      • The Effects of Multicultural Adolescents' Trauma and Hopelessness on Problem Behaviors in South Korea :

        <P>The purpose of this article was to examine the effects of multicultural adolescents' trauma and hopelessness on problem behaviors. The research subjects consisted of 108 adolescents (boys = 54, girls = 54 from multicultural families. They were adolescents aged between 12 and 20. The measures of the current study include scales for assessing adolescents' trauma, hopelessness, and problem behaviors. The data was analyzed using Pearson correlations, SPSS 21.0 program and AMOS 18.0 Structural Equation Modeling (SEM). Results demonstrated that multicultural adolescents' trauma, hopelessness, and problem behaviors were significantly correlated. Also, trauma had an indirect effect through hopelessness, as well as a direct effect on problem behaviors of adolescents. The importance of adolescents' cognition was identified on problem behaviors.</P>

      • Rice <i>OGR1</i> encodes a pentatricopeptide repeat–DYW protein and is essential for RNA editing in mitochondria

        <P>Summary</P><P>RNA editing is the alteration of RNA sequences via insertion, deletion and conversion of nucleotides. In flowering plants, specific cytidine residues of RNA transcribed from organellar genomes are converted into uridines. Approximately 35 editing sites are present in the chloroplasts of higher plants; six pentatricopeptide repeat genes involved in RNA editing have been identified in Arabidopsis. However, although approximately 500 editing sites are found in mitochondrial RNAs of flowering plants, only one gene in Arabidopsis has been reported to be involved in such editing. Here, we identified rice mutants that are defective in seven specific RNA editing sites on five mitochondrial transcripts. Their various phenotypes include delayed seed germination, retarded growth, dwarfism and sterility. Mutant seeds from heterozygous plants are opaque. This mutation, named <I>opaque and growth retardation 1</I> (<I>ogr1</I>), was generated by T-DNA insertion into a gene that encodes a pentatricopeptide repeat protein containing the DYW motif. The OGR1–sGFP fusion protein is localized to mitochondria. Ectopic expression of <I>OGR1</I> in the mutant complements the altered phenotypes. We conclude that OGR1 is essential for RNA editing in rice mitochondria and is required for normal growth and development.</P>

        Ectopic expression of a cold-responsive OsAsr1 cDNA gives enhanced cold tolerance in transgenic rice plants

        <P>The OsAsrt cDNA clone was isolated from a cDNA library prepared from developing seed coats of rice (Oryza sativa L.). Low-temperature stress increased mRNA levels of OsAsr1 in both vegetative and reproductive organs. In situ analysis showed that OsAsr1 transcript was preferentially accumulated in the leaf mesophyll tissues and parenchyma cells of the palea and lemma. For transgenic rice plants that over-expressed full-length OsAsr1 cDNA in the sense orientation, the Fv/Fm values for photosynthetic efficiency were about 2-fold higher than those of wild type-segregating plants after a 24-h cold treatment. Seedlings exposed to prolonged low temperatures were more tolerant of cold stress, as demonstrated during wilting and regrowth tests. Interestingly, OsAsr1 was highly expressed in transgenic rice plants expressing the C-repeat/dehyhdration responsive element binding factor 1 (CBF1), suggesting the regulation of OsAsr1 by CBF1. Taken together, we suggest that OsAsr1 gene play an important role during temperature stress, and that this gene can be used for generating plants with enhanced cold tolerance.</P>

      • Autophagy Stimulation Upregulated Collagen Synthesis in Cultured Human Fibroblast

        Autophagy can be defined as a self-digestive process, targeting internal or damaged organelles and misfolded proteins to lysosomal degradation. While its major role is a survival mechanism under stress conditions, such as nutrient restriction, it can also affect various cellular responses including inflammation, aging, and immune responses. Along with the clinical implications in various human diseases, such as neurodegenerative diseases, infection, and cancer, recent reports also suggested the potential involvement of autophagy in skin aging. In order to investigate the roles of autophagy in skin physiology, amino acid based chemical library was established and potential autophagy stimulating compounds were identified by measuring autophagic vacuole formation in cultured human fibroblast. Upregulation of autophgic flux by screened compounds were further investigated by measuring p62 and LC3-II expressions. Electron microscopic observation was also performed for observing the morphologic changes in cells. Among the screened compounds, S8_049, and S9_067 showed significant stimulating activities in cultured human skin fibroblast. In order to investigate the potential anti-aging effects of the selected compounds, expression of collagen and MMPs were also measured in cultured fibroblasts. As results, increased expression of collagen I was observed in S8_049 treated cells. Full thickness 3D culture model experiments also confirmed the increased expression of collagens in dermal layer in S8_049 treated tissue. Microarray experiments showed the increased expression of several sphingolipids synthesis and degrading enzymes, which suggested that the changes of intracellular sphingolipids levels might be, at least in part, underlying mechanism of autophagy stimulation.

        한국어 속담의 영어 기계번역에 미치는 격조사의 영향 ―구글번역기를 중심으로―

        This study investigates the influence of case markers on the machine-translation of Korean proverbs into English. While grammatical roles are determined by the word order in English, they are indicated by case markers in Korean. The problem is that Korean case markers are optionally deleted. It is, therefore, hypothesized that the presence of case markers in the original text is to be crucially important to the success rate of machine-translation from Korean into English. For this study, the characteristics of Google Translate, the translation system used here, were first surveyed. Then, sentences from three different categories of Korean texts (spoken, written, and proverb texts) were chosen and machine-translated using Google Translate. The findings are as follows. First, the Korean-English machine-translation of three different text types shows that there were more syntactic errors than lexical errors across the categories. Second, there were more errors in complex sentences than in simplex sentences. Third, the error rate was highest in the proverb category and lowest in the written text category. Fourth, across the three categories, the error rate was the highest when deletion of case markers or multi-case markers was involved. Lastly, the supplement of missing case markers to the original Korean proverbs substantially enhanced the success rate of machine-translation. This shows that the presence of case markers plays a crucial role in machine-translation into English, which, in turn, suggests that we can improve the success rate of machine-translation from Korean into English if we add the missing case markers to the Korean input sentences.

      • QFolder : RNA 분자 구조 형성 과정을 모의실험하기 위한 통합 시스템

        RNA 분자의 구조 형성을 완전히 이해하려면, 최종적으로 형성된 구조뿐 아니라 그 구조가 형성되는 과정 자체도 규명되어야 한다. 본 논문은 RNA의 이차 구조 형성 과정을 시뮬레이트하고, 이차 구조를 예측하며, 예측된 구조를 시각화하는 모델링 시스템인 QFolder를 소개한다. QFolder의 시뮬레이션 과정은 두 단계의 신뢰도에 의하여 제어되는데, 이 신뢰도는 동류성 염기 서열의 분석과 단순화된 열역학 및 동역학적 지식을 바탕으로 결정된다. 구조 형성 과정 중에 생성되는 구조들은 그래픽 형태로 보여지고 이 구조들을 편집하거나 보는 다양한 기능도 제공된다. 여러 RNA 분자에 QFolder를 실험한 결과에 의하면, QFolter가 구조 형성 과정 자체를 연구하는데 뿐 아니라 안정적인 이차 구조를 예측하는 데에도 유용한 도구가 될 수 있다는 것을 알 수 있다. Complete understanding of RNA folding requires the determination of both the final structure and the structure formation process. The paper presents a modeling system called QFolder for simulating the secondary structure formation process of RNA molecules, for predicting reliable structures, and for visualizing the predicted structures in a unified framework. The simulation process of QFolder is guided by two levels of certainty factors which are determined by the comparative analysis of homologous sequences, incorporated with simplified knowledge of thermodynamics and kinetics of folding. Intermediate and final structures generated during the simulation process are displayed in graphical form within a friendly interface, which provides various visualization and editing facilities for secondary structures. Experimental results demonstrate the potential of QFolder as a useful tool for predicting reliable secondary structures as well as for the study of the RNA folding process itself.
