      • Effects of Fertilization Time and Culture Medium of Pig Oocytes Matured In Vitro by Liquid Boar Sperm Stored at 4℃

        This study was to investigate the effects of fertilization time and culture medium of pig oocytes matured in-vitro by liquid boar sperm. The sperm rich fraction (30∼60 ml) was slowly cooled to room temperature (20∼23℃) by 2 h after collection. Semen was transferred into 15 ml tubes, centrifuged at room temperature for 10 min 800×g, and the supernatant solution was poured off. The concentrated sperm was resuspended with 5 ml of the LEN diluent to provide 1.0×10^(9) sperm/ml at room temperature. The resuspended semen was cooled in a refrigerator to 4℃. The medium used for oocyte maturation was TCM-199 supplemented with 26.19 mM sodium bicarbonate, 0.9 mM sodium pyruvate, 10μg/ml insulin, 2μg/ml vitamin B_(12), 25 mM HEPES, 10μg/ml bovine apotransferrin, 150μM cysteamine, 10IU/ml PMSG, 10 IU/ml hCG, 10 ng/ml EGF, 0.4% BSA, 75μg/ml sodium penicillin G, 50μg/ml streptomycin sulfate and 10% pFF. After about 22 h of culture, oocytes were cultured without cysteamine and hormones for 22 h at 38.5℃, 5% CO₂in air. Oocytes were inseminated with liquid boar sperm stored at 4℃ for 2 days after collection. Oocytes were coincubated for 1, 3, 6 and 9 h in 500 μl mTBM fertilization media with 1.0×10^(6) sperm/ml concentration, respectively. Thereafter, oocytes were transferred into 500 μl NCSU-23, HEPES buffered NCSU-23, PZM-3 and PZM-4 culture media, respectively, for further culture of 6, 48 and 144 h. The rates of sperm penetration and male pronuclear formation were higher in the fertilization times for 6 and 9 h than in those for 1 and 3 h. The rates of cleaved oocytes were higher in the fertilization times for 6 and 9 h (85.0 and 84.6%) than in those for 1 and 3 h (61.1 and 76.8%). The percentage of blastocyst formation from the cleaved oocytes was highest in the fertilization time for 6 h (33.6%) than in that for 1, 3 and 9 h (11.4, 23.0 and 29.6%). Mean cell numbers per blastocyst were 32.9, 27.6, 26.3 and 24.4 in the fertilization times for 6, 9, 3 and 1 h, respectively. The rate of blastocyst from the cleaved oocytes and the number of cells per blastocyst were higher in HEPES buffered NCSU-23 culture medium than in NCSU-23, PZM-3 and PZM-4 culture media. In conclusion, we found out that liquid boar sperm stored at 4℃ could be used for in-vitro fertilization of pig oocytes matured in-vitro. Also, we recommend the coincubation time of 6 h in 500 μl TBM fertilization medium with 1×10^(6) sperm/ml concentration and the HEPES buffered NCSU-23 culture medium for in-vitro fertilization of pig oocytes matured in-vitro.

      • Interface sulfur passivation using H<sub>2</sub>S annealing for atomic-layer-deposited Al<sub>2</sub>O<sub>3</sub> films on an ultrathin-body In<sub>0.53</sub>Ga<sub>0.47</sub>As-on-insulator

        Atomic-layer-deposited Al<SUB>2</SUB>O<SUB>3</SUB> films were grown on ultrathin-body In<SUB>0.53</SUB>Ga<SUB>0.47</SUB>As substrates for III-V compound-semiconductor-based devices. Interface sulfur (S) passivation was performed with wet processing using ammonium sulfide ((NH<SUB>4</SUB>)<SUB>2</SUB>S) solution, and dry processing using post-deposition annealing (PDA) under a H<SUB>2</SUB>S atmosphere. The PDA under the H<SUB>2</SUB>S atmosphere resulted in a lower S concentration at the interface and a thicker interfacial layer than the case with (NH<SUB>4</SUB>)<SUB>2</SUB>S wet-treatment. The electrical properties of the device, including the interface property estimated through frequency dispersion in capacitance, were better for (NH<SUB>4</SUB>)<SUB>2</SUB>S wet-treatment than the PDA under a H<SUB>2</SUB>S atmosphere. They might be improved, however, by optimizing the process conditions of PDA. The PDA under a H<SUB>2</SUB>S atmosphere following (NH<SUB>4</SUB>)<SUB>2</SUB>S wet-treatment resulted in an increased S concentration at the interface, which improved the electrical properties of the devices.

      • Forecast Driven Simulation Model for Service Quality Improvement of the Emergency Department in the Moses H. Cone Memorial Hospital

        Patient satisfaction with the Emergency Department(ED) in a hospital is related to the length of stay, and especially to the amount of waiting time for medical treatments. ED overcrowding decreases quality and efficiency, therefore affecting hospitals' profitability. This paper presents a forecasting and simulation model for resource management of the ED at Moses H. Cone Memorial Hospital. A linear regression forecasting model is proposed to predict the number of ED patient arrivals, and then a simulation model is provided to estimate the length of stay of ED patients, system throughput, and the utilization of resources such as triage nurses, patient beds, registered nurses, and medical doctors. The near future load level of each resource is presented using the proposed models.

        Purpose: Investigate the level of Internet and smart phone addiction of college students and difference of their perception on the art therapy. Method: Data was collected using 4 categories of questionnaires. Participants of this study were 383 college students who are currently attending universities located in seoul, Kyung-Ki and Incheon. The Chi-square test, One-way Analysis of Variance, Scheffé test were performed using IBM SPSS Statistics 23.0 Result: First, the study has established that the status of attending universities, grade, people who living with, age affected the level of Internet addiction of college students. In terms of the level of smart phone addiction of college students, the status of attending universities, gender, age were the affective factors. Second, there was a significant difference on the perception of the advantages of the art therapy and the level of acknowledging it, depending on the level of Internet addiction. Finally, depending on the level of smartphone addiction, there was a significant difference in the level of perception of the art therapy, expectation toward the art therapy and the helpfulness of art therapy. The more the participants are close to the addicted level, the more they want to experience the art therapy. Conclusion: These results suggest. First, it is necessary to use bigger group of participants. Second, it is necessary to improve the research methods for college students. Third, nurse should offer holistic care toward the patients regarding their general characteristics by adapting this study. Finally, it is necessary to improve the art therapy programs for the college students who are addicted to the Internet and smartphone and to develop researches proving the effectiveness of these programs.

        A microstrip gas chamber(MSGC), applied to digital radiography system, was designed and constructed. The microstrip electrodes, made of aluminum and chromium, were manufactured by using photo-lithography process on glass substrate(Corning 7059) and Si wafer. Anode strip width is 10 μm, cathode 100 μm, and the distance between them is 100 μm. The effective area for detection is 50 x 50mm^(2) and the detection gas is Ar(90%) and CH_(4)(10%). As a result of our experiment, when the anode and drift voltage was 0.5kV and 1.5kV, the gas multiplication rate was about 5 x 10^(3).

      • Sulfonic acid functionalized mesoporous ZSM-5: Synthesis, characterization and catalytic activity in acidic catalysis

        Sulfonic acid functionalized mesoporous ZSM-5 (SO<SUB>3</SUB>H-Meso ZSM-5) were prepared by post-grafting of 3-mercaptopropyltriethoxysilane with various loading in the range of 10-50wt% followed by oxidation of thiol using H<SUB>2</SUB>O<SUB>2</SUB> to generate the corresponding sulfonic acid group. Grafting of organic moieties was evidenced by elemental analysis, <SUP>29</SUP>Si &<SUP>13</SUP>C-NMR and X-ray photoelectron spectroscopy. The SO<SUB>3</SUB>H-Meso ZSM-5 catalyst with improved acidity and mesoporosity exhibited enhanced catalytic activity in acid-catalyzed liquid-phase reaction of 2'-hydroxyacetophenone and benzaldehyde compared with non-functionalized mesoporous ZSM-5 (Meso ZSM-5) and functionalized microporous ZSM-5 (SO<SUB>3</SUB>H-ZSM-5). The higher loading of sulfonic acid functional groups led to higher acidity and consequently resulted in higher yield of products. The catalyst could be reused up to three cycles without significant loss in its catalytic activity.

      • Enhanced H<sub>2</sub>S gas sensing performance of networked CuO-ZnO composite nanoparticle sensor

        <P>CuO-ZnO composite nanoparticles (CuO:ZnO = 4:1 by vol.%) were synthesized via a facile solvothermal route. The CuO-ZnO composite nanoparticle sensor showed significantly enhanced H2S gas sensing performance compared to the pristine CuO and pristine ZnO nanoparticle sensors. The pristine CuO, pristine ZnO and CuO-ZnO composite nanoparticle sensors showed responses of approximately 335%, 161% and 1035%, respectively, to 2 ppm of H2S at 225 degrees C. The CuO-ZnO composite nanoparticle sensor also showed more rapid response to H2S gas than the pristine CuO and ZnO nanoparticle sensors. Both the pristine CuO nanoparticle and CuO-ZnO composite nanoparticle sensors showed selectivity for H2S gas over other gases. The underlying mechanism for the enhanced sensing performance of the CuO-ZnO composite nanoparticle sensor is discussed. (C) 2016 Elsevier Ltd. All rights reserved.</P>

      • Investigation of nitrous acid concentration in an indoor environment using an in-situ monitoring system

        <P><B>Abstract</B></P><P>An in-situ measurement system for the determination of nitrous acid (HONO) was developed and used at an indoor residential environment. The system uses a diffusion scrubber to sample gaseous HONO and the peroxynitrite-induced luminol chemiluminescent method to quantify the amount of HONO. In this system, the detection limit of HONO, estimated as three times the noise level of the scrubbing solution blank, was 120pptv for a 2-min integrated sample. Indoor HONO and NO<I><SUB>x</SUB></I> concentrations were determined for 7 days in the living room of an apartment with a gas range for cooking in the kitchen. Close examination of the relationships among HONO, NO, and NO<SUB>2</SUB> concentrations during both the background and combustion periods confirm that the observed HONO was formed not only by direct emission from gas combustion, but also from heterogeneous reactions of NO<SUB>2</SUB> with H<SUB>2</SUB>O on indoor surfaces. The average ratio of HONO to NO<SUB>2</SUB> over the study period was 0.12±0.05. The HONO/NO<SUB>2</SUB> concentration ratio was 0.04–0.08 during the combustion period, whereas it was 0.10–0.25 after combustion had stopped. This suggests that HONO was generated through different production processes, both during combustion and after the completion of combustion. The controlled combustion experiments indicate that the burning rate is an important factor to determine the peak HONO concentration. In darkness, HONO had a nearly constant removal rate for all of the combustion experiments, whereas the removal rates of NO and NO<SUB>2</SUB> depended on the burning rates of the gas range. Combustion experiments conducted at the fixed burning rate setting show also that ventilation decreased HONO concentration. This indicates that the airflow rate of the range hood fan is an important factor to control the concentration of indoor air pollutants.</P>

        <P>CHDH (choline dehydrogenase) is an enzyme catalyzing the dehydrogenation of choline to betaine aldehyde in mitochondria. Apart from this well-known activity, we report here a pivotal role of CHDH in mitophagy. Knockdown of CHDH expression impairs CCCP-induced mitophagy and PARK2/parkin-mediated clearance of mitochondria in mammalian cells, including HeLa cells and SN4741 dopaminergic neuronal cells. Conversely, overexpression of CHDH accelerates PARK2-mediated mitophagy. CHDH is found on both the outer and inner membranes of mitochondria in resting cells. Interestingly, upon induction of mitophagy, CHDH accumulates on the outer membrane in a mitochondrial potential-dependent manner. We found that CHDH is not a substrate of PARK2 but interacts with SQSTM1 independently of PARK2 to recruit SQSTM1 into depolarized mitochondria. The FB1 domain of CHDH is exposed to the cytosol and is required for the interaction with SQSTM1, and overexpression of the FB1 domain only in cytosol reduces CCCP-induced mitochondrial degradation via competitive interaction with SQSTM1. In addition, CHDH, but not the CHDH FB1 deletion mutant, forms a ternary protein complex with SQSTM1 and MAP1LC3 (LC3), leading to loading of LC3 onto the damaged mitochondria via SQSTM1. Further, CHDH is crucial to the mitophagy induced by MPP+ in SN4741 cells. Overall, our results suggest that CHDH is required for PARK2-mediated mitophagy for the recruitment of SQSTM1 and LC3 onto the mitochondria for cargo recognition.</P>

        Thiacremonone Potentiates Anti-Oxidant Effects to Improve Memory Dysfunction in an APP/PS1 Transgenic Mice Model

        <P>Alzheimer's disease (AD) is pathologically characterized by excessive accumulation of amyloid-beta (A beta) peptide. Evidence suggests that amyloid accumulation can be caused by oxidative stress and inflammatory responses. In this study, we examined neuroprotective effects of thiacremonone, an anti-oxidant and anti-inflammatory compound isolated from garlic. Treatment of thiacremonone significantly attenuated cognitive impairments in amyloid precursor protein (APP)/presenilin 1 (PS1) double-mutant transgenic mice. In addition, activation of nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kappa B) and extracellular signal-regulated kinase (ERK) pathways in the brain was potently inhibited by thiacremonone. We also observed that thiacremonone significantly inhibited activation of NF-kappa B and ERK pathways induced by H2O2 and A beta(1-42) in embryonic neuronal cells. Furthermore, thiacremonone augmented peroxiredoxin 6 (PRDX6) expression in vivo and in vitro associated with reduced oxidative stress of macromolecules such as protein and lipids. This study indicates that thiacremonone might exert memory improvement via stimulating anti-oxidant system. These multiple properties could attenuate A beta accumulation and oxidative stress in Alzheimer's brains. Thus, these results suggest that thiacremonone might be useful to intervene development or progression of neurodegeneration in AD.</P>
