        Preparative separation of minor saponins from Panax notoginseng leaves using biotransformation, macroporous resins, and preparative high-performance liquid chromatography

        Background: Ginsenosides with less sugar moieties may exhibit the better adsorptive capacity and more pharmacological activities. Methods: An efficient method for the separation of four minor saponins, including gypenoside XVII, notoginsenoside Fe, ginsenoside Rd2, and notoginsenoside Fd, from Panax notoginseng leaves (PNL) was established using biotransformation, macroporous resins, and subsequent preparative high-performance liquid chromatography. Results: The dried PNL powder was immersed in the distilled water at 50C for 30 min for converting the major saponins, ginsenosides Rb1, Rc, Rb2, and Rb3, to minor saponins, gypenoside XVII, notoginsenoside Fe, ginsenoside Rd2, and notoginsenoside Fd, respectively, by the enzymes present in PNL. The adsorption characteristics of these minor saponins on five types of macroporous resins, D-101, DA-201, DM-301, X-5, and S-8, were evaluated and compared. Among them, D-101 was selected due to the best adsorption and desorption properties. Under the optimized conditions, the fraction containing the four target saponins was separated by D-101 resin. Subsequently, the target minor saponins were individually separated and purified by preparative high-performance liquid chromatography with a reversed-phase column. Conclusion: Our study provides a simple and efficient method for the preparation of these four minor saponins from PNL, which will be potential for industrial applications.

Calligraphy of Chinese characters is a wonderful work of art in the cultural soil of Chinese characters. This paper expounds the importance of Chinese calligraphy to strengthen the Chinese Nation’s sense of community from the perspectives of the composing method of Chinese characters, the aesthetic ideal of the Chinese nation, the artistic sublimation of Chinese character culture, the typical Chinese traditional culture and the joint creation of the Chinese nation.

        In Situ-DRIFTS Study of Rh Promoted CuCo/Al2O3 for Ethanol Synthesis via CO Hydrogenation

        The promoting effect of rhodium on the structure and activity of the supported Cu-Co based catalysts for CO hydrogenation was investigated in detail. The samples were characterized by DRIFTS, N2-adsorption, XRD, H2-TPR, H2-TPD and XPS. The results indicated that the introduction of rhodium to Cu-Co catalysts resulted in modification of metal dispersion, reducibility and crystal structure. DRIFTS results of CO hydrogenation at reaction condition (P=2 MPa, T=260 oC) indicated the addition of 1 wt % rhodium improved hydrogenation ability of Cu-Co catalysts. The ethanol selectivity and CO conversion were both improved by 1 wt % Rh promoted Cu-Co based catalysts. The alcohol distribution over un-promoted and rhodium promoted Cu-Co based catalysts obeys A-S-F rule and higher chain growth probability was got on rhodium promoted catalyst.

        Complete mitochondrial genome of the meadow moth, Loxostege sticticalis (Lepidoptera: Pyraloidea: Crambidae), compared to other Pyraloidea moths

        The mitochondrial genome (mitogenome) is an important topic for comparative and evolutionary genomics, as well as phylogenetic and population genetics. However, there are limited data regarding the mitochondrial genome available of Pyraloidea, one of the largest superfamilies in Lepidoptera. In this report, we present the complete mitogenome of the meadow moth, Loxostege sticticalis L. (Lepidoptera: Pyraloidea: Crambidae), which is a serious economic pest of both crops and weedsworldwide, thereby enhancing the available genomic information for Pyraloidea. This circular genome is 15,218 bp in length, containing 13 protein-coding genes (PCGs), two rRNA genes (rRNAs), and 22 tRNA genes (tRNAs), with a typical gene orientation and order comparable to other sequenced Pyraloidea insects. The genome composition of the major strand exhibits highly AT bias (80.82%), with a slightly positive AT skew indicating the occurrence of more As than Ts. The L. sticticalis mitogenome has a total of 130 bp of intergenic spacer sequences spread over 15 regions, ranging in size from 1 to 48 bp, of which only two are common among the 23 total Pyraloidea moths that have data collected on the mitogenome (one is located between tRNAGln and ND2 with variation change in length and a limited sequence conservation, and the other is located between tRNASer(UCN) and ND1 with a conserved 6 bp motif ‘ATACTA’). The A + Trich region of 331 bp in the genome is comprised of non-repetitive sequences but contains an ATAGN motif followed by a poly-T stretch of 17 bp, a microsatellite-like (TA)11 element preceded by an ATTTA motif, and a poly-A stretch upstream tRNAMet. These conserved structures identified in the A + T-rich region are presented in all of the sequenced Pyraloidea species. We provide a mitogenome-based phylogeny of Pyraloidea species, in which L. sticticalis shares close ancestry to Ostrinia species with substantial evidence. Our phylogenetic analyses strongly divide Crambidae into two sister lineages, one consisting of Pyraustinae and Spilomelinae, while the other contains Crambinae, Acentropinae, Scopariinae, Schoenobiinae and Glaphyriinae. The mitogenome dataset also supports the basal split between Pyraustinae and Spilomelinae.

        Morphology of the immature stages of Adoretus tenuimaculatus Waterhouse, 1875 (Coleoptera: Scarabaeidae: Rutelinae: Adoretini)

        The immature stages of the brown chafers Adoretus tenuimaculatus Waterhouse, 1875 were investigated morphologically using scanning electron microscopy. The scarabaeiform larvae of A. tenuimaculatus are peculiar for bearing a pair of eye spots on the head capsule, devoid of palidium but numerous hamate setae on the raster. The larvae possess a crown-like apical sensory area on each antenna, maxillary and labial palp. The pupa possesses dorsally six pairs of dioneiform organs on the intersegment regions between the first seven abdominal segments. This study provides the first ultra morphological study of the larvae in Adoretini. Both advantages and disadvantages of the scanning electron microscopy in the larval taxonomy of Scarabaeidae were briefly discussed.

      • Combined Screening of Cervical Cancer, Breast Cancer and Reproductive Tract Infections in Rural China

        Objectives: To investigate the current prevalence and knowledge of cervical cancer, breast cancer and reproductive tract infections (RTIs) in rural Chinese women, and to explore the acceptance and feasibility of implementing a combined screening program in rural China. Methods: A population-based, cross-sectional study was conducted among women aged 30 to 59 years old in Xiangyuan County, Shanxi Province from 2009 to 2010. Socio-demographic characteristics, knowledge of cervical cancer, breast cancer and RTIs, and the attitude toward single or combined screening were collected by an interview questionnaire. Each participant received a clinical examination of the cervix, breast and reproductive tract. Examinations included visual inspection, mammography, laboratory tests and pathological diagnosis. Results: A total of 1,530 women were enrolled in this study. The prevalence of cervical precancerous lesions, suspicious breast cancer, suspicious benign breast disease and RTIs was 1.4%, 0.2%, 14.0% and 54.3%, respectively. Cervicitis, trichomonas vaginitis, and bacterial vaginitis were the three most common RTIs among our participants. Television, radio broadcast, and public education during screening were the major source of healthcare knowledge in rural China. Moreover 99.7% of women expressed great interest in participating in a combined screening project. The affordable limit for combined screening project was only 50 RMB for more than half of the rural women. Conclusion: A combined screening program would be more effective and popular than single disease screening projects, while appropriate accompanied education and a co-pay model for its successful implementation need to be explored, especially in low-resource settings.

        Ferulic acid protects cardiomyocytes from TNF-a/cycloheximide-induced apoptosis by regulating autophagy

        Acute myocardial infarction (AMI) resultsin irreversible cardiac cell damage or death because ofdecreased blood fl ow to the heart. Apoptosis plays an importantrole in the process of tissue damage after myocardialinfarction (MI), which has pathological and therapeuticimplications. Ferulic acid (FA) is a phenolic acid endowedwith strong antioxidative and cytoprotective activities. Thepresent study aimed to investigate whether FA protectscardiomyocytes from apoptosis by regulating autophagy,which is a cellular self-digestion process, and one of thefi rst lines of defense against oxidative stress. Apoptosis wasinduced by TNF-α (10 ng/mL) and cycloheximide (CHX,5 μg/mL) in rat H9c2 cardiomyocytes. FA-inhibited TNF-α/CHX-induced apoptosis was determined by the quantifi cationof TUNEL-positive cells, and the eff ect was associatedwith decreased ROS production and inhibited caspase3activation. FA treatment enhanced autophagy and increasedautophagy-associated protein expression, leading to an inhibitionof mTOR signaling. When co-treated with 3-methyladenine(3-MA), an autophagy inhibitor, the anti-apoptoticeff ect of FA was attenuated. In an in vivo mouse MI model,FA treatment decreased the apoptotic cell number, reducedinfarct size, and improved cardiac performance, as determinedby histological and echocardiographic assessments. Taken collectively, these results suggest that FA could protectcardiomyocytes from apoptosis by enhancing autophagy.
