Rice blast is one of the most destructive diseases of rice worldwide, and the causative agent is the filamentous ascomycete Magnaporthe oryzae. With the successful cloning of more and more avirulence genes from M. oryzae, the direct extraction of M. oryzae genomic DNA from infected rice tissue would be useful alternative for rapid monitoring of changes of avirulence genes without isolation and cultivation of the pathogen. In this study, a fast, low-cost and reliable method for DNA preparation of M. oryzae from a small piece of infected single rice leaf or neck lesion was established. This single step method only required 10 min for DNA preparation and conventional chemical reagents commonly found in the laboratory. The AvrPik and AvrPi9 genes were successfully amplified with the prepared DNA. The expected DNA fragments from 570 bp to 1,139 bp could be amplified even three months after DNA preparation. This method was also suitable for DNA preparation from M. oryzae strains stored on the filter paper. All together these results indicate that the DNA preparation method established in this study is reliable, and could meet the basic needs for polymerase chain reaction-based analysis of M. oryzae.

In a cold rotary forging press with double eccentricity bushings, the rotation speed ratio of double eccentricity bushings has significanteffects on the rocking tool path. Thus, this study examines the effects of the rotation speed ratio of double eccentricity bushings on therocking tool path of cold rotary forging presses. On the basis of kinematics relations, the motion equations of the rocking tool with doubleeccentricity bushings are obtained. The effects of the rotation speed ratio of double eccentricity bushings on the rocking tool path arestudied by using the obtained motion equations.

In this study, the siderophore-producing characteristics and conditions of Bacillus sp. PZ-1 were investigated and the enhancement of siderophores on Pb uptake and translocation in Brassica juncea were determined. Results of single factor experiment showed that glucose, pH, and Pb(NO<sub>3</sub>)<sub>2</sub> could stimulate PZ-1 growth and siderophore production. The maximum siderophore production of 90.52% siderophore units was obtained by response surface methodology optimization at the glucose concentration of 21.84 g/l, pH 6.18, and Pb(NO<sub>3</sub>)<sub>2</sub> concentration of 245.04 μmol/l. The type of siderophore was hydroxamate and its concentration in the fermentation broth amounted to 32.24 μg/ml. Results of pot experiments indicated that the siderophores enhanced B. juncea to assimilate more Pb from soil with the uptake ratio from 1.04 to 2.74, and to translocate more Pb from underground to overground with the TF values from 1.21 to 1.48. The results revealed that Bacillus sp. PZ-1 could produce abundant siderophores and might be potentially used to augment the phytoextraction of Pb from soil.

In this study, an antagonistic bacterium against Fusarium oxysporum was identified and designated as Pseudomonas syringae strain BAF.1 on the basis of 16S rDNA sequence analysis and physiological-biochemical characteristics. It produced catechol-species siderophore at a molecular weight of 488.59 Da and a maximum amount of 55.27 μg/ml with glucose as a carbon source and asparagine as a nitrogen source at a C/N ratio of 10:1, 30°C and pH 7. The siderophore exhibited prominent antagonistic activity against Fusarium oxysporum with a maximum inhibition rate of 95.24% and had also suppressive effects on other kinds of 11 phytopathogenic fungi in the absence of FeCl3·6H2O. Spore germination was completely inhibited by 50 μl of the siderophorecontaining solution, and the ultrastructures of mycelia and spores were also considerably suppressed by siderophore treatment as established by electron microscopy observation. These results indicate that the siderophore produced by Pseudomonas syringae BAF.1 could be potentially used for biocontrol of pathogenic Fusarium oxysporum.

Objective: Two experiments were conducted to investigate the effects of dietary sucralose on diet preference and growth performance of weaned piglets, and a third experiment was a 28-d safety study to examine if high-dose sucralose could affect the health state of weaned piglets. Methods: In experiment one, 48 piglets had free access to a corn-soybean based diet and the same diet supplemented with 150 mg/kg sucralose for 15 d. In experiment two, 180 piglets were blocked into 5 treatments with 6 replications. They were fed basal diets supplemented with 0, 75, 150, 225, and 300 mg/kg sucralose for 28 days. In experiment three, 108 piglets were randomly assigned to 3 treatments and fed diets supplemented with 0, 150 (suitable level), and 1,500 (ten-fold suitable level) mg/kg sucralose for 28 d. Results: The experiment 1 showed that piglets preferred (p<0.05) diets containing sucralose during experimental period. In experiment 2, piglets fed a diet supplemented with 150 mg/kg sucralose had a higher average daily gain (ADG) and average daily feed intake (ADFI) than pigs in the control group and other treatment groups during the experiment period. The concentrations of sucralose over 150 mg/kg may decrease feed intake. However, no difference in feed conversion ratio was observed. In experiment 3, piglets fed diet supplemented with 150 mg/kg sucralose had a higher ADG and ADFI than that of pigs in the control group and 1,500 mg/kg treatment groups during the experiment period. Clinical blood metabolites, organ index and histological morphology were not significantly different between sucralose treatments. Conclusion: Sucralose can promote feed intake and thereby improve growth performance of weaned piglets. Moreover, inclusion of 1,500 mg/kg sucralose was demonstrated to have no observed adverse effects. Supplementing 150 mg/kg sucralose for weaned piglets is recommended in this study.

The Doehlert experimental design was used to optimize the production of mycelial biomass and exopolymer from Hericium erinaceus CZ-2 in this study. Statistical analysis showed that the linear and quadric terms of 3 variables: corn flour, yeast extract, and corn steep liquor had significant effects. The optimized combination of these 3 variables was confirmed through validation experiments. The optimal conditions for higher production of mycelial biomass (19.92 g/L) were estimated when the media composition concentrations were set as: 30.85 g/L,corn flour; 2.81 g/L, yeast extract; 16.9 mL/L, corn steep liquor; 10 g/L, glucose; 1 g/L, KH2PO4; and 0.5 g/L,MgSO4·7H2O; while a maximal exo-polymer yield (1.653g/L) could be achieved when setting concentrations of:32.71 g/L, corn flour; 2.35 g/L, Yeast extract; 14.42 mL/L,Corn steep liquor; 10 g/L, glucose; 1 g/L, KH2PO4; and 0.5g/L, MgSO4·7H2O. The upscale production was also investigated using a 15 L fermentor using the optimized medium.

Roll-forging process is widely used to fabricate the automobile front axle beams. The reasonable selection of the process parameters is significant to ensure the quality and improve the property of the automobile front axle beam owing to the complexity and importance of the preliminary roll-forging process. This paper developed a 3D Finite element (FE) model of the preliminary roll-forging process of the AISI-1045 automobile front axle beam under the Deform-3D platform. A corresponding experiment is subsequently conducted. Good agreement between the experimental and simulative results validates the employed model. Based on this dependable FE model, the influences of initial temperature of billet T 0 , rotational speed of rolls n and friction factor m on the geometric dimension, temperature inhomogeneity, deformation inhomogeneity, and roll-forging load are investigated comprehensively. The results can provide reliable guidelines for the selection of process parameters.
