        Cytotoxicity and L-Amino Acid Oxidase Activity of Animal Venoms

        The cytotoxicity of animal venoms (snakes, insects and marine animals) was measured against SNU-1 (stomach cancer cells) by dye uptake assay (MTT method). And also L-amino acid oxidase (AAO) activity of the venoms was compared. Among them, the venom from Ophiophagus hannah (king cobra) showed a strong AAO activity as well as a high potent cytotoxicity. Cytotoxic protein having a AAO was then partially purified by HPLC-GPC and two fractions (Fr. I and Fr. II) were collected. The $IC_{50}$ values of Fr. I and Fr. II were 0.19 ${\mu}g/ml$ and 1.36 ${\mu}g/ml$, respectively. The results suggested that the cytotoxicity of king cobra venom may be due to its AAO activity.

        동해항 전면 해역에서의 Freak Waves 발생확률

        지난 20년 동안 해양구조물과 선박 등에 예상치 못한 피해를 주는 freak waves에 대한 연구자들의 관심이 증가하고 있다. 다양한 연구결과에도 불구하고 아직 freak waves의 발생원인, 발생 메커니즘, 발생확률 등에 대한 상반된 결과로 인해 합의된 결과가 도출되지 못하고 있다. 본 논문에서는 동해항 전면 해상에서 측정된 파랑자료를 분석하여 freak waves에 대한 발생확률을 추정하였다. 유의파고 2.5 m 보다 크고 Hm/Hs≥2인 freak waves 3개를 발견하였다. Freak waves의 발생확률은 극치파고분포함수를 사용하여 추정하기 적절하며, Mori, Rayleigh, 그리고 Ahn의 극치파고분포함수는 freak wave의 발생확률을 각각 약 O(10<SUP>-1</SUP>), O(10<SUP>-2</SUP>), 그리고 O(10<SUP>-3</SUP><SUP></SUP>) 정도로 추정하였다. 본 논문에서 분석된 파랑자료의 freak waves의 발생확률은 O(10<SUP>-2</SUP>)와 O(10<SUP>-3</SUP>) 사이에 위치하였다. 즉, Rayleigh와 Ahn의 극치파고분포함수에 의해 예측된 발생확률의 중간에 위치하였다. 현재 Rayleigh와 Ahn의 극치파고분포함수 중에 어떤 분포함수가 정확한지에 대한 판단은 좀 더 다양한 해역에서의 freak waves의 발생확률에 대한 분석이 필요하다고 판단된다. Over the last 20 years, freak waves have attracted many researchers because of their unexpected behaviors and damages on offshore structures and vessels in the ocean and coastal waters. Despite many researches on the causes, mechanisms and occurrence of freak waves, we have not reached consensus on the results of the researches. This paper presents the occurrence probability of freak waves based on the analysis of wave records measured at coastal waters of Donghae harbor in the East Sea. Three freak waves were found which satisfied conditions of Hs≥2.5m and Hm/Hs≥2. The occurrence probabilities of freak waves were estimated from extreme distributions by Mori, Rayleigh and Ahn, and found to be on the orders of O(10<SUP>-1</SUP>), O(10<SUP>-2</SUP>), and O(10<SUP>-3</SUP>), respectively. The occurrence probabilities of freak waves measured from waves records were estimated between O(10<SUP>-2</SUP>) and O(10<SUP>-3</SUP>), which were located between predictions by Rayleigh and Ahn’s extreme probability distributions. However, we need more analysis of wave records obtained from diverse field conditions in order to verify the accuracy of the estimation of occurrence probability of freak waves.

        A New Histone Deacetylase Inhibitor, MHY219, Inhibits the Migration of Human Prostate Cancer Cells via HDAC1

        Histone deacetylase (HDAC) inhibitors are considered novel agents for cancer chemotherapy. We previously investigated MHY219, a new HDAC inhibitor, and its potent anticancer activity in human prostate cancer cells. In the present study, we evaluated MHY219 molecular mechanisms involved in the regulation of prostate cancer cell migration. Similar to suberanilohydroxamic acid (SAHA), MHY219 inhibited HDAC1 enzyme activity in a dose-dependent manner. MHY219 cytotoxicity was higher in LNCaP ($IC_{50}=0.67{\mu}M$) than in DU145 cells ($IC_{50}=1.10{\mu}M$) and PC3 cells ($IC_{50}=5.60{\mu}M$) after 48 h of treatment. MHY219 significantly inhibited the HDAC1 protein levels in LNCaP and DU145 cells at high concentrations. However, inhibitory effects of MHY219 on HDAC proteins levels varied based on the cell type. MHY219 significantly inhibited LNCaP and DU145 cells migration by down-regulation of matrix metalloprotease-1 (MMP-1) and MMP-2 and induction of tissue inhibitor of metalloproteinases-1 (TIMP-1). These results suggest that MHY219 may potentially be used as an anticancer agent to block cancer cell migration through the repression of MMP-1 and MMP-2, which is related to the reduction of HDAC1.

        A new plating medium from the extract of Solen gouldi has been developed as alternative to MacConkey agar for the isolation of Shigella. The new medium was tested for the growth of Shigella, Salmonella E. coli, Proteus, Vibrio, Pseudomonas, Staphylococcus and their ability form colonies, and that was compared with MacConkey and Brain Heart Infusion agar media. The following results were obtained. 1. Shigella dysenteriae and Escherichia coli grow best as large colonies on Solen agar medium. 2. Salmonella and Proteus grow well on Solen agar and Brain Heart Infusion agar but MacConkey agar was inhibited. 3. Vibrio parahaemolyticus and Vibric vulnificus was inhibited on Solen agar than thiosulfate-citrate-bilesalt-sucrose agar. 4. The growth of Staphylococcus aureus was highly inhibited on Solen agar media.

        폐좌상시 폐단락률과 PEEP치료의 효과

        To evaluate the physiologic extent of pulmonary contusion and effect of PEEP therapy for pulmonary contusion, we studied 16 patients received PEEP therapy with pulmonary contusion from nonpenetrating chest trauma. Hemodynamic parameters including pulmonary vascular resistance index and intrapulmonary shunt fraction were calculated from standard measurement, and arterial oxygen tension was measured. Pulmonary vascular resistance index and intrapulmonary shunt fraction were increased in patient group. Arterial oxygen tension was decreased with increase of the intrapulmonary shunt fraction(R=0.75). Arter minimal PEEP therap(5-10 ㎝H₂O), pulmonary vascular resistance index was remained unchanged and intrapulmonary shunt fraction was decreased without significant changes of pulmonary hemodynamics. Increment of arterial tension was increased with decrease of intrapulmonary shunt fraction(R=0.43). Decreased stroke volume index suggested of cardiac injury such as cardiac contusion in patient group. These results of our study suggested that increased intrapulmonary shunt fraction caused arterial hypoxia in pulmonary contusion and arterial oxygen tension was increased as a result of reduced intrapulmonary shunt by PEEP therapy.

        일 농촌지역에서 발기부전의 유병률 및 상관 요인

        Objectives : The purpose of this study was to investigate the prevalence of erectile dysfunction (ED) in a rural area of Korea and to examine the relationship between the prevalence and correlates of subjects. Method : This study was done as a part of the chronic disease survey of rural area by the Asan Foundation. Among 30 years or older population from Jung Eup, a rural area of Korea, 711 male subjects were sampled using a cluster probability sampling method. Trained interviewers visited and interviewed subjects aged 30 years or older with a structured interview. ED was categorized as 'none', 'mild', 'moderate', or 'severe' according to the ability to 'attain and/or maintain an erection satisfactory for sexual intercourse'. Response rate was 54.3% (N=386). Results : The age weighted prevalence of ED was 27.2% (minimal 14.1%, moderate 7.6%, severe 5.5%). Prevalence and severity increased with age. Liver disease and underweight were significantly (p<0.05) associated with ED. Smoking seemed to be correlated with ED, but did not reach statistically significant level. Conclusion : ED was highly prevalent in the rural community of Korea. Medical, sociodemographic, and lifestyle variables associated with ED may alert physicians to patients at risk for ED and offer insight to its etiology.

      • 고나트륨혈증후 발생한 중심성 뇌교및 외뇌교 수초 용해증 1예

        중심성 뇌교 수초 용해증은 일반적으로 심한 기저질환을 지닌 환자에서 심한 저나트륨혈증이나 저나트륨혈증을 급속 교정후 오는 것으로 알려져 있다. 저자들은 최근 만성 주정중독환자에서 심한 고나트륨혈증을 서서히 교정후 발생한 중심성 뇌교 및 외뇌교 수초 용해증을 뇌 자기공명 전산화 단층촬영으로 확진하였다. 심한 기저 질환을 가진 환자에서 장기간의 고나트륨혈증의 경우도 중심성 뇌교 수초 용해증을 일으킬 수 있기 때문에 이런 환자에서 저나트륨혈증뿐 아니라 고나트륨혈증도 피해야 될 것으로 사료된다.

      • Antidiabetic effects and gene expression profiling in obese mice treated with Isaria sinclairii over a 6-month period.

        <P>The molecular mechanisms underlying the glucose-lowering effects of Isaria sinclairii (Cicada Dongchunghacho), a fungus cultured on silkworm, are not fully elucidated. Thus the glucose-lowering effects of I. sinclairii as potential an antidiabetic agent were investigated in C57BL/6 obese (ob/ob) mice over a 6-mo period. For a period of 26 wk, ob mice were administered either 5 or 10% (w/w) I. sinclairii powder (IS), 10% dry mulberry leaf powder (ML), or 10% silkworm (SW) powder in the standard diet while a control group received only standard diet. The ML and SW preparations served as positive controls. Isaria sinclairii at 10% in the diet was more effective in reducing body weight compared to 10% ML, 10% SW, or 5% I. sinclairii. The fall in blood glucose levels in the groups treated for 26 wk was greater in both IS groups at 1 mo compared to ML or SW but equal in all groups at 6 mo. Microarray analyses were performed with a mouse 7.4K cDNA clone set array to identify the gene-expression profiles for the IS-, ML-, and SW-exposed ob mouse liver. The 10% IS group, compared to control, showed that 15 genes including glucokinase (Gk-rs1) and LDL receptor relating protein 1 were upregulated and 12 genes including cell translocation gene2 (antiproliferative) and hydroxyprostaglandin dehydrogenase (Hpgd 15) were downregulated. Upregulation of Gk-rs 1 and downregulation of Hpgd 15 were previously shown to occur in drug-induced suppression of diabetes. With ML, Lepr (leptin receptor), Pik3cb (phosphatidylinositol 3-kinase), and Prodh (proline dehydrogenase), related to suppression of diabetes, were upregulated. In the case of SW, the enzymes (G2an, alpha glucosidase 2) and Mmp9 (matrix metalloproteinase 9) involved in elevation of blood glucose levels were both downregulated. Data suggest that I. sinclarii is effective in lowering blood glucose due to the upregulation of glucokinase (Gk-rs1) and downregulation of hydroxyprostaglandin dehydrogenase (Hpgd 15), both associated with suppression of diabetes, indicating that microarray analysis is a useful tool to assess pharmacological potency of therapeutic compounds.</P>

        Electrochemical capacitors based on electrodeposited ruthenium oxide on nanofibre substrates

        <P>Electrodeposition of RuO<SUB>2</SUB> on electrospun TiO<SUB>2</SUB> nanorods using cyclic voltammetry is shown to increase the capacitance of RuO<SUB>2</SUB>. This phenomenon can be attributed to the large surface areas of the nanorods. Among several ranges of deposition, the range from 0.25 to 1.45 V with respect to Ag/AgCl was effective. The electrode deposited with this range exhibited a specific capacitance of 534 F g<SUP>−1</SUP> after deposition for 10 cycles with a scan rate of 50 mV s<SUP>−1</SUP>. The structural water content in RuO<SUB>2</SUB> was quite different depending on the deposition potential range. Higher amounts of structural water increased the charge storage capability. The stability of the electrode was tested using cyclic voltammetry over 300 cycles.</P>
