ZTO/ZrTiOx TFTs showed high mobility of 17.9 cm2 /V and Ion/off of 105 -106 at a low operation voltage of 3 V, indicating that Indium-free ZTO thin films would be potential candidates for low cost, high performance oxide TFT devices.

        Effects of Selenizing Modification on Characteristics and Antioxidant Activities of Inonotus obliquus Polysaccharide

        Excessive reactive oxygen species is implicated in the etiology and pathology of many acute and chronic diseases. Selenium acts as an antioxidant and stimulates the creation of more antioxidants. Selenizing modification polysaccharides potentiate the physiological and pharmacological activities of selenium and polysaccharide. In this study, Inonotus obliquus polysaccharide (IOP) extracted from cultured mycelia was modified into Se-IOP by HNO3- Na2SeO3 method. The characteristics and antioxidant activity of IOP and Se-IOP were comparatively investigated. The result showed that polysaccharide content of purified IOP was 98.9%. The selenium content of Se-IOP was 0.71 mg/g. Both IOP and Se-IOP were homogeneous polysaccharides with a molecular weight of 37.354 and 28.071 kDa, respectively depicted in high performance gel permeation chromatogram (HPGPC). Moreover, both IOP and Se- IOP were composed of Man, Glu, and Gal with a molar ratio of 7.7:32.6:23.3 and 8.3:32.1:22.7, respectively determined by high performance liquid chromatography (HPLC). The characteristic absorption peak of O-Se-O, Se=O and C-O-Se appeared at 1026.75, 772.82, and 652.29 cm-1 respectively in FT-IR spectrum of Se-IOP indicating the success of selenylation. NMR spectrum further confirmed the anomeric carbon signals and chemical shifts in IOP and Se-IOP. These variations induced by selenylation may lead to a higher antioxidant activity of Se-IOP on scavenging hydroxyl, 2,2-diphenyl-1-picryl-hydrazyl (DPPH) and superoxide radical than that of IOP. Furthermore, Se- IOP treatment could also reduce oxidant damage by decreasing the level of malondialdehyde (MDA) and increasing the activities of superoxidedismutase (SOD) and GSH-Px in mice.
