        As an effort to develop noble antitumor lectins from Korean basidiomycetes, wild mushrooms were collected and their cold-water extracts were subjected to a screening test for their lectin activities using hemmagglutination assay on BALB/c mouse erythrocytes. Of the nine species tested, three species, Psathyrella velutina, Fomitella fraxinea and Russula compacta, showed strong hemmagglutinating activities, while the other six species showed minimal or no activities. Of these, partial purification of the lectin components of P. velutina and R. compacta were attempted by precipitation with saturated ammonium sulfate followed strong hemmagglutination activity unlike that of R. compacta, which lost its lectin activity after the purification process. Moreover, the partially purified P. velutina lectin exerted cytotoxic effect on sarcoma 180 cells when assessed by flow cytometric analysis.

        The ether, methanol, and physiological saline extracts of the dried carpophores of ten basidiomycetous fungi, which were collected in the conifer woods of Chungnam National University at Taechon and Keyryoung Mountain Area in Chungcheungnamdo, were subjected to an antibiotic activity test by disc-plate method. Among them, the methanol extracts of Inocybe trechispora, Lactarius velumis, and Laccaria laccata and the ether extracts of I.trechispora showed antibiotic activities against Sarcina lutea; and the methanol extracts of I. trechispora and L. velumis and physiological saline extract of Coriolus versicolor showed antibiotic activities against Staphylococcus aureus; and the methanol extract of L. velumis showed antibiotic activity against Escherichia coli. None of the extracts, however, showed any activity against Serratia marcescens and Proteusvulgaris.

        The ether, methanol, and physiological saline extracts of the dried carpophores of ten basidiomycetes, which were collected from several locations of Korea, were subjected to a screening test for antibiotic activity by disc-plate method using Sarcina lutea, Bacillus subtilis, Escherichia coli, and Serratia marcescens as target organisms. As the result, the methanol extract of five basidiomycetes-Amanita rebescens, Amanita solitaria, Lactarius vellereus, Latarius subzonarius and Scleroderma verrucosum-were found te have antibiotic activity. Among them, the methanol extracts of A. rubescens and L. vellereus exerted strong activity against both of the gram positive and negative bacteria. The ether extracts of A. rubescens and S. verrucosum also showed activity against S. lutea. The physiological saline extract, however, showed no activity at all.

        As an investigation to develop novel antitumor immunotherapeutic agents, four protein-poly-saccharide fractions from Korean wild mushrooms were subjected to a flow cytometric analysis for their in vitro lymphoblastogenic activities on the BALB/c mouse splenic lymphocytes. Of these, the protein-poly-saccharide fractions of Russula alboareolata, Russula compacta and Laccaria amethystea not only showed lymphoblastogenic activity, but also increased the viability of the lymphocytes, while that of Russula nitrata exerted marked cytocidal activity.

      • 영지버섯(Ganoderma Iucidum) 생장점으로부터 분리한 단백다당류의 면역활성

        Two protein-polysaccharide fractions. GLA and GLB, respectively, were prepared from the pileus of the fully grown carpophores and the tips of the growing carpophores of Ganoderma lucidum. At a dose of 100 ㎎/㎏/day ip. GLA and GLB inhibited the growth of sarcoma 180 solid tumor in ICR mice by 56.3% and 81.8%, respectively. In a flow cytometric (FCM) analysis. GLA and GLB enhanced the formation of lymphoblasts of BALB.c splenic leukocytes at a concentration of 100㎍/㎖, by 38.3% and 61.3%, respectively. When ip injected into ICR mice. GLB exerted anti-leukopernic effect against cyclopospamide (75 ㎎/㎏/day ip) in that the leukocyte counts of the peripheral blood of the normal and the cycloposphamide-treated mice, respectively, was (11.1±3.8)×10 exp(3) and (4.0±1.8)×10 exp(3), while the GLB-cyclophosphamide treated mice showed a leukocyte count of (10.8±5.1)×10 exp(3) cells/㎕. These results suggest that GLB is a promising candidate for an effective cancer immunotherapeutic agent.

      • Phellinus baumi 상황버섯의 면역증강활성

        Certain mushrooms belonging to the genus Phellinus, such as P. linteus and P. igniarius, have been known for their antitumor and immunopotentiating activities. These mushrooms have collectively been called "Sang-Whang" in oriental medicine. In Korea, Phellinus baume has been cultivated from mid 1990's and are known as "Sang-Whang" mushrooms. However, its antitumor or immunomodulatory activity has never been carried out. In this study, two protein-polysaccharide fractions, one from hot water extract and the other from alkaline extract, were prepared and their stimulatory activities on the in vitro cultured BALB/c mouse splenic leukocytes were investigated using flow cytometric analysis. As a result, the water soluble protein-polysaccharide fraction, PBD, was found to exert stronger activities than the alkali-soluble protein-polysaccharide fraction, PMA, in in vitro lymphoblastogenic assay. PBD also showed in vitro stimulatory activity on T lymphocytes in that it enhanced the level of CD25 expression of the in vitro cultured splenic leukocytes.

        To ellucidate immunologic activities of ginseng saponin, the splenic leukocytes of BALB/c mice were incubated in RPMI medium for 48hr with or without ginseng total saponin(GTS), and then analyzed flow cytometrically. Upto the concentration of 100㎍/㎖, GTS exherted stimulatory effect on the immune cells most prominently on B cells: the percentage of the surface Immunoglobulin (sIg) -positive cells (B cells) of the control was 47.l%, while that of GTS treated was 58.9%. The percentages of monoclonal antibody(mAb) J1j-reactive cells (T cells) were 28.8% and 3l.3%, those of mAb Gkl.5-reactive cells (CD4' T cells) were l8.5% and l9.l%, and those of mAb M1/70-reactive cells (mostly macrophages) were 4.4 and 5.0% at the concentration of 0 and 100㎍/㎖, respectively. However, at the concentration of 300㎍/㎖ or higher, GTS was found to be toxic to the immune cells especially to B cells in that the flow rate of the cells (gated viable cells) significantly decreased and the percentage of B cells droped from 58.9% to 31.2% and 25.4% at l00, 300, and 1000㎍/㎖, respectivelly. These data strongly suggests that GTS would exhert B-cell mitogen-like activity at physiological concentrations of 100㎍/㎖ or lower.

      • 꽃송이버섯 열수 추출물을 포함하는 조합물들의 항암면역활성

        In order to develope a new composite to be used as immunopotentiating health food, the composites including extracts of Sparassis crispa, Grifola frondosa, Phellinus linteus, red ginseng, and Oxya japonica were prepared and tested for their antitumor immunoactivities. Of these, the composite G3, which contained the extracts of S. crispa (20%), G. frondosa (55%), P. linteus (5%), red ginseng (10%), and O. japonica (10%), showed marked lymphoblastogenic activity on the in vitro cultured BALB/c mouse splenic lymphocytes and successfully suppressed the growth of sarcoma 180 cells implanted into the peritoneum of ICR mice. Moreover, the composite G3 exerted antitumor activity against sarcoma 180 tumor cells subcutaneously implanted into the left groin of ICR mice with the tumor inhibition ratio of 46.2%.

      • Phellinus linteus 균사 배양물로부터 분리한 단백다당체 Kp의 항암활성

        A protein-polysaccaride fraction Kp(53.9% polysaccharide, 14.2% protein) was separated from the shake-cultured mycelia of a basidiomycetous fungus, Phellinus linteus, and its antitumor activity against sarcoma 180 in ICR mice was investigated. When administered after the tumor implantation. Kp exerted antitumor activity by inhibiting the growth of the sarcoma 180 solid tumor by 71.5% and increasing the life span of the sarcoma 180 ascitic mice by 51.5% at 100 ㎎/㎏. In pretreatment tests, in which Kp was administered once daily for 9 days before the tumor implantation. Kp inhibited the growth of the solid and ascites form of sarcoma 180, respectively, by 35.4% and by 80.3% at 100 ㎎/㎏. However, Kp showed no in vitro cytotoxic activity against a murine leukemia L1210 and a human gastric tumor SNU.1 upto the concentration of 200 ㎍/㎖. From these results, it is clear that the antitumor activity of Kp is exerted through its immunomodulating activity on the host's immune system.

        The ether, methanol, and physiological saline extracts of the dried carpophores of ten basidiomycetous fungi, which were collected in the conifer woods of Chungnam National University at Taechon, were subjected to an antibiotic activity test by disc-plate method. Among them, the methanol extracts of Marasmius maximus, Hirschioporus abietinus, and Russula sanguinea and the the ether extracts of M. maximus and H. abietinus showed antibiotic activities against Sarcina lutea; and the methanol and ethanol extract of M. maximus showed antibiotic activity against Staphylococcus aureus. None of the extracts, however, showed any activity against gram negative bacteria, Serratia marcescens and Proteus vulgaris.
