TNBS로 유도된 흰쥐의 대장염(大腸炎)에 대한 노봉방(露蜂房) 약침(藥鍼)의 효과

Objectives: Transient inflammation has been demonstrated to alter visceral sensory function in animal models and acute mucosal inflammation may precede the manifestation of visceral hyperalgesia. Thus in this study we compared effects of herbal acupuncture of Nidus Vespae (NV) applied to the different acupoints in the acute colitis induced by trinitrobenzenesulphonic acid (TNBS) intracolonic injection in rats. Methods: In Male Sprague-Dawley rats, weighing 250~400 g, TNBS (5 mg/kg) was infused intrarectally through a silicon rubber catheter into the anus under isoflurane anaesthesia. Under general anesthesia, acupoints of LI4 (Hapkok), SI25 (Cheonchu), ST36 (Joksamni), BL25 (Daejangsu) were intramuscularly injected by NV. Expressions of cFos protein in the periaqueductal gray (PAG), locus coeruleus (LC), nucleus of solitary tract (Sol), and the 6th lumbar spinal cord (L6 s.c.) were observed at 24 hrs after TNBS induced colitis by immunohistochemistry. Results: The expression of c-Fos protein in L6 s.c., Sol, LC and PAG increased 24 hrs after TNBS injection into colorectum as compared to normal group. NV herbal acupuncture also inhibited the expression of c-Fos protein in Sol but not L6 s.c., LC, and PAG. NV to ST36 inhibited significantly the c-Fos expression in Sol and PAG. NV to ST25 inhibited the c-Fos protein expression all over the observation area. NV to BL25 showed the inhibitory effects in the areas except LC. Whether or not a role of endogenous opioids, intrathecal injection of naltrexone (30 ug/30 ul) was applied before the 2nd herbal acupuncture treatment 24 hrs after TNBS-induced colitis in rat. Naltrexone reversed the inhibition of c-Fos protein expression in the spinal cord and brainstem under different conditions such as type of herbal acupuncture compound and choice of acupoint. Conclusions: In summary, these data show that herbal acupuncture of NV inhibits signal pathways such as spinal cord and brain stem ascending hypersensitivity of colorectum after TNBS induced colitis. This effect may be mediated by acupoints through the endogenous opioid system involving the pain modulation.

Objectives: Ulcerative colitis or Crohn`s disease has been recognized as Ha-ri (下痢) or Jang-Byok (腸벽) in Korean oriental medicine. A purpose of the present study is to investigate the anti-inflammatory effect of moxi-tar herbal acupuncture at LI4 (HapGok) on 2,4,6-trinitrobenzene sulphonic acid (TNBS) induced colitis in rats and further elucidate the possibility of herbal acupuncture on ulcerative colitis which is chronic inflammatory disease of the gastrointestinal tract. Methods: Sprague-Dawley rats, weighing 170~190 g, were subjected to intrarectal injection of either saline (300 ㎕, 500 ㎕) for a control or 2,4,6-trinitrobenzene sulphonic acid (TNBS) (300 ㎕, 500 ㎕) for a colitis. Moxi-tar herbal acupuncture at bilateral LI4 was used as the treatment. A volume 0.4 ㎖of moxi-tar (20 ㎎/㎖) were subcutaneously injected to the LI4 just after the secondary injection time of TNBS in rats. To study the effects of Moxi-tar acupuncture in LI4, body weight, RBC count, WBC count, total protein, Paw edema rate, rate of protein leakage into CMC-pouch fluid, IgG levels and IgM levels were observed. Results: Moxi-tar acupuncture in LI4 on TNBS-induced colitis inhibited the body weight lose rate but not effect RBC, WBC count. In addition, it inhibited the reduction of total protein concentration, paw edema, rate of protein leakage into CMC-pouch fluid, IgG levels and IgM levels. Conclusions: It is suggested that moxi-tar herbal acupuncture at LI4 helps to recover TNBS-induced colitis and plays an important role for an treatment of the irritable bowel syndrome (IBS).
Antioxidant and Antiatherogenic Activity of cis-Hinokiresinol from Trapa pseudoincisa

cis-Hinokiresinol, also known as (+)-nyasol, was isolated for the first time from an aquatic herbaceous plant, Trapa pseudoincisa NAKAI, via silica gel and octadecyl silica gel column chromatographies. The chemical structure was determined via analyses of the spectroscopic data, including NMR, MS and IR. cis-Hinokiresinol was also found to exhibit antioxidant and antiatherogenic activities. The $IC_{50}$ values for the scavenging activities of cis-hinokiresinol on ABTS cation and superoxide anion radicals were 45.6 and $40.5{\mu}M$, respectively. The $IC_{50}$ values for the inhibitory effects on $Lp-PLA_2$, hACAT1, hACAT2 and LDL-oxidation were 284.7, 280.6, 398.9 and $5.6{\mu}M$, respectively.

Background:?Antibiotic susceptibility testing (AST) is essential for choosing an accurate treatment of bacterial infections. Current AST requires at least 48h when a first blood culture (BC) is positive on direct smear examination (DSE). Thus, initial empirical antibiotic treatment is often inadequate, increasing rates of ineffective treatment or unnecessary broad spectrum antibiotic use. Direct rapid antibiotic susceptibility testing (dRAST) based on analyzing change of single bacterial cell under antibiotic condition detects antibiotic resistance in 4-6h after unveiling of DSE results and is expected to help more accurate selection of antibiotic.?Objective:?To assess accuracy of dRAST and evaluate its potential usefulness to improve selection of adequate antibiotic in clinical practice settings.?Material and Methods:?Among patients admitted to Seoul National University Hospital from June 2015 to December 2015, 141 patients with bacteremia were included for analysis. BC bottles from these patients were processed by both current AST method and dRAST. Prescribed antibiotics were recorded. In comparison with current AST, we evaluated whether the faster availability of dRAST results could have led to the initiation of optimal treatments without error. The efficacy of antibiotic treatment was classified as non-, sub-, optimal treatment according to antibiotic susceptibility to isolate.?Results:?Among 141 patients with bacteremia, 37 received non-optimal treatments and 4 had sub-optimal treatments after DSE results were reported. After the full results were known, 66% (27/41) changed non-, sub-optimal treatment into optimal treatment. For 98% (138/141), antibiotics prescribed supposedly in accordance with dRAST results were same with antibiotics prescribed in accordance with current methods. In at least 59% (24/41) patients, adequate treatment could have been done earlier with dRAST results. Unnecessary carbapenem treatments could have been avoided in 64% (9/14) of patients receiving carbapenem.?Conclusions: This study suggests that introduction of dRAST would increase treatment effectiveness and reduce unnecessary broad spectrum antibiotic use in early period of bacteremia.

<P><B>Abstract</B></P> <P>A mesoporous Ni-Al<SUB>2</SUB>O<SUB>3</SUB> aerogel catalyst was prepared by a single-step epoxide-driven sol-gel method and a subsequent supercritical CO<SUB>2</SUB> drying method (NA-ES catalyst). For comparison, a mesoporous Ni-Al<SUB>2</SUB>O<SUB>3</SUB> aerogel catalyst was also prepared by a single-step alkoxide-based sol-gel method and a subsequent supercritical CO<SUB>2</SUB> drying method (NA-AS catalyst). Differences in physicochemical properties and catalytic activities of mesoporous Ni-Al<SUB>2</SUB>O<SUB>3</SUB> aerogel catalysts in the steam reforming of liquefied natural gas (LNG) were investigated. Textural properties of Ni-Al<SUB>2</SUB>O<SUB>3</SUB> aerogel catalysts were affected by the preparation method. Nickel species were highly dispersed in alumina through the formation of nickel aluminate phase in both NA-ES and NA-AS catalysts. However, chemical states of Al atoms in both catalysts were quite different. In addition, nickel species in the NA-ES catalyst exhibited high reducibility and high dispersion compared to those in the NA-AS catalyst. In the steam reforming of LNG, NA-ES catalyst exhibited a better catalytic performance than NA-AS catalyst in terms of LNG conversion and hydrogen yield, although both catalysts showed a stable catalytic performance during the reaction without deactivation behavior. Furthermore, NA-ES catalyst with small average nickel diameter suppressed water-gas shift reaction.
