        Crocin alleviates neurotoxicity induced by bupivacaine in SH-SY5Y cells with inhibition of PI3K/AKT signaling

        Background Bupivacaine, a common local anesthetic, can cause neurotoxicity and permanent neurological disorders. Crocin has been widely reported as a potential neuroprotective agent in neural injury models. Objective The aim of this study was to investigate the role and regulatory mechanism of crocin underlying bupivacaine-induced neurotoxicity. Method Human neuroblastoma SH-SY5Y cells were treated with bupivacaine and/or crocin for 24 h, followed by detecting cell viability using 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide assay. The effect of crocin or bupivacaine on SH-SY5Y cell proliferation was measured by Ki67 immunofluorescence assay. The levels of apoptosis-related proteins and the markers in the PI3K/Akt signaling pathway were examined using western blot analysis. The activities of caspase 3, catalase (CAT), superoxide dismutase (SOD), malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) were tested using respective commercial assay kits. Flow cytometry analysis was executed for detecting SH-SY5Y cell apoptosis. Result Crocin attenuated bupivacaine-induced neurotoxicity in SH-SY5Y cells. Meanwhile, crocin inhibited SH-SY5Y cell apoptosis induced by bupivacaine via repressing the activity of caspase-3, reducing Bax expression, and elevating Bcl-2 expression. Moreover, crocin mitigated oxidative stress in SH-SY5Y cells by increasing the content of CAT, SOD, GSH-Px and reducing the content of MDA. Additionally, crocin protected against bupivacaine-induced dephosphorylation of Akt and GSK-3β. The protective effects of crocin against bupivacaine-induced neurotoxicity in SH-SY5Y cells were counteracted by the Akt inhibitor. Conclusion These results suggested that crocin may exert a neuroprotective function by promoting cell proliferation and suppressing apoptosis and oxidative stress in SH-SY5Y cells. Thus, crocin might become a promising drug for the treatment of bupivacaine-induced neurotoxicity.         Ultimate strength behavior of steel plate concrete composite slabs:An experimental and theoretical study

        Steel plate-concrete composite slabs provide attractive features, such as more effective loading transfer, and more cost-effective stay-in-place forms, thereby enabling engineers to design more high-performance light structures. Although significant studies in the literatures have been directed toward designing and implementing the steel plate-concrete composite beams, there are limited data available for understanding of the composite slabs. To fill this gap, nine the composite slabs with different variables in this study were tested to unveil the impacts of the critical factors on the ultimate strength behavior. The key information of the findings included sample failure modes, crack pattern, and ultimate strength behavior of the composite slabs under either four-point or three-point loading. Test results showed that the failure modes varied from delamination to shear failures under different design factors. Particularly, the shear stud spacing and thicknesses of the concrete slabs significantly affected their ultimate load-carrying capacities. Moreover, an analytical model of the composite slabs was derived for determining their ultimate load-carrying capacity and was well verified by the experimental data. Further extensive parametric study using the proposed analytical methods was conducted for a more comprehensive investigation of those critical factors in their performance. These findings are expected to help engineers to better understand the structural behavior of the steel plate-concrete composite slabs and to ensure reliability of design and performance throughout their service life.

        Benign Thyroid Enlargement Across Racial and Ethnic Groups

        Purpose: The objective of this study is to determine if there is a significant correlation between race/ethnicity and total thyroid volume among patients with benign thyroid disease. Our hypothesis is that patients of racial and ethnic minorities may present with enlarged, non-cancerous thyroids more frequently than white patients. Methods: This is a retrospective cohort study of 457 patients diagnosed with benign thyroid disease between 2008 and 2020 at Montefiore Medical Center. Data were taken from an existing database and the electronic health record. The primary outcome was total thyroid volume (cm3/mL) calculated from ultrasound measurements. The secondary outcome was the maximum dimension (cm) of the largest thyroid nodule. Results: Black patients had a mean thyroid volume (± standard deviation) of 142.7±131.1 cm3/mL and a mean maximum nodule dimension of 4.3±1.9 cm. White patients had a mean thyroid volume of 74.7±128.0 cm3/mL and a mean maximum nodule size of 3.0±1.5 cm. Race was significantly correlated with total thyroid volume (P=0.0007). Black race specifically was significantly correlated with total thyroid volume (95% confidence interval [CI], 0.209 to 0.794; P=0.0008) and maximum nodule dimension (95% CI, 0.006 to 1.278; P=0.05). Conclusion: Black patients had significantly larger total thyroid volumes and nodules compared to white patients. Thyroid volume was not associated with measures of socioeconomic status such as insurance type or estimated income. Although these findings illustrate an important relationship between race and thyroid volume, further research is required to determine the etiology of this discrepancy.

        Methylation-associated silencing of BASP1 contributes to leukemogenesis in t(8;21) acute myeloid leukemia

        The AML1-ETO fusion protein (A/E), which results from the t(8;21) translocation, is considered to be a leukemiainitiating event. Identifying the mechanisms underlying the oncogenic activity of A/E remains a major challenge. In this study, we identified a specific down-regulation of brain acid-soluble protein 1 (BASP1) in t(8;21) acute myeloid leukemia (AML). A/E recognized AML1-binding sites and recruited DNA methyltransferase 3a (DNMT3a) to the BASP1 promoter sequence, which triggered DNA methylation-mediated silencing of BASP1. Ectopic expression of BASP1 inhibited proliferation and the colony-forming ability of A/E-positive AML cell lines and led to apoptosis and cell cycle arrest. The DNMT inhibitor decitabine up-regulated the expression of BASP1 in A/E-positive AML cell lines. In conclusion, our data suggest that BASP1 silencing via promoter methylation may be involved in A/E-mediated leukemogenesis and that BASP1 targeting may be an actionable therapeutic strategy in t(8;21) AML.

        Determination and Variation of Core Bacterial Community in a Two- Stage Full-Scale Anaerobic Reactor Treating High-Strength Pharmaceutical Wastewater

        Knowledge on the functional characteristics and temporal variation of anaerobic bacterial populations is important for better understanding of the microbial process of two-stage anaerobic reactors. However, owing to the high diversity of anaerobic bacteria, close attention should be prioritized to the frequently abundant bacteria that were defined as core bacteria and putatively functionally important. In this study, using MiSeq sequencing technology, the core bacterial community of 98 operational taxonomic units (OTUs) was determined in a twostage upflow blanket filter reactor treating pharmaceutical wastewater. The core bacterial community accounted for 61.66% of the total sequences and accurately predicted the sample location in the principal coordinates analysis scatter plot as the total bacterial OTUs did. The core bacterial community in the first-stage (FS) and second-stage (SS) reactors were generally distinct, in that the FS core bacterial community was indicated to be more related to a higherlevel fermentation process, and the SS core bacterial community contained more microbes in syntrophic cooperation with methanogens. Moreover, the different responses of the FS and SS core bacterial communities to the temperature shock and influent disturbance caused by solid contamination were fully investigated. Co-occurring analysis at the Order level implied that Bacteroidales, Selenomonadales, Anaerolineales, Syneristales, and Thermotogales might play key roles in anaerobic digestion due to their high abundance and tight correlation with other microbes. These findings advance our knowledge about the core bacterial community and its temporal variability for future comparative research and improvement of the two-stage anaerobic system operation.

        MiR-370-3p aggravates blood–brain barrier injury and neuron apoptosis by targeting SMURF1 to activate the TLR4/MyD88/NF-κB signaling in sepsis-associated encephalopathy

        Background Sepsis-associated encephalopathy (SAE) is characterized by blood–brain barrier (BBB) disruption, neuron apoptosis and infl ammation. Studies show that miR-370-3p can be a new diagnostic biomarker for SAE. Objective This study aims to investigate the role of miR-370-3p in SAE and the underlying mechanism. Result An in vitro model of BBB disruption was established in brain microvascular endothelial cells (BMECs) with lipopolysaccharide (LPS). The expression level of RNAs was tested via RT-qPCR. Protein levels were detected by western blotting. Annexin V/PI double staining and CCK-8 assays were conducted to assess the apoptosis and viability of brain microvascular endothelial cells (BMECs). The migration of BMECs was detected by Transwell assay. BBB permeability analysis was performed to assess the relative permeability coeffi cient of BMECs. The in vitro barrier integrity was detected by trans-epithelial/ endothelial electrical resistance (TEER). The binding between miR-370-3p and SMAD specifi c E3 ubiquitin protein ligase 1 (SMURF1) was identifi ed by luciferase reporter assay. In this study, miR-370-3p was upregulated in LPS-treated BMECs. MiR-370-3p knockdown alleviated LPS-induced BBB disruption and apoptosis of BMECs, as well as recovered cell viability and migration. SMURF1 was directly targeted by miR-370-3p. SMURF1 silencing reversed the eff ects of miR-370-3p defi ciency in LPS-stimulated BMECs. Moreover, miR-370-3p activated the TLR4/MyD88/NF-κB signaling via regulating SMURF1 in LPS-stimulated BMECs. Conclusion MiR-370-3p aggravates BBB disruption and neuron apoptosis by targeting SMURF1 to activate the TLR4/ MyD88/NF-κB signaling in SAE.

        Dysfunction of Shh signaling activates autophagy to inhibit trophoblast motility in recurrent miscarriage

        In early pregnancy, the placenta anchors the conceptus and supports embryonic development and survival. This study aimed to investigate the underlying functions of Shh signaling in recurrent miscarriage (RM), a serious disorder of pregnancy. In the present study, Shh and Gli2 were mainly observed in cytotrophoblasts (CTBs), Ptch was mainly observed in syncytiotrophoblasts (STBs), and Smo and Gli3 were expressed in both CTBs and STBs. Shh signaling was significantly impaired in human placenta tissue from recurrent miscarriage patients compared to that of gestational age-matched normal controls. VEGF-A and CD31 protein levels were also significantly decreased in recurrent miscarriage patients. Furthermore, inhibition of Shh signaling impaired the motility of JAR cells by regulating the expression of Gli2 and Gli3. Intriguingly, inhibition of Shh signaling also triggered autophagy and autolysosome accumulation. Additionally, knockdown of BECN1 reversed Gant61-induced motility inhibition. In conclusion, our results showed that dysfunction of Shh signaling activated autophagy to inhibit trophoblast motility, which suggests the Shh pathway and autophagy as potential targets for RM therapy.

        Improving Patients’ Adherence to Physical Activity in Diabetes Mellitus: A Review

        Regular physical activity (PA) is a key element in the prevention and management of type 2 diabetes mellitus (T2DM). Participation in regular PA improves blood glucose control and can prevent or delay T2DM and its complications, along with positively affecting lipids, blood pressure, cardiovascular events, mortality, and quality of life. However, most people with T2DM are not active and show poor adherence. This paper reviews the possible barriers to PA and strategies to improve the adherence to PA. Based on the currently available literature, it is concluded that self-efficacy and social support from family, friends, and health care providers play the important role in adoption and maintenance of regular PA. Here we also highlight some new modern and innovative interventions that facilitate exercise participation and improve the adherence.
