Synthesis and Evaluation of Homoazasugars as Glycosidase Inhibitors

In an effort to develop transition-state mimetics of the glycosidase-catalyzed reaction, five- and six-membered azasugars and their homo-analogs were prepared and tested as inhibitors of glycosidases. Inhibition studies indicate that the fucosyl cationlike, five-membered imine 1 and its reduced form 2 are potent inhibitors of α-fucosidase from bovine kideney with respective K_i values of 160 nM and 2μM. The five-membered homoaminoazasugar 3 is also a potent inhibitor of the enzyme (K_i=1.9×10^-6M), while the glucose and mannose-like six-membered homoaminoazasugars 4 and 5 are less potent than the corresponding 1-deoxyazasugars as inhibitors of α-glucosidase and α-mannosidase, respectively. The primary amino group was placed in an attempt to introduce additional electrostatic interactions in the active site. The inhibitory activities are, however, in the high μM range. Synthesis of homoazasugars structurally related to a disaccharide and a nucleoside is also described.

In order to screen microsatellites for conservation genetics studies of the species, a total of 23 microsatellite loci from Korean goral (Naemorhedus caudatus), including 15 previously developed loci and 8 new loci in this study, were tested. Eleven microsatellites were screened and subjected to cross-species amplification using a test panel of four Caprinae species, Japanese serows (Capricornis crispus), Chinese gorals (Naemorhedus goral), Northern chamois (Rupicapra rupicapra) and domestic goats (Capra hircus). In addition, all eleven microsatellites (SY3A, SY12A, SY12B, SY48, SY58, SY71, SY76, SY84, SY84B, SY112, and SY129) satisfied the criteria to be a core set of microsatellites. This core set of microsatellites and cross-species amplification of Korean goral microsatellites were found to be helpful for high-resolution studies for conservation and management of Korean goral and other endangered Caprinae species.

<P>Abstract</P><P>Microsatellites have been applied in a variety of fields, including conservation genetics. Species-specific microsatellites are considered as more powerful genetic markers to generate an accurate genetic composition of a species itself. Accordingly, we characterized 15 polymorphic microsatellite loci from Korean goral, <I>Nemorhaedus caudatus</I>, one of the most endangered species in South Korea. The new markers should benefit future studies of the endangered species of other Asian gorals and their relatives for the study of genetic diversity and potential conservation management.</P>

Background: Unusually high amounts of N region addition and CDR3 length diversity were found in immunoglobulin (Ig) light chain Vk and Jk joins in patients with rheumatoid arthritis (RA). We sought to determine whether this finding is due to excessive activity of the enzyme responsible for N region addition (terminal deoxynucleotidyl transferase [TdT]) in B lineage cells in bone marrow or from positive antigenic selection of B cells with long CDR3 lengths. Methods: We used FACS to isolate $IgM^+/IgD^+$ B cells (predominantly naive) and $IgM^-/IgD^-$ B cells (predominantly class-switched) B cells from peripheral blood of a patient with RA known to have enrichment for long Vk CDR3s and from that of two normal controls. RT-PCR of VkIII transcripts was performed, followed by sequencing of individual cDNA clones. We analyzed the CDR3 lengths and N region additions in 97 clones. Results: There was enrichment for long CDR3 lengths (11 or 12 amino acids) in both $IgM^+/IgD^+$ and $IgM^-/IgD^-$ B cells in RA compared to B cell subsets in the normal controls. The $IgM^+/IgD^+$ B cell subset in RA was markedly enriched for N region addition and was similar to that seen in the $IgM^-/IgD^-$ subset. Conclusion: These data suggest that enrichment for N region addition and long CDR3 lengths in RA may result from unusually high or prolonged activity of TdT in bone marrow.
On the Role of Chromium in Dynamic Transformation of Austenite

The effect of Chromium (Cr) on the dynamic transformation (DT) of austenite to ferrite at temperatures up to 430 °C aboveAe3was studied in a medium-carbon low-alloy steel. Hot compression tests were performed using Gleeble 3800® thermomechanicalsimulator followed by microstructural examinations using electron microscopy (FESEM-EBSD). Driving forcecalculation using austenite flow stress and ferrite yield stress on an inverse absolute temperature graph indicated that Crincreases the driving force for the transformation of austenite to ferrite; however, when the influence of stress and thermodynamicanalysis are taken into account, it was observed that Cr increases the barrier energy and therefore, emerges as abarrier to the transformation. An analysis, based on lattice and pipe diffusion theories is presented that quantifies the role ofstress on the diffusivity of Cr and is compared with other the main alloying elements such as C, Si and Mn and its impact,positive or negative, on the DT barrier energy. Finally, a comparison is made on the differential effects of temperature andstress on the initiation of DT in medium-carbon low-alloy steels.
